Polymorphism of the Promoter Region of Hsp70 Gene and Its Relationship with the Expression of HSP70mRNA, HSF1mRNA, Bcl-2mrna and Bax-AMrna in Lymphocytes in Peripheral Blood of Heat Shocked Dairy Cows

  • The blood samples were collected from dairy cows at the same milking stage. The single-strand conformation polymorphism (PCR-SSCP) method was used to analyze for polymorphism at the 5'flanking region of the hsp70 gene. The mRNA expression levels of HSP70, HSF1, Bcl-2 and Bax-$\alpha$ at different daily-mean-temperature were analyzed by relative quantitative RTPCR. The DNA content, cell phase and the ratio of apoptosis of lymphocytes in peripheral blood of dairy cattle at different daily-meantemperature were determined by FCM. The PCR-SSCP products of primer pair 1 showed polymorphisms and could be divided into four genotypes: aa, ab, ac, cc, with the cis-acting element (CCAAT box) included. Mutations in the hsp70 5'flanking region (468-752 bp) had different effects on mRNA expression of HSP70, HSF1, Bcl-2 and Bax-$\alpha$. The ac genotypic cows showed higher expressions of HSP70mRNA, HSF1mRNA and Bcl-2mRNA/Bax-$\alpha$mRNA and lower ratio of apoptosis. These mutation sites can be used as molecular genetic markers to assist selection for anti-heat stress cows.

Development of Rapid Detection Method for Unfolded Protein Response in the Mammalian Cells

  • The mammalian unfolded protein response (UPR) protects the cell. against the stress of unfolded or misfolded proteins in the endoplasmic reticulum (ER). It has recently demonstrated that IRE1, PERK, ATF6, and X-box protein 1 (XBP-l) directly or indirectly participate in this process. Upon accumulation of unfolded/misfolded proteins in the ER lumen, release of BiP from Ire1p permits dimerization and autophosphorylation to activate its kinase and endoribonulease activities to initiate XBP-1 mRNA splicing. Spliced XBP-1 mRNA removed middle part of 23 bp and encodes a potent transcription factor, XBP-l protein that binds to the unfolded protein response element (UPRE) or endoplasmic reticulum stress element (ERSE) sequence of many UPR target genes and produces several kind of ER chaperones. In this study, we described both the result and the detailed experimental procedures of XBP-1 mRNA splicing induced by ER stress, this result might help to elucidate the roles of the UPR and early diagnosis in a number of human diseases involving endoplasmic reticulum storage disease (ERSD).

대장균의 xylA 프로모터 영역의 조절 특성 (Regulatory Characterization of xylA Promoter Region in Escherichia coli)

  • xylA 유전자의 프로모터상에서 조절양상을 조사하기 위하여 xylA 유전자의 프로모터(Pxyl)와 lacZ 유전자를 연결한 Pxyl-lacZ 융합 유전자를 제작하여 xylose에 의한 ${\beta}-galactosidase$ 생산의 조절양식을 조사하였다. xylA 프로모터 부위를 분리하여 lac 프로모터가 없는 고복제수의 lac 오페론 백터인 pMC1403에 클로닝시켜 pMCX191을 제작하여 reading frame에 변화가 없는 Pxyl-lacZ 융합 유전자를 만들었으며 이 벡터에서 Pxyl-lacZ 단편을 분리한 후 저복제수 벡터인 pLG339에 클로닝시켜 pLGX191을 제작하였다. 상기 플라스미드들을 xylA 변이주인 DH77에 형질전환시켜 Pxyl-lacZ 융합 유전자에서 ${\beta}-galactosidase$의 발현조절을 조사한 결과 xylose 농도에 따른 유도, glucose에 의한 발현억제 및 cAMP에 의한 억제해제 양상 등이 염색체상의xylA 유전자의 발현조절과 같은 경향을 나타내었다. pMCX191과 pLGX191을 이용하여 유전자 투여 량 효과를 본 결과도 복제수에 따른 차이가 크지 않았다. xylA 프로모터 부위내 조절영역를 추정하기 위해 구조유전자 상류 -209 bp를 포함한 xylA 유전자를 pUC19에 클로닝시킨 pUX30에서 프로모터 부위가 부분결손된 벡터들을 제작하여 결손부위의 염기서열을 확인하였다. 이들 부분결손 xylA 프로모터를 가진 xylA 유전자에서 xylose isomerase의 발현을 조사한 결과, 번역 개시점에서 -166 bp 이상의 영역을 결손시킨 pUX31과 pUX32의 경우pUX30과 비슷한 발현 양상을 보인 반면 -120 bp 이상의 영역을 결손시킨 pUX33과 pUX34에서는 모벡터에 비해 약 30% 수준의 발현을 보였다. 또한 pUX33과 pUX34에서는 xylose 유도시 cAMP에 의한 발현 촉진효과도 볼 수 없었다. -59 bp 이상의 부위가 결손된 pUX35의 경우에는 전혀 xylA 유전자의 발현이 일어나지 않았다. 이러한 결과로 볼때 xylA 프로모터내의 조절부위는 -165 bp에서 -59 bp 사이에 존재하는 것으로 추정되었다.

Intestinal segment and vitamin D3 concentration affect gene expression levels of calcium and phosphorus transporters in broiler chickens

  • Two experiments were conducted in this research. Experiment 1 investigated the spatial expression characteristics of calcium (Ca) and phosphorus (P) transporters in the duodenum, jejunum, and ileum of 21-day-old broilers provided with adequate nutrient feed. Experiment 2 evaluated the effects of dietary vitamin D3 (VD3) concentration (0, 125, 250, 500, 1,000, and 2,000 IU/kg) on growth performance, bone development, and gene expression levels of intestinal Ca and P transporters in 1-21-day-old broilers provided with the negative control diet without supplemental VD3. Results in experiment 1 showed that the mRNA levels of calcium-binding protein 28-kDa (CaBP-D28k), sodium-calcium exchanger 1 (NCX1), plasma membrane calcium ATPase 1b (PMCA1b), and IIb sodium-phosphate cotransporter (NaPi-IIb) were the highest in the broiler duodenum. By contrast, the mRNA levels of inorganic phosphate transporter 1 (PiT-1) and 2 (PiT-2) were the highest in the ileum. Results in experiment 2 showed that adding 125 IU/kg VD3 increased body weight gain (BWG), feed intake (FI), bone weight, and percentage and weight of Ca and P in the tibia and femur of 1-21-day-old broilers compared with the negative control diet (p < 0.05). The rise in dietary VD3 levels from 125 to 1,000 IU/kg further increased the BWG, FI, and weights of the bone, ash, Ca, and P (p < 0.05). No difference in growth rate and leg bone quality was noted in the broilers provided with 1,000 and 2,000 IU/kg VD3 (p > 0.05). Supplementation with 125-2,000 IU/kg VD3 increased the mRNA abundances of intestinal Ca and P transporters to varying degrees. The mRNA level of CaBP-D28k increased by 536, 1,161, and 28 folds in the duodenum, jejunum, and ileum, respectively, after adding 1,000 IU/kg VD3. The mRNA levels of other Ca and P transporters (PMCA1b, NCX1, NaPi-IIb, PiT-1, and PiT-2) increased by 0.57-1.74 folds by adding 1,000-2,000 IU/kg VD3. These data suggest that intestinal Ca and P transporters are mainly expressed in the duodenum of broilers. Moreover, the addition of VD3 stimulates the two mineral transporter transcription in broiler intestines.

LC-MS/MS 시스템을 이용한 모유 중 파라벤류 분석법 확립 (Analysis Method of Parabens in Human Breast Milk by LC-MS/MS System)

  • 파라벤은 화장품, 식품, 의약품 등에 첨가되는 살균성 보존제로 미생물의 성장을 억제한다. 그러나 체내에서 에스트로겐과 유사한 작용을 하는 내분비계 교란물질이며, 특히 독성물질에 민감한 영유아 및 태아에게 노출될 경우 생식기 기형, 고환암, 성조숙증 등 성장발달에 큰 영향을 미친다. 따라서 본 연구에서는 영아가 주로 섭취하는 모유에서의 파라벤 수준을 확인하기 위하여 모유 중 4가지 파라벤(methyl paraben, ethyl paraben, propyl parabne, butyl paraben)의 분석법을 확립하였다. 시료는 액체상추출(Liquid-liquid extraction, LLE)을 통하여 전처리 한 후, 액체크로마토그래피 텐덤 질량분석기(liquid chromatography tandem mass spectrometry, LC-MS/MS)를 사용하여 정량분석 하였다. 확립된 분석조건에서 얻어진 검량선의 직선성은 r2 > 0.999이고, 검출한계(LOD)는 0.019~0.044 ng/mL이었다. 또한 정확도는 85.3~105.9%, 정밀도는 10% 이하의 값을 나타내었다. 10개의 모유시료 중 파라벤류의 평균농도 ± 표준편차는 MP 0.660 ± 0.519 ng/mL, EP 1.631 ± 2.081 ng/mL, PP 0.326 ± 0.320 ng/mL이었고, BP는 불검출 되었다.

소 반추위 메타게놈에서 비배양 세균의 α-amylase 유전자 클로닝 (Cloning of α-Amylase Gene from Unculturable Bacterium Using Cow Rumen Metagenome)

  • 미생물 메타게놈은 특이한 생체촉매의 다양한 원료로 제공된다. 한우의 반추위에서 게놈 DNA를 분리한 후 메타게놈 은행을 구축하고 $\alpha$-amlylase를 암호화하는 유전자를 클로닝하여 DNA 및 아미노산 서열을 밝히고 생화적 특징을 조사하였다. amyA유전자는 1,893 bp로 631개의 아미노산 잔기를 가진 단백질을 암호화였으며 효소의 분자량은 단백질 전기영동 결과 약 71,000 Da으로 확인되었다. 이 효소를 다른 아밀라제와 비교한 결과 $21-59\%$의 상동성을 보였다. AnyA는 pH $6.40\%$에서 최적 활성을 나타내었고, pl값은 5.87이었다. E. coliDH5$\alpha$에서 발현된 AmyA의 활성은 $\Mg^{2+}$(20mM), $Ca^{2+}$ (30 mM) 존재 시 그 활성이 증가하였고, $Fe^{2+}$, $Cu^{2+}$ 존재 시 저해되었다. amyA 유전자의 internal primer를 사용하여 인공적으로 배양할 수 있는 49종의 반추세균에서 분리한 게놈 DNA을 주형으로 PCR분석한 결과 해당하는 벤드를 확인할 수 없었다. AmyA는 현재로 배양할 수 없는 반추 미생물에서 온 것으로 추정된다.

Age quadratically affects intestinal calcium and phosphorus transporter gene expression in broiler chickens

  • Objective: This research aimed to evaluate the effects of age on growth, tibia development, and intestinal calcium (Ca) and phosphorus (P) transporter gene expressions in broiler chickens. Methods: A total of 224 male Arbor Acres broilers were fed with nutrient-adequate diets and reared in eight cages (28 broilers per cage). Eight broilers (one broiler per cage) were selected and killed at 5, 10, 15, 20, 25, 30, 35, and 40 days of age, respectively. Results: Body weight continuously increased with age of broiler chickens from 5 to 40 days. The bone weight, ash weight, diameter, and length of the tibia also increased with broiler age. By contrast, the tibia ash, Ca, and P percentages quadratically changed with age (p<0.001), and the highest values of mineral contents were observed at 20, 25, and 25 days of age, respectively. The mRNA abundances of calcium-binding protein 28-kDa (CaBP-D28k), sodium-calcium exchanger 1 (NCX1), and plasma membrane ATPase 1b (PMCA1b) increased from 5 to 25 days and then decreased up to 40 days. Similar results were noted in the mRNA abundances of IIb sodium-phosphate cotransporter (NaPi-IIb), inorganic phosphate transporter 1 (PiT-1), inorganic phosphate transporter 2 (PiT-2), nuclear vitamin D receptor (nVDR), and membrane vitamin D receptor (mVDR). The mRNA abundances of Ca and P transporters and VDRs were the highest at 25 days of age. Conclusion: These data indicate that age quadratically affects intestinal Ca and P transporter gene expression and mineral absorption capacity in broiler chickens.

Molecular Cloning and Functional Expression of esf Gene Encoding Enantioselective Lipase from Serratia marcescens ES-2 for Kinetic Resolution of Optically Active (S)-Flurbiprofen

  • An enantioselective lipase gene (esf) for the kinetic resolution of optically active (S)-flurbiprofen was cloned from the new strain Serratia marcescens ES-2. The esf gene was composed of a 1,845-bp open reading frame encoding 614 amino acid residues with a calculated molecular mass of 64,978 Da. The lipase expressed in E. coli was purified by a three-step procedure, and it showed preferential substrate specificity toward the medium-chain-length fatty acids. The esf gene encoding the enantioselective lipase was reintroduced into the parent strain S. marcescens ES-2 for secretory overexpression. The transformant S. marcescens BESF secreted up to 217kU/ml of the enantioselective lipase, about 54-fold more than the parent strain, after supplementing 3.0% Triton X-207. The kinetic resolution of (S)-flurbiprofen was carried out even at an extremely high (R,S)-flurbiprofen ethyl ester [(R,S)-FEE] concentration of 500 mM, 130 kU of the S. marcescens ES-2 lipase per mmol of (R,S)-FEE, and 1,000 mM of succinyl ${\beta}-cyclodextrin$ as the dispenser at $37^{\circ}C$ for 12h, achieving the high enantiomeric excess and conversion yield of 98% and 48%, respectively.

Tissue Distribution, SNP Detection and Association Study with Immune Traits of Porcine LBP and CD14 Genes

  • Lipopolysaccharide binding protein (LBP) and CD14 protein play important roles in the defense against infection of Gram-negative bacteria. In the present study, tissue distribution and polymorphism of porcine LBP and CD14 genes were analyzed. Real-time PCR results showed that the porcine LBP gene was especially highly expressed in liver, while CD14 gene was highly expressed in liver and spleen tissues. A 1,732 bp cDNA fragment of porcine LBP gene and a 1,682 bp genomic DNA fragment of CD14 gene were isolated. Polymorphisms were identified in these two fragments and showed that there were 14 potential SNPs in the porcine LBP gene and 3 potential SNPs in the porcine CD14 gene. Three SNPs, 292G/A (Gly/Ser), 1168G/A (Ala/Thr) of the LBP gene and -61G/A of the CD14 gene, were genotyped using restriction fragment length polymorphism (RFLP) method. Association analyses indicated that polymorphism of the 292G/A locus was significantly associated with porcine immune traits hematocrit (HCT), IgG and delayed-type hypersensitivity (DTH) (p<0.01), and the 1168G/A locus was significantly associated with HCT and mean corpuscular volume (MCV) traits (p<0.05). No significant association was found between the -61G/A locus and immune traits of the pig. Our data indicated that the LBP gene was significantly associated with immune traits of pig. Also, we identified some SNPs which may be useful markers for disease-resistant breeding of pigs.

대장균 Leucine-Responsive Regulatory Protein의 정제 및 형광분광학적 특성 분석 (Purification and Fluorometric Analysis of Leucine-Responsive Regulatory Protein from Escherichia coli)

  • Leucine-responsive regulatory protein (Lrp)는 대장균 (Escherichia coli)에서 발견된 '글로벌 조절자(global regulator)'로서 Lrp-regulon이 leucine에 의하여 상이한 형태의 조절 양상을 나타낸다. 6XHis-tag 시스템으로 제조한 야생형 Lrp (Lrp Wt)와 돌연변이 Lrp (Lrp R145W) 단백질을 정제하여 그들의 생화학적 성질을 조사하였다. 이들은 gel retardation assay를 통하여 ilv 오페론의 프로모터 영역 consensus 염기서열인 21bp의 이중가닥 DNA와 결합하여 복합체를 형성하는 것을 확인 하였다. 형광성 아미노산인 tryptophan을 지닌 Lrp R145W은 단백질의 농도가 증가함에 따라 형광이 커졌으며, 아미노산 leucine에 의하여 형광성의 변화가 관찰되었다. 즉 1 ${\mu}M$의 Lrp R145W 단백질에 leucine을 첨가하여 결합시키면 약 20 ${\mu}M$까지는 형광이 증가하다가 그 이상의 농도에서는 감소하는 양상을 얻었다. 이들 실험 결과는 leucine과 Lrp의 결합 양상 및 구조변이에 관한 심층연구에 있어서 Lrp의 고유 형광성이 요긴하게 쓰일 수 있음을 시사한다.
