Effect of Maternal Factors on the Concentrations of Minerals and Immunological Substance in Breast Milk (모유의 무기질과 면역물질함량에 영향을 미치는 요인에 관한 연구)

  • The purpose of this study was to evaluate the effect of maternal nutritional status and health behaviors on the concentrations of minerals (Zn, Fe, Ca) and the immunological substances (lactoferrin, sIgA, Iysozyme) in breast milk. Breast milk was collected from 193 healthy Korean women from obstetric clinics and postpartum care centers in Seoul. : 99 colostrum (1 - 5 days postpartum), 33 transitional milk (6 - 10 days postpartum), 61 mature milk (11 - 50 days postpartum). The concentrations of minerals and immunological substance were highest in colostrum and decreased with lactational period. Concentrations of Zn and Fe reduced significantly from colostrum to mature milk, however, Ca concentration stayed constant throughout the lactational period. Contents of lactoferrin, sIgA, and lysozyme were significantly lower in mature milk than in colostrum. Mother's nutritional status, assessed by prepregnancy BMI, had an effect only on colostrum, but not on transition and mature milk. Fe concentration of colostrum was significantly lower in underweight (prepregnancy BMI < 18.5) than in overweight mothers (prepregnancy BMI $\geq$ 23.0). Also lower tendency was observed for sIgA and lysozyme contents, even though the difference was not statistically significant. Pregnancy weight gain had no effect on the breast milk component. Since nutritional factors had some effect on colostrum, the health behaviors of mothers providing colostrum were assessed. The mother's behavior of smoking, drinking, morning sickness, parity, disease, nutrient supplement use had no significant effect on the breast milk component, however, Zn, sIgA, and lysozyme were the somewhat affected components by maternal health behavior.

Indirect ELISA Method for Measurement of Lactoperoxidase using IgY Antibody (IgY 항체를 이용하여 Lactoperoxidase 정량을 측정하기 위한 Indirect ELISA 방법의 개발)

  • To determine the concentration of Lactoperoxidase (LPO), an indirect enzyme-linked immunosorbant assay(ELISA) was developed. Anti-LPO egg yolk immunoglobulin(IgY) was transferred to egg yolk by immunizing of Brown hens with LPO. The titer of purified anti-LPO IgY was 1: 520,000. The immunological response of anti- LPO IgY with ${\alpha}$-lactalbumin, ${\beta}$-lactoglobulin, casein and lysozyme were evaluated, resulting that the anti-LPO IgY found to be a specific antibody toward LPO and no cross-reaction was observed against ${\alpha}$-lactalbumin, ${\beta}$-lactoglobulin, casein, and lysozyme in double immunodiffusion test and ELISA test. In indirect ELISA method, coating concentration of LPO and dilution rate of anti-LPO IgY was 0.25$\mu\textrm{g}$/mL and 1:8,000 respectively. Sensitivity in the standard curve of LPO was ranged from 0.01 to 1$\mu\textrm{g}$/mL using anti-LPO IgY.

Physiological Characteristics of Fusants by Interspecific Protoplast Fusion of the Genus Cellulomonas (Cellulomonas 속 종간 원형질 융합체의 특성)

  • In order to investigate physiological characteristics of fusants by interspecific protoplast fusion of the genus Cellulomonas, protoplasts of Cellulomonas flavigena NCIB 12901 and Cellulomonas bibula NCIB 8142 were fused and cell wall regenerated. To give gene maker, C. bibula was treated with 500 ug/ml NTG for 1 hr and arginine requiring auxotrophic mutants were isolated. Protoplasts of the genus Cellulomonas were obtained by treatment with $600{\mu}{\textrm{g}}$/ml lysozyme, and 0.5M sorbitol was optimal for osmotic stabilizer on protoplast fromation. Protoplast fusion was enhanced by 40% PEG)M.W.6,000) containing 25 mM $CaCl_{2}$ at $30^{\circ}C$ for 30 min and fusion frequency between C. bibula and C. flavigena was $5\times 10^{-4}$. Processes of protoplast formation, cell wall regeneration and protoplast fusion were obsdrved by scanning electron microscope. By comparing enzyme activities of cellulase, exocellobiohydrolase, .betha.-glucosidase of the parent strains of Cellulomonas with those of thier mutants and fusants, fusants with increased enzyme activity were obtained. By the studies on nutritional requirement, antibiotic resistance, cellulolytic enzyme activities, type of peptidoglycan and motility of two mutants and fusants, fusants were proved to be recombinant of both mutant strains.

Improvement of the Strains Degrading Recalcitrant Aromatic Compounds by Cell Fusion Between Arthrobacter spp. and Pseudomonas putida (Arthrobacter spp. 와 Pseudomonas putida 의 세포융합에 의한 난분해성 방향족 화합물 분해세균의 균주개량)

  • To develope the new strains of microorganisms having the degradative ability for various aromatic hydrocarbons. spheroplast cell fusions were performed with Arthrobacter spp. degrading phthalate ester and Pseudomonas putida degrading alkylbenzen sulfonate(ABS) and the characteristics of the fusants were investigated. The spheroplasts of P. putia KUD15 and Arthrobacter sp. were formed effectively by lysozyme-EDTA treatment and by Ampicillin-lysozyme-EDTA treatment. respectively. The Spheroplast formation frequency and the regeneration frequency of the strains were 98-99% and 5-8%, respectively. For cell fusion. 40% PEG6000 was used as a fusogenic agent and the formation frequencies of fusion product were $1.8{\times}10^{4}-$2.9{\times}10^{4}$ Most of the fusants, which were selected in complemented antibiotics media showed the degradative ability in minimal selective medium added phthalate ester or ABS as sole carbon source. ABS degradation by fusant strain was increased about 20% with compared with the parental strain, while the degradative ability of phthalate ester was simillilar to that of parental strain.

Effect of Dietary Protein Level on Immune Substances in Milk and its Transfer to Pups in Rats (흰쥐에서 식이 단백질 수준이 수유를 통한 수동면역에 미치는 영향)

  • This study was performed to determine the effect of maternal protein intake on 1) the concentration of immune substances in milk 2) degree of passive immunity to pups via lactation, and 3) specific antibody production to a specific antigen, $\beta$-lactoglobulin(BLG). 4) the effect of passive immunity that pups received from mother during lactation on the production of antibodies when the pups were challenged to the same antigen. Part of the female rats were immunized with BLG before and during pregnancy. The pregnant rats were placed into either 25% or 10% isolated soy protein diet throughout gestation and lactation. After weaning, pups from each group continued to be fed the same diet. At 18 weeks of age, all the pups were challenged with BLG. Total IgA and IgG, lysozyme, BLG-specific IgA and IgG were measured in dam's serum, dam's milk, and pup's serum. Total IgG, and lysozyme in dam's serum and milk were higher in high protein group. Total IgA and IgG in pup's serum remained higher in high protein group from 5 to 18 weeks of age. BLG-specific antibodies were found in the milk and serum of immunized dams, and in serum of pups born to immunized dams but not in the non-immunized group. BLG-specific IgA and IgG were again higher in high protein group and declined with time. The concentration decreased faster in the low proetein group than in the high protein groups. After immunization the pups with LBG, serum BLG-specific antibodies were not differ between rats born to immunized dams and those born to non-immunized dams. Therefore passive immunity rats received via milk as a pup had no effect on the BLG-specific antibody production later in life. This study shows the importance of protein status of mother and strongly support to the endorsement of breast feeding.
