Comparison of Fermentation Characteristics of Italian Ryegrass (Lolium multiflorum Lam.) and Guineagrass (Panicum maximum Jacq.) during the Early Stage of Ensiling

  • The fermentation characteristics and mono- and di-saccharides compositions during the early stage of ensiling were studied with a temperate grass, Italian ryegrass (Lolium multiflorum Lam.) and a tropical grass, guineagrass (Panicum maximum Jacq.). The laboratory silos were kept in the room set at 25$^{\circ}C$, and then were opened on 0.5, 1, 2, 3, 5 and 7 days (14 days in Italian ryegrass) after ensiling, respectively. The Italian ryegrass silage showed a fast and large pH decrease caused by a fast and large production of lactic acid during the first 5 days of ensiling and succeeded to achieve lactic acid type fermentation; high lactic acid/acetic acid and lactic acid content at the end of ensiling (14 days), low values of pH (3.74), acetic acid, ethanol and ammonia-N/total nitrogen, none or only small amounts of Butyric acid, valeric acid and propionic acid. The guineagrass silage showed a slow decrease in pH and a slow increase in lactic acid content during the full ensiling period, causing a high final pH value, low contents of lactic acid, acetic acid, total volatile fatty acids and total organic acids. In Italian ryegrass silage, mono- and di-saccharides compositions decreased largely within the initial 0.5 day (12 h) of ensiling. Sucrose disappeared rapidly within the initial 0.5 day of ensiling, but fructose and glucose contents showed an initial rise by the activity of enzymes in plant tissues, and then decreased gradually. On the other hand, the contents of monoand di-saccharides in guineagrass showed the largest decreases due mainly to plant respiration within the initial 0.5 day of ensiling, and no initial rises in fructose and glucose contents during the early stage of ensiling because of the absence of fructans which are hydrolyzed into fructose and glucose in temperate grasses. In both silages, the rate of reduction in mono- and di-saccharides compositions within the initial 5 days of ensiling was ranked in the order of glucose>fructose>sucrose, suggesting that glucose and fructose might be more favorably utilized than sucrose by microorganisms and glucose is the first fermentation substrate. It was concluded that the silage made from Italian ryegrass with high moisture content had a good fermentation quality owing to the dominance of lactic acid bacteria and active lactic acid fermentation during the initial stage of ensiling. These results can be explained by rapid plant sap liberation and the high activity of plant enzyme hydrolyzed fructans into fructose and glucose within the initial 2 days of ensiling, which stimulate the homofermentative lactic acid bacteria growth. In ensiling a temperate grass, the physical characteristics may ensure the rapid onset of fermentation phase, which results from the smaller losses of water-soluble carbohydrates during the initial stage of ensiling and providing sufficient water-soluble carbohydrates for lactic acid bacteria. The silage made from guineagrass with intermediate dry matter and high initial mono- and di-saccharides content was stable silage. This could be explained by the higher incorporation of air during the very early stage of ensiling and the restriction of cell breakdown and juice release due to the properties of a tropical grass with coarse porosity and stemmy structures. These physical characteristics delayed the onset of lactic acid bacteria fermentation phase by extending the phases of respiration and aerobic microorganisms activity, causing the higher loss of water-soluble carbohydrates and the shortage of lactic acid bacteria fermentation substrates.

A Co-inhibitory Molecule, B7-H4, Synergistically Potentiates Oral Tolerance by Inducing CD4+CD25+FoxP3+ T Cells

  • Background: A co-inhibitory molecule, B7-H4, is believed to negatively regulate T cell immunity by suppressing T cell proliferation and inhibiting cytokine production. However, the mechanism behind B7-H4-mediated tolerance remains unclear. Methods: Balb/c $(H-2^d)$ mice were fed with dendritic cell line, DC2.4 $(H-2^d)$ every day for 10 days. Meantime, mice were hydrodynamically injected with recombinant plasmid expressing B7-H4 fusion protein (B7-H4.hFc) or hFc via tail vein. One day after last feeding, mice were immunized with allogeneic B6 spleen cells. 14 days following immunization, mice were challenged with B6 spleen cells to ear back and the ear swelling was determined the next day. Subsequently, a mixed lymphocyte reaction (MLR) was also performed and cytokines profiles from the reaction were examined by sandwich ELISA. Frequency of immunosuppressive cell population was assayed with flow cytometry and mRNA for FoxP3 was determined by RT-PCR. Results: Tolerant mice given plasmid expressing B7-H4.hFc showed a significant reduction in ear swelling compared to control mice. In addition, T cells from mice given B7-H4.hFc plasmid revealed a significant hyporesponsiveness of T cells against allogeneic spleen cells and showed a significant decrease in Th1 and Th2 cytokines such as IFN-${\gamma}$, IL-5, and TNF-${\alpha}$. Interestingly, flow cytometric analysis showed that the frequency of CD4+CD25+FoxP3+ Tregs in spleen was increased in tolerant mice given recombinant B7-H4.hFc plasmid compared to control group. Conclusion: Our results demonstrate that B7-H4 synergistically potentiates oral tolerance induced by allogeneic cells by increasing the frequency of FoxP3+ CD4+CD25+ Treg and reducing Th1 and Th2 cytokine production.

The Effect of 1,25-Dihydroxyvitamin D3 on the Viability of Periodontal Ligament Cells and the Experimental Tooth Movement in Rats (1,25-Dihydroxyvitamin D3가 치주인대세포활성 및 실험적 치아이동에 미치는 영향에 관한 연구)

  • Vitamin D is known to exert its action by activating DNA and RBA within target cells to produce proteins and enzymes that can be used in bone resorption process. Particularly, the active form of vitmain D, 1,25-dihydroxycholecalciferol $[1,25-(OH)_2D_3]$, is considered to be one of the most potent stimulators of osteoclatic acitivity in vitro. The purpose of this study was to evaluate the effect of 1,25-Dihydroxyvitamin $D_3$ on the avtivity of periodotal ligament cells and, the experimental tooth movement. Human periodontal ligament cells were collected from the first premolar tooth extracted for the orthodontic treatment, and were incubated in the environment of $37^{\circ}C$, 5% $CO_2$ and 95% humidity. Microtitration(MIT) assay was done at 10, 25, 50 and 100ng/ml of 1,25-Dihydroxyvitamin $D_3$. 21 Sprague-Daft rats were divided into a control gmup(3), and experimental groups(18) where 100g of force from helical spring was applied across the maxillary incisors 1,25-Dihydroxyvitamin $D_3$ was injected into periodontal ligament at the mesial or distal surface of maxillary incisors so that we can compare the control side and the experimental side. Expreimental groups were sac rifled at 12, 24, 36, 48, 72hours and 7 days after force application, respectively. And the obtained tissues were evaluated histologically. The observed results were as follows. 1. The activity of periodontal ligament cells in l0ng/ml or 25ng/ml of 1,25-Dihydroxyvitamin $D_3$ 1,25-Dihydroxyvitamin $D_3$ was not significantly different to the control at the cultivation of 1, 2 and 3 days. 2. The activity of periodontal ligament cells was significantly increased at 3 days in 50 ng/ml of 1,25-Dihydroxyvitamin $D_3$ and 2, 3 days in 100g/ml of 1,25-Dihydroxyvitamin $D_3$. 3. Up to 7 days after force application, there was no difference in osteoblastic activity, tearing of periodontal ligament and proliferation of capillary at tension side between 1,25-Dihydroxyvitamin $D_3$ injection side and the control side. 4. The osteoclastic activity and the resorption of alveolar bone was greater in 1,25-Dihydroxyvitamin $D_3$ injection side than the control side at 36 hours after force application.

Synergistic Inhibition of Aronia melanocarpa and Moringa oleifera Seed Extract on Experimental Atopic Dermatitis (아로니아 및 모링가 종자 복합물의 항아토피 상승효과)

  • Atopic dermatitis is a chronic, relapsing inflammatory skin disease. This study aimed to investigate the therapeutic benefits of Aronia melanocarpa (AM) and Moringa oleifera seed extract (MO) on experimental atopic dermatitis. We examined the effects of AM or MO and their combination on 2,4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis in BALB/c mice as well as tumor necrosis factor $(TNF)-{\alpha}$ and interferon $(IFN)-{\gamma}-stimulated$ HaCaT keratinocytes. Mice were orally treated with extract during repeated application of DNCB to shaved dorsal skin. Our results show that treatment with AM and MO in combination reduced histological manifestations such as epidermal hyperplasia and inflammatory cell infiltration. Furthermore, it significantly decreased skin thickness and serum immunoglobulin E (IgE) level compared to the AM or MO alone treated group. Combined extract of AM and MO suppressed expression of $TNF-{\alpha}/IFN-{\gamma}-induced$ T helper 2 (Th2) chemokines such as thymus and activation-regulated chemokine and macrophage-derived chemokine. To sum up, combination of AM and MO suppressed the inflammatory response and serum IgE as an indicator of several allergic diseases in DNCB-induced experimental atopic dermatitis and Th2 chemokine expression in HaCaT cells. This result suggests that combination of AM and MO could be a valuable strategy to improve atopic dermatitis.

Soil Microbial Flora and Chemical Properties as Influenced by the Application of Pig Manure Compost (돈분퇴비의 시용이 배추재배지 토양의 미생물상 및 화학성에 미치는 영향)

  • Studies were conducted during 2 months from May of 1997 to evaluate the effects of pig manure compost(PMC) on soil microbial flora. To do so, a field experiment of Chinese cabbage(Brassica campestris L.) was conducted in a randomized block design on a sandy loam soil and microbial floral characteristics in soils were analyzed. Treatments to control included the application of PMC at (A) $8Mg\;ha^{-1}$CM-8), (B) $29Mg\;ha^{-1}$(CM-2,9), and (C) $57Mg\;ha^{-1}$(CM-57), and of chemical fertilizer(D) at $320N-80P_2O_5-200K_2O\;kg\;ha^{-1}$(NPK). In each treatment, the rhizosphere and non-rhizosphere soils were tested for the analysis of microbial populations. The populations of bacteria, actinomycetes, and fungi increased in soils with the applications of PMC and chemical fertilizer, but that of Bacillus sp. decreased. However, the population of fluorescent Pseudomonas sp. was reduced in NPK plots only. With increasing application rates of PMC, the number of colony forming units(cfu) of bacteria (Pseudomonas sp. and actinomycetes) and fungi increased. in all PMC-treated plots, the population density peaked at early growth stage for bacteria(including Bacillus sp.), at late growth for fluorscent Pseudomonas sp., and at harvest for fungi and actinomycetes. The rhizosphere effect was greatest for fluorscent Pseudomonas sp. As the application rates of PMC increased, Total N, organic matter, available phosphate, and exchangeable -K, -Ca, and -Mg increased compared to control, but soil pH was lowered. In NPK plots, EC was 3.4-fold and exchangeable K was 5-fold higher than control.

Elimination of Lily Symptomless Virus by In Vitro Scaling and Reinfection Rates under Various Culture Conditions in Korean Native Lilies (한국 자생나리의 기내 인편삽에 의한 Lily Symptomless Virus 제거 및 구근 재배조건에 따른 재감염 분석)

  • The lily symptomless virus (LSV) is the most common virus in Korean native lilies and causes various types of damage to overall plant growth. This study was carried out to investigate the elimination rate of the LSV by the in vitro scale culture (scaling) method in Korean native lilies and to test reinfection rates of the LSV under several field culture conditions of bulb production. Four Korean native lilies (Lilium dauricum, L. distichum, L. lancifolium, and L. maximowitzii) were used and their scales were cultured in vitro for micro-scale formation. The micro-scales were subcultured repeatedly using MS culture medium supplemented with 30 or $90g{\cdot}L^{-1}$ sucrose. The culture conditions were $24{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD with 16 hour daylength using fluorescent lamps and maintained at $22{\pm}2^{\circ}C$. The virus-free bulblets were grown for one to three years in the greenhouse and transplanted to the field in October or March. Virus infection rates were investigated by direct tissue blotting immunobinding assays and measurement of chlorophyll and protein contents. Virus-free plants could be obtained from the 5th subculture of micro-scales in L. lancifolium and L. maximowitzii or from primary culture in L. dauricum and L. distichum. LSV-free plants were reinfected during bulb production in the field. Reinfection rates were higher at older bulb ages and under higher planting density. The plants planted in October and at inland Gyeongsan had higher infection rates than those planted in March and at coastal area Pohang. The reinfection rate of L. maximowitzii was higher than those of L. dauricum and L. lancifolium. The LSV-infected plants had lower chlorophyll contents and unchanged protein contents compared to virus-free plants.

Productivity of Mixed Pasture as Affected by White Clover Varieties and Plant Height in Cutting Dates (화이트 클로바 품종 및 예취시 초장이 혼파 초지의 생산성에 미치는 영향)

  • To find out the way to be able to prolong the grazing years of mixed pasture under Cheju summer-hot weather, an experiment designed with split plot (main plot : 3 cutting-plant height such as 15cm, 30cm and 45cm and sub plot : 4 white clover varieties such as Regal, Tahora, Brown and SRVR) was carried out at Cheju Agricultural Experiment Station during the period from Sept. 1994 to Oct. 1996 Dry matter yield in 15cm plant height in cutting dates has shown significantly lower increase than those in 30 and 45cm(P<0.05). There was highiy significant increase in dry matter yield of Brown plot when compared with other varieties of white clover in 2nd year(P<0.01). There was no difference on botanical composition of mixed sward as affected by plant height in cuttings dates in 2nd year. The percentage of white clover tends to lower in white clover varieties Brown and SRVR when compared with Regal and Tahora. The rate of weed was not affected by plant height in cutting dates and white clover variety in mixed pasture at last cutting time of 2nd year. In 15cm plant height, concentration of ADE and NDF was the lowest one while P contents was the highest one(P<0.01). Potassium contents was the lowest one in 30cm plant cutting height(P<0.05), and nitrogen showed statistically interaction between plant cutting height and white clover variety(P<0.01). White clover variety was no effective on concentration of constituents in mixed pastures. In conclusion, white clover variety SRVR harvested at 15cm plant height is effective on nutrient value of pasture and balance of grass and white clover in botanical composition.

Seasonal Diversity, Production and Nutrients Concentration of Mushrooms in an Oak Forest (상수리나무림의 계절별 버섯 생산량 및 영양염류 함량)

  • Seasonal production and nutrient concentrations of mushrooms in an Quercus acutissima forest were studied from 1999 to 2000. Thirty 2×2m quadrats were established randomly in the study area. 114 species of mushrooms were identified during the study period. Although mushrooms occurred from May to November in each year, the highest biomass production occurred in August. Seasonal dominant species was Lactarius violascens in June, R. bella and L. violascens in July, Marasmius siccus in August, Cortinarius tenuipes in September, Lepista nuda and C. tenuipes in October, respectively. Annual production of mushrooms in 1999 and 2000 were 84.8 kgDW·ha/sup -1/·yr/sup -1/ and 86.7 kgDW·ha/sup -1/·yr/sup -1/, respectively. Seasonal production was27.1kg/ha for July, 35.9kg/ha for August and 17.1 kg/ha for September, 3.7kg/ha for October, respectively. Seasonal dominant species in biomass was Russula alboareolata in June, R. bella in July, R. pseudodelica in August and September, Lepista nuda in October, respectively. R. pseudodelica showed the highest biomass in the study area, which is 17.5% of the total biomass. Average concentration of nutrients in mushrooms was 45.2mg/g for N, 1.5 mg/g for P, 29.6 mg/g for K, 0.2 mg/g for Ca and 0.8 mg/g for Mg, respectively, which were much higher than those in the leaf litter. Nutrients absorbed by mushrooms in 2000 was 4,163.1 g·ha/sup -1/·yr/sup -1/ for N, 130.1 g·ha/sup -1/·yr/sup -1/ for P, 2,568.2 g·ha/sup -1/·yr/sup -1/ for K, 12.4 g·ha/sup -1/·yr/sup -1/ for Ca, 68.5 g·ha/sup -1/·yr/sup -1/ for Mg, respectively.

Effect of Collection Time in Oocyte Recovery from Superovulated Korea Native Goats (재래산양의 과배란처리에 있어서 회수시간이 난자의 회수율에 미치는 영향)

  • The purpose of the present study was to examine whether collection time affects results of oocyte recovery from superovulated goats. Fiftyty-one mature Korean native goats, maintained in a pen under natural day length and fed hay ad libitum, were pretreated with progestagen impregnated CIDR for 10 days and then the goats were divided into two groups. One group of the goats received a single intramuscular injection of 1,000 IU PMSG on Day 8 of CIDR insertion. The other group of the goats received twice daily intramuscular injections of a total of 70 mg FSH for 3 days from Day 8 of CIDR. All the gonadotropin treated goats were injected with 10 mg $PGF_{2\alpha}$ on Day 8 and 400 IU hCG in the afternoon on Day 10. For oocyte recovery, donor goats were fasted 24 h before operation. Anesthesia was induced by intravenous injection of 2% xylazine(0.2 mg/kg body weight) and ketamin(11 mg/kg body weight). In vivo oocytes were recovered by follicle aspiration or oviduct flushing at 29 to 34, 35 to 40 and 41 to 50 h after hCG injection through mid-ventral incision. There was no significant difference in the mean number of CL and oocytes recovered. Oocyte collection at 29 to 40 h after hCG increased(P<0.05) the recovery rate of ovulated oocytes in oviducts compared to 41 to 50 h. The same results were also observed in the recovery of follicular oocytes. Oocyte grade was not affected by collection time. When oocytes were collected from follicular oocytes at 41 to 50 h after hCG, the recovery rate of Grade II oocytes was the lowest(P<0.05). From these results, it is suggested that oocyte recovery at 35 to 40 h after hCG will be successful for further use.

Identification of Genes Connected with the Sensitivity to 5-FU and Cisplatin in Squamous Cell Carcinoma Cell Lines (편평세포암 세포주에서 5-FU와 Cisplatin에의 감수성과 관련된 유전자의 동정)

  • Squamous cell carcinoma (SCC) in head and neck show a variability in the response to chemotherapy, even when it present with similar histological tumor type, grade, and clinical stage. The purpose of present study it to identify predictive bio-marker for the sensitivity or resistance to conventional chemotherapeutic agents, 5-fluorouracil (5-FU) and Cisplatin Oral cancer cell lines were used in present study. MTT assay was performed to evaluate the sensitivity and/or resistance to 5-FU and Cisplatin. And RT-PCR was carried out for evaluation of the mRNA expressions of various genes associated with mutation, inflammation (COX pathway), cell cycle, senescence and extracellular matrix (ECM). The molecules which are correlated with the sensitivity to 5-FU are XPA, XPC, OGG, APEX, COX-2, PPAR, Cyclin E, Cyclin B1, CDC2, hTERT, hTR, TIMP-3, TIMP-4 and HSP47. And the molecules are correlated with the sensitivity to Cisplatin are COX-1, iNOS, eNOS, PCNA, collagen 1 and MMP-9. Taken together, when choosing the appropriate chemotherpeutic agents for patients, considering the molecules which are correlated or reversely correlated is helpful to choose the resonable agents for cancer patients.
