Dietary proteins influence colorectal cancer (CRC) risk, depending on their quantity and quality. Here, using pyrosequencing, we compared the fecal microbiota composition in Balb/c mice fed either a normal protein/carbohydrate diet (ND, 20% casein and 68% carbohydrate) or a high-protein/low-carbohydrate diet (HPLCD, 30% casein and 57% carbohydrate). The results showed that HPLCD feeding for 2 weeks reduced the diversity and altered the composition of the microbiota compared with the ND mice, which included a decrease in the proportion of the family Lachnospiraceae and Ruminococcaceae and increases in the proportions of the genus Bacteroides and Parabacteroides, especially the species EF09600_s and EF604598_s. Similar changes were reported in patients with inflammatory bowel disease, and in mouse models of CRC and colitis, respectively. This suggests that HPLCD may lead to a deleterious luminal environment and may have adverse effects on the intestinal health of individuals consuming such a diet.
Protein hydrolysates and krill meal (KM) are used as protein sources in aquafeeds. The study was conducted to examine the supplemental effects of shrimp protein hydrolysates (SH) or KM in a high-plant-protein diet for red seabream (Pagrus major). A fish meal (FM)-based diet (40%) was considered as the high-FM diet (HFM) and a diet containing 25% FM and soy protein concentrate, in the expense of FM protein from HFM diet, was considered as the low fish meal (LFM) diet. Two other experimental diets (SH and KM) were prepared by including SH and KM into LFM diet at 5% inclusion levels in exchange of 5% FM from the LFM diet. A feeding trial was conducted for fifteen weeks using triplicate group of fish (Initial mean body weight, 8.47 ± 0.05 g) for a diet. Growth performance and feed efficiency of fish were significantly enhanced by HFM, KM and SH supplemented diets over those of fish fed LFM diet. Interestingly, these parameters of fish fed SH diet showed better performance than KM and HFM groups. Liver IGF-I expression of fish fed SH diet was comparable to HFM group and higher than KM and LFM diets. Protein digestibility of SH diet was significantly higher than KM, HFM, and LFM diets. Dry matter digestibility of SH diet was comparable to HFM diet and significantly higher than KM and LFM diets. Nitro blue tetrazolium and superoxide dismutase activities of HFM, SH and KM groups were significantly elevated than the LFM group and SH diet increased catalase and glutathione peroxidase activities of fish compared to KM and LFM groups. Hemoglobin level and hematocrit of fish fed SH and KM diets were significantly higher than LFM group. A diet containing 20% FM with KM is comparable to a HFM diet which contains 40% FM for red seabream. SH can be used to replace FM from red seabream diet down to 20% and fish performance can be improved better than a diet containing 40% FM. Overall, it seems that SH is more effective ingredient in red seabream diet compared to KM.

The variations of growth gain and the composition of free amino acid levels in plasma and erythrocytes of young rats(wistar strain male) were determined by microbioassay method, feeding diets of rice group and 7% casein group as a control for three weeks. The results were as follows; 1. The growth gain of control diet group was higher than the rice diet group. 2. The contents of free tryptophan, lysine, and threonine levels in plasma and erythrocytes on rats of 7% casein group were higher than the rice group. 3. In the 7% casein diet group and the rice group, these free amino acids were included more in erythrocytes than in plasma. 4. Therefore, generally feeding by high protein score diet was included more Try, Lys, Thr in plasma and erythrocytes than feeding by low protein score diet. So the high and low of protein score was assumed by the contents of Try, Lys, Thr in plasma and erythrocytes on rats.

Effects of dietary protein levels on the manifestations of the toxicity of gramoxone, a bipyridine herbicide, in the liver of rats were investigated. The addition of gramoxone, with regard to the body weight and feed efficiency ratio of rats, had a move dramatic effect on animals fed a low or intermediate protein diet than for those similarly treated among rats fed a relatively high protein diet. Lipid content in the rat liver tended to increase with the addition of gramoxone into each protein diet, with the exception of the high protein-gramoxone diet. The addition of gramoxone tended to increase hepatic TBA value significantly in rats, especially among those fed the low protein-gramoxone diet or the control-gramoxone diet. Significant morphological changes, including fat changes of hepatic cells and increases in the number of Kupffer cells, were found both in rats fed the low protein diet and those fed any of the gramoxone-treated diets. fat changes within hepatic cells were found to be especially severe in rats fed the low protein-gramoxone diet. Distributions of glycogen in rat liver appeared to increase in rats fed any of the diets to which gramoxone had been added.

Obesity can be defined as a metabolic disease due to an increased fat accumulation in the body caused by an imbalance of calorie intake and output The prevalence of obesity has increased substantially over the past 2-3 decades in developed and developing countries. The health impact of weight gain is so marked that obesity has now been classified as a major global public health problem In order to investigate the effect of diet conversion and oral administration of Platycodon grandiflorum extracts on the treatment of obesity, male Spraque-Dawley rats were divided into four groups: a group converted to normal diet (Control group), a group maintained high fat (30%) diet (H), and two groups with Platycodon grandiflorum extract added to the previously mentioned two groups. All animals were fed high fat diet for 7 weeks to induce the obesity. Then they were divided as mentioned above. Animals were fed experimental diet and Platycodon grandiflorum extract (150 mg/ml/rat/day) for 7 weeks. Body weight, adipose tissue weight (subcutaneous, epididymal, peritoneal fat pads) and serum lipids (total cholesterol and triglyceride) showed some differences among groups. The Platycodon grandiflorum feeding markedly decreased both body weight and adipose tissue weight in control group compared to H, high fat diet maintaining, group. Platycodon grandiflorum extracts significantly decreased the concentrations of serum lipids compared to H group. Fat cell numbers and sizes were significantly reduced in the oriental medicinal herb extract administrated group. Increased fatty acid binding protein (FABP) expression in high fat diet group was decreased by the dietary conversion to normal diet and the oral administration of Platycodon glandiflorum extracts. In contrast, there was no significant effect on FABP expression in the high fat maintenance group. In this study, the conversion from high fat diet to low fat or normal diet had a beneficial effect on body weight loss and serum lipid profiles. Dietary Platycodon glandiflorum extracts had an additive beneficial effect on the prevention and treatment of obesity.
The effects of diet induced hyperlipidemia on kidney function were studied in Sprague-Dawley rats fed high fat diet containing 20% beef tallow and high cholesterol diet containing 5% cholesterol for 8, 12, 16 weeks, respectively. The concentrations of serum total lipid, total cholesterol and LDL-cholesterol were significantly high cholesterol diet groups during all experimental periods (p<0.05). HDL-cholesterol concentration was the lowest value in high cholesterol diet group of 16 weeks(p<0.05). Triglyceride concentration was not affected by experimental diets. Serum total protein, albumin and creatinine concentration tended to higher in high cholesterol diet groups than those in high fat diet groups. And serum urea-N concentration was higher in high fat diet group of 16 weeks than that in other diet groups. Urinary total protein and urea-N were higher in high cholesterol diet groups than those in other diet groups regardless of experimental period period. There was no significant difference in urinary creatinine concentratin among diet groups(p<0.05). GFR was lower in high cholesterol diet groups than that in high fat diet groups at 8, 16 weeks, respectively. Wet weight per body weight, total lipid, triglyceride, total cholesterol concentations of liver tissue were apparently high in high cholesterol diet groups(p<0.05). Kidney wet weight per body weight wer not affected by experimently diets, total lipid concentration of kidney tissue was significantly high in high fat diet groups of 12 weeks(p<0.05), kidney tissue triglyceride concentrations of high cholesterol diet groups of 12, 16 weeks apparently low, and total cholesterol concentration of kidney tissue was higher in experimental diet groups than that of control groups at 12, 16 weeks(p<0.05). Fecal excretion, total lipid, triglyceride and cholesterol concentrations of feces were markedly high in all high cholesterol diet groups except high fat diet group of 16 weeks. The results of light microscopic examination indicated that glomerulosclerosis was not obsrved in rats fed experimental diets.
Schisandra chinensis extract (SCE) protects against hypocholesterolemia by inhibiting proprotein convertase subtilisin/kexin 9 (PCSK9) protein stabilization. We hypothesized that the hypocholesterolemic activity of SCE can be attributable to upregulation of the PCSK9 inhibition-associated low-density lipoprotein receptor (LDLR). Male mice were fed a low-fat diet or a Western diet (WD) containing SCE at 1% for 12 weeks. WD increased final body weight and blood LDL cholesterol levels as well as alanine transaminase and aspartate aminotransferase expression. However, SCE supplementation significantly attenuated the increase in blood markers caused by WD. SCE also attenuated WD-mediated increases in hepatic LDLR protein expression in the obese mice. In addition, SCE increased LDLR protein expression and attenuated cellular PCSK9 levels in HepG2 cells supplemented with delipidated serum (DLPS). Non-toxic concentrations of schisandrin A (SA), one of the active components of SCE, significantly increased LDLR expression and tended to decrease PCSK9 protein levels in DLPS-treated HepG2 cells. High levels of SA-mediated PCSK9 attenuation was not attributable to reduced PCSK9 gene expression, but was associated with free PCSK9 protein degradation in this cell model. Our findings show that PCSK9 secretion can be significantly reduced by SA treatment, contributing to reductions in free cholesterol levels.
This study was performed to investigate the effects of dietary protein and calcium levels on calcium metabolism in eight healthy Korean adult females. The 2-day metabolic study consisted of a 2 day adaptation period and three 6-day experimental periods. Three experimental diets were low protein low calcium(LPLCa : protein 44g, Ca 422mg), higher protein low calcium(HPLCa : protein 85g, Ca 365mg), and high protein high calcium (HPHCa : protein 84g, Ca 727mg). The apparent calcium absorption was likely to be affected by the calcium intake rather than by the protein intake. Average calcium absorption rate was about 23-29% of calcium intake. The calcium balance was -21.44mg for LPCa, -25.02mg for HPLCa, and -3.22mg for HPHCa. Avergae urinary calcium excretion was 127.7mg for LPLCa, 108.6mg for HPLCa, and 215.4mg for HPHCa. Urinary calcium excretion was more closely related to the changes of calcium intake rather than of protein intake. These results seemed to be due to the interactions between the high phosphours contained in the high protein diet and the little discrepancy of protein intake levels.
This study were performed to investigate effect of dietary cadmium(Cd) and protein levels on growth, body protein metabolism and Cd toxicity in growing rats. Forty eight male rats of Sprague-Dawley weighing 113$\pm$2g were blocked into 6 groups accoridng to body weight. Dietary protein were given at the levels of 7, 15 and 40% of diet and Cd (200ppm)were either added or not. The result obtained were summerized as follow; 1) Food intake, weight gain, FER PER, liver and kidney weight, weight and length of bones, hematocrit, and hemoglobin content in Cd-added groups were low than those in Cd-free groups. 2) Serum total protein showed no significant difference with Cd addition, but it was significantly lower in low protein diet groups. Liver protein in Cd-added groups was lower than Cd-free groups, and was tend to be increased with increasing dietary protein level. 3) Daily urinary and fecal nitrogen excretions in Cd-added groups were lower than Cd-free groups, and were increased with increasing dietary protein level. 4) Cadmium contents in blood, liver, kidney, and femur were tend to be decreased with increasing dietary protein level. Especially, Cd content in kidney of Cd-added groups was significantly decreased with increasing dietary protein level. 5) Daily urinary and fecal Cd excretions were tend to be increased with increasing dietary protein level, and Cd-added-high protein diet group showed the highest Cd excretion among the Cd-added groups, Cd absorption ration and Cd retention ratio were tend to be decreased with increasing dietary protein level.
The present study was undertaken to determine the metabolic effect of various levels of fat in the diet. Forty males and the same number of females weighing $35{\pm}29$ were divided into three experimental groups and one control group, 10 rats each in both sexes. The dietary lipid contents were included in three levels, 2% as low, 30% as high and fat free diet in order to reflect the lipid consumption of present Korean diet. 20% sugar casein diet were employed as standard for control animals. This study was carried for 16 weeks. After these period animals were sacrificed to collect the internal organs and blood samples by heart puncture. In the result of this study, high fat diet group is lower than low fat diet group in the body weight gain, food efficiency ratio, protein efficiency ratio, urinary nitrogen and organ nitrogen contents were same trend but there is no significant difference in these respects. It was noteworty that high, and free fat diet group revealed more glucose total protein, albumin and globulin contents in the serum than other compared groups. It can be concluded that fat content whether low or high are free in the diet did not show significant effect on body nitrogen metabolism. But 30% high fat diet increased the total lipid and total cholesterol contents in the liver and the serum. This fact can be interpreted that 2% low fat diet and fat free diet do not need to increase the amount of fat content in the diet as high as 30% fat diet. This result might indicate the one possible reason to decide the recommanded dietary fat levels in Korean diet.
