Studies on the Acute Toxicity of an Insecticide Cartap to Several Species of Freshwater Animals (몇가지 담수산동물(淡水産動物)에 대한 살충제 Cartap의 급성독성(急性毒性)에 관한 연구(硏究))

  • The acute toxicity of an insecticide cartap to several species of freshwater animals was evaluated in the laboratory with special reference to the species specificity, effects of water temperatures and pH values. The aquatic animals tested were the Carassius auratus L., Aphyocypris chinensis $G{\"{U}}NTHER$, Misgurus anguillicaudatus CANTOR, Moina macrocopa STRAUS. The susceptibility of aquatic animals to cartap was different with the species of animals. At the water temperature of $25^{\circ}C$ and pH 7, TLm values of the insecticide to the C. auratus L., A. chinensis G. and M. anguillicaudatus were 0.88, 0.26 and 0.13 ppm in 48 hours, respectively, and to Moina macrocopa S., 306 ppm in 3 hrs. In the case of the three species of fish, TLm 48 values were significantly decreased with rise in temperature. In the case of water flea, where TLm value was 107 ppm at $20^{\circ}C$, there was no consistent response to temperature change, with the highest figure at $25^{\circ}C$ than at either 20 or $30^{\circ}C$. and the susceptibility of C.auratus L. and A. chinensis G. greatly decreased with the increase of pH in water. The toxicity to M. anguillicaudatus and M. macrocopa was significantly higher at pH 9 than at pH 6 or 7. In conclusion, the toxicological reactions of the freshwater animals to cartap were variably influenced by the water temperatures and pH values of water and species of animals.

Influence of Farming Practices on Weight-Length Relationship of the Loach (Misgurnus anguillicaudatus) Population in Rice Paddy Fields (영농방법이 논에 서식하는 미꾸리 개체군의 체중-체장 관계에 미치는 영향)

  • The length frequency and weight-length relationships within the loach population of Misgurnus anguillicaudatus were investigated in environment-friendly (EFP) and conventional paddy fields (CP) in South Korea, in order to identify the influences of farming practices to the loach population living in paddy ecosystem. The size-frequency of distribution of M. anguillicaudatus populations noted to differ significantly when the loach populations of the EFP and the CP were compared. The mean weight and length of the loach population in EFP was greater than those of the loaches in CP. The equations based on weight and total length relationship in EFP and CP were $W=0.000004L^{3.0747}$ and $W=0.000002L^{3.2106}$, respectively. The condition factor (K) against total length of loaches at two paddy field types with different farming practices indicates that the loach population in EFP(mean K=0.95) had access to better nutritional conditions than those in CP (mean K=0.67). It therefore appears that the size and structure of loach populations in rice paddy fields might be affected by farming practices.

Molecular Phylogeny of Korean Loaches Inferred from Mitochondrial DNA Cytochrome b Sequences (Mitochondrial Cytochrome b gene의 분석에 의한 한국산 미꾸리과 어류(Cobitidae)의 계통)

  • Phylogenetic relationships between 8 species Korean loaches (Cobitidae) were investigated by comparing mitochondrial cytochrome b gene sequences. However our results are in accordance with finding observed using other morphological studies, new interesting interspecific variation in Korean loaches were found. Orthrias and Lefua appeared to be paraphyletic in Cobitidae observed. Their sequence divergence value was agreed with interfamilic sequence divergences between Cobitidae and Cyprinidae ranged from 0.184 to 0.272. Otherwise, the present results support that two species of Iksookimia and Cobitis melanoleuca were early diverged respectively. And another remarkable result was sequence divergence between Misgurnus anguillicaudatus from China and M. anguillicaudatus from Yongdok, Korea. That was 0.099, which was interspecific value. Also the phylogenetic location of some Iksookimia species was suggested as the cobitid intergeneric hybrid origin.

Triploid hybridization as a reproductive containment method of genetically modified fish, exemplified by fast-growing transgenic mud loach

  • Transgenic triploid hybrid between fast-growingtransgenic mud loach (Misgurnus mizolepis) males and cyprinid loach (M. anguillicaudatus) females were generated and their performance on growth, feed conversion ability and reproduction were evaluated. Although the growth accelerations of diploid and triploid transgenic hybrids were not as much as those of original transgenic mud loaches, they still represented persistent growth stimulation ranging 11 to 28 fold when compared to their non-transgenic counterparts, with significantly improved feed conversion efficiency up to 2-fold (compared to non-transgenic hybrid) and 1.5-fold (compared to non-transgenic mud loach) in maximum. The gonad development of diploid hybrids was fertile in histological views regardless of transgenic genotypes but the extent of developmentin hybrid fish were less than mud loach diploids at the same age. On the other hands, very stringent sterility was obtained in both sexes of the triploid hybrid transgenics: ovary and testis from transgenic triploid hybrids were significantly depressed and any notable sign for maturation to ovum or spermatids was not detected. No viable embryo was obtained in a fertilization trial using the suspension prepared from the minced testes of transgenic triploid hybrids. This study may indicate the potential usefulness of triploid hybridization as a mean for reproductive containment of transgenic mud loach.

Effect of Transgenic Genotype on Transgene Expression in Mud Loach (Misgurnus mizoIepis): I. Copy Number-Dependent Expression in Gynogenetically Derived Homozygous Transgenics

  • To examine the effect of copy number-dependent transgenic genotype on the expression of foreign gene, stable hemizygous and homozygous transgenic breeding line was established using artificial parthenogenesis. For this purpose, induced diploid gynogenetic transgenesis was optimized in mud loach (Misgurnus mizolepis) using UV-irradiated cyprinid loach (M. anguillicaudatus) sperm and thermal shocks. Optimum UV range for inactivation of cyprinid loach sperm was between 3,150 to $4,050\;ergs/mm^2$ The UV-irradiated sperm were inseminated into eggs from recessive color strain (yellow) or heterozygous transgenic mud loach containing CAT gene. Cold shock at $2^{\circ}C$ for 60 min, 5 min post fertilization successfully restored the diploidy of eggs inseminated with UV-irradiated sperm. Restoration to diploidy was confirmed by flow cytometry and gynogenetic status was verified by examining maternal exclusive inheritance of multi-locus DNA fingerprints, body color and transgenic marker. Putative isogenic transgenic fish clearly showed homozygous status at trans gene locus based on Southern blot hybridization and progeny testing. Further, such homozygous gynogenetic diploids revealed the increased levels of transgene expression, when compared to those of heterozygous (hemizygous) transgenic fish.

Experimental and epidemiological studies on the life cycle of Echinostoma hortense Asada, 1926(Trematoda: Echinostomatidae) (남한강류역(南漢江流域)의 호르텐스극구흡충(棘口吸蟲) 감염실태(感染實態)와 생활사(生活史)에 관(關)한 연구(硏究))

  • Recently there have been some reports on human infections of Echinostoma hortense in Korea. It was found that a few species of freshwater fishes were playing the role of the second intermediate host of E. hortense. However, molluscan intermediate host has not been identified yet in Korea. The present study aimed to establish the life cycle of E. hortense in laboratory. Experimental studies such as egg production from the rat, development of the eggs in vitro, exposure of miracidia to freshwater snails, shedding pattern of cercariae from infected snails, morphology of cercariae, cercarial infection to the second intermediate host and infection of metacercariae to the definitive hosts were done. In addition, epidemiological surveys on the infection status in inhabitants and house rats, and on the natural infection of larval echinostomes in the snails and fishes were carried out along the South Hangang-river. The results obtained were as follows: 1. The eggs deposited from adults in physiological saline were cultivated at room temperature($20{\sim}24^{\circ}C$). The miracidia were firstly observed on 8 days after cultivation, and 85.5% of the eggs contained the mature miracidia on 11 days after cultivation. More than 90% formed the miracidia when cultivated at temperature $22{\sim}27^{\circ}C$. Hatching of the miracidia began on 12 days after cultivation and continued for a week. The size of the miracidia was $103.0{{\times}}51.4{\mu}m$ in average. The motility of the miracidia were active up to 8 hours after shedding, but they were all dead within 10 hours after shedding. 2. A freshwater snail, Radix auricularia coreana was cultivated in aquaria. A hatched $F_1$ snails from the egg masses were exposed to 20 miracidia respectively. Escape of cercariae started on 15 days after infecton. Radix auricularia coreana was experimentally identified as the first intermediate host of E. hortense in Korea. 3. Cercarial shedding started on $15{\sim}20$ days after infection by snail, continued for about 10 days (8.8 days in average). Infected snails were dead within 32 days after the miracidial infection. About 1,335 cercariae($328{\sim}1,994$) per snail were shed in its life, and 119 cercariae in average per snail per day were shed. The cercariae were motile for more than 24 hours, and then squirming at the bottom until death. The body and tail sizes of cercariae were $356{\times}186{\mu}m$ and $510{\times}68{\mu}m$ in average, respectively. The rediae parasitized in the snail hosts were found mainly around the pericardial regions, and their size was $1,575{\times}258{\mu}m$ in average. The numbers of developing cercariae in a mature redia were 14 in average ($7{\sim}20$ in range). The numbers of rediae in a snail were 102 in average on 15 days after miracidial infection and 221 in average on 28 days. 4. Three uninfected Misgurnus anguillicaudatus, less than 6.5cm long were used in for the cercarial infection. They were all exposed with 755 cercariae, and examined at 5-day intervals starting from 10 days after infection. All the fishes were infected with metacercariae of E. hortense and a total of 275 was found infected(36.4%). The metacercariae were fed to rats and the adult worms were obtained on 15 days after infection. 5. The infected rats began to deposit the eggs on 11 days after infection. The number of eggs deposited per day per worm (EPD/worm) was $400{\sim}500$ on 3 weeks after infection and was increased to $1,000{\sim}1,500$ on 4 to 17 weeks, then decreased to 800 on 21 weeks after infection. 6. A total of 745 stool specimens collected from 576 male and 169 female residents of 8 different villages along South Hangang basin was examined. Out of 745 specimens, the eggs of Echinostoma sp. were found in 2 cases (0.3%). Of 34 house rats one showed egg-positive (2.9%). 7. Total 971 Radix auricularia coreana collected from 7 sampling stations were examined for shedding of cercariae. Three snails (0.3%) shed the cercariae of E. hortense. A total of 119 out of 542 freshwater fishes (22.0%) had the metacercariae of E. hortense. The fishes parasitized with the metacercariae were 4 out of 14 examined species. The infection rates of 4 species were 34.1% (106 out of 311) in Misgurnus anguillicaudatus, 30.4% (7 out of 23) in Misgurnus mizolepis, 4.3% (2 out of 46) in Moroco oxycephalus and 22.2% (4 out of 18) in Odontobutis obscura interrupta. In summarizing the above results, the first intermediate host of E. hortense was found as Radix auricularia coreana in Korea. Also, it took about 46 days for the shortest completion of a life cycle of E. hortense in summer; that is, 10 days for miracidial development in eggs, 15 days for cercarial development in the snail, about 10 days for metacercarial development in the second intermediate hosts, and 11 days for the maturation as the adults in the definitive hosts. The natural infection rates of E. hortense in the intermediate hosts were relatively high but those in the definitive hosts were low in the middle areas of South Hangang basin.

The Ichthyofauna and Community Structure of Fish at Wondong Marsh in the Kyeongnam Province, Korea (원동습지의 어류상과 군집구조)

  • The ichthyofauna and the structure of fish community were surveyed from June to August in 2000 at four stations of the Wondong marsh, Yangsan city in Kyungnam province. During the study period, 24 species belonging to eight families were collected. Cyprinid fish occupied 62.5% (15 species) and cobitid and centrarchid fish 8.3% (each 2 species). According to relative abundance in the whole marsh, Z. platypus (23.2%) was dominant species and S. gracilis majimae (14.2%) was subdominant species. M. salmoides (9.6%), C. auratus (8.3%), P. herzi (8.3%), M. yaluensis (6.5%), M. anguillicaudatus (4.0%), L. macrochirus (3.4%), P. parva (2.8%), R. ocellatus (2.5%), C. brevicauda (2.5%), C. sinensis (2.5%) and O. platycephala (2.2%) were common species and C. carpio, A. rhombeus, H. labeo, S. variegatus wakiyae, S. chankaensis tsuchigae, Z. temmincki, O. bidens, S. asotus, C. herzi, R. brunneus, C. argus were rare specieswhich occupied less than 2.0% in relative abundance. Six species of S. variegatus wakiyae, S. chankaensis tsuchigae, S. gracilis majimae, M. yaluensis, C. herzi and O. platycephala in observed species were known as endemic species. By analyzing of fish community in the surveyed area, the species diversity and evenness of the fishes were high but dominance was low. And the similarity of the fish species among the sites was mid-grade as more than 0.55 in index.

The Risk Assessment of Butachlor for the Freshwater Aquatic Organisms (Butachlor의 수서생물에 대한 위해성 평가)

  • To assess the effect of butachlor on freshwater aquatic organisms, acute toxicity studies for algae, invertebrate and fishes were conducted. The algae grow inhibition studies were carried out to determine the growth inhibition effects of butachlor (Tech. 93.4%) in Pseudokirchneriella subcapitata (formerly knows as Selenastrum capriconutum), Desmodesmus subspicatus (formerly known as Scendusmus subspicatus), and Chlorella vulgaris during the exposure period of 72 hours. The toxicological responses of P. subcapitata, D. subspicatus, and C. vulgaris to butachlor, expressed in individual $ErC_{50}$ values were 0.002, 0.019, and $10.4mgL^{-1}$, respectively and NOEC values were 0.0008, 0.0016, and $5.34mg\;L^{-1}$, respectively. P. subcapitata was more sensitive than any other algae species. Butachlor has very high toxicity to the algae, such as P. subcapitata and D. subspicatu. In the acute immobilisation test for Daphnia magna, the 24 and $48h-EC_{50}$ values were 2.55 and $1.50mg\;L^{-1}$, respectively. As the results of the acute toxicity test on Cyprinus carpio, Oryzias latipes and Misgurnus anguillicaudatus, the $96h-LC_{50}s$ were 0.62, 0.41 and $0.24mg\;L^{-1}$, respectively. The following ecological risk assessment of butachlor was performed on the basis of the toxicological data of algae, invertebrate and fish and exposure concentrations in rice paddy, drain and river. When a butachlor formulation is applied in rice paddy field according to label recommendation, the measured concentration of butachlor in paddy water was $0.41mg\;L^{-1}$ and the predicted environmental concentration (PEC) of butachlor in drain water was $0.03 mg\;L^{-1}$. Residues of butachlor detected in major rivers between 1997 and 1998 were ranged from $0.0004mg\;L^{-1}$ to $0.0029mg\;L^{-1}$. Toxicity exposure ratios (TERs) of algae in rice paddy, drain and river were 0.004, 0.05 and 0.36, respectively and indicated that butachlor has a risk to algae in rice paddy, drain and river. On the other hand, TERs of invertebrate in rice paddy, drain and river were 3.6, 50 and 357, respectively, well above 2, indicating no risk to invertebrate. TERs of fish in rice paddy, drain and river were 0.58, 8 and 57, respectively. The TERs for fish indicated that butachlor poses a risk to fish in rice paddy but has no risk to fish in agricultural drain and river. In conclusion, butachlor has a minimal risk to algae in agricultural drain and river exposed from rice drainage but has no risk to invertebrate and fish.

Distribution of Fish in Paddy Fields and the Effectiveness of Fishways as an Ecological Corridor between Paddy Fields and Streams (소규모 어도 설치에 따른 논 주변 생태계의 연계성 평가)

  • Agriculture modern environments can vary due to factors such as land consolidation and ditch enhancement projects. But, these improvements projects can include cover-ing-up of irrigation ditches with concrete, increasing the drop between paddy field and drainage ditches, which might decrease the abundance and diversity of fish fauna around paddy fields. In this study, for the management of agrobiodiversity on fish in paddy fields, we installed a small-scale fishways between paddy fields and drainage ditches, and evaluated the effects on the eco-connection of the paddy fields, ditches and stream. Five fish species were recovered at the drainage ditches. The species exhibited characteristics spawning and growth based on the paddy field. The results indicate that the five fish species could ascend the paddy fields through the small-scale fishways. There are no difference of species numbers at ditches of environment-friendly agriculture paddy fields (A) and good agricultural practices (B) region, but individual numbers were higher at the B region. This result could be interpreted as indicating that ditch diversity was affected by positional properties rather than farming practices because the water flow and connection to adjacent stream of B region were better than A region. After ascertaining the fish species capable of ascending in a pre-survey of the drainage ditches, we set up small-scale fishways at the drop between paddy fields and drainage ditches. Three species of fish (Aphyocypris chinensis, Misgurnus anguillicaudatus, M. mizolepis) arrived at the paddy fields via small-scale fishways. The main movement time was from 18:00~24:00 and 00:00~06:00, indicating a preference for the night time period rather than the day period for migration. Concentrating the operation time from night to dawn seems prudent for effective management of small-scale fishways.
