Development and Validation of HPLC Method for Pharmacokinetic Study of Promethazine in Human (염산프로메타진 체내동태 연구를 위한 혈청 중 프로메타진의 HPLC 정량법 개발 및 검증)

  • A rapid, selective and sensitive reversed-phase HPLC method for the determination of promethazine in human serum was developed, validated, and applied to the pharmacokinetic study of promethazine. Promethazine and internal standard, chlorpromazine, were extracted from human serum by liquid-liquid extraction with n-hexane containing 0.8% isopropanol and analyzed on a Capcell Pak CN column with the mobile phase of acetonitrile-0.2 M potassium dihydrogen phosphate (42:58, v/v, adjusted to pH 6.0 with 1 M NaOH). Detection wavelength of 251 nm and flow rate of 0.9 mL/min were fixed for the study. The assay robustness for the changes of mobile phase pH, organic solvent content, and flow rate was confirmed by $3^{3}$ factorial design using a fixed promethazine concentration (10 ng/mL) with respect to its peak area and retention time. In addition, the ruggedness of this method was investigated at three different laboratories using same quality control (QC) samples. This method showed linear response over the concentration range of 1-40 ng/mL with correlation coefficients greater than 0.999. The lower limit of quantification using 1 mL of serum was 1 ng/mL, which was sensitive enough for pharmacokinetic studies. The overall accuracy of the quality control samples ranged from 96.15 to 105.40% for promethazine with overall precision (% C.V.) being 6.70-11.22%. The relative mean recovery of promethazine for human serum was 63.54%. Stability (freeze-thaw and short-term) studies showed that promethazine was stable during storage, or during the assay procedure in human serum. However, the storage at $-80^{\circ}C$ for 4 weeks showed that promethazine was not stable. Extracted serum sample and stock solution were not allowed to stand at ambient temperature for 12 hr prior to injection. The peak area and retention time of promethazine were not significantly affected by the changes of mobile phase pH, organic solvent content, and flow rate under the conditions studied. This method showed good ruggedness (within 15% C.V.) and was successfully used for the analysis of promethazine in human serum samples for the pharmacokinetic studies of orally administered Himazin tablet (25 mg as promethazine hydrochloride) at three different laboratories, demonstrating the suitability of the method.

  • Pressure-balanced external Ag/AgCl electrode has been extensively used for both Pressurized Water Reactor (PWR) and Boiling Water Reactor (PWR) environments. The use of KCI-based buffer solution often becomes the source of electrode potential drift due to slow leakage through its porous plug, typically made of zirconia. It is reported that results of our effort to improve the stability of electrode potential by using high purity water as the filling solution in which $Cl^-$ ion activity can be established and maintained at the solubility of AgCl even with the sustained leakage for a long period. Stability tests have been made in boron and lithium mixture solution at $288^{\circ}C$. The electrode potential remained stable within 10 mV over one week period. And after a thermal cycle between 288 to $240^{\circ}C$ the potential shift of Ag/AgCl electrodes did not exceed 15 mV By using the limiting equivalent ionic conductances and Agar's hydrodynamic theory, the thermal liquid junction potential (TLJP) of the electrode has been predicted. The calculated values for the water-fiued Ag/AgCl electrode potential, in which the chlorine concentration in the filling solution was derived from the measured data at ambient temperature, had a good agreement with the experimental values.

Effects of Freezing and Thawing Methods on the Quality of Dongchimi (동결 및 해동 방법이 동치미의 품질에 미치는 영향)

  • Dongchimi (Korean-style fermented radish with juice) products were frozen to prevent further acidification and softening of texture by restraining microbial growth and enzyme activity during storage. Dongchimi juice and radish were separated prior to freezing process. Dongchimi radish was frozen at $-20^{\circ}C,\;-70^{\circ}C$ and immersed in liquid nitrogen and dongchimi juice was frozen at $-20^{\circ}C\;and\;-70^{\circ}C$. Frozen dongchimi samples were thawed with ambient temperatures of $4^{\circ}C\;and\;27^{\circ}C$ and with 915 MHz-microwave, respectively. Dongchimi radish immersed in liquid nitrogen and thawed with 915 MHz-microwave showed the highest pectinesterase activity and hardness, and the lowest polygalacturonase activity and color change, indicating that this quick freezing-quick thawing method can be used for the long-term storage of dongchimi products. Dongchimi juice frozen at $-70^{\circ}C$ and thawed with 915 MHz-microwave retained its pH and titrable acidity, and showed a largest reduction in total aerobic count and lactic acid bacteria.

Along and across-wind vibration control of shear wall-frame buildings with flexible base by using passive dynamic absorbers

  • A flexible-base coupled-two-beam (CTB) discrete model with equivalent tuned mass dampers is used to assess the effect of soil-structure interaction (SSI) and different types of lateral resisting systems on the design of passive dynamic absorbers (PDAs) under the action of along-wind and across-wind loads due to vortex shedding. A total of five different PDAs are considered in this study: (1) tuned mass damper (TMD), (2) circular tuned sloshing damper (C-TSD), (3) rectangular tuned sloshing damper (R-TSD), (4) two-way liquid damper (TWLD) and (5) pendulum tuned mass damper (PTMD). By modifying the non-dimensional lateral stiffness ratio, the CTB model can consider lateral deformations varying from those of a flexural cantilever beam to those of a shear cantilever beam. The Monte Carlo simulation method was used to generate along-wind and across-wind loads correlated along the height of a real shear wall-frame building, which has similar fundamental periods of vibration and different modes of lateral deformation in the xz and yz planes, respectively. Ambient vibration tests were conducted on the building to identify its real lateral behavior and thus choose the most suitable parameters for the CTB model. Both alongwind and across-wind responses of the 144-meter-tall building were computed considering four soil types (hard rock, dense soil, stiff soil and soft soil) and a single PDA on its top, that is, 96 time-history analyses were carried out to assess the effect of SSI and lateral resisting system on the PDAs design. Based on the parametric analyses, the response significantly increases as the soil flexibility increases for both type of lateral wind loads, particularly for flexural-type deformations. The results show a great effectiveness of PDAs in controlling across-wind peak displacements and both along-wind and across-wind RMS accelerations, on the contrary, PDAs were ineffective in controlling along-wind peak displacements on all soil types and different kind of lateral deformation. Generally speaking, the maximum possible value of the PDA mass efficiency index increases as the soil flexibility increases, on the contrary, it decreases as the non-dimensional lateral stiffness ratio of the building increases; therefore, there is a significant increase of the vibration control effectiveness of PDAs for lateral flexural-type deformations on soft soils.

Estimation of Source Apportionment for Semi-Continuous PM2.5 and Identification of Location for Local Point Sources at the St. Louis Supersite, USA (미국 St. Louis Supersite에서의 준 실시간 PM2.5에 대한 기여도 추정 및 지역 규모 오염원의 위치 파악)

  • In this study, 1-hour integrated $PM_{2.5}$ mass and chemical composition concentrations were monitored at the St. Louis-Midwest Supersite in Illinois. Time-resolved samples were collected one week in each of June 2001 (22 June to 28 June), November 2001 (7 November to 13 November), and March 2002 (19 March to 25 March). A total of 427 samples were collected by CAMM (continuous ambient mass monitor) and 15 compounds were analyzed by AAS, PILS (particle-into-liquid sampler), and TOT (thermal optical transmittance) method. PMF was applied to identify the sources and apportion the $PM_{2.5}$ mass to each source for highly time resolved data. In addition, the nonparametric regression (NPR) was applied to identify the predominant directions of local sources relative to wind direction. Also, this study performed compare the NPR analysis and location of actual local point sources at the St. Louis area. The PMF modeling identified nine sources and the average mass was apportioned to gasoline vehicle, road dust, zinc smelter, copper production, secondary sulfate, diesel emission, secondary nitrate, iron+steel, and lead smelter, respectively. These results suggested that this study results will be help for $PM_{2.5}$ source apportionment studies at similar metropolitan area, establish $PM_{2.5}$ standard, and establish effective emissions reduction strategies in Korea.

Development of Helmholtz Solver for Thermo-Acoustic Instability within Combustion Devices (연소시스템의 열음향 불안정 예측을 위한 Helmholtz Solver 개발)

  • In order to effectively predict thermo-acoustic instability within real combustors of rocket engines and gas turbines, in the present study, the Helmholtz equation in conjunction with the time lag hypothesis is discretized by the finite element method on three-dimensional hybrid unstructured mesh. Numerical nonlinearity caused by the combustion response term is linearized by an iterative method, and the large-scale eigenvalue problem is solved by the Arnoldi method available in the ARPACK. As a consequence, the final solution of complex valued eigenfrequency and acoustic pressure field can be interpreted as resonant frequency, growth rate, and modal shape for acoustic modes of interest. The predictive capabilities of the present method have been validated against two academic problems with complex impedance boundary and premixed flame, as well as an ambient acoustic test for liquid rocket combustion chamber with/without baffle.

  • We report a fuel cell power supply unit for mobile phone which operates at room temperature and ambient pressure using liquid methanol and air. The unit consists of a direct methanol fuel cell (DMFC) and a back-up battery connected parallely to the fuel cell. DMFC supplies half of the required power and the back-up battery supplies the other half during talk mode. In standby mode, DMFC covers $100\%$ of the required power and charges the back-up battery as well, Eight unit cells, each having $9 cm^2$ of active area, were connected in series in order to raise the output volotage to $2.5\~3.9V$, which is typical for most mobile phones.

  • The purpose of this analytic study is to design and examine an efficient hydrogen liquefaction cycle by using a pre-cooler. The liquefaction cycle is primarily comprised of a pre-cooler and a refrigerator. The fed hydrogen gas is cooled down from ambient temperature (300 K) to the pre-cooling coolant temperature (either 77 K or 120 K approximately) through the pre-cooler. There are two pre-cooling methods: a single pre-coolant pre-cooler and a cascade pre-cooler which uses two levels of pre-coolants. After heat exchanging with the pre-cooler, the hydrogen gas is further cooled and finally liquefied through the refrigerator. The working fluids of the potential pre-cooling cycle are selected as liquid nitrogen and liquefied natural gas. A commercial software Aspen HYSYS is utilized to perform the numerical simulation of the proposed liquefaction cycle. Efficiency is compared with respect to the various conditions of the heat exchanging part of the pre-cooler. The analysis results show that the cascade method is more efficient, and the heat exchanging part of the pre-coolers should have specific UA ratios to maximize both spatial and energy efficiencies. This paper presents the quantitative performance of the pre-cooler in the hydrogen liquefaction cycle in detail, which shall be useful for designing an energy-efficient liquefaction system.

A Study on the Change of Condensable Particulate Matter by the SO2 Concentration among Combustion Gases (연소 배출가스 중 SO2 농도에 따른 응축성먼지 변화에 관한 연구)

  • Particulate matter (PM) emitted from fossil fuel-combustion facilities can be classified as either filterable or condensable PM. The U.S. Environmental Protection Agency (EPA) defined condensable PM as material that is in the phase of vapor at the stack temperature of the sampling location which condenses, reacts upon cooling and dilution in the ambient air to form solid or liquid in a few second after the discharge from the stack. Condensable PM passed through the filter media and it is typically ignored. But condensable PM was defined as a component of primary PM. This study investigates the change of condensable PM according to the variation in the sulfur dioxide of combustion gas. Domestic oil boilers were used as the source of emission ($SO_2$) and the level of $SO_2$ concentration (0, 50, 80, and 120 ppm) was adjusted by diluting general light oil and marine gas oil (MGO) that contains sulfur less than 0.5%. Condensable PM was measured as 2.72, 6.10, 8.38, and $13.34mg/m^3$ when $SO_2$ concentration in combustion gas were 0, 50, 80, and 120 ppm respectively. The condensable PM tended to increase as the concentration of $SO_2$ increased. Some of the gaseous air pollutants emitted from the stack should be considered precursors of condensable PM. The gas phase pollutants which converted into condensable PM should reduced for condensable PM control.

Application of Analytical Instrument Method for Determining Level of Malodorous Sulfur Compounds. (악취성분중 황화합물에 대한 기기분석법의 적용)

  • The analytical instrument method was applied to analyze malodorous sulfur compounds emitted from industrial fields. Six factories and two sites which release malodorous substances into ambient air were selected to determine the level of hydrogen sulfide($H_2S$), methylmercaptan(MeSH), dimethyl sulfide($Me_2S$), and dimethyl disulfide($Me_2S_2$) using automated thermal desorption system (STD400) and GC-FPD in summer and fall seasons of 1999. The Air sampler for ATD400 uses a small pump to draw sample and a mass flow controller to adjust sample amount without using a dilution apparatus. The trap temperature of ATD400 reached to $-80^{\circ}$ by supplying liquid nitrogen and $H_2S$ can be analyzed under this condition. The recovery rates of $H_2S$, MeSH, $Me_2S$, and $Me_2S_2$ of odorous sulfur compounds standard were shown 98.2%, 93.6%, 98.2%, 99.4% respectively. The concentrations of $Me_2S$ at outside boundary of G market, L factory, and J factory were 0.018ppm, 0.021ppm, 0.032ppm in summer, respectively. The concentration of $H_2S$ at Nanjido landfill was 1.167ppm in summer, but that of $H_2S$ was not detected in fall because of soil covering. The concentration of H2S and $Me_2S_2$ at inside of Chonggye stream were 0.564ppm and 1.045ppm in summer, while those of H2S and Me2S2 were 0.285ppm and 0.465ppm in fall, respectively.
