시험관내에서 홍화의 물 추출물이 T 및 B 림프구의 활성에 미치는 영향 (Effect of Water Extract of Carthamus tinctorious L. on In Vitro Activity of T and B Lymphocytes)

  • Based on the traditional application of Carthamus tinctorious L. (CF) as a component of Korean medicinal decoctions, in the present study, we investigated in vitro an immunomodulatory activity of water extract of CF(WECF). Water extract of CF significantly increased the in vitro proliferative responses of spleen cells (SPC). However, addition of WECF during anti-CD3 activation resulted in a significant decrease in SPC proliferation. Flow cytometric analysis showed that WECF addition chanced T and B cell frequencies in anti-CD3-activated spleen cell populations. Using purified cells, it was revealed that WECF is mitogenic to B cells but rather inhibitory to T cell Proliferation. Upon anti-CD3 stimulation, high concentration (1 mg/ml) of WECF significantly inhibited T cell proliferation until day 2 of stimulation. At day 3, anti-CD3-activated cells exposed to WECF recovered their proliferation to the level comparable to control. Although B cell proliferation was also inhibited in proliferation at day 1, it recovered sooner and then was rather augmented by WECF at day 3. These data indicate that WECF down-regulates lymphocyte proliferation at early phase of activation but T cells are more vulnerable than B cells to WECF, However, CD4+ and CD8+ T cells did not differ in WECF-mediated immunotoxicity. Data of propidium iodide (PI) staining showed that WECF accelerates activated T cell, but not B cell, apoptosis and WECF concurrently inhibited cytokine production of activated T cells. Taken together, WECF exhibits B cell mitogenic activity and differential toxicity more pronounced to T cells, suggesting a possible in vivo application of WECF for specific control of T cells without alteration of B cell activity.

개의 아토피성 피부염에 대한 집먼지 진드기 항원 특이적 IgE와 림프구 증식 반응의 증가 (Increased Allergen-specific IgE Values and Lymphocyte Proliferation Response to House Dust Mites in Dogs with Atopic Dermatitis)

  • 혈중 항원 특이적 IgE 검사와 피내시험으로 집먼지 진드기 (house dust mites, HDM)에 양성 반응을 보인 20두의 아토피성 피부염으로 진단된 개를 대상으로, 말초혈단핵구 (peripheral blood mononuclear cells, PBMC)를 채취하여 HDM항원에 대한 반응을 검토하였다. PBMC를 분리하여 HDM항원으로 자극한 결과 20두 중 13두 (65%)에서 항원 특이적이 림프구의 증식반응을 확인할 수 있었다. HDM 항원에 증식반응은 아토피성 피부염군에서 대조군에 비해 유의적으로 높은 반응이 확인되었다 (P=0.007). 또한, HDM에 대한 반응은 혈중의 IgE 농도와 유의적으로 상관관계를 나타내었으며 (P=0.035), 이는 체내에서 항원 특이적인 IgE의 생산을 촉진하는 작용을 반영하는 지표가 될 수 있다고 생각되었다. 이러한 결과로, 말초혈액중에 존재하는 HDM 항원 특이적인 림프구는 개의 아토피성 피부염의 병태생리에 관여하고 있는 것으로 시사되었다.

십전대보탕가미방(十全大補湯加味方)의 면역조절효과(免疫調節效果)에 관한 실험적(實驗的) 연구(硏究) (Immunomodulatory effects of Kamishipjundaebotang(KSDBT))

  • This study was done to evaluate the immunomodulatory effects of KSDBT. KSDBT significantly enhanced the lymphocyte proliferation through 3H thymidine uptake and also increased T and T helper cell. It upregulated IL-2, IL-12(p35, p40) and INF-${\gamma}$. NO and carbon clearance were significantly increased by KSDBT. These results indicate KSDBT exert antitumor activity by immunomodulation of cytokines.

  • B 세포이 가장 중요한 기능은 항체를 생산하여 체액성 면역반응을 조절하는 일이다. B 세포가 항체를 생산해 내는 형질세포(plasma cell)로 분화하기 위해서는 다음의 몇 가지 단계를 거쳐야 한다. 즉 resting 상태에서 mitogen이나 항원의 자극에 의한 활성화(activation)가 첫번째 단계이고 두번째로는 증식(proliferation)이며 마지막 단계가 분화(differentiation)이다. B 세포가 일단 mitogen이나 항원에 의해 활성화가 되면 활성화 된 clone의 증식과 분화는 T 세포에서 생산되는 여러 종류의 림포카인(lymphokine)의 영향하에서 이루어진다. 따라서 B 세포 기능에 영향을 미치는 림포카인의 종류를 알아내고 최근 새롭게 발견되는 림포카인의 B 세포에 대한 영향을 연구함이 중요하리라 생각된다. 여기에서는 B 세포 기능에 영향을 미치는 여러 림포카인의 종류의 그 기능에 관해서 간략하게 기술하고자 한다.

항암제 Mitomycin C의 부작용에 대한 수종 복합생약의 영향 (Effect of Several Combined Preparation of Crude Drugs on the Adverse Effects of Anticancer Agent-Mitomycin C)

  • The studies were conducted to investigate the combined effects of several combined preparation of crude drugs and mitomycin C(MMC). The combined effects on the proliferation of HepG2, A549, KHOS-Np, A431 and HeLa cells were estimated by MTT colorimetric assays. Sa Kunja Tang(SKT), Boyang Hwanoh Tang(BHT) and Hyulbu Choogo Tang(HCT) inhibited the proliferation of A549 and HeLa cell. The inhibitory action of MMC was increased by the combined treatment of SKT and MMC, and Sa Mul Tang(SMT) and MMC, respectively. When the mice were treated by MMC, the number of leukocyte was decreased significantly at the 3rd day, but recovered at the 7th day. In the groups of MMC treated with SKT or HCT, the number of leukocyte was increased significantly that the group of MMC treated only at the 1st and 3rd day. The combined treatment of SKT, SMT, BHT, HCT and MMC retained the spleen weight of mice at the level of normal mice, but decreased the thymus weight of mice. The combined treatment of SKT, SMT, BHT, HCT and MMC increased the number of PFC significantly than the MMC treated group. The combined treatment of SKT, SMT, BHT, HCT and MMC increased the T cell proliferation significantly thant the MMC treated group.

귀전우 Petroleum Ether 추출물의 세포독성 (Cytotoxicity of Petroleum Ether Extract of Euonymus alatus)

  • The purpose of this research was to investigate the effects of petroleum ether extract of Euonymus alatus (EAP) on the proliferation of human tumor cells. EAP inhibited the proliferation of HeLa, Hep G2, KHOS/NP and A431 cells. The cytotoxicity of doxorubicin on human tumor cells and Balb/c 3T3 cells were increased by the combination of EAP. EAP did not affect the proliferation of Balb/c 3T3 cells, mouse spleen cells and human lymphocytes. These results suggest that EAP has the cytotoxicity on human tumor cells without cytotoxicity on Balb/c 3T3 cells, mouse spleen cells and human lymphocytes, and increase the cytotoxicity of doxorubicin.

Expression and Characterization of Purinergic Receptor, $P2Y_{10}$ in Hematopoietic Stem Cells

  • Hematopoietic stem cells (HSC) are multipotent cells that reside in the bone marrow and replenish all adult hematopoietic lineages throughoutthe lifetime. In this study, we analyzed the expression of receptors of $P2Y_{10}$, purinergic receptor families in murine hematopoietic stem cells, hematopoietic progenitor cells. In addition, the biological activity of $P2Y_{10}$ was investigated with B lymphocyte cell line, Ba/F3 in effect to cell growth and cell cycle. From the analysis of expression in hematopoieticstem cell. and progenitor with RT-PCR, $P2Y_{10}$ was strongly expressed in murine hematopoieticstem cells (c-kit+ Sca-l+ Lin-) and progenitor cell population, such as c-kit- Sca-l+ Lin-, c-kit+ Sca-l- Lin- and c-kit- Sca-l- Lin-. To investigate the biological effects by $P2Y_{10}$, retroviral vector from subcloned murine $P2Y_{10}$ cDNA was used fur gene introduction into Ba/F3 cells, and stable transfectant cells were obtained by flow cytometry sorting. In cell proliferation assay, the proliferation ability of $P2Y_{10}$ receptor gene­transfected cells was strongly inhibited, and the cell cycle was arrested at G1 phase. These result suggest that the $P2Y_{10}$ may be involved the biological activity in hematopoietic stem cells and immature B lymphocytes.

Immunostimulatory Effects of ${\beta}$-glucan Purified from Paenibacillus polymyxa JB115 on Mouse Splenocytes

  • We investigated the effects of ${\beta}$-glucan purified from Paenibacillus polymyxa JB115 on the viability and proliferation of splenocytes. Splenocytes play a critical role in host immunity. MTT assays and trypan blue exclusion tests revealed that ${\beta}$-glucan significantly promoted the viability and proliferation of splenocytes over a range of concentrations. However, there was no specific subset change. ${\beta}$-glucan protected splenocytes from cytokine withdrawal-induced spontaneous cell death. For further mechanistic studies, ELISA assay revealed that ${\beta}$-glucan enhanced the expression of anti-apoptotic molecules and interleukin 7 (IL-7), a cytokine critical for lymphocyte survival. We also investigated the IL-2 dependency of ${\beta}$-glucan-treated splenocytes to determine if treated cells could still undergo clonal expansion. In flow cytometric analysis, ${\beta}$-glucan induced increased levels of the activation marker CD25 on the surface of splenocytes and ${\beta}$-glucan-treated splenocytes showed higher proliferation rates in response to IL-2 treatment. This study demonstrates that ${\beta}$-glucan can enhance the survival of splenocytes and provides valuable information to broaden the use of ${\beta}$-glucan in research fields.

Effect of mucilage from yam on activation of lymphocytic immune cells

  • The immunostimulating activities of mucilage fraction from yam were investigated. The proliferation of BSA-primed lymph node cells was enhanced between 4.1- to 10.9-fold compare to control, when cultured with 1 to $25{\mu}g/mL$ of yam-mucilage fraction. It showed strong immunostimulating activity than ginseng extract and as remarkable as Bifidobacterium adolescentis M101-4 known as a positive immunostimulator. Mitogenicity to lymph node cells was fully induced by concanavalin A and lipopolysaccharide. The proliferation of splenocytes and Peyer's patch cells was enhanced between 5.0- to 14.1-fold and 2.4- to 6.4-fold, respectively, when cultured with 1 to $25{\mu}g/mL$ of yam-mucilage fraction. It enhanced the production of cytokines such as tumor necrosis $factor-{\alpha}$ and IL-6 in the culture of RAW 264.7 macrophage cells. In the culture of lipopolysaccharide-stimulated RAW 264.7 cells, production of cytokines was as similar as compared to controls. In unstimulated RAW 264.7 cells, both tumor necrosis $factor-{\alpha}$ and IL-6 production were enhanced between 15.6- to 60.1-fold and 2.3- to 9.1-fold, respectively. Mucilage fraction from yam is expected to be a safe immunopotentiator to maintain the host immunity and develop a physiologically functional food.
