Evaluation of Micronucleus Frequency in Cytokinesis-blocked Bovine Lymphocytes from Regions around Wolsong Nuclear Power Plant (세포질 분열 차단 림프구를 이용한 월성원자력발전소 주변 소의 미소핵 발생 평가)

  • Cytogenetic and hematological analysis was performed in bovine peripheral blood from the regions around Wolsong nuclear power plant and control area. The frequency of micronuclei (MN) in peripheral blood lymphocytes from cattle was used as a biomarker of radiobiological effects resulting from exposure to environmental radiation. An estimated dare of radiation was calculated by a best fitting linear-quadratic model based on the radiation-induced MN formation from the bovine lymphocytes exposed in vitro to radiation over the range from 0 Gy to 4 Gy. MN rates in lymphocytes of cattle from Wolsong nuclear power plant and control area were 9.87/1,000 and 9.60/1,000, respectively. There were no significant differences in MN frequencies and hematological values in cattle between Wolsong and control area. The study indicates that the MN assay is a rapid, sensitive and accurate method that can be used to monitor a large population exposed to radiation.

  • Objectives: The purpose of this study was to verify the effect of Palmiboshinwhan (PMBSW) in methotrexate (MTX)-induced immunosuppressed SD rats. Methods: The test articles were once a day dosed for 14 days by gastric gavage from 2 days after last MTX-dosing, and changes in body weight, spleen weight and total blood leukocyte numbers were observed with total lymphocyte numbers, B and T lymphocyte percentages, CD3+CD4+, CD3+/CD8+, CD4+/CD8+ T lymphocyte percentages in the blood and spleen, the serum interleukin (IL)-2 levels and the productivity of IL-2 of splenic cells. Result & Conclusion: It is concluded that PMBSW has relatively good immunostimulating effect in the MTX-induced immunosuppressed SD rats. Theefficient dosage was considered above 500mg/kg. In addition, it is considered that the immunostimulating effect of PMBSW was mediated to both the B and T lymphocytes. The more favorable effects were detected in T lymphocytes rather than B lymphocytes, and PMBSW showedrelatively good stimulating potential against CD4+ T lymphocytes but not any stimulating effect against CD8+ T lymphocytes in the present study.

  • We previously examined extracts, isolated from Scutellaria baicalensis (SB), chemical mediators, and IgE by mesenteric lymph node (MLN) lymphocytes in rats. The present study was to evaluate the effects of extracts of SB on the MLN lymphocytes function of mice given orally by 20 mg/kg for 2 weeks with dextran sulfate sodium (DS)-induced colitis. Results show that IgE levels in MLN lymphocytes was low, while IgA was high, in mice given SB compared to that fed water. Concentrations of $Inteferon-{\gamma}$ and interleukin (IL)-2 of T cells by concanavalin A treatment was significantly higher in the SB fed group than the normal group. Activation-induced IL-4 and IL-10 secretion was lower in SB fed mice compared control mice after DS-induced colitis. These results suggested that SB suppresses the inflammation in DS-induced colitis through the modulation of Th1/Th2 balance to down-regulate $Th_2$ response in MLN lymphocytes.

Stimulatory Effect of Saponin from Panax ginseng on Function of Lymnphocytes in the Elderly

  • We used the saponin Rgi extracted from Panax ginseng to study its effects on lymphocytes of 10 young and 19 elderly persons. The proliferate response of Iymphocytes cocultured for 72h with PHA and saponin was measured by using MTT method and the'H-TdR incorporation procedure. PHA and Rgl had stimulative effects on the phenotype of Iymphocytes (p<0.001). Rgl also increased the fluidity of lymphocyte membrane of the aged (p<0.001). The CD2s and CDfsRA positive cells of Iymphocytes in the elderly were lower than those of the young people,8.6clo $\pm$ 2.7olo vs 10.43% : 3.5%, 20.95% $\pm$ 15.5clQ vs 50.86% :4.3olo, respectively. More CDfsRO positive cell lymphocy populations were seen in the aged. The CEfsRO positive cells of the young people were 39.63% $\pm$ 3.2%. We discussed the cause of declined immune function of Lymphocytes of aged person and the mechanism of the effect of P. ginseng on Lymphocytes. Use of Frequencies of Micronuclei as Biological Dosimetry in Korean Native Cattle and Goat Lymphocytes after Irradiation in vitro (한우 및 한국재래산양 유래 말초혈액 림프구의 미소핵을 이용한 방사선 피폭의 생물학적 선량측정)

  • The frequencies of gamma-ray-induced micronuclei (MN) in cytokinesis-blocked (CB) lymphocytes at several doses were measured in three donors of Korean native cattle and Korean native goat. Measurements performed after irradiation showed a dose-related increases in MN frequency in each of the donors studied. When analysed by linear-quadratic model the line of best fit was cattle : y : 0.1016D +$0.0118D^2$+0.0147, goat : y = 0.1353D +$0.0043D^2$+0.0087 (y : number of MN/CB cells and D = irradiation dose in Gy). The relative sensitivity of goat lymphocytes compared with cattle lymphocytes was estimated by best fitting linear-quadratic model based on the radiation-induced MN data over the range from 0 Gy to 4 Gy. In the case of MN frequency with 0.05, 0.1, 0.2, 0.4 and 0.8, the relative sensitivities of goat lymphocytes were 1.106. 1.166. 1.140, 1.069 and 0.976 respectively. Our in vitro radiobiological study confirmed that the cytogenetic response obtained in blood from cattle and goat can be utilized for application in environmental studies.

Effect of Bambusae Caulis in Liquamen on T-lymphocytes and Peritoneal Macrophages (죽력(竹瀝)이 T-lymphocytes 및 복강(腹腔) Macrophage에 미치는 영향(影響))

  • The purpose of this research was to investigate effects of Bambusae Caulis in Liquamen(BCL) on T-lymphocytes and peritoneal macrophages in mice. The apoptosis and subpopulation of T-lymphocytes were tested using a flow cytometer. The phagocytic activity of mouse peritoneal macrophage was tested using a luminometer. Nitric oxide production was tested using a Griess reagents. BCL induced T-lymphocytes apoptosis. BCL increased $T_H$ cells population and decreased $T_C$ cells population of T-lymphocyte, but did not affect splenocytes subpopulation. BCL increased nitric oxide production and phagocytic activity of peritoneal macrophage in mice. These results suggest that BCL regulates the immune system in consequence of an increase in helper T cell population and macrophages activation.

Experimental Trichothecene(T-2) Toxicosis in Korean Native Goats (한국 재래산양에서의 실험적 Trichothecene(T-2) 독소중독증)

  • To investigate the effects of T-2 toxin on the blastogenesis of lymphocytes, pathology, hemogram and blood chemistry in the goat, the korean native goats were treated orally with T-2 toxin for 21 days with a dosage of 0.6mg per kg body weight. The results were as follows: 1. The total count of leukocytes and lymphocytes decreased significantly from 14 to 21 days after treatment. 2. Mryeloid: erythroid ratios increased significantly on days 12 after treatment. 3. Delayed-type hypersensitivity skin reactions to tuberculin were reduced predominantly. 4. T-2 toxin induced prolonged prothrombin time. 5. Mitogenic responses of lymphocytes to both lipopolysaccharide and phytohemagglutinin were significantly depressed on days 7 and 14 after treatment. 6. Treatment of T-2 toxin caused marked depletion of lymphocytes in the thymus, mesenteric lymph node, peyer's patchs and spleen.

Effect of Korean Mistletoe Lectin on Gene Expression Profile in Human T Lymphocytes: A Microarray Study

  • Korean mistletoe has a variety of biological effects, such as immunoadjuvant activities. This study investigates the effects of Korean mistletoe lectin (Viscum album L. var. coloratum agglutinin, VCA) on human T lymphocytes to determine whether VCA acts as an immunomodulator. Purified human T-lymphocytes were cultured with VCA and RNA from each point was analyzed using Affymetrix human genome chips containing 22,500 probe sets which represents more than 18,000 transcripts derived from 14,500 human genes. As a result, there was a striking upregulation of genes coding for chemokines. Seventeen genes out of 50 coding for proteins with chemokine activity were upregulated including CXCL9 and IL-8 which are related to the treatment of cancer. In addition, 28 cytokine genes were upregulated including IL-1, IL-6, IL-8, IFN-$\gamma$, and TNF-$\alpha$. Taken together, the data suggest that Korean mistletoe lectin, in parallel with European mistletoe, has an ability to modulate human T cell function.

  • Warthin's tumor is a benign salivary gland tumor, occurring primarily in the parotid gland. It's incidence is 4% of all salivary gland tumors. The cytologic finding of Warthin's tumor is characteristic and it consists of sheets of oncocytes and scattered lymphocytes in the background of granular material. We reports three cases of Warthin's tumor showing characteristic cytologic findings. The patients consisted of two males and a female and their ages were 60, 73, and 69 years, respectively. All the lesions were in the parotid gland. The cytologic findings showed sheets of oncocytic epithelial cells and scattered lymphocytes in the back-ground of granular materials and many red blood cells. In one case, the back-ground was composed of many Inflammatory cells including lymphocytes, neutrophils and macrophages.

Xylene Induces Oxidative Stress and Mitochondria Damage in Isolated Human Lymphocytes

  • Xylene is a cyclic hydrocarbon and an environmental pollutant. It is also used in medical technology, paints, dyes, polishes and in many industries as a solvent; therefore, an understanding of the interaction between xylene and human lymphocytes is of significant interest. Biochemical assessment was used to demonstrate that exposure of lymphocytes to xylene induces cytotoxicity (at 6 hr), generates intracellular reactive oxygen species, collapse of mitochondrial membrane potential, lysosomal injury, lipid peroxidation and depletion of glutathione (at 3 hr). The findings show that xylene triggers oxidative stress and organelle damage in lymphocytes. The results of our study suggest that the use of antioxidant, mitochondrial and lysosomal protective agents can be helpful for individuals subject to chronic exposure to xylene.
