Development of a spawning method using liquid inoculum of Agaricus bisporus (양송이 액체접종원을 이용한 종균 제조 방법 개발)

  • Currently, the spawn of the mushroom Agarcus bisporus is produced by a method developed in the 1980s, and anew spawning method needs to be developed to improve the quality of the spawn. In this study, the condition for a maximum mycelium weight(5.92±0.52 g/L) was shaking culture (24 hours/day) at 24℃ and 120 rpm in CDB (compost dextrose broth). Based on this, the ventilated liquid culture method (2.5 L/min) was cultured for 10 days. This method was appropriate, andwhen the inoculum was cultured at 50 g/mL for about 10 days, it was cultured well without agglomeration and shaking of seed.

Condition of mycelial culture and inoculum volume of spawn on cultivation of Agrocybe cylindracea (버들송이의 균사배양조건 및 최적 접종량 설정)

  • Studies were made to optimize the media composition and cultural condition for mycelial growth of Agrocybe cylindracea. Sawdust spawn of media composition for optimal growth was found to be pine sawdust combination of 30% wheat bran and poplar sawdust combination of 20% corn bran were the best of the optimal combination. The optimal concentration of white sugar was 1.0~1.5%. The nitrogen sources was found to be yeast extract and soybean powder. Also, optimal concentration were $0.7g/{\ell}$ and $0.1g/{\ell}$, respectively. The mineral sources of optimal medium compositions were $MgSO_4{\cdot}7H_2O\;0.3g/{\ell}$, $KH_2PO_4\;0.5g/{\ell}$ and $K_2HPO\;1.2g/{\ell}$. Optimal amount of inoculum for cultivation of A. cylindracea were $20{\sim}25g/850m{\ell}$ and $25m{\ell}/850m{\ell}$ in the sawdust spawn and liquid spawn, respectively.

Physico-chemical characteristics and antioxidant activities in oyster mushroom (Pleurotus ostreatus) cultivated with liquid spawn (액체종균으로 배양된 느타리버섯(Pleurotus ostreatus)의 이화학적 특성 및 항산화 활성)

  • The physicochemical characteristics of oyster mushrooms (Pleurotus ostreatus) cultivated using liquid spawn (MLS) were compared with those of commercial mushrooms cultivated using solid spawn. The color intensity of the two types of mushrooms showed no remarkable difference. The hardness of the MLS-cultivated mushrooms was significantly higher, but their moisture content (86.80%) was significantly lower than that of the commercial mushrooms. Mineral contents in MLS-cultivated mushrooms (421.17 mg/100 g) were significantly higher than those in the commercial mushrooms (333.26-362.78 mg/100 g); in particular, the potassium (K) content was the most abundant in the former. The amino acid content in the MLS-cultivated mushrooms (4,695.22 mg/100 g) was about 1.4-2.0 times that in the commercial mushrooms. The essential amino acid contents and sum of aspartic acid and glutamic acid were higher in the MLS-cultivated mushrooms than in the commercial mushrooms. The ${\beta}-glucan$ content in the MLS-cultivated mushrooms was 1.1-2.3 times higher than that in the commercial mushrooms. The total phenol and flavonoid contents and the DPPH and ABTS radical-scavenging activities of the MLS-cultivated mushrooms were significantly higher than those of the commercial mushrooms; however, the reducing power showed an opposite trend. Therefore, MLS-cultivated mushrooms contained higher amounts of valuable components and higher antioxidant activities than commercial mushrooms.

Characteristics of sawdust cultivation of Lentinula edodes with different methods of spawn inoculation

  • This study was carried out to investigate the management characteristics and growth performance of L. edodes from the cooling stage to incubation. Bags of different heights and weights are available for bagging. When the medium size of $17{\times}13cm$ was used and the size of the inoculation hole was changed from 1/3 to 2/3, the browning period was shortened to 30 days. Mycelial growth was evaluated according to the cooling temperature after sterilization. It was observed to be the highest at 122 mm/15 days at $10^{\circ}C$ and 114 mm/15 days and 117 mm/15 days at $15^{\circ}C$ and $20^{\circ}C$, respectively. The contamination rate of the sawdust media before inoculation was measured as 0, $4.5{\times}10$, $1.3{\times}10^2$, $4.0{\times}10^3cfu$ at $5^{\circ}C$, $10^{\circ}C$, $15^{\circ}C$, and $24^{\circ}C$ respectively. The average of $1.6{\times}10^8$ colony forming units (cfu) of microorganisms was observed in the sawdust that had been piled for six months outdoors. In summer, the sawdust has to be used immediately after mixing. The sterilized medium had an average of $4{\times}10^3cfu$ of microorganisms at $24^{\circ}C$ and $1.3{\times}10^2cfu$ at $15^{\circ}C$. After 15 days of inoculation in vitro, the growth conditions of the sawdust was the best at 132 mm, followed by grain and liquid. When inoculated with liquid spawn, the moisture content of the substrate should be adjusted between 50% and 55% in advance.

Studies on collection and spawn manufacture of Armillaria spp. for production of Gastrodia tuber (천마생산을 위한 천마버섯균의 수집과 우량종균 제조에 관한 연구)

  • Armillaria isolates (KNU-A110, KNU-A234, KNU-A1022 and KNU-A1030) were excellent isolates for producing Gastrodia tuber in farm cultivation. Depth of soil between $10\;cm{\sim}19\;cm$ was favorable for producing Gastrodia tuber and rhizomorph at cultivated area. Eighty nine isolates were collected from 5 countries; 16 from Japan, 22 from USA, 26 from France, 4 from Africa and 21 from Korea. Mycelial fan of most isolates were better formed on basal medium with lemon extract than without lemon.A. mellea (KNU-A997) were strongly pathogenic to Gastrodia but A. gallica (KNU-A110) were excellent symbiotic to Gastrodia tuber. Mycelial growth were good on basal medium containing $0.25%{\sim}0.5%$ ethanol and sawdust spawn added wheat bran and corncob. Liquid culture inoculation were not only fast growth of mycelium but also reduction of contamination.

Effect of Perenniporia fraxinea on Eliminating Urushiol from Rhus verniciflua Stokes Stem Bark (아까시재목버섯이 옻나무 껍질의 urushiol 제거에 미치는 영향)

  • This study evaluated the raw materials for spawning Perenniporia fraxinea, to eliminate urushiol. The growth rates of spawns on grains of millet, brown rice, and wheat were 4.92±0.05, 2.20±0.03, and 1.93±0.03 mm/day, respectively, and the laccase activity was 0.86±0.02, 0.04±0.01, and 0.01±0.00 U/mL, respectively. These observations revealed millet as the most appropriate grain for spawn production in terms of growth rate and enzyme activity. Inoculation of lacquer tree (Rhus verniciflua Stokes) stem bark with millet spawns of P. fraxinea resulted in a reduction of its urushiol contents, up to 86.6% on the third day and up to 98.5% on the seventh day. The optimal period of cultivation to eliminate urushiol was three days with 68% of residual flavonoids and 42% of phenolic components. When compared to the product cultivated from liquid spawn, the millet spawn reduced the cultivation period from 10 days to 3 days for eliminating urushiol.

Growth Condition of Liquid Culture by Pleurotus ostreatus (액체배양에서 느타리버섯균의 적합한 생장조건 구명)

  • For the practical using of liquid spawn we carried out selection test of nutrient sources, cultural methods and cultural apparatus for liquid spawn production of oyster mushroom(ASTI 2001, ASTI 2018, ASTI 2072, ASTI 2016, ASTI 2070, ASTI 2180, ASTI 2183, ASTI 2042). The optimal temperature and pH range for mycelial growth of Pleurotus species were $25^{\circ}C\;to\;30^{\circ}C$ and 5.5 to 6.5, respectively. The effect of carbon sources, nitrogen sources and mineral salts on the mycelial growth was studied using petridish culture. Generally, the disaccharides and polysaccharides showed good effect for mycelial growth of Pleurotus species, and the polysaccharides were superior to the other classes of carbon sources for mycelial growth. For the mycelial growth of the 8 oyster mushroom stains, soybean flour was superior to the other kinds of nitrogen sources. On the other hand, addition of mineral salts did not affect, and even poor under certain mineral salts, the mycelial growth of the 8 oyster mushroom stains. The brown sugar selected out the carbon source of the agricultural medium. Also the soybean flour selected out the nitrogen source of agricultural medium. In the medium selection, we selected out agricultural optimum medium composed of brown sugar 3%, soybean flour 0.3%, potassium phosphate 0.05%, magnesium sulfate 0.05%. Under the 250 ml erlenmeyer culture, the effects of such factors as the inoculum rate, the working volume, cultural method and flask shapes on the mycelial growth were examined. The optimal inoculum rate and working volume on the mycelial growth of oyster mushroom was 2 mycelial disk (diameter 6 mm) and 100 ml, respectively. The shake flask culture was enhanced the mycelial growth than at the erlenmeyer flask. Pulp form growth of mycelium in the erlenmeyer flask culture was obtained in the culture with glass rod of length 50 mm, diameter 10 mm.

  • Sparassis latifolia is one of the most expensive mushrooms in Korean market owing to its high ${\beta}$-glucan content and immunoactivity. However, because of the long cultivation period and high contamination rates, it has low production efficiency. Therefore, we first need to establish the optimum conditions for liquid spawn production to increase its production efficiency. As a result of experiments, molasses culture medium was selected for mycelial growth. Also, the optimum sugar content for molasses and amount of aeration used were approximately 8 Brix% and 0.3~0.6 vvm, respectively. Mycelial dry weight increases, while the medium decreases, as the incubation period increases. Therefore, to achieve maximum production efficiency, the incubation period of 9 to 11 days is appropriate.

Fruit body formation on rice brown and timber log using liquid spawn of Phellinus linteus (목질진흙버섯(Phelinus linteus) 액체배양에 의한 현미와 원목에서 자실체 형성)

  • The main objectives of this study were to fruit body of Phellinus linteus using liquid spawn. Highest mycelial growth and in vitro fruiting was observed when $180m{\ell}$ of water was added to 200g of brown rice in $1000m{\ell}$ PP bottle. Optimum inoculum amount was 100ml per bottle. Better mycelial growth and fruiting were observed when the amount of brown rice was lowered. For timber log culture of P. linteus, low contamination was observed when logs were sterilized for as low as 60 mins. But, best mycelial growth was observed when the logs were sterilized for 120mins. Among different log types, highest mycelial growth and fruiting were observed in oak and mulberry. Only few fruiting could be observed in birch, chestnut, alder, while no fruiting was produced in Aspen. Best fruiting was produced from isolate PH-211, although other isolates also produced few fruiting.
