Intestinal trematode infections in the villagers in Koje-myon, Kochang-gun, Kyongsangnam-do, Korea (경상남도 거창군 고제면 주민들의 장흡충 감염상)

  • An epidemiological study fur the Intestinal trematode infection of the villagers was done In Koje-myon, Kochang-gun, Kyongsangnam-do, Korea in March, 1994. Of 116 stool specimens examined, total helmlnthic ova positive cases were 13 (11.2%) and cumulative ova positive cases 21 (18.1%): Echinostoma hortense 11 cases (9.5%), Metngonimus sp. 6 cases (5.2%), and Clonorchis sinensis 4 cases (3.4%). After the treatment and purgation, variable numbers of E. hokene, 6 to 227 per person, were collected from 7 echlnostome egg Positive cases, together with M. tokohoshii In 6 cases. Three adult flukes of E. cinetorchis were collected from one person and one Stellantchasmw jnlcatus was recovered from another case. This mountainous village was proved to be one of the endemic foci of echlnostomlasis In Korea.

Whole-Genome Characterization of Alfalfa Mosaic Virus Obtained from Metagenomic Analysis of Vinca minor and Wisteria sinensis in Iran: with Implications for the Genetic Structure of the Virus

  • Alfalfa mosaic virus (AMV), an economically important pathogen, is present worldwide with a very wide host range. This work reports for the first time the infection of Vinca minor and Wisteria sinensis with AMV using RNA sequencing and reverse transcription polymerase chain reaction confirmation. De novo assembly and annotating of contigs revealed that RNA1, RNA2, and RNA3 genomic fragments consist of 3,690, 2,636, and 2,057 nucleotides (nt) for IR-VM and 3,690, 2,594, and 2,057 nt for IR-WS. RNA1 and RNA3 segments of IR-VM and IR-WS closely resembled those of the Chinese isolate HZ, with 99.23-99.26% and 98.04-98.09% nt identity, respectively. Their RNA2 resembled that of Canadian isolate CaM and American isolate OH-2-2017, with 97.96-98.07% nt identity. The P2 gene revealed more nucleotide diversity compared with other genes. Genes in the AMV genome were under dominant negative selection during evolution, and the P1 and coat protein (CP) proteins were subject to the strongest and weakest purifying selection, respectively. In the population genetic analysis based on the CP gene sequences, all 107 AMV isolates fell into two main clades (A, B) and isolates of clade A were further divided into three groups with significant subpopulation differentiation. The results indicated moderate genetic variation within and no clear geographic or genetic structure between the studied populations, implying moderate gene flow can play an important role in differentiation and distribution of genetic diversity among populations. Several factors have shaped the genetic structure and diversity of AMV: selection, recombination/reassortment, gene flow, and random processes such as founder effects.

Cryopreservation of Mulberry Germplasm Core Collection and Assessment of Genetic Stability through ISSR Markers

  • A simple and reliable cryo technique using desiccation and slow freezing of winter dormant buds was employed for 238 core collection of mulberry germplasm collected from diverse geographical regions and maintained under tropical conditions in the ex situ field gene bank to develop long-term biodiversity conservation for ensuring sustainable utilization of these valuable resources. Desiccation and freezing tolerance of bud grafts and excised shoot apices in the axillary buds of different Morus species under in vivo and in vitro condition indicated species-specific variation and most of the wild Morus species were found sensitive. In vitro regeneration and cryopreservation($-196^{\circ}C$) protocols using differentiated bud meristem like axillary winter dormant buds were worked out for a wide range of Morus species, land races, wild and cultivated varieties. Successful cryopreservation of mulberry winter dormant buds of different accessions belonging to M. indica, M. alba, M. latifolia, M. cathayana, M. laevigata, M. nigra, M. australis, M. bombycis, M. sinensis, M multicaulis and M. rotundiloba was achieved. Among wild species Morus tiliaefolia, and M. serrata showed moderate recovery after cryopreservation. Survival rates did not alter after three years of cryopreservation of different Morus species. ISSR markers were used to ascertain the genetic stability of cryopreserved mulberry, which showed no difference detected among the plantlets regenerated from frozen apices in comparison to the non-frozen material.

Comparison of Plant Community Structures in Cut and Uncut Areas at Burned Area of Mt. Gumo-san (금오산(金烏山)의 산화지(山火地)에서 벌목지(伐木地)와 비벌목지(非伐木地)의 식물(植物) 군집구조(群集構造) 비교(比較))

  • This is a report on the early vegetation, plant community structure, and secondary succession of cut and uncut sites of burned areas in Mt. Gumo-sun. The forest fire occurred on April, 1994 and the pine forest and its floor vegetation were burned down. The investigation was carried out from April, 1995 to October, 1996. The results are summarized as follows : The floristic composition of cut and uncut sites of burned area and unburned area were composed of 32, 36, and 34 kinds of vascular plants respectively. The biological spectra showed the $H(G)-D_1-R_5-e$ type, $H(M)-D_1-R_5-e$ and $M(N)-D_1-R_5-e$ in cut, uncut, and unburned site respectively. The dominant species based on $SDR_3$ of the cut site were Miscanthus sinensis var. purpurascens(100.00). Caret humilis(52.27), Quercus serrata(51.19) and Lysimachia clethroides(39.40), however, in the uncut sites the dominant species were Quercus acutissima(56.91), Pinus densiflora(26.83) in the tree layer, Quercus serrata(50.43), Lindera glauca(40.51), Lespedeza bicolor(37.85) in the shrub layer, and Miscanthus sinensis var. purpurascens(72.27), Pteridium aquilium var. latiusculum(60.92), Carex humilis(63.63) in the herb layer. Pinus densiflora(99.88), Miscanthus sinensis var. purpurascens(82.74), Quercus serrata(77.47) and Carex humilis(74.02) were dominant in the unburned site. The species diversity(H) and evenness index(e) were 1.05, 0.70 and 1.32, 0.85 in the cut and uncut site, respectively and 0.22, 0.63 in the unburned site. Dominance index(C) was 0.15, 0.06 and 0.96 in the cut, uncut site and unburned site, respectively. Degree of succession(DS) was 345.19, 747.47 and 674.34 in cut, uncut and unburned site, respectively. The index of similarity(CCs) was 0.66 between cut and uncut sites, 0.50 between unburned and cut sites and 0.61 between unburned and uncut sites. The amount of exchangeable sodium, calcium, magnesium and soil pH were increased, but the amount of organic matter, available phosphous, total nitrogen, total carbon and exchangeable potassium were decreased in cut site after fire.

  • 1. Claviceps species or Graminaceae were investigated from Kyonggi-Do Korea including Kwang-neung, Anseung, Pyongtack, Chingpyong Yichun and Suwon from June to November, 1969. 2. Sclerotia of Claviceps species were collected from 16 species of Graminaceae consisted of 11 genera. 3. New hosts of Claviceps species including 15 species in 10 genera were reported in Korea, 4. Claviceps pucilla Ces. on Themeda riandra FORSK. var. japonica M. was recorded in addition to Claviceps purpurea (Fr.) Tul. 5. Claviceps Purpurea (FR.) Tul. on Miscanthus Sinensis ANDERS occured at 449m above sea level near Suwon.

  • This study was conducted to investigate the growth characteristics of artificially planted trees on top of the underground parking lots. The trees were planted 17 years ago and are now in good condition. The survey results are as follows. The planting depth of the trees was 0.9 - 1.3m. The horizontal roots of the trees were distributed in the depth of 0.2m ~ 0.6m, Growth condition was also considered to be good. Also, in the case of the straight root of the trees, the pine tree(Pinus densiflora) grew near the bottom but tree root was very thin, and the Japanese hackberry tree(Celtis sinensis) and the zelkova tree(Zelkova serrata) were seemed to bend at the bottom and to grow by changing direction. When trees were planted on artificial ground, the roots grew well horizontally, and the forces of growing vertically were much weakened. because the plants were planted in earth ball state. As a result, it was considered that the roots would hardly penetrate the bottom-pressing concrete floor.

A Study on the Growth Characteristics of Native Plants by Seeding Amounts of Cool-season Turfgrasses on the Disturbed Slope (훼손비탈면의 한지형 잔디 파종량에 따른 자생식물의 생육특성에 관한 연구)

  • This research was initiated to investigate the revegetation characteristics under the seeding amount treatments of cool-season turfgrasses on the disturbed slope. 4 different seeding amounts of cool-season turfgrasses (0.0 $g/m^2$, 1.5 $g/m^2$, 3.0 $g/m^2$, 6.0 $g/m^2$) with the same seeding amounts of native woody (Rhus chinensis, Albizzia julibrissin) and herbaceous plants (Lotus corniculatus var. japonicus, Dianthus sinensis, Aster yomena) were treated with 3 replications on the experimental disturbed slope. Data such as soil hardness (mm), moisture content (%), surface coverage rate (%), the number of each germinating plant and plant height were analyzed. There were no statistic differences observed in the soil hardness and the soil moisture content while a significant difference was observed in the surface coverage rates. The surface coverage rates were higher in 3.0 $g/m^2$ and 6.0 $g/m^2$ plots compared to the rest two plots of cool-season turfgrasses. However, the emergence of native woody and herbaceous plants was relatively low in 3.0 $g/m^2$ and 6.0 $g/m^2$ plots of high seeding amounts of cool-season turfgrasses. This result indicated that high seeding amounts of cool-season turfgrasses could decrease the emergence of native woody and herbaceous plants. We concluded that 1.5 g~3.0 $g/m^2$ seeding amounts of cool-season turfgrasses was optimum level for the balanced emergence of cool-season turfgrasses, native woody and herbaceous plants in the viewpoint of the ecological disturbed slope revegetation.

  • We investigated in the existential forms of heavy metals and their distribution in plant and soil surrounding ruined mine in Daduk area. The content of heavy metals in soils was more plentiful in A soil than that in another soil, excepting only Mn. The content of Mn in soils was greater in B soil than those in another soils. The content of each heavy metals, according to existential form, had different patterns on the comparision of each elements ; organic-form on Cd and As, Carbonate-form Cu, Pb and Mn and Residual-form on Fe and Zn. The popularity ration of M. senensis and J. decipiens were about 65% and 72%, respectively, and which means that these plants had resistances to heavy metals. The contents of heavy metals absorbed in root was higher than those of stem except Mn, In M. sinensis, the contents of Zn in root was similar to that of stem. To study the heavy metal contents in water, the highest content of heavy metal, mainly Fe, Mn and Zn, were calculated in extracting water.

Optimization of the re-extraction process of ethanol from catechin components from Korean green tea extract (한국산 녹차추출물 catechin 성분의 ethanol 재추출 공정 최적화)

  • The purpose of this study was to determine the optimum re-extraction conditions of ethanol from catechin in Korean green tea (Camellia sinensis L.) using response surface methodology (RSM). The experiments were carried out according to a five level and two variable central composite design (CCD). The two independent variables were solvent ratio to sample content (1, 4, 7, 10, 13 mL/g) and extraction temperature (-20, -10, 0, 10, $20^{\circ}C$) on the dependent variables including yield, epigallocatechin (EGC), epicathchin (EC), epigallocatechingallate (EGCG), epicatechingallate (ECG), total catechin and caffeine. ANOVA results showed that Coefficients of determination ($R^2$) of estimated models for dependent variables were ranged from 0.9054~0.9778, while $R^2$ of caffeine were estimated 0.8770. The optimum ranges for the maximized extraction including yield, EGC, EC, EGCG, ECG, caffeine and total catechin were 4.5~7.5 mL/g in ratio of ethanol to sample and $-8{\sim}8^{\circ}C$ in extraction temperature. The actual values of yield, EGC, EC, EGCG, ECG, caffeine and total catechin, respectively, at the optimized conditions were 35.02%, 13.31%, 3.978%, 19.11%, 4.29%, 5.30% and 40.68%.

An Unrecorded Species of Pratylenchus Ioosi Loof (Tylenchida: Pratylenchidae) from Tea in Korea (한국산기종 차나무뿌리썩이선충, Pratylenchus Ioosi Loof(참선충 목: 뿌리썩이선충科))

  • Pratylenchus loosi Loof, 1960 is recorded for the first time from Korea. The nematode was isolated from the roots of tea (Thea sinensis L.) and soil around the roots from Yeongam-gun, Jeo-llanam-do and Namjeju-gun, Jeju-do, Korea. This species has two lip annuli. The female is 433-646 $\mu\textrm{m}$ long: a= 29.1-37.5, b = 5.1-6.4, c = 15.0-21.3, vulva(%) = 73.0-85.4. The male body length is 408-512$\mu\textrm{m}$ long: a=36.1-40.0, b=4.8-6.7, c =17.0-19.0, and spicule = 14.1-18.0 $\mu\textrm{m}$. The stylet is 11.6-18.0 $\mu\textrm{m}$ long and the number of incisures is four. The shape of spermatheca filled with sperm is broadly rounded, oval and quadrangular The shape of tail is narrowly round to subacute.
