SCYL1BP1 has Tumor-suppressive Functions in Human Lung Squamous Carcinoma Cells by Regulating Degradation of MDM2

  • SCY1-like 1-binding protein 1 (SCYL1BP1) is a newly identified transcriptional activator domain containing protein with many unknown biological functions. Recently emerging evidence has revealed that it is a novel regulator of the p53 pathway, which is very important for the development of human cancer. However, the effects of SCYL1BP1 on human lung squamous carcinoma cell biological behavior remain poorly understood. In this study, we present evidence that SCYL1BP1 can promote the degradation of MDM2 protein and further inhibit the G1/S transition of lung squamous carcinoma cell lines. Functional assays found that reintroduction of SCYL1BP1 into lung squamous carcinoma cell lines significantly inhibited cell proliferation, migration, invasion and tumor formation in nude mice, suggesting strong tumor suppressive function of SCYL1BP1 in lung squamous carcinoma. Taken together, our data suggest that the interaction of SCYL1BP1/MDM2 could accelerate MDM2 degradation, and may function as an important tumor suppressor in lung squamous carcinomas.

$I{\kappa}B{\alpha}$-SR 유전자이입이 Cisplatin, Paclitaxel에 대한 폐암세포주의 감수성에 미치는 영향 (The Effect of $I{\kappa}B{\alpha}$-SR Gene Transfer on the Sensitivity of Human Lung Cancer Cell Lines to Cisplatin and Paclitaxel)

  • 연구배경 : 종양 세포들이 항암제에 대하여 저항을 나타내는 기전인 'apoptosis에 대한 저항'에 NF-${\kappa}B$의 활성이 중요한 역할을 하리라 생각되고 있다. 즉, NF-${\kappa}B$가 종양세포의 apoptosis를 억제하는 작용을 일으킴으로써 종양 세포의 생존에 유리하게 작용하고 있음이 제시되 고 있다. 방 법 : 본 연구에서는 비소세포폐암에 대한 항암 치료의 내성에 대하여 외부 자극으로 분해되지 않는 $I{\kappa}B{\alpha}$-SR의 삽입으로 NF-${\kappa}B$의 활성을 억제시키고 이로 인해 항암제에 대한 폐암세포주의 감수성이 증가하는지 여부를 밝혀 보고자 하였다. 결 과 : 비소세포폐암 세포주로 NCI H157, NCI H460 세포주를 이용하였고, Ad-$I{\kappa}B{\alpha}$-SR를 transduction 한 후 cisplatin을 처치한 군에서 NF-${\kappa}B$의 핵 내로의 이동이 억제되었으며 대조군과 비교시 $IC_{50}$이 2-3배 정도 유의하게 낮아짐을 관찰하였다. 또한 paclitaxel의 경우에도 Ad-$I{\kappa}B{\alpha}$-SR로 감염된 폐암세포주는 대조군과 비교시 IC50이 2배 정도 유의하게 낮아짐을 관찰하였다. 이러한 항암제에 대한 감수성의 증가의 기전으로 cisplatin의 경우는 $I{\kappa}B{\alpha}$-SR의 이입이 NF-${\kappa}B$의 활성을 억제 함으로 인한 apoptosis의 증대 때문인 것으로보인다. 결 론 : 폐암세포주에서 Ad-$I{\kappa}B{\alpha}$-SR transduction은 Clsplatin, paclitaxel에 대한 폐암세포주의 감수성을 증가시킴으로 앞으로 폐암을 치료하는데 있어 새로운 치료 방법이 될 수 있을 것으로 보인다.

Distinct Pro-Apoptotic Properties of Zhejiang Saffron against Human Lung Cancer Via a Caspase-8-9-3 Cascade

  • Lung cancer is the leading cause of cancer-related death worldwide. Here we investigated the antitumor effect and mechanism of Zhejiang (Huzhou and Jiande) saffron against lung cancer cell lines, A549 and H446. Using high performance liquid chromatography (HPLC), the contents of crocin I and II were determined. In vitro, MTT assay and annexin-V FITC/PI staining showed cell proliferation activity and apoptosis to be changed in a dose- and time-dependent manner. The inhibition effect of Jiande saffron was the strongest. In vivo, when mice were orally administered saffron extracts at dose of 100mg/kg/d for 28 days, xenograft tumor size was reduced, and ELISA and Western blotting analysis of caspase-3, -8 and -9 exhibited stronger expression and activity than in the control. In summary, saffron from Zhejiang has significant antitumor effects in vitro and in vivo through caspase-8-caspase-9-caspase-3 mediated cell apoptosis. It thus appears to have more potential as a therapeutic agent.

유황오리 추출물의 각종 암세포에 대한 생육억제 효과 (Growth Inhibition of Extract from Sulfur fed Duck Carcass against Various Cancer Cell Lines)

  • 유황오리 추출물의 각종 암세포에 대한 생육억제효과를 MTT assay를 이용하여 검토한 결과 10 mg/$m\ell$의 농도에서 KB(구강상피암) 89.5$\pm$0.7%, SNU-1(위암) 69.8$\pm$1.7%, K-562(백혈병) 79.8$\pm$2.8%, Farrow(흑색종) 82.7$\pm$2.6%, WiDr 결장암) 76.3$\pm$2.5%, SK-MES-1(폐암) 59.2 $\pm$4.4%, HL6O(백혈병) 60.5 +3.5%, Calu-3(폐암) 53.2$\pm$1.6%, HEP-2(후두암) 80.7$\pm$0.5%, P388(마우스 백혈병) 79.9$\pm$3.7%, 3LL(마우스 폐암) 87.2$\pm$3.3%의 효과가 있다는 사실이 판명되었다. 그리고 유황오리 추출물의 HP-20 column chromatography에서는 100% methyl alcohol 용출물이 HEP-2에 대한 생육억제효과가 있었으며, 10 mg/$m\ell$에서 99.1$\pm$0.4%의 생육억제효과가 나타났다. 또한 일반오리 추출물과 유황오리 추출물의 각종 암세포에 대한 생육억제효과를 비교하였을 때 거의 모든 암세포에서 유황오리 추출물의 효과가 더 높았다.

Effect of 5-FU and MTX on the Expression of Drug-resistance Related Cancer Stem Cell Markers in Non-small Cell Lung Cancer Cells

  • Cancer stem cells (CSCs) are often characterized by the elevated expression of drug-resistance related stem-cell surface markers, such as CD133 and ABCG2. Recently, we reported that CSCs have a high level of expression of the IL-6 receptor (IL-6R). The purpose of this study was to investigate the effect of anticancer drugs on the expression of the drug resistance-related cancer stem cell markers, ABCG2, IL-6R, and CD133 in non-small cell lung cancer (NSCLC) cell lines. A549, H460, and H23 NSCLC cell lines were treated with the anticancer drugs 5-fluorouracil (5-FU; $25{\mu}g/ml$) and methotrexate (MTX; $50{\mu}g/ml$), and the expression of putative CSC markers was analyzed by fluorescent activated cell sorter (FACS) and the gene expression level of abcg2, il-6r and cd133 by reverse transcriptase-polymerase chain reaction (RT-PCR). We found that the fraction of ABCG2-positive(+) cells was significantly increased by treatment with both 5-FU and MTX in NSCLC cells, and the elevation of abcg2, il-6r and cd133 expressions in response to these drugs was also confirmed using RT-PCR. Also, the number of IL-6R(+) cells was increased by MTX in the 3 cell lines mentioned and increased by 5-FU in the H460 cell line. The number of CD133(+) cells was also significantly increased by both 5-FU and MTX treatment in all of the cell lines tested. These results indicate that 5-FU and MTX considerably enhance the expression of drug-resistance related CSC markers in NSCLC cell lines. Thus, we suggest that antimetabolite cancer drugs, such as 5-FU and MTX, can lead to the propagation of CSCs through altering the expression of CSC markers.

비소세포 폐암세포에서 Uteroglobin의 이입에 의한 cPLA2와 COX-2 발현 및 ERK 활성의 변화 (The Effect of Uteroglobin on cPLA2, COX-2 Expression and ERK Activation in NSCLC Cells)

  • 연구배경 : Uteroglobin은 정상 폐상피세포에서 발현되는 단백질로 비소세포암 조직이나 세포주에서는 발현이 저하되어있다. 항염증작용을 하며 암세포에 과발현 시키면 암의 형질이 소실됨이 밝혀지고 있다. 역시 염증작용과 관련이 있는 cPLA2와 COX-2도 암과의 관련성이 밝혀지고 있고, 암억제나 MMP의 억제 등의 공통점을 가지고 있으나 이들의 관련성에 대해서는 밝혀진 바가 없다. 또한, COX-2의 암과의 관련성을 설명하는 기전으로 ERK 활성화의 관련 가능성이 있으나, uteroglobin과 ERK의 관련성도 아직 밝혀지지 않고 있다. 비소세포폐암 세포주에 uteroglobin을 과발현시킨 후, cPLA2와 COX-2의 발현, 그리고 MMP-2, MMP-9, ERK의 활성화가 어떻게 변화하는지에 대해 실험하였다. 방 법 : 폐선암세포주인 A549와 NCI-H460 세포주에 adenovirus-uteroglobin, adenovirus-null을 각각 20,100,200 moi로 transduction 시킨 뒤, 48시간 배양한 후에 단백질을 추출하였다. Uteroglobin의 발현을 확인한 후, cPLA2, COX-2, pERK, total ERK에 대해 Western blot을 시행하였고, 배양액으로 zymography를 시행하였다. 결 과 : A549 세포주와 NCI-H460 세포주에서 uteroglobin의 발현을 확인한 세포주에서 cPLA2와 COX-2의 발현이 감소함을 Western blot으로 확인하였고, pERK가 증가함을 Western blot으로 보았고, ERK의 활성화가 증가함을 확인하였다. MMP-9은 활성이 저하되었고, MMP-2는 변화가 없었다. MEK inhibitor인 U0126을 이용하여 ERK의 활성화를 저해시킨 후, uteroglobin의 발현에는 영향이 없었고, MMP-9의 활성저하효과가 소실되었다. 결 론 : 폐암세포주에서 uteroglobin의 항암작용기전에 cPLA2 와 COX-2의 발현의 감소와 ERK의 활성화가 기여할 것으로 사료된다.

Cannabidiol Induces Cytotoxicity and Cell Death via Apoptotic Pathway in Cancer Cell Lines

  • In view of obtaining potential anticancer compounds, we studied the inhibitory activity and the cytotoxic effects of a candidate compound in cancer cells. The cytotoxic effects of cannabidiol (CBD) in vitro were evaluated in NIH3T3 fibroblasts, B16 melanoma cells, A549 lung cancer cells, MDA-MB-231 breast cancer cells, Lenca kidney cells and SNU-C4 colon cancer cells. The cells were cultured in various concentrations of CBD for 48 h and 25 ${\mu}$M of CBD for 6-36 h. The cells were observed to exhibit inhibitory effects of the cell viability in their growth, and then cytotoxicity was estimated. The inhibitory activity of CBD was increased in all cancer cells and showed especially strong increment in breast cancer cells. The cytotoxicity of CBD increased in a dose- and time-dependent manner with growth inhibition in all cancer cell lines. Also, to assess the membrane toxicity induced by CBD, we investigated lactate dehydrogenase (LDH) release. After treatment with various concentrations of CBD, LDH release rate of cancer cells was accelerated. On the other hand, in the induction of cell death, caspase-3, -8 and -9 activations were detected in cancer cells after treatment with various concentrations of CBD, and CBD effectively induced activity of caspase-3, -8 and -9 in A549 lung cancer cells, MDAMB-231 breast cancer cells and Renca kidney cells. Therefore these results suggest that CBD has a possibility of anticancer agents and anticancer effects against cancer cells by modulation of apoptotic pathway in the range of 5-80 ${\mu}$M concentration.

An Aqueous Extract of a Bifidobacterium Species Induces Apoptosis and Inhibits Invasiveness of Non-Small Cell Lung Cancer Cells

  • Chemotherapy regimens for non-small cell lung cancer (NSCLC) have various adverse effects on the human body. For this reason, probiotics have received attention regarding their potential value as a safe and natural complementary strategy for cancer prevention. This study analyzed the anticancer effects of aqueous extracts of probiotic bacteria Bifidobacterium bifidum (BB), Bifidobacterium longum (BL), Bifidobacterium lactis (BLA), Bifidobacterium infantis 1 (BI1), and Bifidobacterium infantis 2 (BI2) on NSCLC cell lines. When the aqueous extracts of probiotic Bifidobacterium species were applied to the NSCLC cell lines A549, H1299, and HCC827, cell death increased considerably; in particular, the aqueous extracts from BB and BLA markedly reduced cell proliferation. p38 phosphorylation induced by BB aqueous extract increased the expression of cleaved caspase 3 and cleaved poly (ADP-ribose) polymerase (PARP), consequently inducing the apoptosis of A549 and H1299 cells. When the p38 inhibitor SB203580 was applied, phosphorylation of p38 decreased, and the expression of cleaved caspase 3 and cleaved PARP was also inhibited, resulting in a reduction of cell death. In addition, BB aqueous extracts reduced the secretion of MMP-9, leading to inhibition of cancer cell invasion. By contrast, after transfection of short hairpin RNA shMMP-9 (for a knockdown of MMP-9) into cancer cells, BB aqueous extracts treatment failed to suppress the cancer cell invasiveness. According to our results about their anticancer effects on NSCLC, probiotics consisting of Bifidobacterium species may be useful as adjunctive anticancer treatment in the future.

  • 연구배경 : IGFs는 다양한 종양세포에서 세포분열 및 성장에 관여하는 것으로 알려진 펩티드로써 폐암 조직에서 IGF-1에 대한 항체를 이용하여 면역조직화학염색을 실시하여 폐암세포에서 이의 발현 및 조직학적 형태에 따라 발현의 정도를 비교해 보고자 하였다. 방 법 : 15명의 소세포성 폐암 환자와 42명의 비소세포성 폐암 환자를 대상으로 IGF-1에 대한 면역조직화학적 염색을 실시하였다. 결 과 : 모든 폐암 조직애서 IGF-1의 발현을 보였고 비소세포성 폐암조직은 소세포성 폐암조직보다 IGF-1에 대한 발현의 정도가 유의하게 증가되어 있었다. 결 론 : 폐암세포는 IGF-1의 발현을 보이며 이에 대한 면역조직화학염색은 폐암세포의 조직학적 형태를 감별하는데 도움을 줄 수 있다.

Effects of OH Radical Density from Atmospheric Plasma to Induce Cell Death in Lung Cancer and Normal Cells

  • Atmospheric plasma's electron temperature is less than thermal plasma, so it is useful at bio experiment. We have investigated the optical emission spectroscopy (OES) lines by spectrometer during Atmospheric plasma bombardment onto the PBS surface by using an Ar gas flow. Also we have measured the OH radical density inside the solution induced by the Atmospheric plasma bombardment. OH radical species are appeared at 308 nm and 309 nm. Densities of OH radical species has been found to be significantly decreased versus depth of the solution from 2 mm to 6 mm. OH radical density inside the PBS is measured to be about $1.87{\times}1016cm-3$ downstream at 2 mm from the surface under optimized Ar gas flow of 200 sccm in Atmospheric plasma. Also we have investigated cell viability of lung cancer and normal cell after Atmospheric plasma treatment for fixed exposure time in 60 seconds, but different depths. We used SEM, we observed change of cell morphorogy, did experiment about FDA & PI Staining method. It is found that there is selectivity between the lung cancer and lung normal cell, in which cancer cell definitely has higher cell death ratio more than normal cell. We have investigated change of bond structure in FT-IR spectroscopy, the following peaks were observed: and intense O-H peak at 3422 cm-1 and at 2925 cm-1 corresponds to C-H stretch vibrations of methylene group.
