Lysozyme has been reported to be present in the secretory granules of the Paneth cell, and lysozyme immunoreactivity has been detected by immunogold method in Paneth cells of the intestine of human, mouse and rat. The present study was aimed at clarifying the intracellular distribution and changes of the lysozyme immunoreactivity in rabbit Paneth cell after common bile duct ligation of rabbit, using the electron microscope immunogold technique. Healthy adult rabbits weighing about 2kg body weight were divided into normal and bile duct ligated groups. Common bile duct ligation was performed aseptically under ether anesthesia. Experimental animals were sacrificed on the 1st, the 3rd, the 5th, the 7th and the 14th day after the operation. Mucosal specimens from the intestinal gland of ileum were fixed in 2.5% glutaraldehyde-1.5% paraformaldehyde, followed by 1% osmium tetroxide, embedded in araldite mixture, cut with LKB-V ultratome. Ultrathin sections were placed on parlodion coated nickel grids (200mesh). The section-bearing grids were floated upside down on the added substance in a moist chamber at room temperature except for the primary antibody step, which was at $4^{\circ}C$. Sections were etched with a saturated solution of sodium m-periodate for 60min. After etching, sections were pretreated with 0.02M tris buffered saline (TBS), pH 8.4, with 1% bovine serum albumin (BSA, Sigma) for 60min, then treated polyclonal rabbit anti-human lysozyme (Dakipatts) diluted 1 : 50 in TBS with 0.1% BSA for 20hr. Subsequently, grids were incubated 60min in biotinylated goat anti rabbit IgG (Amersham) diluted 1 : 100 in TBS with 0.1% BSA. After this, sections were incubated 60min on streptavidin gold G10 (Amersham) diluted 1 : 50 in TBS with 0.1% BSA. After each step, the grids were briefly rinsed with TBS with 0.1% BSA. After the strepavidin gold step, the sections were jet washed with distilled water. Counterstain of the sections performed by uranyl acetate and lead citrate, and observed with JEM 100 CX II electron microscope. Observed results were as follow; 1. Secretory granules of mouse Paneth cells have a lysozyme immunoreactivity and also eosinophil leucocyte of rabbit applied for the positive-control stain, are well labeld with gold particles. 2. Normal rabbit Paneth cells have a lysozyme immunoreactivity restricted on the secretory granules. 3. Amount lysosomes containing myelin figures in the Paneth cells were significantly increased from 5th day after the common bile duct ligation. 4. Immunoreactivity of Paneth cell secretory granules were more activated on the 3rd day after the common bile duct ligation as compared with those of the normal animal. But the lysozyme immunoreactivity were decreased from the 5th day after the common bile duct ligation. 5. Considering the above finding, lysozyme contained Paneth cell are affected following of common bile duct ligation, whereas lysosomes containing myelin-figure do not exhibit any immunoreactive relationship with those of secretory granules.
톡소플라즈마 과면역(過免疫) 우혈청(牛血淸)에서 유래된 면역증강제인 obioactin으로 처리한 마우스 복강 macrophages내(內)에서의 톡소플라즈마 증식억제 활성을 검토하였다. obioactin 및 lonomycin A로 처리한 macropohages에서는 첨가농도의 증가에 따라 세포내의 톡소플라즈마 증식이 현저하게 억제되었다. 그러나 macrophages 활성물질인 muramyl dipeptide(MDP)는 톡소플라즈마의 증식억제 효과가 없었다. 이와같이 obioactin 및 lonomycin A의 첨가에 의해 macrophages내(內)에서 톡소플라즈마의 증식이 억제되는 기전의 일부를 해명하기 위한 일환으로 활성산소 중간체 및 lysozyme 분비량을 검토하였다. obioactin과 MDP로 처리한 macrophages에서는 활성산소 중간체인 superoxide anion($O_2{^-}$)과 hydropen peroxide($H_2O_2$)의 생산은 첨가농도에 의존해서 증가하였으나 lonomycin A 첨가군에서는 대조군과 차이가 없었다. 한편 세포내에서 분비되는 lysozyme의 양은 obioactin, lonomycin A 및 MDP를 첨가한 각각의 macrophages에서 첨가농도의 증가에 따라 무처지 대조군에 비해 감소되었다. 이러한 결과로 부터 obioactin은 macrophages를 활성화시켜 세포내에서 활성산소 중간체($O_2{^-}$ 및 $H_2O_2$)를 발생시켜 이것들에 의해 톡소플라마즈의 증식이 억제되는 것으로 사료되었다. 그러나 macrophages내에서 분비되는 lysozyme은 톡소플라즈마의 증식억제와는 무관하였다.
This study examined the effects of the dietary inclusion of various concentrations of red ginseng byproduct (RB) and a mixture containing red ginseng byproduct, garlic extract, yeast and filler (CR) on the growth, body composition, serum chemistry, and lysozyme activity of juvenile olive flounder (Paralichthys olivaceus). Juvenile fish (n= 630) weighing 5.0 g were randomly distributed into 21 180 L flow-through tanks (30 fish/tank). Seven experimental diets were prepared in triplicate: a control diet without additive, and diets containing 0.5, 1 and 2% concentrations of RB (RB-0.5, RB-1, RB-2) and CR (CR-0.5, CR-1, CR-2) at the expense of wheat flour. After an 8-week feeding trial, serum chemistry and lysozyme activity of fish were measured. Mean weight gain was significantly higher in fish fed the control diet than in fish fed the RB and CR diets. The dietary inclusion of RB and CR reduced feed utilization. Mean serum glucose and triglyceride (TG) levels were higher in fish fed the control diet than in fish fed the other diets. Mean glutamate pyruvate transaminase (GPT) levels of fish fed the control and RB-2 diets were higher than those of fish fed the RB-0.5, RB-1, CR-1, and CR-2 diets. Mean lysozyme activity levels of fish fed the RB-0.5 and RB-1 diets were higher than those of fish fed the control and CR diets. The results of this study indicate that red ginseng byproduct may be utilized as an immunostimulant rather than as a growth promoter for juvenile olive flounder. Dietary inclusion of 0.5% red ginseng byproduct effectively improved serum glucose, GPT, TG, and lysozyme activity of the fish in this study.
Purpose : The purpose of this study was to evaluate the clinical effect of Vitamin C, Vitamin E, lysozyme, carbazochrome complex medicine($IGATAN^{(R)}$) for periodontal disease. Material and Methods : The study was performed by double blinded, ramdomized method. Forty two subjects diagnosed as chronic incipient or moderate periodontitis at department of periodontology, Yonsei Dental Hospital were included in the study. This study was approved by Institutional Review Board, Yonsei University Hospital. All subjects received scaling at their first examination and second examination was scheduled after 2 weeks. At second examination, periodontal parameters such as plaque index(PI), gingival index(GI), probing depth(PD), bleeding on probing(BOP), gingival recession(GR) and clinical attachment level(CAL) were recorded(Baseline) with prescription of Vitamin C, Vitamin E, lysozyme, carbazochrome complex($IGATAN^{(R)}$) (Experimental group 23 subjects) or placebo medicine(Control group; 19 subjects). The subjects were recalled after 4 weeks for periodontal parameters measurement. Results : In the experimental groups, PI, GI, CAL and BOP scores were significantly reduced at 4 weeks compared to baseline. A statistically significant decrease in or and BOP scores were observed in the experimental group compared to the control group. Conclusion : It can be concluded that Vitamin C, Vitamin E, lysozyme, carbazochrome complex medicine($IGATAN^{(R)}$) have an effect in reducing gingival bleeding and improving periodontal inflammatory condition inchronic incipient- moderate periodontitis.
난백에 함유된 lysozyme은 용균작용을 하며 ovalbumin은 면역계에서 항원으로써 역할을 한다. 난백분석에는 일반적으로 전기영동, 겔 투과 크로마토그래피, reversed-phase HPLC(RP-HPLC)를 사용하는데 신속한 분석을 위해 RPHPLC가 사용된다. HPLC 컬럼으로 C18 컬럼(Agilent, USA)을 사용하였다. 최적의 분리조건을 찾기 위해 이동상의 조성과 온도를 변화하였고 용량인자, 분리도를 계산하여 그 값을 비교하였다. 등용매 용리에서 acetonitrile(ACN), distilled ater(DW), trifluoroacetic acid(TFA)의 이동상 부피비를 30/70/0.1~60/40/0.1로 ACN 부피비를 10%씩 증가시키며 실험하였고 이 때 온도는 $20^{\circ}C$이었다. 기울기 용리에서는 ACN과 증류수의 비율을 20분 동안 10/90~60/40으로 증가시켰고 20, 30, $40^{\circ}C$의 온도변화를 주었다. 등용매 용리에서 이동상 조성이 50/50/0.1일 때 세 개의 피크로 분리되었고 개의 피크 중 첫 번째 피크가 lysozyme, 세 번째 피크가 ovalbumin임을 확인하였다. 기울기 용리에서는 온도가 $30^{\circ}C$일 때 네 개의 피크가 나왔으며 네 개의 피크 중 첫 번째 피크가 lysozyme, 세 번째 피크가 ovalbumin임을 확인하였다.
Our purpose is to develop an efficient synthetic method of obtaining oligosaccharide unit in sufficient amounts to study functions of glycans. Many exoglycosidases have been used as tools for the oligosaccharide synthesis. In contrast, a limited number of reports are available on the utilization of endoglycosidases. We describe herewith the efficient synthetic method of useful oligosaccharides and derivatives as biomaterials utilizing lysozyme, cellulase, and lacto-N-biosidase-mediated transglycosyltions.
