Alfalfa (Medicago sativa L.) is an important forage legume grown in Kansas, USA and its productivity with cool-season grasses however is unknown. The objective of this study was to determine the dry matter yield (DMY) and forage nutritive value of alfalfa-grass mixtures compared to those of alfalfa and grasses grown in monoculture with and without nitrogen fertilization. Three different alfalfa varieties were planted (reduced-lignin alfalfa, Roundup Ready, and conventional alfalfa) and two kinds of cool-season grasses (smooth brome, Bromus inermis Leyss, and tall fescue, Festuca arundinacea Schreb) were planted as a monoculture or in alfalfa-grass mixtures. Nitrogen fertilizer (urea) was applied at green-up at a rate of 56 kg/ha and after the second cutting at a rate of 56 kg/ha in 2016 and 2017, respectively. and control treatments received no nitrogen. DMY was significantly higher in monoculture alfalfa and alfalfa-grass mixtures than in grass monocultures. Between alfalfa monoculture and alfalfa-grass mixtures, no significant differences in DMY were found. For all treatments, nitrogen application significantly increased DMY compared to the control. In 2016 and 2017, the low-lignin alfalfa monoculture had the lowest acid detergent fiber (ADF) and the grass monocultures had the highest ADF. In 2016 and 2017, neutral detergent fiber (NDF) in smooth bromegrass and tall fescue was higher than in other species treatments. A low-lignin alfalfa monoculture had significantly lower NDF concentration compared to alfalfa-grass mixtures. When averaged over 2016 and 2017, relative feed value (RFV) was highest in low-lignin alfalfa and lowest in the grass monocultures. In both years, nitrogen fertilizer application did not affect nutritive values.

In this study, we separated the lignin from the wood by using the high boiling point solvent for developing more environmental friendly pulping method. High boiling point solvents as Ethers, glycols and ketones were used to remove the lignin in the pine wood meals. The Yield and lignin content of residual wood meals was reduced according to the input of the catalyst. Me-C, E-Ca, TEG and MIBK had the best delignification rate of 9 kinds of high-boiling point solvents. At the hydrolysis ratio of the selected solvents, The TEG was highest remain ratio of carbohydrates and the E-Ca was lowest remain ratio of lignin. And the Me-C was most excellent lignin hydrolysis ratio at the low catalyst. The selectivity of delignification of Me-C, E-Ca, TEG and MIBK solvents were 49.6, 49.9, 53.8 and 53.1%, respectively, and its values were similar to those of the commercial Kraft Pulp.
Background: The patatin-related phospholipase AIII family (pPLAIIIs) genes alter cell elongation and cell wall composition in Arabidopsis and rice plant, suggesting diverse commercial purposes of the economically important medicinal ginseng plant. Herein, we show the functional characterization of a ginseng pPLAIII gene for the first time and discuss its potential applications. Methods: pPLAIIIs were identified from ginseng expressed sequence tag clones and further confirmed by search against ginseng database and polymerase chain reaction. A clone showing the highest homology with pPLAIIIβ was shown to be overexpressed in Arabidopsis using Agrobacterium. Quantitative polymerase chain reaction was performed to analyze ginseng pPLAIIIβ expression. Phenotypes were observed using a low-vacuum scanning electron microscope. Lignin was stained using phloroglucinol and quantified using acetyl bromide. Results: The PgpPLAIIIβ transcripts were observed in all organs of 2-year-old ginseng. Overexpression of ginseng pPLAIIIβ (PgpPLAIIIβ-OE) in Arabidopsis resulted in small and stunted plants. It shortened the trichomes and decreased trichome number, indicating defects in cell polarity. Furthermore, OE lines exhibited enlarged seeds with less number per silique. The YUCCA9 gene was downregulated in the OE lines, which is reported to be associated with lignification. Accordingly, lignin was stained less in the OE lines, and the expression of two transcription factors related to lignin biosynthesis was also decreased significantly. Conclusion: Overexpression of pPLAIIIβ retarded cell elongation in all the tested organs except seeds, which were longer and thicker than those of the controls. Shorter root length is related to auxinresponsive genes, and its stunted phenotype showed decreased lignin content.
This study presents a conceptual design of vanillin production from Kraft lignin. Most of the existing Kraft lignin is used as low-quality boiler fuel or discarded as wastewater, and only 2% or less of lignin has been refined into high-quality products. We propose the process developed in this study to utilize discarded Kraft lignin. The existing vanillin production concept process consisted of alkali oxidation using NaOH, filtration, chromatography, and crystallization. Chromatography, which is difficult to commercialize, was changed to a solvent extraction process. The recovery rate of vanillin of the proposed solvent extraction process is 92.9%, and the purity is 99.5%, which is similar to the existing chromatography process. The reason why the solvent extraction process showing similar results to chromatography can replace the existing chromatography process was analyzed.

Effect of excessive addition of water reducing agent was examined by concrete tests. Water reducing agents of lignin or naphtalene base were used in the experiment. Setting of concrete was retarded according to the increase of dosage of water reducing agent and was extremely delayed at the 3 times or more use of lignin base agent and near 5 times or more use of naphtalene base agent respectively. When water reducing agent was used more than 6 times of standard dosage, early strength of concrete was very low and the strength reduction was very severe at all test ages in the concrete using lignin base agent.
This study was carried out to analyze the organic acids, polyphenols, pigments and fiber materials concentration with a different stalk position and grade of korean leaf tobaccos. Eight kinds of flue-cured leaf tobaccos which were different stalk position and grade were used for this study. Three kinds of major organic acids(citric, malic and oxalic), 2 kinds of polyphenols(chlorogenic acid and rutin), 3 kinds of pigments($\beta$-carotene, chlorophyll-a and chlorophyll-b), and 2 kinds of fiber components(pectin and lignin) were analyzed. All of these chemical components were changed with a different stalk position. When the citric acid, malic acid, $\beta-carotene$, chlorophyll-a, and lignin concentration were low in the middle stalk position and high in both bottom and upper position, oxalic acid and chlorogenic acid show the highest concentration in the middle stalk position. All of these chemical components also changed with a different grade of leaf tobaccos. As the citric acid, malic acid, $\beta-carotene$, chlorophyll-b, and lignin concentration decreased as the grade ascended, the oxalic acid and chlorogenic acid concentration increased as the grade ascended. This results assumed that the quality of korean leaf tobacco was directly proportional to oxalic acid and chlorogenic acid concentration but it was inversely proportional to citric acid, malic acid, $\beta-carotene$, chlorophyll-b and lignin concentration.
The purpose of this was to investigate the degradation efficiency of phenol compounds(catechol, ferulic acid, protocatechuic acid, syringic acid, vanillic acid) by Streptomyces halstedii scabies SAI-36, Streptomyces avendulas SA2-14, and Strptomyces badius(ATCC 39117, control group). The results were as follows: Catechol showed the degradation efficiency that is lower than 50% in three strains. Ferulic acid and vanillic acid showed high degradation efficiency of 98.8% and 94.5% respectively by Streptomyces lavendulas SA2-14. protocatechuic acid and syringicacid showed high degradation efficiency of 89.6% and 77.9%. The degradation efficiency of catechol by Streptomyces halstedii scabies SAI-36, Streptomyces lavendulas SA2-14 and Streptomyces badius(ATCC 39117) was low as 49.2%, 40.2% and 20.2% respectively. But the degradation of other phenolic compoumds except catechol by Streptomyces laven-dulas SA2-36 and Streptomyces badius(ATCC 39117). The results demonstrated that two experimental strains are superior ability to control group in degradation of phenol compounds and Streptomyces lavendulas SA2-14 was superior of two experimental strain. This results were consistent with previous research results that Streptomyces lavendulas SA2-14 was the best strain in degradation ability for lignin, decoloration abilities for variousdyes, and various enzyme production abilities. Therefore, it is suggested that lignin can be used as a indicator when selecting Actinomycetes for degradation of non-degradable materials such as phenol compounds.
Lignocellulose ($2^{nd}$ generation) is difficult to hydrolyze due to the presence of lignin and the technology developed for cellulose fermentation to ethanol is not yet economically viable. However, recent advances in the extremely new field of biotechnology for the ethanol production are making it possible to use of agriculture residuals and nonedible crops biomass, e.q., rice straw and miscanthus sinensis, because of their several superior aspects as agriculture residual and nonedible crops biomass; low lignin, high contents of carbohydrates. In this article, as the basic study of AP(Ammonia Percolation), the properties and the optium conditions of process were established, and then the overall efficiency of AP was investigated. The important independent variables for AP process were selected as ammonia concentration, reaction temperature, and reaction time. The percolation condition for maximizing the content of cellulose, the enzymatic digestibility, and the lignin removal was optimized using RSM(Response Surface Methodology). The determined optimum condition is ammonia concentration; 11.27%, reaction temperature; $157.75^{\circ}C$, and reaction time; 10.01 min. The satisfying results were obtained under this optimized condition, that is, the results are as follows: cellulose content(relative); 39.98%, lignin content(relative); 8.01%, and enzymatic digestibility; 85.89%.
Lignin, a prominent constituent of woody biomass, is abundant in nature, cost-effective, and contains various functional groups, including hydroxyl groups. Owing to these characteristics, they have the potential to replace petroleum-based polyols in the polyurethane industry, offering a solution to environmental problems linked to resource depletion and CO2 emissions. However, the structural complexity and low reactivity of lignin present challenges for its direct application in polyurethane materials. In this study, Kraft lignin (KL), a representative technical lignin, was fractionated with ethanol, an eco-friendly solvent, and mixed with conventional polyols in varying proportions to produce polyurethane films. The results of ethanol fractionation showed that the polydispersity of ethanol-soluble lignin (ESL) decreased from 3.71 to 2.72 and the hydroxyl content of ESL increased from 4.20 mmol/g to 5.49 mmol/g. Consequently, the polyurethane prepared by adding ESL was superior to the KL-based film, exhibiting improved miscibility with petrochemical-based polyols and reactivity with isocyanate groups. Consequently, the films using ESL as the polyol exhibited reduced shrinkage and a more uniform structure. Optical microscope and scanning electron microscope observations confirmed that lignin aggregation was lower in polyurethane with ESL than in that with KL. When the hydrophobicity of the samples was measured using the water contact angle, the addition of ESL resulted in higher hydrophobicity. In addition, as the amount of ESL added increased, an increase of 7.4% in the residual char was observed, and a 4.04% increase in Tmax the thermal stability of the produced polyurethane was effectively improved.
Organic by-products derived from cellulose and lignin during organosolv pretreatments of yellow poplar wood (Liriodendron tulipifera) in the presence of $H_2SO_4$ and NaOH as catalysts, respectively, were subjected to various analyses to elucidate their effects on further performance of biological ethanol fermentation and provide preliminary data for the structure and utilization of organosolv lignin. Monomeric sugars amounted to ca. 2.2-7.7% in the organosoluble fraction of the organosolv pretreatment with $H_2SO_4$, while significantly low amount of sugars (0.2-0.3%) were determined in that of the organosolv pretreatment with NaOH. In case of addition of $H_2SO_4$ during organosolv pretreatment of biomass, a fermentation of the organosoluble fraction could be considered as an essential process to increase an efficiency of biomass utilization as well as yield of bioethanol. Precipitates, insoluble by-products in the solvent mixture, were also cficiency oed by diverse analytical methods and revealed that these were typically composed of a lignin moiety regardless of catalyst. According to the results of nuclear magnetic resonance (NMR), Fourier Tcinsform Infrared Spectroscopy (FT-IR) and Gel permeation chromatograp r (GPC), the main components of precipitates seem to be lignin polymers. However, their structures could be slightly modified during pretreatment and mixed with some carbohydrates by chemical bonds and/or physical associations.
