  • 본 연구자들은 이전에 cellulase, xyalnase 및 lichenase의 다기능 효소활성을 지니는 절단된 Cel44C-$Man26A_{P558}$${\beta}$-glycosyl hydrolase를 보고하였다. 본 연구에서는 절단된 Cel44C-$Man26A_{P558}$ 효소의 다기능성 ${\beta}$-glycosyl hydrolase 활성을 증가시키기 위해 DNA shuffling을 시도하였다. DNA shuffling에 의해 단일변이(P438A)를 가진 M2Cel44C-$Man26A_{P558}$와 이중변이(A273T 및 P438A)를 가진 M21Cel44C-$Man26A_{P558}$를 얻었다. 이중변이를 가진 M21Cel44C-$Man26A_{P558}$은 단일변이를 가진 M2Cel44C-$Man26A_{P558}$ 보다 효소활성이 낮게 나타났으나, M2Cel44C-$Man26A_{P558}$와 M21Cel44C-$Man26A_{P558}$은 대조구인 Cel44C-$Man26A_{P558}$ 보다 약 1.3에서 2.2배 정도 높은 효소활성을 나타내었다. 특히, 단일변이를 가진 M2Cel44C-$Man26A_{P558}$는 대조구인 Cel44C-$Man26A_{P558}$보다 cellulase, xylanase 및 lichenase 효소활성이 약 1.5에서 2.2배 정도 높게 나타났다. ${\beta}$-Glycosyl hydrolase의 cellulase, linchenase 및 xylanase 최적 효소활성은 각각 pH 7.0, 7.0 및 6.0에서 이었다. 이러한 결과는, 아미노산 잔기인 Ala438이 다기능성 ${\beta}$-glycosyl hydrolase 활성을 증가시키는 중요한 역할을 한다고 추정할 수 있다.

  • 탄소원으로 Avicel이 첨가된 배지에서 공주에 소재한 밤 나무 농장의 토양 미생물을 증균 배양하여 mannanase를 생산하는 미생물을 분리하였다. 분리균 YB-43의 16S rDNA 서열은 Cellulosimicrobium 속 균주와 유사도가 99.6% 이상으로 가장 높게 나타났다. Locust bean gum (LBG)이나 konjac이 첨가된 배지에서 분리균의 mannanase 생산성이 크게 증가하였다. 0.7% LBG가 첨가된 LB 배지에서 Cellulosimicrobium sp. YB-43이 균체외로 생산한 mannanase를 DEAE-Sepharose와 Q-Sepharose 컬럼 크로마토그래피로 부분 정제하여 mannanase A (ManA)와 mannanase C (ManC)로 분획하였다. ManA는 $55^{\circ}C$와 pH 6.5, ManC는 $65^{\circ}C$ pH 7.5에서 최대활성을 보였으며, ManA는 $40^{\circ}C$ 이하에서 1시간 동안 실활되지 않았으나 ManC는 $20^{\circ}C$에서도 상당량 실활되었다. 또한 ManA와 ManC는 기질특이성과 mannooligosaccharides의 최종 분해산물에도 차이가 있는 것으로 나타났다. 이로 보아 Cellulosimicrobium sp. YB-43은 특성이 서로 다른 2종류 mannanases를 생산하는 것으로 판단된다.

Paenibacillus woosongensis으로부터 Mannanase 26AT 유전자의 클로닝과 유전자 산물의 분석 (Cloning a Mannanase 26AT Gene from Paenibacillus woosongensis and Characterization of the Gene Product)

  • Paenibacillus woosongensis의 유전체 부분 염기서열로부터 mannanase를 코드하는 것으로 유추되는 open reading frame을 중합효소연쇄반응으로 증폭하여 대장균에 클로닝하고 염기서열을 결정하였다. Mannanase 유전자는 man26AT로 명명하였으며 1,053 아미노산으로 구성된 단백질을 코드하는 3,159 뉴클레오티드로 이루어졌다. 아미노산 잔기배열을 분석한 결과 Man26AT는 glycosyl hydrolase family 26의 mannanase와 상동성이 높은 활성영역, 탄수화물 결합영역 CBM27과 CBM11로 구성되어 있었다. Man26AT의 아미노산 배열은 P. ihumii의 유추된 mannanase와 상동성이 81%이고 다른 Paenibacillus 속 균주의 여러 mannanases와 57% 이하의 상동성을 보였다. man26AT 유전자를 함유한 재조합 대장균의 균체 파쇄상등액은 $55^{\circ}C$와 pH 5.5에서 최대의 mannanase 활성을 보였고, $50^{\circ}C$에서 1시간 열처리한 후에 80% 이상의 잔존활성을 보였다. Man26AT는 locust bean gum (LBG) galactomannan과 konjac glucomannan에 대한 분해활성이 유사하였으며, carboxymethylcellulose, xylan과 para-nitrophenyl-${\beta}$-mannopyranoside는 분해하지 못하였다. Man26AT에 의해 mannotriose, mannotetraose, mannopentaose와 mannohexaose 등의 만노올리고당이나 LBG로부터 공통의 최종 가수분해 산물로 mannose, mannobiose와 mannotriose가 생성되었다. 또한 mannotriose 보다 큰 만노올리고당이 LBG와 guar gum의 분해산물로 각각 생성되었다. 그러나 Man26AT는 mannobiose를 분해하지는 못하였다. 활성염색을 통해 Man26AT는 균체 내에서 3개 이상의 크기가 다른 활성 단백질로 분해된 것이 확인되었다.

  • A review of this literature and discussions reveal a development of ideas concerning the elements of nursing models. The elements of a nursing model are the nurses view of the human being, nursing's goal, and nursing activities. It has long been recognized that human beings, at one time or another, require nursing care. Varieties of literature were reviewed in regard to the human being as recipient of nursing care through the theory development in nursing. Florence Nightingale initiated the modern era of nursing and described more clearly man as the recipient of nursing care. She looked at man as responding to the laws of nature whether the person was healthy or sick. Henderson added to Nightingale's concept of man , the recipient of nursing care by emphasizing that man is a whole, complete, and independent being. Her view is further specified by her enumeration of the activities the human being must perform. Johnson has developed a very comprehensive view of man as the recipient of nursing care. Man is a behavioral system which has a tendency to achieve and maintain stability in patterns of functioning. Like Nightingale, Johnson sees that similar patterns occur in both health and illness. Johnson postulates that the whole behavioral system of the human is composed of eight sub-systems: affiliative, achievement, aggressive, dependency, eliminative, ingestive, restorative, sexual. Roger's main contribution to the development of nursing models was her emphasis upon unitary man. She pointed out that man is a unified whole, possessing his own integrity and manifesting characteristics that“are more than and different from the sum of his parts.”Rogers focuses on the life processes of the human and points out that these processes have the following characteristics. Wholeness, openness, unidirectionality, pattern and organization, sentence, and thought. According to Roy, man is a biopsychosocial being in constant interaction with a changing environment. To cope with this changing environment, man has certain innate and acquired mechanisms. Man's ability to respond positively or to adapt, depends upon the degree of the change taking place and the state of the person coping with the change. When she analyzes man as an adaptive organism she further describes man as being composed of four adaptive modes: physiological needs, self-concept, role function, and interdependence. Based on the literary review through the theory development in nursing, general approach by a unified nursing model to a view of the recipient of nursing care may be stated as follows: Man is a unified whole composed of subsystems with a flexible and normal line of defense; his internal regulating mechanisms help him to cope with a changing environment; he functions by the principles of homeodynamics.

  • As a result I get the fallowing conclusions;1. Oriental medicine and confucianism have similar point about heart because oriental medicine and confucianism have same philosophical backing 2. Oriental physitian read the books complated by confucianism by confuncianism physitian because racial characterristics are based on confucian thoughts, and my race make much of the heart. So to use confucian thought to phychotheraphy need to study confucian In(仁) thought.3. Thought that oriental medicine refers man as small university and thought that man and heaven are same based on the ethical doctrine that man's inborn nature is good are same connection. but oriental medical aim is agreement external features of man with university, confucian aim is agreement heart of man with university4. Confucianism will be used in phychotheraphy because phychotheraphy require close relationship between psychiatrist and patient and confucianism gives importance to man's relationship5. Oriental medical phychotheraphy deal prevention and cure at same time, but, confucianism deal prevention entirely. so confucianist target a saint by training at ordinary times.

자동생산체계에서 인간-기계 시스템의 안전도측정과 안전작업설계에 관한 연구 (A Study on Safety Assessment and Design of the Safe Task in Automated Man-Machine System)

  • Some problems to assess the safety of automated man-machine system are studied in many ways. The difficulty occurred in this system is the vagueness of human behavior. Fuzzy set theory is used to assess the human behavior in safety analysis. The unsafe behavior listed top 10 in accident statistics would be explained as the factors of human vagueness. Three cases are considered, which consist of man-machine system as man-man, man-machine, machine-machine types. For the design of safe task, using characteristics of work performance, each motion cycle time is required to know the rate of learning. Approach of human behavior to the standard motion means more safe motion. It is important to design the works as to minimize the time performance to the standard motion's, which utilize the control of risk potential with easy. In that process, use of fuzzy set theory is appropriate to analyze the human behavior to identify its vagueness.
