Effect of Cyclohexane Treatment on the Liver Damage in CCl4-Pretreated Rats (CCl4전처치한 흰쥐에 Cyclohexane 투여가 간손상에 미치는 영향)

  • TO evaluate an effect of cyclohexane treatment on the degree of liver damage, rats were induced liver damage with 10 or 17 times $CCl_4$ injection (0.1 m1/100 g body wt., 50% $CCl_4$ dis-solved in olive oil) at intervals of every other day. Cyclohexane (1.56 g/kg body wt., i.p.) was administrated to the animals at 48 hours after the last pretreatment of $CCl_4$ . Rats were sacrificed at 4 hours after injection of cyclohexane. On the basis of histopathological findings, liver weight/body weight (LW/ BW, %), activities of serum alanine aminotransferase (ALT), xanthine oxidase (XO) and akaline phosphatase (ALP), and contents of liver protein and manlondialdehyde (MDA), $CCl_4$ -pretreatment induced liver damage. And $CCl_4$ 17 times treated group showed more severe liver damage than $CCl_4$ 10 times treated group. Administration of one dose of cyclohexane to $CCl_4$ 10 times treated animals resulted in the enhanced liver damage; liver necrosis with proliferation of fibroblast and bile duct abnormality, and increase in hepatic MDA content and the activities of serum ALP and ALT, But the enhanced liver damage was not found in $CCl_4$ 17 times treated animals. Serum cyclohexanone concentrations at 4 or 8 hours after injection of cyclohexane were higher in all liver damaged groups than normal group and were somewhat higher In $CCl_4$ 17 times treated animals than $CCl_4$ 10 times treated ones. Among the oxygen free radical metabolizing enzymes, hepatic cytochrome P45O dependent aniline hydroxylase (CYPdAH) activity in cyclohexane metabolizing enzyme system was meaningfully increased by the injection of cyclohexane to the liver damaged rats, with increased Vmax and high affinity to aniline. LW/BW (%) and activities of serum XO and ALT were more significantly increased in liver damaged groups than normal group by administration of cyclohexanone. In conclusion, it is assumed that an enhancement of liver damage by injection of one dose of cyclohexane to liver damaged animals might be caused by oxygen free radicals and cyclohexanone.

A Biopharmaceutical Study on Ampicillin in Pathological Animals (병태 동물에서 암피실린의 생물약제학적 연구)

  • It was to investigate the absorption, excretion, protein binding of ampicillin in the pathological animals pretreated with carbon tetrachloride and mercuric chloride. The absorption of ampicillin was not affected in rats with damaged liver and kidney as compared with that of normal rats. The blood level of ampicillin after oral administration was increased significantly in rabbits with damaged kidney and liver. The blood level of ampicillin in rabbit with damaged kidney was more increased than that in rabbits with damaged liver. In severly damaged rabbits, it was more increased than that of mildly damaged rabbits. Urinary excretion of ampicillin in pathological animals was more inhibited than that of ampicillin in normals. Hepatic excretion of ampicillin was accelerated in rabbits with damaged kidney. However, in rabbits with damaged liver, it was inhibited as compared with that in normals. Protein binding of ampicillin was slightly enhanced by the various concentration of carbon tetrachloride and mercuric chloride, respectively. The results suggest that the increase of blood level of ampicillin in pathological animals was due to the inhibition of renal excretion.

  • Objectives : This study was designed to investigate effects of Sayeoksanhap-Pyeongweisan-Gambang (SPG) on gene expression in rats damaged by CCl4 Methods : We investigated the effects of SPG on gene expression in terms of microarray methods in rat liver which were obtained from rats damaged by CCl4. Results : Decreased gene expressions, which were induced by single injection of CCl4, were restored to those in normal rats by administration of SPG or herbal acupuncture. In acupuncture group, gene expressions were restored by 80% of those in control group. In oral administration group and combination group, gene expressions were restored above 90% of those in cuntrol group. Conclusion : These results suggest that oral administraion of SPG was useful to protect liver against CCl4 by its restoration of gene expressions in liver resected from rat damaged by CCl4.

  • To explore the effect of monosodium-L-glutamate(MSG) on CCI$_{4}$-damaged liver in Wister male rat, 5% MSG solution as drink water were administered after S.C. injection of 0.1 mg/kg CC1$_{4}$ twice a week for 4 weeks. After last administration of MSG, heptic glutathione(GSH) dependent system was assayed. It showed that MSG increased significanly hepatic glutathione(GSH) and glutathione peroxidase(GSH$_{px}$), but decreased glutathione-S-transferase(GST) acivity in normal rats. MSG increased significantly the GSH$_{px}$ and GST activities in rats with CCI$_{4}$-induced liver damage. These results indicate that decrease of GSH dependent systems in CC1$_{4}$ liver injury might be partially elevated by coadministration of MSG.

Effect of Cyclohexane Treatment on Serum Level of Glutathione S-Transferase Activity in Liver Damaged Rats ($CCl_4$ 에 의한 간손상 모델 실험동물에 있어서 cyclohexane 투여가 혈청 glutathione S-transferase 활성에 미치는 영향)

  • To evaluate the effect of cyclohexane(CH) treatment on the serum levels of glutathion S-transferase(GST) activity in liver damaged animals, damaged liver was induced with pretreatment of 50% $CCl_4$ dissolved in olive oil (0.1 m1/100g body weight) intraperitoneally 17 times every other day. To $CCl_4$-treated rats, CH (1.56 g/kg body weight, i.p) was injected once and then the animals were sacrificed at 4 hours after injection of CH. The $CCl_4$-treated animals were identified as severe liver damage on the basis of liver functional findings, 1,e, increased serum levels of alanine aminotransferase(ALT), alkaline phosphate(ALP) and xanthine oxidase(XO) activities. On the other hand, $CCl_4$-treated animals injected with CH once($CCl_4$-pretreated animals) showed more decreased serum levels of ALT and XO, and more increased those of ALP rather than $CCl_4$-treated animals. In case of comparing the GST with ALT activity in liver, both $CCl_4$-treated and pretreated animals showed similar changing pattern of enzyme actvity. Especially $CCl_4$-pretreated animals showed significantly increased serum level of GST actvity compared with the $CCl_4$-treated those, whereas those of ALT showed reversed tendency. In aspects of GST enzyme kinetics, $CCl_4$-pretreated animals showed higher Vmax of liver GST enzyme than $CCl_4$-treated animals. In conclusion, injection of CH to the liver damaged rats led to enhanced liver damage and more increased activity of serum GST which may be chiefly caused by the enzyme induction.

Effects of Houttuynia cordata Thunb on Lipidperoxide and Cholesterol in 2,3,7,8-TCDD-damaged Rats (2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD)로 처치된 흰쥐에서 어성초가 과산화지질 및 콜레스테롤에 미치는 영향)

  • TCDD, one of the notorious toxic environmental pollutants, damages various organs including liver and is regarded as an endocrine disrupter. To investigate the effects of Houttuynia cordata Thunb (HCT) on the biochemical parameters of function, liver and serum of TCDD-treated rats were used. Seven days after the injection of TCDD (1 $\mu\textrm{g}$/kg), HCT (200 mg/kg) was administered to rats on every other day for four weeks. The lipidperoxide activity was examined by measuring the level of total cholesterol, HDL-cholesterol, LDL-cholesterol, total lipid and triglyceride (TG) in serum, and malondialdehyde (MDA) in liver tissue of rats. Result showed that lipidperoxidation was inhibited In the significant level when 2,3,7,8-TCDD-damaged rats were treated with HCT.

Effects of Epimedium Koreanum Extract on the Carbon Tetrachloride-Induced Liver Damage and Its Related Organ Damages in Rats (음양곽 추출물이 사염화탄소 투여로 유발된 간손상 및 연관된 장기 손상의 회복에 미치는 영향)

  • To evaluate hepatoprotective effect of Epimedium Koreanum nakai (EKN) on liver-damaged animal model, rats were intoxicated with carbon tetrachloride (1 ml/kg) for 9 weeks orally. Liver-damaged rats were divided into 2 groups: liver-damaged control (LDC) group and EKN group were administered vehicle (saline), EKN extract per os for 4 weeks respectively. Normal control (NC) group was administered saline as the same process of LDC group. The weights of prostate (absolute), testis (relative), epididymis (relative) and packed cell volume (PCV), mean corpuscular volume (MCV) of EKN group significantly (P < 0.05) increased compared with LDC group. But Mean corpuscular hemoglobin (MCH), mean corpusulcar hemoglobin concentration (MCHC) and aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) were significantly (P < 0.01, P < 0.05) decreased. Fibrotic regions in hepatic parenchyma of EKN group significantly (P < 0.01) decreased compared with LDC group and mean diameters of hepatic lobules significantly (P < 0.01) increased. Percentages of degenerative kidney regions and number of degenerative kidney tubules, number of vasodilated atrophic glomerulus of EKN group was significantly (P < 0.01, P < 0.05) decreased compared with LDC group. Number of atrophic seminiferous tubules and epididymal tubules showing oligospermatozoa of EKN group were significantly (P < 0.01) decreased compared with LDC group. In conclusion, EKN extract has a favorable effect on the $CCl_4$-induced liver damage.

The Effects of Gardeniae Fructus Aqua-Acupuncture on Liver Injury of Rats Induced by CCI4 (치자약침(梔子藥鍼)이 실험적(實驗的) 백서(白鼠)의 간손상(肝損傷)에 미치는 영향(影響))

  • This is the study of the effects of Aqua-acupuncture with Gardeniae Fructus on thc recovery of rat's liver which was damaged by 0.3ml/ea of $CCI_4$. Rats were divided into 4 groups; Normal-group(None treated group), Control-group(Not treated after $CCI_4$-intoxicated), Exp. I(Treated with Saline Aqua-acupuncture after $CCI_4$-intoxicated) and Exp. ll(Treated with Gardeniae Fructus Aqua-acupuncture after $CCI_4$-intoxicated). Biochemical assays for each serum enzyme activities of AST, ALT, Albumin, LDH, ${\gamma}$-GT, TG and Total cholesterol were performed. The results were summarized as follows: 1. AST activities in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. Albumin in serum increased in the Gardeniae Fructus Aqua- acupurkture treated group after $CCI_4$-intoxicated. 4. LDH in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after CClcintox icated, the Gardeniae Fructus Aqua acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. 5. ${\gamma}$-GT In serum significantly decreased In the Gardeniae Fructus Aqua-acupuncture trea ted group after $CCI_4$-intoxicated. TG in serum significantly decreased in the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after $CCI_4$-intoxicated, the Gardeniae Fructus Aqua -acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. 7. Total cholesterol in serum decreased in the Gardenias Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated. In comparison with Saline-treated group after $CCI_4$-intoxicated, the Gardeniae Fructus Aqua-acupuncture treated group after $CCI_4$-intoxicated worked highly effectively to rat's damaged liver. The results from ahove show that the Gardeniae Fructus Aqua- acupuncture has highly effects on the damaged liver caused by $CCI_4$. Therefore it is expected that the Gardeniae Fructus Aqua- acupuncture could be used to cure the damaged liver.

To investigate the effects of Houttuynia cordata Thunb (HCT) on the biochemical parameters of function, liver and serum of TCDD-treated rats were used. After 7 days from TCDD (1 $\mu\textrm{g}$/kg) injection, HCT (200 mg/kg) was administered into rats intraperitoneally for 4 weeks. The lipidperoxide content was examined by measuring the level of total cholesterol, HDL-cholesterol, LDL-cholesterol, total lipid and triglyceride (TG) in serum, and malondialdehyde (MDA) in liver tissue of rats. Serum Levels of Xanthine Oxidase Activities in Cyclohexanone-Treated Rats Pretreated with Carbon Tetrachloride

  • To investigate an effect of cyclohexanone (CHO) treatment on the serum levels of xanthine oxidase (XO) in liver damaged animals, the rats were intraperitoneally pretreated with 50% carbon tetrachloride ($CCl_4$) in olive oil (0.1 mL/ 100 g body weight) 14 times every other day. To the $CCl_4$-pretreated rats, CHO (1.56 g/kg body weight) was injected once and then the animals were sacrificed at 4 hours after CHO treatment. The increasing rate of serum and liver XO activities to the control was higher in CHO-treated animals pretreated with $CCl_4$ than the $CCl_4$-pretreated those. Concomitantly CHO injection to the $CCl_4$-pretreated animals showed somewhat higher Vmax and lower Km value in the kinetics of liver XO enzyme. Furthermore, increasing rate of hepatic malonedialdehyde content to the control was also higher in CHO-treated animals pretreated with $CCl_4$ than $CCl_4$-pretreated those. On the other hand, the injection of CHO to the $CCl_4$-pretreated animals showed the more enhanced liver damage on the basis of liver function finding; liver weight per body weight (%), serum levels of alanine aminotransferase activity and hepatic glucose-6-phosphatase activity. In conclusion, injection of CHO to the $CCl_4$-pretreated rats led to more increased activity of serum XO and it may be caused by acceleration of hepatocyte membrane permeability and induction of enzyme protein.
