Antioxidant activity and improvement effect of Acer tegmentosum Maxim of dietary fatty liver in rat fed on a high-fat diet (벌나무(Acer tegmentosum Maxim) 추출물의 항산화 활성 및 고지방식이를 급이한 흰쥐의 지방간 개선 효과)

  • The effects on the radical scavenging activities and hepatic lipid levels in rats fed a high-fat diet (HFD) in the 70% ethanolic extract from Acer tegmentosum Maxim (ATM) were evaluated. Total phenol content of ATM was 168.60 mg catechin/g in the 70% ethanolic extract of Acer tegmentosum. The DPPH and ABTS radical scavenging activities were 18.32 mM TE/g and 32.25 mM TE/g, respectively. Food efficiency ratio was lower significantly in supplemented group with 150 mg/kg BW/day during 5 weeks (HFD+ATM) compared to HFD. Total cholesterol and triglyceride contents in liver tissue of HFD+ATM were lower significantly compared to those of the HFD. Supplementation of ATM significantly decreased lipid peroxide contents and increased radical scavenging activity in the liver tissue compared with that of HFD group. Moreover, the hepatocytes of HFD rats showed a typical fatty liver morphology showing the presence of cytoplasmic lipid droplets, whereas administration of ATM attenuated the number and the size of lipid droplets. In the liver tissue of ATM administrated HFD group, the mRNA levels of SREBP-1c, $PPAR{\gamma}$, and FAS were decreased. Therefore, these results suggest that Acer tegmentosum extracts could have antioxidant activities and the hypolipidemic effects in liver tissue by its phenolic compounds.

Folic acid supplementation prevents high fructose-induced non-alcoholic fatty liver disease by activating the AMPK and LKB1 signaling pathways

  • BACKGROUND/OBJECTIVES: The present study aimed to evaluate the effects of folic acid supplementation in high-fructose-induced hepatic steatosis and clarify the underlying mechanism of folic acid supplementation. MATERIALS/METHODS: Male SD rats were fed control, 64% high-fructose diet, or 64% high-fructose diet with folic acid for eight weeks. Plasma glutamate-pyruvate transaminase, glutamate-oxaloacetate transaminase, lipid profiles, hepatic lipid content, S-adenosylmethionine (SAM), and S-adenosylhomocysteine (SAH) were measured. RESULTS: The HF diet significantly increased hepatic total lipid and triglyceride (TG) and decreased hepatic SAM, SAH, and SAM:SAH ratio. In rats fed a high fructose diet, folic acid supplementation significantly reduced hepatic TG, increased hepatic SAM, and alleviated hepatic steatosis. Moreover, folic acid supplementation in rats fed high fructose enhanced the levels of phosphorylated AMP-activated protein kinase (AMPK) and liver kinase B (LKB1) and inhibited phosphorylation of acetyl coenzyme A carboxylase (ACC) in the liver. CONCLUSIONS: These results suggest that the protective effect of folic acid supplementation in rats fed high fructose may include the activation of LKB1/AMPK/ACC and increased SAM in the liver, which inhibit hepatic lipogenesis, thus ameliorating hepatic steatosis. The present study may provide evidence for the beneficial effects of folic acid supplementation in the treatment of non-alcoholic fatty liver disease.

  • Dibutyl phthalate (DBP) is used extensively in the plastic industry and has been known as an endocrine disruptor. Present study was undertaken to examine whether DBP can induce oxidative stress in mice. In this study, oxidative stress was measured in terms of the modification of lipid peroxidation and gamma-glutamyl transferase (GGT) activity. The serum toxicity index, level of lipid peroxidation and triglyceride (TG), and activity of GGT were measured in male ICR mice after a single administration of DBP (5 g/kg, po). DBP did not alter serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine, glucose and cholesterol level. However, the treatment with DBP was found to significantly increase the level of lipid peroxidation in liver and lung. The TG content and activity of GGT in the liver of DBP-exposed animals was also increased. These results indicate that DBP can induce mild oxidative stress in mice. The GGT activity is considered to be increased as one of the adaptive defense mechanisms to oxidative stress induced by DBP.

Hepatoprotective Effect of Stamen Extracts of Mesua ferrea L. against Oxidative Stress induced by $CCl_4$ in Liver Slice Culture Model

  • Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.

Inhibitory Effect of Angelica keiskei Koidz Green Juice on the Liver Damage in CCl4-Treated Rats (신선초 녹즙이 사염화탄소 투여에 의한 흰쥐의 간 손상에 미치는 영향)

  • To investigate effects of Angelica keiskei Koidz green juice on the liver damage of CCl4-treated tats, Sprague-Dawley male rats weighing 80~100g were divided into 4 groups of control group(CON), Angelica keiskei Koidz green juice-treated group(ANJ), CCl4-treated group(CCL) and Angelica keiskei Kodiz green juice and CCl4-treated group(ACL). Each group was sacrified after feeding for 4 weeks and examined the activities of transminase (sGOT, sGPT), superoxide dismutase (SOD), catalase and glutathione peroxidase(GSH-Px), and contents of lipid peroxide and glutathione in liver. The activities of sGOT and sGPT, and content of lipid peroxide after CCl4 treatment were markedly increased, compared to CON, but those levels were significantly decreased by the pretreatment of Angelica keiskei Koidz green juice as compared to CCL. The activities of SOD, catalase and GSH-Px were elevated by CCl4-treatment as compared to control group, and concomitant treatment of Angelical keiskei Koidz and CCl4 decreased those levels significantly except the activity of catalase. The hepatic content of glutathione was decreased by CCl4 and increased more abundant by Angelica keiskei Koidz administration than CCl4 treated group. These results suggest that Angelica keiskei Koidz green juice is believed to have a possible protective effect for the carbon tetrachloride-induced hepatotoxicity in rats.

Effect of Several Carbohydrate Sources on Lipid Metabolism in Cholesterol Fed Rats (탄수화물급원의 차이가 흰쥐 체내 지질함량에 미치는 영향)

  • The effects of several carbohydrate sources on plasma, liver and fecal lipid contents of rats fed diet containing sucrose, corn starch, brown rice, milled rice or wheat flour with 0.3% cholesterol for four weeks were investigated. Plasma triglyceride and total cholestrol increased significntly in sucrose group, and decreased significantly in milled rice group compared to other three groups. There were no significant differences in plasma lipid contents among corn starch, brown rice and wheat flour groups. The contents of liver triglyceride and cholesterol in brown rice group decreased significantly compared to there four groups. Fecal excretion of total cholesterol in rice and wheat flour groups was significantly higher than in sucrose and corn starch groups. The contents of fecal bile acid increased significantly in milled rice and brown rice groups compared to other three groups. Fecal bile acid content of wheat flour group was similar to those of sucrose and corn starch group. From these results, it is suggested that rice has a possibility of hypolipidemic effect in cholesterol fed rat.

The Effects of Dietary Shellfish on Lipid Metabolism in Rats Fed High-Fat Supplement (고지방식이를 급여한 흰쥐의 패류식이 급여 효과)

  • This study was conducted to investigate the effects of shellfish on lipid metabolism in rats fed high fat supplements. Male sprague-dawley rats weighting approximately 165g were fed a basal diet, a high fat diet, or a high fat diet plus shellfish for 4 weeks. The shellfishes on the were oyster, hard-shelled mussel, little neck clam, and march clam. Alfter 4 weeks high fat diet, supplementation of 20% lard significantly increased plasma. GOT, GPT, ${\gamma}$-GTP , and liver triglyceride (TG). Plasma GOT, GPT, ${\gamma}$-GTP , triglyceride, and total cholestrerol levels were significantly lower in shellfish groups than in basal and high-fat groups regardless of high-fat supplementation (p<0.05). The total lipid and cholesterol content in liver showed similar results(p<0.05). There were no differences in glucose, HDL-cholesterol in plasma and total cholesterol and total lipid in liver between basal and high-fat supplemented diets. Liong chain fatty acids, specific components of shellfishes group, were exclusively higher than in basal and high-fat diets, and were most well-reflected in liver and plasma. From the above results, the hypolipidemic effects of shellfish were detected in the process of inducing hyperlipidemia by high-fat supplement.

Effects of Dietary Lipid Source and Level on Growth Performance, Blood Parameters and Flesh Quality of Sub-adult Olive Flounder (Paralichthys olivaceus)

  • This study was conducted to investigate the effects of dietary lipid source and level on growth performance, blood parameters, fatty acid composition and flesh quality of sub-adult olive flounder Paralichthys olivaceus. Eight experimental diets were formulated to contain 5% squid liver oil (SLO), 5% linseed oil (LO), 5% soybean oil (SO), a mixture of 1% squid liver oil, 2% linseed oil and 2% soybean oil (MIX), no lipid supplementation with high protein level (LL-HP), 10% squid liver oil (HL-SLO), a mixture of 1% squid liver oil, 4.5% linseed oil and 4.5% soybean oil (HL-VO), and 1% squid liver oil with high starch level (LL-HC), respectively. Two replicate groups of fish (average initial weight of 296 g) were fed the diets for 17 wks. After 5 wks, 11 wks and the end of the feeding trial, five fish from each tank were randomly sampled for analysis of body composition. At the end of the feeding trial, final mean weight of fish fed the LL-HP diet was significantly (p<0.05) higher than that of fish fed the HL-VO diet, but did not differ significantly from those of fish fed the SLO, LO, SO, MIX, HL-SLO and LL-HC diets. Fish fed the LL-HP diet showed significantly higher feed efficiency than fish fed the LO, HL-SLO and HL-VO diets. Feed efficiency of fish fed the LO, SO and MIX diets were similar to those of fish fed the SLO and HL-SLO diets. Fish fed the HL-SLO diet showed significantly higher total cholesterol content in plasma compared with other diets. Fatty acid composition of tissues was reflected by dietary fatty acid composition. The highest linoleic (LA) and linolenic acid (LNA) contents in the dorsal muscle were observed in fish fed the SO and LO diets, respectively, regardless of feeding period. The highest eicosapentaenoic acid (EPA) content in the dorsal muscle was observed in fish fed the LL-HP and LL-HC diets after 11 and 17 weeks of feeding, respectively. Fish fed the SLO and HL-SLO diets showed higher docosahexaenoic acid (DHA) content than that of other treatments after 11 and 17 weeks of feeding, respectively. Dietary inclusion of vegetable oils reduced n-3 HUFA contents in the dorsal muscle and liver of fish. The n-3 HUFA contents in tissues of fish fed the SLO and HL-SLO diets were higher than those of fish fed other diets, except for the LL-HP and LL-HC diets. Hardness, gel strength, chewiness and cohesiveness values of dorsal muscle in fish were significantly affected by dietary lipid source. The results of this study indicate that fish oil in fish meal based diets for sub-adult olive flounder could be replaced by soybean oil and linseed oil without negative effects on growth and feed utilization.

The Effects of Juglandis Semen Herbal Acupuncture on t-Butylhydroperoxide-induced Liver Damage (호도약침액(胡桃藥浸液)이 t-Butylhydroperoxide에 의한 간손상(肝損傷)에 미치는 영향(影響))

  • Objectives : This study was undertaken to examine whether Juglandis Semen herbal acupuncture (JGA) exerts protective effect against oxidant-induced cell injury in rabbit liver. Methods : The cell damage was estimated by measuring lactate dehydrogenase (LDH) release, and lipid peroxidation was estimated by measuring malondialdehyde (MDA) in rabbit liver slices. Results : t-Butylhydroperoxide (tBHP) caused an increase in LDH release and lipid peroxidation in a dose-dependent manner over concentrations of 0.5-2 mM, which were prevented by addition of 0.05% JGA. The protective effect of JGA was dose-dependent in concentration range of 0.005 to 0.1%. The concentrations of 0.005 and 0.1% JGA completely prevented the LDH release and lipid peroxidation by 1 mM tBHP. When liver tissues were exposed to 1 mM tBHP, alanine aminotransferase (ALT) activity in the medium was significantly increased, which was prevented by 0.05% JGA. tBHP (2 mM) decreased GSH content and the effect was prevented by 0.05% JGA. Conclusion : These results suggest that JGA exerts protective effect against oxidant-induced cell injury by antioxidant action resulting from enhancement of GSH content in the liver.

Effects of Alcohol Intoxication and Moldy Rice Consumption on the Liver Lipid Content in Rat (Alcohol과 쌀 곰팡이의 Toxicity가 간(肝)의 지방(脂肪) 축적(蓄積)에 미치는 영향(影響))

  • For the elucidation of the metabolic effects due to alcoholic drink and moldy rice intake author designed two experiments as follows. In the first experiment, sixty male and female rats, divided into six groups, 10 rats each in both sexes, were given 7.5% sucrose-30% alcohol solution (Jinro-Soju) and diets varied of fat content (2%, 7%, 30% of diet) and protein quality (casein, bean, anchovy) for 6 weeks. In the Second experiment, six groups of rats, 10 male rats each, were fed the diets containing zero, 5%, 10%, 25%, 50%, and 100% moldy rice contaminated Penicillium Islandicum and the experimental periods were 4 weeks and 6 weeks. The results of these studies due to the toxicity of alcohol consumption and moldy rice ingestion of the fatty liver production and the other effects are as follows. 1. Food intake of alcohol consumed groups decreased to 50%-70% of that of standard group. In the second study, there was no significant differences on food intake due to the different contents of moldy rice and experimental period. 2. On the view paint of body weight gain, the body weight of alcohol consumed groups gained much less than that of standard group, because food consumption was decreased due to alcohol ingestion. 3. In comparison of F.E.R. and P.E.R. between standard and experimental groups, there was no significant differences in this study. 4. As a matter of fact, there was no significance on the nitrogen balance in both studies. 5. From the result of hematology, R.B.C. counts, W.B.C. counts, hemoglobin, and hematocrit showed normal values in all groups including in this study. So we might conclude that the toxicity of alcohol and moldy rice do not effect significantly on blood picture. 6. The larger organs shrank on the range of 20%-70% of that against standard group in this study. The major reason for the shrinkage of organs might be account of decreased food consumption due to alcoholic drink. There was no great changes on the organ weight due to the ingestion of moldy rice. 7. The nitrogen content in various organs in both experiments was revealed at the normal level for the worst condition in terms of the least food intake. In other word, it was noteworthy that the concentration of nitrogen in various organs was kept at the normal ratio as standard groups under the circumstances of this study. 8. The lipid content in the liver of rats fed alcohol and diets containing either various lipid contents or protein quality did not increase. Hepatic lipids accumulation due to the dietary fat content was observed, but there wvas no significances among the compared groups. In the second experiment, the difference of hepatic lipid content between the moldy rice groups and standard group was not showed. In addition to the result of total lipid, hepatic free cholesterol, free fatty acid, and triglyceride did not change in both studies, we might conclude that the toxicity of alcohol and moldy rice could not effect on the hepatic lipid contents. 9. There was no significant differences on the serum glucose level between alcohol groups and standard group. In the second experiment, serum glucose level increase in 6th week compared with in 4th week, but there was no significant differences.
