The Experimental Study on Biochemical Changes of Rat Liver Following Single Irradiation of High Energy X-ray (고(高)에너지 방사선(放射線)을 1회조사(一回照射)한 흰쥐 간(肝)의 생화학적(生化學的) 변화(變化))

  • In order to investigate radiation effects on the liver, functional changes of liver were analyzed after irradiation. Doses of 10 Gy, 15 Gy and 20 Gy were exposed partially to the liver of male rats(Sprague-Dawley) with X-ray(4MV linear accelerator) at room temperature. On 1, 2, 4 and 8 weeks after irradiation, liver tissues and sera of the animals were compared with those of unirradiated animal by liver function tests. Enzyme activities in sera such as alanine aminotransferase, aspartate aminotransferase, malondialdehyde. The content of malondialdehyde in the activities of many enzymes including alanine aminotransferase, aspartate aminotransferase in sera were increased slightly with increasing exposure dose in all experiments and the activities of these enzymes increased markedly in 20 Gy irradiated groups. From these above results, functional changes of the liver were induced in all irradiated groups. Damaged liver was recovered along with time collapse after irradiation to the doses of 10 Gy and 15 Gy while no recovery was deteced within 8 weeks after irradiation to 20 Gy. These results suggest that careful attention must be paid to liver not to be included in exposure field in radiation therapy.

Effect of Green Tea on Gene Expression of Superoxide Dismutase and Glutathione Peroxidase in Rat Liver Exposed to Microwaves (녹차가 전자파 조사 흰쥐 간조직의 Superoxide Dismutase 및 Glutathione Peroxidase 유전자 발현에 미치는 영향)

  • The purpose of this study was to investigate the effects of green tea on gene expression of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in rat liver exposed to microwave. Sprague-Dawley male rats with 200$\pm$10g body weight were assigned to normal and microwave exposed groups : microwave exposed groups ; microwave exposed groups were divided two groups : microwave(MW) group which was administrated the distilled water and green tea(GT) group which was administrated the green tea extracts. The rats were irradiated with microwave at frequence of 2.45 GHz for 15 min and then the gene expression in the damaged tissue were investigated at 0.1, 3, 4,6 and 8 days after the microwave irradition to compared with the normal group. The level of SOD gene expression in MW group was lower than the normal group within 6 days but that of GT group as higher than MW group. These results may imply that green tea stimulates SOD expression and there by protecting tissues from free radicals. The GSH-Px gene was expressed a little bit lower than the normal group but that of GT group was expressed to higher lever than MW group from 4 days after irradiation. These results suggest that the administration of green tea extract may activate antioxidative gene expressions such as SOD and GSH-Px in rat and that may help to recover liver tissues from microwave damage by removing hazardous free radicals and oxidized by products from cells.

Effects of the Low Power He-Ne IR Laser Treatment on the Liver Damage Induced with $CCI_4$ in Rats

  • This study was investigated the effects of the lower power He-Ne IR laser treatment on the changes of blood biochemical components in the rat liver damaged by the carbon tetrachloride (CCl₄). The twenty one Sprague-Dawley adult male rats weights (260±18.6 g) were designed to the three groups: one control group and two experimental groups, the experimental groups were divided into the CCl₄-treated groups and the laser therapy group (CCl₄+ Laser). The experimental groups were injected twice with CCl₄(1.0 ml/kg body weight) intraperitoneal for two days. Each group was sacrificed after two weeks irradiated with the lower power He-Ne IR laser for ten minutes per every day. The activity of alanine aminotransferase (ALT), lactic dehydrogenase (LDH), alkaline phosphatase (ALP) and the concentration of serum glucose treated with He-Ne IR laser groups was significantly decreased to the conrtol (treated with carbon CCl₄) group. The activity of aspartate aminotransferase (AST) was decreased in the laser group but not significantly, the concentration of the serum cholesterol in the laser group was significantly increased comparing with the control and case control groups. In conclusion, the effect of the lower power He-Ne IR laser treatment is believed to be a possible protective effects for CCl₄ induced acute hepatotoxicity in rats.

Inhibitory Effect of Leek Greek Juice on $CCl_{4}$-induced Hepatotoxicity in Rats (부추녹즙이 사염화탄소 투여에 의한 흰쥐의 간손상에 미치는 영향)

  • To investigate the effects of leek green juice on the damaged liver of $CCl_{4}$-treated rats, Sprague-Dawley male rats weighing about 100g, were divided into 4 groups ; control group (CON), leek green juice-administered group (LGJ), $CCl_{4}$-treated group (CCL) and leek green juice and $CCl_{4}$-treated group (LCL). After 6 weeks, the activities of sGPT and sGOT, and content of hepatic TBA-reactants, elevated by $CCl_{4}$ treatment, were markedly decreased by administering leek green juice, compared to CCL. It was also observed that activities of hepatic SOD and GSH-Px were elevated by $CCl_{4}$-treatment as compared to CON, but concomitant treatment of leek green juice and $CCl_{4}$ decreased those levels adjacent to CON, whereas catalase activity did not show significant decreasing effects compared to CCL. The hepatic content of glutathione, decreased by $CCl_{4}$, was more abundantly increased by leek green juice administration than by CCL. These results suggest that leek green juice is believed to b a possible protective effect for the carbon tetrachloride-induced hepatotoxicity in rat liver.

Protective Effect of Semisulcospira libertina Extract on Induced Hepatitis in Rats (다슬기 열수 추출물이 간독성이 유도된 흰쥐에 미치는 보호 효과)

  • This study aimed to investigate the protective effect of Semisulcospira libertina extract on liver injury induced by D-galactosamine in rats. After the administration of S. libertina extract, the local fat degeneration and infiltration of inflammatory cells in liver tissues were significantly decreased and peripheral hemorrhages around portal triads and central necrosis around central veins were found to be protective. The elevated levels of plasma ALT, AST, and LDH, the ALP activation lipid peroxidation, and the lipid contents of a damaged liver were recovered in experimental rats administrated with S. libertina extract, suggesting its role in blood enzyme activation and lipid content restoration within damaged rat liver tissues. Moreover, the expression rate of TNF-α, which accelerates inflammation and induces tissue damage and necrosis, was significantly decreased. In addition, the activities of antioxidant enzymes were more effectively upregulated compared to those of the control group induced hepatotoxicity. All data showed that S. libertina extract has a preventive role against liver damages, such as inflammation and tissue necrosis, as instigated with D-galactosamine by improving the activities of blood enzymes and antioxidant enzymes and modulating the expression of inflammation factor, suggest S. libertina extract is an effective medicinal resource for the restoration of hepatotoxicity.

Effects of Hanbag Mushroom(Grifola frondosa) on Oxidative Stress in Diabetic Rats (당뇨유발 흰쥐에 있어서 산화적 스트레스에 대한 함박잎새버섯의 효과)

  • This research was carried out to investigate the effects of Hambag mushroom on the oxidative stress in diabetic rats, Sprague-Dawley. The diabetic rats induced by streptozotocin were fed with hambag mushroom-powder(G. frondosa) for 6 weeks. For the level of oxidative stress in liver and pancreas tissues, it was studied by measuring LPO (lipid oxide) level as an indicator of lipid peroxidation, XOD(xanthine oxidase) as one of important sources for free radicals and the levels of GSH and GST as anti-oxidant systems. Also, as an indicator of liver damaged by oxidative stress, the activities of serum ALT and AST were measured. It was observed that the levels of ALT, AST, LPO and XOD were higher by about two times in both tissues from diabetic rats than in those from control rats. This indicates that the oxidative stress induced by diabetes caused the tissues damages. However, these levels were decreased in the tissues from rats with hambag mushroom-powder. Futhermore, the activity of GST were higher in both tissues from diabetic rats fed with hambag mushroom-powder than in those from diabetic rats. Thus, it is considered that the hambag mushroom-powder decreases the level of oxidative stress by increasing activity of anti-oxidant system such as GSH and GST. It is suggested that the hambag mushroom-powder can be useful for preventing the tissues damaged by diabetes-induced oxidative stress.

Effects of Herbal-Acupuncture with Inzinsammultang on the Recovery of Liver Injury in Rats (인진삼물탕(茵蔯三物湯) 약침처리(藥針處理)가 백서(白鼠)의 간손상(肝損傷) 회부(回復)에 미치는 영향(影響))

  • These studies were carried out to investigate the effect of Herbal-acupuncture with Inzinsammultang extract on the recovery from liver injury of rats. The liver injury of rats induced with 0.3ml/ea carbon tetrachloride. The Herbal-acupunture with Inzinsam multang extract solution inserted into corresponding focus of Kansu(BL18) in rats. In this study, SD-Rats(Sprague-Dawley) were divided into 4 groups ; Normal group (None treated group), Control-group(The group not treated after $CCI_4$-intoxication), Treated group; Example I-group(Saline-injected groups after $CCl_4$-intoxication) and Example II-group(Inzinsammultang-injected groups after $CCl_4$-intoxication). Through histological observation, Example II-group sho-ws that liver injury is weaker than Control group.(p<0.05) Biochemical assays for each serum enzyme activities of AST, ALT and LDH, levels of albumin, ${\gamma}$-GT, TG, total cholesterol were performed. The results were summarized as follows 1. AST(Aspartate aminotransferase) activity in serum significantly decreased in the Inzinsammultang-injected groups after $CCl_4$-intoxication. Inzinsammultang-injected groups after $CCl_4$-intoxication showed significantly higher AST activities, compared with the Saline-injected groups after $CCl_4$-intoxication.(p<0.05) 2. ALT(Alanine aminotransferase) activity in serum significantly decreased in the Inzinsammultang-injected groups after $CCl_4$-intoxication. The Inzinsammultang-injected groups after $CCl_4$-intoxication showed significantly higher AST activities, compared with the Saline-injected groups after $CCl_4$-intoxication(p<0.05) 3. Level of Albumin in serum significantly increased in the Inzinsarnmultang-injected groups after $CCl_4$-intoxication. 4. LDH(Lactate dehydrogenase) activities in serum significantly decreased in the Inzinsammultang-injected groups after $CCl_4$-intoxication.(p<0.05) 5. ${\gamma}$-GT(Glutamyl transferase) in serum significantly decreased in the Inzinsammultang-injected groups after $CCl_4$-intoxication(p<0.05) 6. As for the TG(Triglyceride) levels in serum, Inzinsammultang-injected groups after $CCl_4$-intoxication is no significant differences compared with the Saline-injected groups after $CCl_4$-intoxication.(p<0.05) 7. Total cholesterol in serum significantly increased in the Inzinsarnmultang-injected groups after $CCl_4$-intoxication.(p<0.05) Those results indicate that the Inzinsammultang Herbal-acupuncture have significant effects on the liver injury induced by $CCl_4$. So it is expected that the Inzinsammultang Herbal-acupuncture can be used to cure inflammations and recover the functions of damaged liver cells.

Radioprotective Effects of Blueberry on the Liver of Radiation Irradiated Rats (방사선에 조사된 쥐 간에서 블루베리의 방사선방호효과)

  • Radiation were seriously damaged on liver functions. Blueberry was fruits that contains Vit A, Vit c, Vit E, follic acid, ${\beta}$-carotene, and anthocyanin. The purpose of this study was to evaluate the protection effects of blueberry the liver functions. Irradiation dose was used to 4 Gy (Linac 6 Mev) X-ray Treatment device Experiment animals was used to 7 rats in each groups. It was investigated liver functions that contains TP, ALB, GLOB, ALT, ALKP and CHOL. We showed that Blueberry was not recovery effects on radiation-induced liver functions. But, Statistically significant value was showed ALB (p>0.01) and ALT (p>0.1). It was concluded that blueberry was not used to recovery materials on radiation-induced liver functions.

Protective Effects of Some Plant Extracts on Lipids Contents of Rats Treated with Carbon Tetrachloride (사염화탄소를 투여한 흰쥐의 지질농도에 미치는 식물추출물의 보호효과)

  • This study was performed to investigate the possible effects of some plants protecting intact rat liver damaged by $CCl_4$. The extract of mugwort (Artemsiae capillaris), soybean sprout and pine leaf (Pinus strobus) inhibited markedly the in vitro activities of rat liver fatty acid synthase, whereas those of shiitake (Lentinus ododes), Houttuynia cortata, Acanthopanacis cortex and buckwheat leaves had less effects. Treatment with the water extract of pine leaf and soybean sprout caused a marked decrease in the $CCl_4$-induced toxicity in rat liver, judged from their effects on the levels of glutamic oxaloacetic transaminase (GOT) and glutamate pyruvic transaminase(GPT) in the serum. The extract of mugwort and soybean sprout reduced markedly the content of liver microsomal peroxides induced by $CCl_4$ treatment and serum TBA values, respectively. The extract of soybean sprout decreased efficiently the content of liver triglyceride elevated by $CCl_4$ treatment. Nevertheless, the extracts did not exert the supression of hepaticmegaly induced by $CCl_4$. The results suggest that soybean sprout and pine leaf may be potential sources improved the biochemical parameters like as peroxidation value or serum GOT and GPT, although these extracts had minimal effects in the increase of liver size induced by carbon tetrachloride.

Effect of Water Soluble Extract of Lichens on Weights of Various Organs, Blood Components and Activities of Transaminases and 5-Nucleotidase in Rat (지의류(地衣類)의 수용성 추출물이 흰쥐의 각 장기무게, 혈액성분 및 Transaminases와 5-Nucleotidase 활성에 미치는 영향(I))

  • In order to evaluate the effect of water soluble extract of lichens (Physcia, Parmelia and Clandonia species) on liver damage, activities transaminase(GPT) and 5'-nucleotidase in serum and liver were measured in rats fed lichens extract. DNA and RNA were measured in liver and spleen, as well as various organ weights and blood components. Control group was fed water to compare with the lichen group. Three sets of experiments were conducted: the first set was done with normal rats, the second one with rats with liver damage induced by $CCl_4$ injection was divided into three subgroups. i.e. no treatment group, pre-treatment group and post-treatment group, and the third one was with rats with acute and chronic liver damage. In normal rats, lichens extract feeding reduced serum GOT and GPT activities. In liver damaged rats, both pre-and post-treatment had suppressing effect against increase of serum enzymes. In rats with acute and chronic liver damage, lichens fed group had lower activities of serum GOT, GPT and 5'-nucleotidase but higher activities of liver enzymes than control group. This effect was more pronounced in rats with acute liver damage. Liver weight increased considerably with lichens intake. Hemoglobin and hematocrit levels were also higher in lichens fed group. Nucleic acid contents in spleen but not in liver were increased in lichens fed group. The latter increase was more significant with chronic liver damage. It is suggested from the present study that water soluble lichens extract play protective and therapeutic roles in organs against infection and atrophic disease.
