Safety Assessment of Potential Lactic Acid Bacteria Bifidobacterium longum SPM1205 Isolated from Healthy Koreans

  • The safety assessment of Bifidobacterium longum SPM1205 isolated from healthy Koreans and this strain's inhibitory effects on fecal harmful enzymes of intestinal microflora were investigated. The overall safety of this strain was investigated during a feeding trial. Groups of SD rats were orally administered a test strain or commercial reference strain B. longum $1\times10^9\;CFU/kg$ body weight/day for four weeks. Throughout this time, their feed intake, water intake and live body weight were monitored. Fecal samples were periodically collected to test harmful enzyme activities of intestinal microflora. At the end of the four-week observation period, samples of blood, liver, spleen, kidney, and gut tissues were collected to determine for hematological parameters and histological differences. The results obtained in this experiment demonstrated that four weeks of consumption of this Bifidobacterium strain had no adverse effects on rat's general health status, blood biochemical parameters or histology. Therefore, it is likely to be safe for human use. Fecal harmful enzymes such as $\beta-glucosidase,\;\beta-glucuronidase$, tryptophanase and urease, were effectively inhibited during the administration of the B. longum SPM1205. These results suggested that this B. longum SPM 1205 could be used for humans as a probiotic strain.

  • Disease associated with microorganisms are far from resolved by current therapeutics. One of effective approach to health maintenance and disease control is the use of dietary bacterial and carbohydrate supplements. This comprises use of probiotics and prebiotics. Probiotics mean the live microorganisms, which when administered in adequate amounts confer a health benefit on the host. Prebiotics mean a nondigestible food ingredient that beneficially affects the host by selectively stimulating the growth and/or activity of one or a limited number of bacteria that can Improve the host health. Especially, probiotics has the relation which is close with innate immunity and adaptive immunity. And probiotics has the clinical value with many disease like lactose intolerance, constipation, acute gastroenteritis, food hypersensitivity and allergy, atopic dermatitis, crohn's disease, rheumatoid arthritis, pelvic radiotherapy, intestinal inflammation and chemical exposure, colon cancer, inhibitory effect of Helicobacter pylori and lowering the level of cholesterol. We use jointly korean medicine and probiotics and it has the more therapeutic effect in the many disease.

In vivo evaluation of preventive effect of Lactobacillus reuteri on porcine epidemic diarrhea in suckling piglets

  • Lactic acid bacteria have been reported their beneficial roles on host including reduction of infectious diarrhea problems. In this study, preventive effect of Lactobacillus (L.) reuteri HY25101 and L. johnsonii HY25103 on porcine epidemic diarrhea virus (PEDV) was investigated in suckling piglets. Two groups of one day old PEDV naïve piglets were orally administered L. reuteri HY25101 and L. johnsonii HY25103 for three days respectively before challenge with lethal dose of PEDV. In second experiment, passive immunized one day old piglets using colostrums containing PEDV specific IgA were used. The survival rates of the L. reuteri HY25101 administered group were significantly higher than that of L. johnsonii HY25103 administered group and viral shedding was rapidly diminished in L. reuteri HY25101 administered group. Interestingly piglets born from the sow immunized with attenuated PEDV vaccine were not completely protected from PEDV challenge, however coadministeration of L. reuteri HY25101 and colostrums containing PEDV specific IgA were more effectively prevent PEDV infection. These results suggested that dietary treatment using L. reuteri HY25101 could reduce diarrheal problem and mortality rate caused by PEDV in suckling pigs. In addition, L. reuteri HY25101 could be used as one of effective compensation treatment with attenuated live vaccine for PED.

Efficiency of a Lactobacillus plantarum-Bacillus subtilis Combination on Growth Performance and Fecal Microflora Populations of Broiler Chickens

  • A study was investigated the effects of the probiotic mixture (PM) contained Lactobacillus plantarum (L. plantarum) SY-99 $2.4{\times}10^9CFU/kg$ and Bacillus subtilis (B. subtilis) SJ-61 $2.2{\times}10^9CFU/kg$ on growth performance and fecal microflora counts of broiler chickens. Referred to the previous studies, a total of 160 one-day-old male broiler chicken (Ross 308) were randomly assigned to four experimental groups, which were a basal diet alone (control) and supplemented with PM 0.1 g/kg feed (T1), PM 0.2 g/kg feed (T2), and PM 0.4 g/kg feed (T3). After 28 days of administration, the average daily feed intake of T1 and T3 was significantly different compared to that of control (p<0.05). In addition, the feed conversion ratio of T2 and T3 was significantly improved compared to that of control (p<0.05). On the 28th day after administration, all treatment groups were significantly increased fecal lactic acid bacteria (LAB) counts and decreased fecal Enterobacteriaceae (ENT) counts compared to T1 (p<0.05). The results of this study indicated that the combination of L. plantarum and B. subtilis strains in the range of 0.2-0.4 g/kg feed could be used for the improvement of growth performance and fecal microflora in broiler chickens.

분말형 프로바이오틱스 섭취방법에 따른 구강 내 pH 변화 (Changes in pH values in the oral cavity according to the intake method of powdered probiotics)

  • Objectives: The purpose of this study was to investigate the changes in pH in the oral cavity using the probiotic intake method. Methods: A total of 109 participants were enrolled and randomly assigned to three groups. Participants in the control group did not ingest powdered probiotics, those in experimental group 1 ingested powdered probiotics by dissolving them on the tongue, and those in experimental group 2 dissolved powdered probiotics on the tongue and rinsed with water. pH values were measured 5 times in all. The significance of each group was examined by the Kruskal-Wallis test. The trend over time was expressed as a graph with groupwise means and confidence intervals, considering repeated measurement data. Results: A significant difference was found between the control group and experimental group 1 at two time-points, i.e., immediately after intake and 3 min after ingestion. As a result of the time trend, the pH value of experimental group 2 was smaller than that of experimental group 1, compared to the control group. Conclusions: Studies have shown that taking probiotics with water may help reduce changes in oral pH. Probiotics should be aware of live bacteria and provide consumers with more detailed information on proper dosage and precautions.

  • Cow's milk and dairy products are elements of the human diet that could play an important role in improving human health. The macronutrients and micronutrients found in milk could supply the nutrients required to maintain human health. Among them, milk-derived bioactive peptides have been identified as potential ingredients found in health promoting functional foods. These bioactive peptides target diet-related chronic diseases, particularly non-communicable ones such as cardiovascular disease, diabetes and obesity. Additionally probiotics such as lactic acid bacteria (LAB) are can be considered live microorganisms that confer health benefits for the host-, when administered in adequate amounts. Further, the calcium, vitamin D, and protein content of milk and dairy products could play a role in proving bone health. The effect of milk and calcium on bone mineral density could prevent against fracture, osteoporosis and rickets. Furthermore, milk and dairy products also contain which factors that, which protect against dental caries (anti-cariogenic properties). This paper reviews the various nutritional functions of milk and dairy products in improving human health.

소아 영양 및 유아식 응용을 위한 신바이오틱스의 잠재력: 총설 (Potentials of Synbiotics for Pediatric Nutrition and Baby Food Applications: A Review)

  • 프로바이오틱스, 프리바이오틱스 및 신바이오틱스 등의 물질과 미생물을 조제분유에 강화하여 건강증진 기능성 유산균, 특히 비피더스균과 유산간균의 성장을 선택적으로 자극함으로써 장내균총에 유익한 효과를 나타내고자 하는 노력이 경주되고 있다. 지난 10년 동안, 모유 수유가 장내균총에 미치는 유용 효과를 도모하기 위해서 프로바이오틱스와 프리바이오틱스를 함유하는 새로운 신바이오틱 조제분유가 제안되었다. 새로운 신바이오틱스는 상승 효과로 인해서 프로바이오틱스 및 프리바이오틱스를 개별적으로 사용하는 경우 보다 더 도움이 되는 것으로 기대된다. 한편, 조제분유 및 건조 제품의 제조에 사용되는 가공 기술 및 저장 조건 등이 프로바이오틱스의 생존에 악영향을 미치게 된다. 충분한 유산균수($10^8cfu/g$)로 장내 예정된 부위에 도달하여 숙주에 긍정적인 효과를 나타내기 위해서는 식품 제조공정 및 위장관 통과 시 프로바이오틱스의 생존성을 유지하는 것이 가장 중요한 요소이다. 일반적으로 안전하다고 인정되며, 높은 생물학적 가치를 가지고 있는 물질을 이용하여 적절하고 비용 효율적인 마이크로캡슐화 기술이 개발되면 조제분유의 품질 향상이 가능하게 된다. 프로바이오틱스는 복합적인 효과를 나타낸다. 하나는 살아 있는 생균체가 장내균총에 유익하게 영향을 미쳐 면역조절 효과를 보여주는 것이며, 다른 하나는 사균체가 항-염증 반응을 나타내는 것이다. 최근에, 사균체 또는 정제되지 않은 미생물 분획물이 건강에 긍정적인 영향을 미친다는 것을 명확하게 정의할 새로운 용어에 대한 필요성이 대두되었다. 그 결과, 충분한 양을 경구 또는 국소적으로 투여했을 때 인간과 동물에게 이로운 작용을 하는 사균체(손상되지 않거나 또는 파손된)나 정제되지 않은 세포 추출물(즉, 복잡한 화학적 조성)을 정의하기 위해 paraprobiotics라는 용어가 제안되었다. 프로바이오틱 균주 또는 프리바이오틱스를 조제분유에 강화했을 때 조제분유 수유아의 분변 미생물균총이 조정된다. 즉, 비피더스균 및 유산간균과 같은 건강 유익한 세균을 증가 시킴으로써, 프리바이오틱스는 분변 미생물균총의 조성을 변경하고, 이에 따라서 면역계의 활성을 조절한다. 따라서, 품질 향상을 위한 조제분유의 개발은 토착 장내균총의 대사 활성과 증식을 선택적으로 자극하는 비피더스균 등의 특정 프로바이오틱스 및 이눌린과 올리고당 등의 프리바이오틱스를 사용하여, 모유의 건강 유용 효과와 유사하게 접근하는 것에 초점을 맞추고 있다.

양식업에서 바실러스 폴리퍼멘티쿠스 콩발효물의 상업적 응용 (Industrial Application for Aquaculture of Fermented Soybean with Bacillus polyfermenticus SCD)

  • 바실러스 폴리퍼멘티쿠스 에스씨디는 활성아포를 형성하여 장까지 도달하는 특성 때문에 장기적인 장 질환 치료에 사용되어져 왔다. 뿐만 아니라 강력한 항생물질을 생산하는 장점을 가지고 있다. 현 연구는 바실러스 폴리퍼멘티쿠스 에스씨디와 콩을 함께 발효시킨 것 (BPFS)과 그것의 생균제 특성에 관하여 조사하였다. 바실러스 폴리퍼멘티쿠스 에스씨디는 와 BPFS는 병원성 그람양성 (스트렙토코커스 파라우베리스, 스트렙토코커스 이니에, 락도코커스 가르비에), 그람 음성 (플렉시박터 트렉튜오서스, 비브리오 하베이, 비브리오 불니피쿠스, 비브리오 오다리), 곰팡이 (아스파라질러스 니거, 아스파라질러스 오리자에)에 이르기까지 넓은 범위의 항균성을 가졌다. BPFS에 의한 생존율 증가와 부영양화로 인한 수질오염의 정화작용을 관찰하기 위해서 통영 앞바다 양식장에서 조피볼락을 사용해서 현장 실험을 하였다. 그 결과, 해양 병원성 유해 박테리아에 노출시키고 일반 사료만 복용시킨 군에서는 99%의 폐사율을 보였으나, BPFS를 사료와 함께 복용시킨 군에서는 먹이 유인효과에 의한 성장률의 증가와 부영양화에 의한 수질오염을 정화하는 작용이 관찰되었다. 따라서 BPFS는 양식업에서 항생물질 대체제, 사료첨가제, 부영양화에 의한 정화제로서 개발하여 산업화하기 용이한 후보물질이라고 사료된다.
