Effects of Zinc on the Activity and Conformational Changes of Arginine Kinase and Its Intermediate

  • The effects of zinc on arginine kinase and its collapsed-state intermediate were studied. Both arginine kinase and the collapsed-state intermediate were inactivated in the presence of zinc, following a biphasic kinetic course. The corresponding apparent rate constants of inactivation at different zinc concentrations and conformational changes in the presence of 0.5 mM zinc were obtained. The conformational changes of arginine kinase and the collapsed-state intermediate were followed by fluorescence spectra and circular dichroism spectra. Comparison of the results for arginine kinase and the collapsed-state intermediate showed that the collapsed-state intermediate was more susceptible to zinc, which indicated that the collapsed-state intermediate was more flexible and unstable than arginine kinase. The special structure of arginine kinase might explain these diverse phenomena.

  • PYP consists of a water-soluble apoprotein and 4-hydroxycinnamyl chromophore bound to Cys69 via thiolester linkage, Upon absorption of a photon, the photocycle is initiated, leading to formation of several photo-intermediates. Among them, M intermediate is important to understand the signal transduction mechanism of PYP, because it is a putative signaling state. As well known, the dynamics of a protein is closely correlated with the occurrence of its function. Here we report the results of IO ns molecular dynamics (MD) simulation for the M intermediate in aqueous solution and discuss the characteristic feature of this state from a viewpoint of structural fluctuation.

  • In the multi-pass shape drawing process, it is important to design the intermediate dies for producing sound products. Up to now, the design of the intermediate dies is mainly carried out by the industrial experts based on their experience. In this study, a design program was developed to design the intermediate dies for multi-pass shape drawing process. The program was programmed by using VisualLISP. In this program the intermediate dies can be designed by using the initial material shape and the final product shape. In order to verify the effectiveness, the program was applied to design the intermediate dies of multi-pass shape drawing for producing four teeth spline and gun slide. Finally, FE analysis and shape drawing experiment were performed to verify the effectiveness of the designed intermediate dies. As a result, it was possible to produce the drawn products with the required dimensional accuracy.

  • The multi-stage shape drawing is used to obtain long shaped products with high levels of dimensional accuracy and quality. It is important to design the cross sectional shapes of the intermediate passes to meet the required dimensional accuracy of the final product in the multi-stage shape drawing. Until now, the cross sectional shapes of the intermediate passes have been designed by the experiences. It is still remained unsolved problem to design the cross sectional shapes of intermediate pass drawing dies in the multi-pass shape drawing. In this study, a new technique is proposed to design the cross sectional shapes of intermediate passes. The proposed method is applied to a multi-stage shape drawing for a LM-guide which is one of the representative shape drawing products. In order to verify the effectiveness of the proposed method, FE-simulation and experiments have been carried out. The dimensional accuracy of the proposed method is compared with that of the conventional shape drawing process designed by the industrial engineers.

  • Receptor-mediated endocytosis has been suggested for a stage-specific transport mechanism of vitellogenin into the oocytes of a sabellid poly chaete, Pseudopotamilla occelata. Membrane proteins of oocytes of three size classes, including small (30-70 $\mu\textrm{m}$ in diameter), intermediate (70-140 $\mu\textrm{m}$ in diameter) and large (180-200 $\mu\textrm{m}$ in diameter), showed a atage-specific variation. Coelomic fluid proteins (CP), ass$\mu\textrm{m}$ed to be vitellogenin, consists of several proteins, which showed quite a different pattern from that of yolk proteins. Incorporation of $^{125}I$-CP into the oocytes of the intermediate size class almost linearly increases with time, showing a contrast to the pattern of the large size class, in which the incorporation is low and approaches a plateau, suggesting the vitellogenin transports by a regulated process only in the intermediate size class. Vitellogenin receptor proteins were identified to be 60 kDa and 68 kDa only in the intermediate size class by a ligand blotting test.

Phenotypic Analysis of Neurofilament Light Chain E397K Mutant in Cultured Cells

  • Charcot-Marie-Tooth disease (CMT) is blown as one of the inherited disorder of peripheral nervous system. Recently, it was found that point mutations in the neurofilament light subunit (NF-L) gene cause CMT. Neurofilaments (NFs) are heteropolymers consist of NF-L, NF-M and NF-H. To assess the relationship between CMT and NF-L mutation in cellular level, we performed phenotypic analysis of the mutant NF-L (E397K) using cultured cell lines. Vimentin-deficient human adrenal carcinoma SW13 (Vim-) cells have a potential to form the intermediate filaments when the cells are expressing both NF-L and NF-M. Our results show that co-expression of wild type NF-L with NF-M showed intermediate filament formation in SW13 (Vim-) cells, while E397K with NF-M did not. This result means that E397K mutant lost its ability to form the intermediate filament in vivo, and further suggests that the E397K mutation is closely related to CMT.

  • Peroxidase activity of cytochrome P-450 was examined using N, N-dimethylaniline (NDA) as a substrate and cumene hydroperoxide (CHP) as an oxidant. The initial rates of the N-demethylation for varied concentrations of NDA (0.05-0.5 mM) by P-450 at different fixed concentrations of CHP (0.02-0.2 mM) were determined. The results suggest that P-450 proceeds its peroxidative reaction by the rapid equilibrium random bi bi mechanism to form a ternary complex with substrate and oxidant as an active intermediate.

  • Objectives : This study was carried to identify the component of the Pericardium Meridian Muscle in human. Methods : The regional muscle group was divided into outer, middle, and inner layer. The inner part of body surface were opened widely to demonstrate muscles, nerve, blood vessels and to expose the inner structure of the Pericardium Meridian Muscle in the order of layers. Results We obtained the results as follows; He Perfcardium Meridian Muscle composed of the muscles, nerves and blood vessels. In human anatomy, it is present the difference between terms (that is, nerves or blood vessels which control the muscle of the Pericardium Meridian Muscle and those which pass near by the Pericardium Meridian Muscle). The inner composition of the Pericardium Meridian Muscle in human is as follows ; 1) Muscle P-1 : pectoralis major and minor muscles, intercostalis muscle(m.) P-2 : space between biceps brachialis m. heads. P-3 : tendon of biceps brachialis and brachialis m. P-4 : space between flexor carpi radialis m. and palmaris longus m. tendon(tend.), flexor digitorum superficialis m., flexor digitorum profundus m.   • In the multi-pass shape drawing process, the appropriate process design is very important to produce sound products. The reduction ratio, die angle, and the intermediate die shape are very important process variable of the multi-pass shape drawing. The aim of this study is the determination of the reduction ratio, die angle, and the intermediate die shape of the 2 pass shape drawing process for producing steering spline shaft. In this study, FE analysis, Taguchi method, and ANN(artificial neural network) were applied to determine the appropriate reduction ratio, die angle, and intermediate die shape. After the determination of the process variables, FE analysis and drawing experiment were performed to evaluate the effectiveness of the determined process variables. The dimensional accuracy of the final drawn spline shaft was evaluated by using 3D surface profiler and 3D laser digitizing system.

  • The intermediate M420 formed in a solution of firefly luciferin in deoxygenated dimethyl-sulfoxide added potassium f-butoxide was observed to emit yellow-green and red light by filter-photometry. By H-NMR, M420 was found to be deprotonated at the site where luciferin reacts with oxygen.
