금은화 추출물을 이용한 미생물 발효 생성물의 항산화 특성 (Antioxidant Properties in Microbial Fermentation Products of Lonicera japonica Thunb. Extract)

  • 최근에 합성 항산화제를 대체할 천연물에서 새로운 항산화 물질을 찾는 연구가 활발하게 진행되고 있다. 천연물 중에 Lonicera japonica Thumb은 항균, 항염증, 항바이러스, 해독작용, 혈중 지질 감소, 해열작용 등 약리적인 효능뿐 아니라 다양한 항산화 물질을 포함하고 있는 것으로 보고되어 있다. 본 연구는 금은화 열수, 알코올 추출물을 기질로 전통발효식품에서 분리한 Lactobacillus plantarum NHP1 균주로 발효과정을 진행한 후 항산화 물질 및 래디컬 소거능이 변화하는지를 관찰하기 위해 진행되었다. 전통 발효식품으로부터 발효를 위한 균주를 분리한 후 16S rDNA 염기서열을 비교하여 동정한 결과 Lactobacillus plantarum NHP1로 명명하였다. 발효를 위한 기질인 Lonicera japonica Thumb는 열수 및 70% ethanol을 이용하여 추출하였다. 열수 및 ethanol 추출물의 발효는 2 L 발효기에 1.2 L 추출물을 함유한 배지를 공급하고, $30^{\circ}C$ 및 100 rpm의 조건으로 48 시간 수행하였다. 발효 전 후의 총 페놀, 총 플라보노이드, DPPH 소거능 및 ABTS 변화를 측정한 결과, 발효 전 총 페놀양은 열수 추출물과 ethanol 추출물에서 $56.5{\pm}4.9$ GAE mg/g, $72.7{\pm}5.3$ GAE mg/g이었고, 발효 후 각각 30.2%, 12.9% 증가되었다. 총 플라보노이드 함량은 모든 용매에서 발효 후에 유의적으로 약 75% 이상 증가되었다. 발효 후 총 페놀 함량과 플라보노이드 함량이 증가한 것에 비해 DPPH 소거능은 비례적으로 증가되지 않았으나, ABTS 항산화능은 발효에 의해 50% 증가되었다. 그동안 선행 연구에서 금은화의용매 추출물에 대한 기능성연구를 진행한 것이 대부분인 것에 비해, 본 연구 결과를 바탕으로 금은화 추출물에 발효기술을 접목한다면 항산화 기능 및 다른 기능성이 대폭 증가되어 기능성 건강식품 및 미용식품의 소재로의 사용이 활성화 될 수 있을 것이라 사료된다.

  • Lonicerae Folium et Caulis, the folium and stem of Lonicera japonica Thunb., has been used as diuretic, stomachic, antipyretic, analgesic and anti-inflammatory agent in Korea. We isolated a main iridoid, loganin which has some important biological effects from the folium and stem of this plant. Generally, it is known that iridoid compounds have variable contents by the collecting time and a part of plant. The content of main compound is important to evaluate its quality. In order to evaluate the quality of Lonicerae Folium et Caulis, the method of quantitative determination of loganin as a reference standard compound has been developed. (omitted)

  • A tyrosinase inhibitor, Lonjapoin, was isolated from the ethanolic extracts of Flos Lonicera by activity-guided fractionation. Lonjapoin ($IC_{50}$/; 2.07 $\mu\textrm{g}$/mL) was found to be potentially as effective inhibitor of production of melamin. Flos Lonicera (Jinyinhua) is the dry flower buds of Lonicera japonica. Thunb. It is used as an antibacterial and antiphlogistic agent in the treatment of abscess, laryngeal catarrh, erysipelas, dysentery, cold and fever. In our continuing search for natural source as cosmetic ingredients from Chinese medicinal herbs and found the extracts of Flos Lonicera showed the significant tyrosinase inhibition activity. In this study the active constituent of Lonicera japonica. obtained with the process of tyrosinase activity assay and column chromatography.

Distribution and Habitat Chracteristics of Lonicera japonica Thunb. in the Inland and the Seashore Areas of Korea

  • This study was conducted to evaluate the status of habitat distribution, environmental characteristics and plant species growing with Lonicera japonica in its habitats. In the distribution of plant coverage below 30%, it was 84.6% in the inland area and 80.7% in the seashore area. There was similar coverage in both inland and seashore areas. But the average relative coverage in the seashore area was 26.1%, which is a little higher than that of the inland area (22.5%). In the habitats, both inland and seashore areas showed a high distribution ratio below 10 degrees. But among them, inland areas showed a high distribution ratio (62.4%) below 10 degrees, which was about twice as much as the seashore areas (32.2%). Habitat distribution was most often found on the southern slopes in both inland and seashore areas. The average soil pH of Lonicera japonica habitat was 5.2 in the inland areas and 6.9 in the seashore areas. Also, the seashore areas showed little content of $P_2O_5$, but high content of K, Mg, Na on the other hand compared to the inland area. Miscanthus sinensis was the highest importance value (16.4) among plants growing with Lonicera japonica in their habitats in both inland and seashore areas. But in the inland areas, Artemisia princeps showed the highest importance value, and Miscanthus sinensis was the highest in the seashore areas.

Discrimination of Lonicera japonica and Lonicera confusa using chemical analysis and genetic marker

  • Objective : Lonicera japonica THUNB. a traditional herbal medicine, has been commonly used anti-inflammatory disease. It has been very complicated with respect to its sources on the market. The significant selection of medicine depends on its origin. However, it is difficult to discrimination criteria for confirming L. japonica authenticity using the senses. This study was performed to determine the discriminant analysis of L. japonica and L. confusa. Methods : The identification of L. japonica and L. confusa were performed by the classification and identification committee of the national center for standardization of herbal medicines. And we examined its differences using HPLC and genetic marker analysis. Results : The analytical pattern of High Performance Liquid Chromatography was determined from the corresponding peak curves ((E)-aldosecologanin, chlorogenic acid, luteolin 7-O-glucoside, sweroside). For L. japonica, additional unknown peaks were detected at 13.8 min, 20.6 min, and 36.9 min. And, we developed genetic marker using the the tRNA-Leu gene, trnL-trnF intergenic spacer and tRNA-Phe region of chloroplast DNA. By the method, 164 bp PCR product amplified from L. confusa was distinguished into L. japonica and L. confusa efficiently. Conclusion : Base on these results, two techniques provide effective approaches to distinguish L. japonica from L. confusa.

Lonicera Japonioa Inhibits the Production of NO through the Suppression of NF-kB Activity in LPS-stimulated Mouse Peritoneal Macrophages

  • The flowers of Lonicera japonica Thunb. (Caprifoliaceae) has been used as anti-inflammatory drug in the folk medicine recipe and been proved its anti-inflammatory effect in the oriental medicine. However, the action mechanism of Lonicera japonica that exhibits anti-inflammatory effects has not been determined. Since nitric oxide (NO) is one of the major inflammatory parameter, we studied the effect of aqueous extracts of Lonicera japonica (AELJ) on NO production in lipopolysaccharide (LPS)-stimulated mouse peritoneal macrophages. NO and inducible NO synthase (iNOS) level were significantly reduced in LPS-stimulated macrophages by AELJ compared to those without Electrophoretic mobility shift assay (EMSA) indicated that AELJ blocked the activation of nuclear factor kappa B (NF-kB), which was considered to be a potential transcription factor for the iNOS expression. AELJ also blocked the phosphorylation and degradation of inhibitor of kappa B-alpha (IkB-${\alpha}$). Furthermore, IkB kinase alpha (IKK${\alpha}$), which is known to phosphorylate serine residues of IkB directly, is inhibited by AELJ in vivo and in vitro. These results suggest that AELJ could exert its anti-inflammatory actions by suppressing the synthesis of NO through inhibition of NF-kB activity.

It has been known as an anti-inflammatory agent in Korea from ancient times and is used widely for treating upper-respiratory tract infections, diabetes mellitus and rheumatoid arthritis. Lonicera Japonioa Suppresses the Mast Cell-Mediated Immediate Allergic Reaction

  • The flowers of Lonicera japonica Thunb. (Caprifoliaceae) have been used as a traditional effective drug in treating bacillary dysentery, purulence. However, the exact role of Lonicera japonica in allergic reaction has not been clarified yet. Immediate hypersensitivity, popularly known as allergy, is a major clinical problem in humans. It has been found that the histamine release from mast cells is an essential step in the pathological process of immediate hypersensitivity. In this study, the effect of aqueous extract of Lonicera japonica (AELJ) on allergic reaction was investigated. AELJ inhibited the compound 48/80-induced anaphylactic reactions and antigen-induced passive cutaneous anaphylaxis (PCA). AELJ in vitro exhibited a dose-dependent inhibition of degranulation in RPMC stimulated by compound 48/80. AELJ also suppressed the morphological changes and the increase of intracellular free calcium level induced by compound 48/80. These results suggest that inhibitory effect of AELJ on allergic reaction may be mediated through the decrease of intracellular free calcium levels, and AELJ importantly contributes to the treatment of anaphylaxis and may be useful for other allergic disease.

Lignans from Lonicerae caulis

  • A new 2,7'-cyclolignan named lonicerinol (1) along with eight known lignans, (-)-epipinoresinol (2), (-)-pinoresinol (3), $9{\alpha}$-hydroxypinoresinol (4), 7R,8S-dihydrodehydrodiconiferyl alcohol (5), ($\pm$)-neo-olivil (6), (+)-isolariciresinol (7), 3-methoxy-8,4'-oxyneoligna-3',4,7,9,9'-pentol (8), and (-)-pinoresinol 4-O-glucoside (9), were isolated from the caulis of Lonicera japonica THUNB. (Caprifoliaceae). All of these constituents except for (-)-pinoresinol (3) and $9{\alpha}$-hydroxypinoresinol (4) are reported for the first time from the genus Lonicera. The structures and absolute configurations of these compounds were determined on the basis of spectroscopic analysis, including CD and 1D- and 2D-NMR techniques and chemical methods.

인동의 성분연구 (3) - Iridoid 화합물 (Phytochemical Studies on Lonicera Caulis (3) - Iridoids)

  • Six iridoids were isolated from the 70% ethanol extract of Lonicera Caulis (Caprifoliaceae) and their structures were identified as epialyxialactone (1), secologanin dimethyl acetal (2), sweroside (3), loganin (4), loganic acid (5) and demethylsecologanol (6). The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. This is the first report of the iridolactone, epialyxialactone (1), from the Caprifoliaceae plants and loganic acid (5) and demethylsecologanol (6) from Lonicera Caulis.
