Characterization of Chitinase in Oak Tissues and Changes in Its Activity Related to Water Stress and Inoculation with Hypoxylon atropunctatum

  • Chitinase activities from Shumard oak tissues were determined to study changes in chitinase activities related to water stress. The enzyme extracted in sodium acetate buffer (0.1M, pH 4.5) was assayed by a colorimetric method. In addition, the fungal hyphae of Hypoxylon atropunctatum in xylem tissues of oak were observed through scanning electron microscopy. The enzyme in oak tissues was mainly endochitinase, and optimum pH for enzyme activity was 5. Specific chitinase activities from both of stems held under high relative humidity (ranges of 0.63-1.11 pKatal/$\mu\textrm{g}$ of protein) and stems held under low relative humidity (ranges of 0.41-0.99 pKatal/$\mu\textrm{g}$ of protein) were significantly increased following fungal inoculation with H. atropunctatum. However, there was no significant difference in chitinase activities between tissues held under high and low humidities, which might be due to fungal chitinase. Scanning electron microscopy showed holes in fungal hyphae in the xylem tissues of stems held under high humidity but not in the stems held under ow humidity, suggesting that hyphae might be hydrolyzed by plant hydolases such as chitinase.

Influence of Freezing Rate on the Aroma Retention in a Freeze Drying System (동결건조 시스템에서 동결속도가 향미물질 보존에 미치는 영향)

  • The objective of this study was to investigate the effects of freezing rate on aroma retention and to examine the mechanism of aroma retention during freeze drying process. Our experiments were carried out with self-manufactured freeze-dryer. Gelatin gels (2% w / w, 80${\times}$20mm) containing diacetyl(2mg/ml) were frozen unidirectionally (Neumann's model) from the bottom at -45, -30, -20, and -15$^{\circ}C$ and followed with freeze-drying. Under the upper conditions we measured freezing rate and the change of temperature and pressure during freeze drying. Freeze-dried gelatins were cut horizontally into 5 mm thickness from the bottom measured and diacetly contents. Besides, we observed the effect of the relative humidity of the diacetyl contents freeze-dried gelatin during storage. The retained diacetyl content was increased at high freezing temperature and in order of 0∼5, 5∼10, 10∼15, 15∼20 mm section from the bottom of the sample. It was observed that the retained diacetyl content was high in 15∼20 mm section. The retained diacetyl content and freeze-dried gelatin stored in the condition of high relative humidity was decreased significantly but in the low relative humidity case, was it decreased in small amount. The results of our experiment resents that the low temperature freezing and low humidity storing condition is effective for preserving aroma compound in food.

  • The low level atmospheric obsewation carried out to Investigate magnitude, formation and dissipation of nocturnal surface Inversion layer, also to survey relation to each meterological parameter In Inversion layer at Pusan power plant for Oct. 13, 1996. As coastal area, the surface Inversion layer height was relatively high(186m), and after sunset unstable layer formated firom surface to around 40m, and the Inversion layer was left still in the upper layer. The surface Inversion layer dissipated at 0920LST perfectly. The layer that strong Inversion layer was formated, showed steep variation of potential temperature and wand speed and relative humidity.

  • This research was carried out study the effect of several relative humidities on the cracking of kernels of chestnuts. The moisture contents of total kernel increased at the higher humidity over 88% relative humidity. but the moisture contents of inner layer was decreased at low relative humidity below 76 %. The starch contents of kernel decreased, but free stgars increased during the total period of storage. The tannin contents increased during total of storage, and the tannin contents of out layer was higher than inner layer because of tannin transfer from astringent shell. As we considered the sprouting ratio, the rotting ratio and the cracking ratio, the optimal relative humidity of chestnut storage was proved 88% relative humidity.

  • Paulownia coreana has various advantages as a convenient workability, aesthetic outlook, beautiful patterns, low weight and high strength, and its permeability. P. coreana has been widely used for storage but there are no empirical researches proving its functionality in a field of conservation science until now. In this study, the seasonal and daily temperature and relative humidity control, and microbe and insect repellent activity were evaluated under the controlled and uncontrolled circumstances from 2015 to 2016. The results showed to be mainly excellent in relative humidity control and the buffering effect was good to adjust the average daily relative humidity range from the outside. With respect to the antimicrobial properties of P. coreana, we observed that its water-soluble extract produced visible zones of inhibition against five bacteria. However, it was difficult to predict the antimicrobial and/or insecticidal properties.

  • This paper examined the possibility of NET application for a relative weather stress index in Korea. The characteristic of NET distribution used temperature, relative humidity, wind speed which forecasting at Korean Meteorological Administration were analyzed. Regional critical values of daily maximum NET of stress index for summer resembled the distribution of daily maximum temperature because were not impacted wind and humidity but temperature. Regional critical values of daily minimum NET of stress index for winter distributed variously compared with summer. The highland region and the northern region of Seoul were impacted of low temperature and coastal region which strong wind. The occurrences of stressful days did not vary in summer, but obviously increased in winter after mid-1990s.

  • Washing machine manufacturers typically advise consumers to maintain the relative humidity in the drum less than 80% for three days after the termination of a washing cycle in order to prevent bacteria proliferation. A vent installed in the back of a washing machine is used to release moisture to satisfy this condition. Up to now, the design and installation of the vent have been based on experiments without understanding its roles and physical phenomena. In this study, various CFD results are presented in order to explain the physical mechanism of moisture release in a washing machine. Two methods of moisture release (diffusion and convection) were studied; diffusion was found to be the dominant process in removing moisture. Experiments were also performed to validate this behavior. In addition, this study will aid in the efficient design of vents to keep the relative humidity low inside the drum.

Performance Analysis of a Wet Air-Cycle Refrigeration System (습공기사이클 냉동시스템의 성능해석)

  • The objective of this study is to theoretically analyze the performance of an open wet air-cycle refrigeration system, which nowadays is increasingly generating environmental concern. The temperature and relative humidity of the outside air are selected as the most important parameters. As the temperature and relative humidity of the outside air increase, the pressure ratio of the ACM compressor is determined to be nearly constant, the air temperature at the exit of the system increases, and the amount of condensed water, the cooling capacity, the COP, and the total entropy production rate increase overall. The effects of the effectiveness of the heat exchanger and the efficiency of the turbine on the performance are greater than that of the efficiency of the ACM compressor. Also, the performance of the wet air-cycle refrigeration system with two heat exchangers is enhanced, with a high COP and low total entropy production rate, compared to the system with a single heat exchanger.

  • The purpose of this Study was to determine thermal sensation and physiological responses for men in summer indoor environment, under various air temperature and relative humidity, with male university students. Subjective Evaluation, Heart Rate Variability(HRV), Electroencephalogram(EEG) were examined. We found that comfort of people was achieved at 50% R.H., 24C, and the difference of skin temperature was found at the calf area as air temperature changes. At low air temperature and low humidity, heart rate was decreased, but there was no change at brain wave, keeping a-wave.

  • During PEMFC operation, OCV(open circuit voltage) and low humidity conditions accelerate the degradation of perfluorosulfonic acid membrane. There have been no studies that clearly explain why these conditions accelerate the membrane degradation. In this study, the hydrogen permeability through the membrane, I-V polarization of MEA, fluoride emission rate(FER) and $H_2O_2$ concentration in condensed water were measured during cell operation under OCV and low relative humidity(RH). The experimental results were evaluated with oxygen radical mechanism the most commonly known for membrane degradation. It seems that low RH of anode is a good condition for $H{\cdot}$ radical formation on the Pt catalyst and the OCV condition accelerate the $H{\cdot}$ to form $HO_2{\cdot}$ radical attacking the polymer membrane.
