Isolation and partial haracterization of rat LDH A-genomic sequences

  • As a part of the study to elucidate the mechanism by which transcription of LDH A-gene is regulated by cAMP, we aimed to isolated rat LDH A gene and characterize cAMP-reponsive element (CRE). We have screened $1.2{\times}10^6$ recombinant phages of rat Charon 4A genomic library and isolated 33 positive clones among which we identified 12 different LDH A gene-related clones. By the results of restriction enzyme mapping, Southern blotting, and nucleotide sequence analyses, we concluded that the 12 LDH A gene-related clones were intronless and frequently mutated LDH A-pseudogenes. In this report, we present the characteristic features of the 12 rat liver LDH A-pseudogenes.

Inhibition of C-terminal O-Methyltransferase by a Rat Liver Cytosolic Peptide

  • The activity of SD-framesylcysteine O-methyltransferase was assayed by incubating the enzyrne with a synthetic in vitro substrate, [N-acetyl-S-trans, trns-famesyl-L-cysteine (AFC)], together with S-adenosyl-L-[emthyl-$_{14}$C)ester(AFCME)], was then analyzed either directly on HPLC or by converting the AFC[$methyl^{14}C$]ME to [$methyl^{14}C$] aclcohol by basehydrolysis. Employing these two analytical methods, it was established that a peptide purifed from rat liver cytosol fraction [Int. J. Biochem., 25, 1157 919930] strongly inhibited the above enzyme activity with $IC_{50}\; of\; 7.1\times 10^{-8}$ M. Also, the S-famesylcysteine O-methyltransferase from several human colon cancer cells was equally inhibited by the peptide.

Inhibitory effect of honokiol and magnolol on cytochrome P450 enzyme activities in human liver microsomes

  • Honokiol and magnolol, the major bioactive neolignans of magnolia officinalis, are the most important constituents of the crude drug prescriptions that are used in the therapy of neuroses and various nervous disorders. There have been limited reports on the effects of neolignoid compounds on human cytochrome P450 activity. Therefore, the inhibitory effects of honokiol and magnolol on seven human cytochrome P450 s were evaluated in human liver microsomes. Honokiol and magnolol showed the most potent inhibition of CYP1A2-mediated phenacetin O-deethylase activity ($IC_{50}$ values of 3.5 and 5.4 mM, respectively) among the seven P450s tested. These in vitro data indicate that neolignan compounds can inhibit the activity of CYP1A2 and suggest that these compounds should be examined for potential pharmacokinetic drug interactions in vivo.

고들빼기 첨가 식이가 알콜투여 흰쥐의 지방대사와 간기능에 미치는 영향 (Effects of Ixeris Sonchifolia H. Fiet on Lipid Metabolism and Liver Function of Rats Administered with Ethanol)

  • To investigate the effects of Ixeris sonchifolia Hance diets on serum and hepatic lipid levles and enzyme activities in rats administered with ethanol chronically, Sprague-Dawley male rats were AIN-76 diet(control), control diet plus ethanol, control plus Ixeris sonchifolia Hance diet, or control plus Ixeris sonchifolia Hance diet plus ethanol for 30 days. Ixeris sonchifolia Hance diets significantly decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and GOT levels that were increased due to the chronic ethanol administration. In addition, Ixeris sonchifolia Hance diets significantly decreased the liver triglyceride and total lipid levels that were increased due to the ethanol administration. The present findings, combined with previous data showing differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the serum Y-GPT activity of rats(Cha and OH[2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505) raise the possibility that GABA in plants could have a nytraceutical role in the recovery of chronic alcohol-related diseases. 저산소/재관류로부터 청폐사간탕의 PC12 세포 보호 효과 (Protective Effect of Metabolized Chungpesagan-tang on Hypoxia/Reperfusion Induced-PC12 Cell Damage)

  • This research was performed to investigate the protective effect of Chungpesagan-tang (CST) from hypoxia/reperfusion induced-PC12 cell damage. To elucidate the mechanism of the protective effect of CST, cell viability, changes in activities of superoxide dismutase, glutathione peroxidase, catalase, caspase 3 and the production of malondialdehyde were observed after treating PC12 cells with CST which was metabolized by rat liver homogenate. Pretreatment of CST with liver homogenate appeared to increase its protective effect against hypoxia/reperfusion insult. The result showed that CST exhibited the highest protective effect against hypoxia/reperfusion at the dose of $1\;{\mu}g/ml$ in PC12 cells, probably by recovering the redox enzyme activities and MDA to control level.

Isolation of Human CYP4F2 genomic DNA and its $5^I$ End Regulatory Region Structure

  • Human cytochrome P450 4F2 shows high regioselectivity in hydroxylation of stearic acid and leukotriene $ B_4.$ As a first step of its regulation study, human cytochrome P450 4F2 genomic DNA was isolated from liver of a person who was administered clofibrate for 10 years. From Southern hybridization, restriction enzyme digestion and sequencing experiments, isolated genomic DNA fragment was found to contain around 32 Kb DNA and more than 20 Kb of $5^I$ end regulatory region. Sequences of the structural gene region revealed exon 1 and exon 2. Further regulation studies would elucidate the feedback mechanisms of the oxidative degradation of fatty acids, inflammatory response and the clearance of leukotriene B4 in the liver. Furthermore, regulation study of this gene could explain the species difference in responses to peroxisome proliferator and help in the safety evaluation of peroxisome proliferating chemicals to human being.

개에 있어서 면역매개성 용혈성빈혈 일례 (Immune Mediated Hemolytic Anemia in a Dog)

  • A 3-year-old, 4.0 kg, intact male shih-tzu dog with anorexia, depression, pale mucous membranes, tachypnea, tachycardia was referred to the Veterinary Medical Teaching Hospital. Autoagglutination was observed by naked eye when blood was collected in an EDTA-tube and many spherocytes were found on a Diff-Quik stained blood smear. PCV was 6% and indirect bilirubin was increased markedly. So the immune-mediated hemolytic anemia was diagnosed. Autoagglutination was too severe to perform cross-matching test. Blood was not transfused as it might accelerate or precipitate hemolytic crisis, and regeneration of erythrocytes was very good. Thus corticosteroid of immunosuppressive dose and fluid were administered and PCV was monitored. Although blood was not transfused, PCV increased from 6 to 15.9% in a day and to 30% 7 days later. Therapy for liver was concurrently conducted because liver enzyme activities were high. Corticosteroid tappering therapy was conducted for 75 days and PCV was recovered to 46% after 4 months form start of the treatment.

부자를 포함한 한약처방이 간효소치에 미치는 영향 -임상 6예를 중심으로 (The Pilot Study of the Hepatotoxicity of Aconiti Radix Lateralis Preparata Contained Herbal Medicine in 6 Cases)

  • It is considered that herbal medicine may also have hepatotoxicity and Aconiti Radix Lateralis Preparata is thought to be the most poisonous. We investigated the changes of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and ${\gamma}-glutamyltranspeptidase{\;}({\gamma}-GT$)$ in 6 cases administrated herbal medicine containing Aconiti Radix Lateralis Preparata and tried to evaluate its influence on liver enzymes. Before administration, the mean values of AST and ALT were $30.5{\pm}12.4${\;}U/L{\;}and{\;}30.7{\pm}22.5{\;}U/L$. After administration, those of AST, ALT and ${\gamma}-GT{\;}were{\;}25.7{\pm}8.7 {\;}U/L,{\;}28{\pm}14.8{\;}U/L{\;}and{\;}34.3{\pm}19.4{\;}U/L$ respectively. Therefore, it is indirectly assured that herbal medicine containing Aconiti Radix Lateralis Preparata didn't influence liver enzymes such as AST, ALT, ${\gamma}-GT$.

Effect of Scutellariae Radix Extract on Human CYP450 Mediated-Drug Metabolism

  • Scutellariae Radix is widely used in the traditional herbal medicine for the treatment of fever, cough, dysentery, hepatitis and hypertension in Korea, China and Japan. In this study, we investigated the effects of 70% ethanolic extract of Scutellariae Radix (SRE) on CYP450-mediated drug metabolism in the in vitro systems using human liver microsomes and hepatocytes. The microsomal incubation assay showed that SRE inhibited the drug metabolism reactions catalyzed by CYP1A2, CYP2C8 and CYP2C9 in a dose-dependent manner. In particular, SRE was shown to strongly inhibit the metabolic activity of CYP1A2 with an $IC_{50}$ value of 4.6 ${\mu}g/mL$. When SRE was evaluated for its effect on the induction of CYP450 enzyme activities in cryopreserved human hepatocytes, SRE did not exhibit any effect.

어류 Metallothionein에 대한 특이적 면역 효소 측정법의 확립에 관한 연구 (The Study on the Establishment of Specific ELISA for the Detection of Fish Metallothionein)

  • The purpose of this experiment is to estabilish the sensitive and specific ELISA (enzyme linked immunosorbent assay) system for the detection of fish metallothionein (MT). Silver carp were injected with CA of 1-8mg/kg body wt. 4 times during 10 days. Silver carp was very tolerant species to CA. Cd induced MT in liver was seperated and purified by gel filtration chromatography and ion exchange chromatography and identified by spectrophotometry, native gel electrophoresis and western blot analysis. The rabbit antiserum was produced by immunizing rabbit with lyophilized MT, and the competitive ELISA system was estabilished for the detection of fish MT. In the present ELISA system, the detection limit was about 33 ng/ml. When this ELISA system was employed to determine the MT level in the supernatant sample of fish liver homogenate, the reaction curve showed a good parallel corelationship with the calibration curve over a certain dilution range. The results indicate that the competitive ELISA can be a useful tool for the detection of fish MT in the toxicological study and the evaluation of water pollution.
