Effect of Diets with Job's Tears and Cudrania tricuspidata Leaf Mixed-powder Supplements on Body Fat and Serum Lipid Levels in Rats Fed a High-Fat Diet (율무와 꾸지뽕잎 혼합물이 고지방식이를 급여한 흰쥐의 체지방 및 혈청 지질농도에 미치는 영향)

  • This study investigated the effects of diets with a supplement of Job's tears (JT) and Cudrania tricuspidata leaf (CTL) powder as a functional food ingredient on obesity and lipid profiles in diet-induced obese rats. Male Sprague-Dawley rats were fed a high-fat diet (D12492, 45 kcal fat) for 5 weeks during the first phase. In the second phase, which lasted 5 weeks, the rats were divided into four experimental groups. The groups were a high-fat diet group as a control (HFD), a high-fat diet with 10% JT powder supplement group (JT-10), a 10% CTL powder supplement group (CTL-10), and a 20% JT and CTL 1:1 mixed-powder supplement group (MIX-20). The adipose tissue (RFP, EFP) weights were significantly decreased in the CTL-10 and MIX-20 groups than in the HFD group (p<0.05). The fecal weight produced by the MIX-20 group was highest among all experimental groups (p<0.05), and it was significantly increased during the second phase compared to the first phase (p<0.05). The serum total cholesterol content was significantly decreased in the CTL-10 and MIX-20 groups, and the LDL-cholesterol content was significantly low in the JT-10 and MIX-20 groups than in the HFD group (p<0.05). The blood glucose levels were significantly decreased in all experimental diet supplement groups than in the HFD group (p<0.05). The findings show that a diet of mixed JT and CTL could improve lipid metabolism.

  • The Present study was conducted to investigate the effects of Ginseng extract on the tension-area curve for stearic acid monolayer. At the same time, the effects of Ginseng extract on osmotic and mechanical fragility of human red cells and histamine release from rabbit leukocytes were studied, The results are summarized as follows. 1. The Ginseng alcohol extract was found to expand liquid expanded phase of stearic acid monolayer, thus it is speculated that this agent may be acting as a surface active substance. 2. Osmotic hemolysis was inhibited by the Ginseng alcohol extract and the same effect was also observed in the presence of Ginseng saponin. However, the Ginseng alcohol extract was found to decrease hematocrit ratio of the RBC suspension, therefore, the inhibition of the osmotic hemolysis by this agent may be secondary effect to the reduced cell volume. 3. The mechanical hemolysis was also inhibited by the Ginseng alcohol extract but the inhibition was independent of changes in hematocrit ratio. 4. Histamine release from rabbit leukocytes was significantly increased in vitro in the presence of the Ginseng alcohol extract.(p<0.05)

  • The biosynthesis of phospholipid and the composition of fatty acid in C. ellipsoidea mitochondria treated with antibiotics(cycloheximide, nalidixic acid) during the culture analyzed. The growth of Chlorella and the contents of total lipid in mitochondria treated with antibiotics were lower than those of the control. The synthesis of PC (phosphatidylcholine) and PI(phosphatidylinostiol) were inhibited in the nalidixic acid treatment and also the contents of PC(phosphatidylcholine), PE (phosphatidylethanolamine), PG(phosphatidylglycerol) and PI(phosphatidylinositol) in the cycloheximide treatment were also inhibited. The major fatty acids utilized for the various phospholipids formation in each antibiotics treatment were analyzed stearic acid, myristic acid, palmitic acid, oleic acid, linoleic acid and linolenic acid at the late phase of the culture.

Effects of Vitamins C and E on Hepatic Drug Metabolizing Function in Nypoxia/Reoxygenation (저산소 및 산소재도입시 vitamin C와 E가 간장 약물대사 기능에 미치는 영향)

  • Liver isolated from 18 hours fasted rats was subjected to $N_2$hypoxia (for 45 min) followed by reoxygenation (for 30 min). The perfusion medium used was Krebs-Henseleit bicarbonate buffer (pH 7.4, $37^{\circ}C$). Vitamin C (0.5 mM) and trolox C (0.5 mM), soluble vitamin E analog, were added to perfusate. Lactate dehydrogenase (LDH), total glutathione, oxidized glutathione, lipid peroxide and drug-metabolizing enzymes were measured. After hypoxia LDH significantly increased but this increase was attenuated by vitamin C and combination of vitamin C and E. Total glutathione and oxidized glutathione in perfusate markedly increased during hypoxia and this increase was inhibited by vitamins C, E and its combination. Similarly; oxidized glutathione and lipid peroxide in liver tissue increased after hypoxia and reoxygenation and this increase was inhibited by vitamin I and combination of vitamin C and E. Hepatic drug metabolizing function (phase I, II) were suppressed during hypoxia but improved during reoxygenation. While vitamins C and E only increased glucuronidation, the combination of vitamin C and E increased the oxidation, glucuronidation and sulfation. Our findings suggest that vitamins C and E synergistically ameliorates hepatocellular damage as indicated by abnormalities in drug metabolizing function during hypoxia/reoxygenation and that this protection is in major part, caused by decreased oxidative stress.

  • This study described formation and dispersion of stable lamellar structure containing ceramide. To characterize of the lamellar structure containing ceramide, we used differential scanning calorimetry, x-ray diffraction, and FT-IR spectra, which enabled us to demonstrate that ceramide with appropriate amounts of lipid, oil and surfactant can assemble to form a stable lamellar-phase. These approaches to use ceramide offers useful means to fabricate a variety of biocompatible emulsion and liposome, which enlarge their applicability in the fields of drug delivery, dermatology and cosmetics.

DPPH Radical Scavenging Effect and in vitro Lipid Peroxidation Inhibition Activity of Portulaca oleracea and Its Active Principles (쇠비름(Portulaca oleracea) 추출물의 DPPH radical 소거능과 in vitro 지질과산화 억제 효과와 활성성분)

  • An antioxidative activity of Portulaca oleracea was tested by in vitro experimental models. The antioxidative activities were determined by evaluation the DPPH radical scavenging activity and by measuring lipid peroxide using 2-thiobarbituric acid (TBA). The crude extract was sequentially partitioned with n-hexane, 15% aq. MeOH, EtOAc, n-BuOH, $H_2O$. Among them, a remarkable antioxidative effect was observed in the fractions of EtOAc and n-BuOH. The DPPH radical scavenging effect $(IC_{50}=17.90{\mu}g/ml)$ of the n-BuOH soluble fraction was comparable with that of natural antioxidant, ${\alpha}-tocopherol(IC_{50}=\;6.99{\mu}g/ml)$ and the inhibitory effect of lipid peroxidation in mouse liver homogenate was similar to that of natural antioxidant, L-ascorbic acid at a concentration of 0.1mg/ml to 5mg/ml. From the BuOH soluble fraction yielded two biophenolic glycosides, 3-hydroxy-1-(2-hydroxyethyl)phenyl-4-O-${\beta}$-D-glucopyranoside(1) and 2-(3,4-dihydroxyphenyl)ethyl-O-${\beta}$-D-glucopyranoside(2) using column chromatography and revered-phase HPLC. In particular, the DPPH radical scavenging activity of 2 was comparable to that tocopherol$(IC_{50}=6.59{\mu}g/ml)$.

  • Lipid extraction preceding fatty acid methyl esters preparation for gas-liquid chromatography is time-consuming and cumbersome. We performed one-step extraction/methylation method with a mixture of methanol-heptane-benzene-DMP-H$_2$SO$_4$ without prior fat extraction. The simultaneous digestion and lipid transmethylation takes place at 8$0^{\circ}C$ in a single phase. After cooling till room temperature, two phases are formed. The upper one of the phases contains the fatty acid methyl esters ready for GLC. The fatty acid composition of major industrial crops obtained by the one step extraction/methylation method (method 1 and 2) was almost identical with the fatty acid composition of the pure fats extracted with hexane by the Soxtec instrument (method 3). Due to its simplicity, speed, and reduced organic solvent the one-step extraction/methylation method (method 1 and 2) should be useful to determine overall fatty acid composition, especially in situations where many samples have to be analyzed.

  • Based on reaction time and substrate molar ratio, structured lipid (SL-corn) was produced at 1:2:2(corn oil/capric acid/CLA) and 4% immobilized lipase from Rhizomucor miehei (RM IM). Reaction was carried out for 24 hr at $55^{\circ}C$ in 1-L stirred-batch reactor. After reaction, 13.3 mol% capric acid and 8.9%, CLA were incorporated into corn oil. Iodine and saponification values of SL-corn were 68 and 202, respectively. Tocopherol content decreased after reaction (about 39%). SL-corn showed more yellowish color than corn oil (p<0.05). Reversed-phase HPLC indicated triacylglycerol species containing capric acid in SL-corn resulted in faster crystallization than that of corn oil.

  • 1-Furan-2-yl-3-pyridin-2-yl-propenone (FPP-3) has recently been synthesized and characterized to have an anti-inflammatory activity through the inhibition of the production of nitric oxide. In the present study, adverse effects of FPP-3 on hepatic functions were determined in female BALB/c mice. When mice were administered with FPP-3 at 125, 250 or 500 mg/kg for 7 consecutive days orally, FPP-3 significantly increased absolute and relative weights of liver with a dose-dependent manner. In addition, FPP-3 administration dramatically increased the hepatotoxicity parameters in serum at 500 mg/kg, in association of hepatic necrosis. FPP-3 significantly induced several phase I enzyme activities. To elucidate the possible mechanism(s) involved in FPP-3 induced hepatotoxicity, we investigated the hepatic activities of free radical generating and scavenging enzymes and the level of hepatic lipid peroxidation. FPP-3 treatment significantly elevated the hepatic lipid peroxidation, measured as the thiobarbituric acid-reactive substance, and the activity of superoxide dismutase. Taken together, the present data indicated that reactive oxygen species might be involved in FPP-3-induced hepatotoxicity.

Bioremedation of petrolium pollution (유류오염의 미생물학적 제어)

  • As basic study for purpose bioremedation in oil-contaminated environment, Primarily, we isolated biosurfactant producer- strains utilized of oil-agar plate, and measured surface tension and emulsifying activity. We investigated in oil-contaminated soil and sea water. In this laboratory, Pseudomonas sp. EL-012S strain isolated from oil-contaminated soil was able to product novel biosurfactant under the optimal culture condition. Its condition was n-hexadecane 2.0%, NH$_4$NO$_3$0.4%, Na$_2$HPO$_4$0.6%, KH$_2$PO$_4$0.4%, MgSO$_4$.7$H_2O$ 0.02%, CaCl$_2$.2$H_2O$ 0.001%, FeSO.7$H_2O$ 0.001%, initial pH 7.0 and aeration at 3$0^{\circ}C$, respectively. This biosurfactant was produced in both late-exponential and early-stationary phase. The biosurfactant from Pseudomonas sp. EL-012S was composed of carbohydrate, lipid and protein. The purified-biosurfactant was examined two (biosurfactant type I, II) with the silica gel G60 column chromatography and the purified biosurfactant confirmed thin layer chromatography, high performed liquid chromatography and gas chromatography. The biosurfactant type I involved in carbohydrate-lipid-protein characteristics lowered surface tension of water to 27dyne/cm and interfacial tension 4.5dyne/cm aginst to n-hexadecane and the biosurfactant type B involved in carbohydrate lipid characteristics lowered surface tension of water to 30dyne/cm and interfacial tension 8dyne/cm against to n-hexadecane. Specially type I had the properties such as strong emulsifying activity, emulsion stability, pH-stability, thermo-stability, high cleaning activity and forming ability.
