목 적: 인간 배아줄기세포는 재생 의학이나 조직공학에 있어서 큰 잠재적인 능력을 가지고 있는 것으로 알려져 있다. 이들은 다양한 growth factors 처리나 유전자 발현을 변화시켜 특정 세포로 유도 분화 및 분리가 가능하지만 그 효율성은 아직까지 낮은 상태이다. 본 연구에서는 인간 배아줄기세포로부터 비특이적으로 분화된 세포들을 특정 세포 표면 항체를 이용한 magnetic cell sorting (MACS) 방법으로 분리, 배양하여 그들의 특성을 살펴보았다. 연구방법: 미분화 배아줄기세포주(Miz-hESC4)를 물리적인 방법으로 계대 배양하였으며, 부유 배양법으로 배아체 형성을 유도하였다. 배아체의 자발적인 분화를 위해 DMEM에 10% FBS를 첨가하여 2주 동안 배양하였다. 이렇게 분화된 세포들을 CD34, human epithelial antigen (HEA), human fibroblast (HFB)에 대한 항체를 이용한 MACS system으로 각각의 항체에 대한 양성 또는 음성 세포를 분리하였다. 이러한 MACS 분리 세포를 4주 동안 배양하면서 형태적인 변화를 관찰하고 특이 유전자의 발현 양상을 분석하였다. 결 과: 분리 배양한 CD34 양성 세포들은 배양 초기에는 둥근 형태를 나타내다가 배양 후기에는 작은 다각형의 형태로 관찰되었으며, HEA 양성 세포들은 큰 다각형의 형태를 나타내었고, HFB 양성 세포들은 전형적인 방추체 형태로 관찰되었다. 특이 유전자에 대한 RT-PCR 결과에서, CD34 양성 세포들과 HFB양성 세포들에서는 내배엽과 중배엽 관련 유전자의 발현하는 것을 확인할 수 있었고, HEA 양성 세포들에서는 외배엽 관련 유전자인 NESTIN과 NF68KD의 발현을 관찰할 수 있었다. 배양기간이 경과함에 따라 CD34 양성 세포의 특이 유전자 발현 양상이 변화되었다. 결 론: 이상의 결과는 비특이적으로 분화된 인간 배아줄기세포로부터 특이 세포를 MACS 방법을 이용하여 성공적으로 분리할 수 있음을 보여주었다. 따라서, MACS 방법과 특이 세포에 대한 항체는 인간 배아줄기세포의 유도 분화와 특이 세포의 분리에 매우 유용할 것으로 생각된다.
이 연구는 뇌성마비 아동의 손 기능을 평가하기 위해 개발된 손 기능 분류 체계(Manual Ability Classification System: MACS)를 이용하여 경직형 뇌성마비 아동의 손 기능을 평가하고 손 기능과 기능적 수행도 평가인 일상생활활동평가(Functional Independence Measure of Children: WeeFIM), 대운동 기능 분류체계(Gross Motor Function Classification System: GMFCS)의 관계를 알아보고, 이를 통해 뇌성마비 아동의 손 기능 평가 체계로써의 MACS에 대한 기초의 자료를 제공하는데 그 목적이 있었다. 경직형 뇌성마비 아동 60명을 대상으로 아동의 손 기능과 대동작 기능, 일상생활활동을 평가하였다. 평가한 자료를 바탕으로, MACS와 WeeFIM 및 GMFCS와의 관계와 손 기능 수준에 따른 일상생활활동의 차이를 알아보았다. 그 결과, MACS와 GMFCS의 상관은 유의수준.05에서 유의한 것으로 나타났으며, 그 정도는 r =.659이었다. WeeFIM과의 상관을 알아본 결과에서도 총점과의 상관은 유의수준.05에서 유의한 것으로 나타났으며, 그 정도는 r = -.576이었다. 손 기능 분류 체계로 평가한 수준에 따른 일상생활 활용의 차이를 알아본 결과 유의한 차이가 있는 것으로 나타났다(p <.05). 뇌성마비 아동의 손 기능 평가 도구로써 MACS는 임상적으로 유용한 평가 체계로 사용될 수 있음을 알 수 있다.
Collection of large number of gonadal Primordial germ cells(gPGCs) is a prerequisite factor for improving germline transmission efficacy in the aves, In this study a magnetic-activated cell sorter(MACS) was applied for improving retrieval efficacy of quail gPGCs and the migration capacity of MACS-separated gPGCs was further examined after being transplanted to recipient embryos. We also induced germline chimeras by transfer of MACS-separated quail gPGCs at the efficiency of 17.4% on average.
이 연구는 뇌성마비 아동의 기능적 수준 분류 체계의 유용성을 알아보기 위해 실시되었다. 이를 위해 2008년 9월부터 2010년 8월까지의 기간 동안 뇌성마비 아동 217명을 대상으로 대동작 기능 분류체계(GMFCS), 손 기능 분류 체계(MACS), 일상생활 활동을 측정하고 도구 간의 관계를 알아보았다. 그 결과, 대동작 기능 분류체계와 일상 생활 활동은 모든 하위 영역 및 총점과 유의한 상관이 있는 것으로 나타났다(p<.05). 손 기능 분류체계와 일상생활 활동은 이상운동형 아동에서 일상생활 하위 영역 중 의사소통과 상관이 유의하지 않은 것을 제외하고 모든 하위 영역 및 총점과 유의한 상관이 있는 것으로 나타났다(p<.05). GMFCS와 MACS의 관계를 알아본 결과, 가장 많은 분포를 나타낸 것은 GMFCS의 경우 1수준(20.3%)과 5수준(40.6%)이었고, MACS의 경우는 2수준(48.8%)과 5수준(16.6%)이었다. 결론적으로, 뇌성마비 아동의 기능적 수준 분류 체계인 GMFCS와 MACS는 임상적으로 유용한 평가 체계로 사용될 수 있을 것으로 생각된다.
This study investigated the relationship between function classification systems and the Pediatric Evaluation of Disability Inventory (PEDI) functional skills in children with cerebral palsy (CP). Two hundred and eleven children with CP participated in this study. The Korean-Gross Motor Function Classification System (K-GMFCS), Korean-Manual Ability Classification System (K-MACS), Korean-Communication Function Classification System (K-CFCS), and self-care, mobility, and social function domains of the Korean-Pediatric Evaluation of Disability Inventory (K-PEDI) functional skills were measured by physical therapists or occupational therapists. All of the function classification systems were significantly correlated with PEDI functional skills ($r_s$=-.549 to -.826) (p<.05). Especially, K-GMFCS, K-MACS, and K-CFCS were correlated significantly with mobility, self-care, and social function, respectively. Using stepwise multiple regression analysis, we established that K-GMFCS, K-MACS, and K-CFCS were predictors of self-care skills (74.3%) and mobility skills (79.5%) of the K-PEDI (p<.05). In addition, K-CFCS and K-MACS were predictors of social function (65.9%) of the K-PEDI (p<.05). The information gathered in this study using the levels measured in the function classification systems may be useful to clinicians for estimating the PEDI functional skills in children with CP.

Pigs are considered as optimal donor animal for the successful xenotransplantation. To increase the possibility of clinical application, genetic modification to increase compatibility with human is an important and essential process. Genetic modification technique has been developed and improved to produce genetically modified pigs rapidly. CRISPR/Cas9 system is widely used in various fields including the production of transgenic animals and also can be enable multiple gene modifications. In this study, we developed new gene targeting vector and enrichment system for the rapid and efficient selection of genetically modified cells. We conducted co-transfection with two targeting vectors for simultaneous inactivation of two genes and enrichment of the genetically modified cells using MACS. After this efficient enrichment, genotypic analysis of each colony showed that colonies which have genetic modifications on both genes were confirmed with high efficiency. Somatic cell nuclear transfer was conducted with established donor cells and genetically modified pigs were successfully produced. Genotypic and phenotypic analysis of generated pigs showed identical genotypes with donor cells and no surface expression of ${\alpha}$-Gal and HD antigens. Furthermore, functional analysis using pooled human serum revealed dramatically reduction of human natural antibody (IgG and IgM) binding level and natural antibody-mediated cytotoxicity. In conclusion, the constructed vector and enrichment system using MACS used in this study is efficient and useful to generate genetically modified donor cells with multiple genetic alterations and lead to an efficient production of genetically modified pigs.
PURPOSE: The purpose of this study was to investigate the difference and relationship between sensory processing abilities, gross motor and fine motor capabilities in children with cerebral palsy. METHODS: 104 children with cerebral palsy participated in the study. Sensory processing abilities of the subjects were measured by Short Sensory Profile (SSP). Gross and fine motor abilities were each measured using the Gross Motor Function Classification System (GMFCS) and Manual Ability Classification System (MACS), respectively. RESULTS: There were significant correlations between SSP level and GMFCS (R=.72, p<.00) or MACS (R=.77, p<.00) levels. Significant differences were showed each gross motor (p=.01) and fine motor level (p=.00) among sensory processing level of children. In addition, sub-items of sensory processing as Tactile sensitivity, Movement sensitivity, Auditory filtering and Low energy/Weak were significantly were showed significant correlations gross motor and fine motor level (p=.01). Also, multiple regression result was showed that as MACS level and GMFCS level were higher, the SSP total score was higher all of participants (adjusted $R^2=.62$). CONCLUSION: Sensory processing abilities of children with cerebral palsy were related with gross motor and fine motor capabilities. Also gross motor and fine motor capabilities are as higher, the sensory processing skill was well of cerebral palsy.
The new controller developed here, which is the facility with only one measurement, is a new concept for the level controller of the existing nuclear steam generator. A MACS (Microcomputer-based Adaptive Control System of a Steam Generator) is quite practical and efficient, and has also simple structure and higher flexibility in the installment for actual plant. A key ingredient of this system is adaptive regulator which can calculate adaptive, optimal valve position in response to changes in the dynamics of the process and the disturbances. In spite of many difficulties in the steam generator water level control at low power, it can be concluded from the experimental and simulation results, that the MACS can provide optimal, robust steam generator level control from zero to full power. The amount of the control input effort can be reduced by adjusting the weighting factor. However, the steady state water level errors are generated. To avoid the steady errors, the different adaptive algorithm should be investigated in the future. The 3 second sampling time is acceptable for this system. However, action should be taken to shorten the sampling time for better digital control.
본 논문은 이온 선택성 전극을 모듈화한 MACS를 사용하여 시스템의 크기를 축소할 수 있었고, PDA를 사용함으로써 측정된 데이터를 장소에 구애받지 않고 분석할 수 있는 휴대용 전자혀 시스템을 개발하였다. MACS는 ${NH_4}^+$, $Na^+$, $Cl^-$, ${NO_3}^-$, $K^+$, $Ca^{2+}$, $Na^+$, pH의 7종의 이온 선택성 전극을 이용하여 구성하였으며, 초기화 및 교정과정과 완충용액에 의한 안정화 과정을 거친 후 MACS로 시료에 대한 각각의 이온선택성 전극의 변화를 측정한다. 이렇게 각 전극으로부터 측정된 데이터를 이용하여 신경회로망 알고리즘으로 측정된 시료의 종류를 구분할 수 있다. 실험은 분류가 어렵다고 알려진 고급양주와 저급양주를 분류하는 것으로 진행되었으며, 성공적이며 우수한 실험 결과를 얻었다 이로부터 사용된 알고리즘이 휴대용 전자혀 시스템에 적절히 사용될 수 있음을 밝혔으며, 실제 휴대용 전자혀 시스템에 간단한 학습에 의해 적용될 수 있을 것으로 생각된다.

The objective of this study was to establish a selection process for high quality sperm in bovine semen using sperm separation by magnetic activation (MACS). For this, semen from 21 Nellore bulls was collected using an artificial vagina. To guarantee the presence of pathologies in the ejaculate, animals previously declassified in four consecutive spermiogram were used. Semen was analyzed in five statuses: (1) fresh semen (fresh); (2) density gradient centrifugation (DGC), percoll column; (3) non-apoptotic fraction after separation by MACS (MAC); (4) apoptotic fraction from the separation (MACPOOR); and (5) MAC followed by DGC (MACDGC). Using a computerized analysis system (CASA), motility was measured. The sperm morphology was evaluated by phase contrast, and the supravital test was completed with eosin/nigrosin staining. For DGC, 20 × 106 cells were used in a gradient of 90% and 45% percoll. MACS used 10 × 106 cells with 20 μL of nanoparticles attached to annexin V, and filtered through the MiniMACS magnetic separation column. Membrane integrity was assessed with SYBR-14/IP and mitochondrial potential with JC-1 by flow cytometry. Processing sperm by MACDGC, was more effective in obtaining samples with high quality sperm, verified by the total of abnormalities in the samples: 35.04 ± 2.29%, 21.50 ± 1.47%, 17.30 ± 1.10%, 30.68 ± 1.94% and 10.50 ± 1.46%, respectively for fresh, DGC, MAC, MACPOOR, and MACDGC. The subpopulation of non-apoptotic sperm had a high number of live cells (82.65%), membrane integrity (56.60%) and mitochondrial potential (83.98%) (p < 0.05). These findings suggest that this nanotechnological method, that uses nanoparticles, is efficient in the production of high-quality semen samples for assisted reproduction procedures in cattle.
