Effects of CoCl2 on multi-lineage differentiation of C3H/10T1/2 mesenchymal stem cells

  • Mesenchymal stem cells (MSCs) in the bone marrow and other somatic tissues reside in an environment with relative low oxygen tension. Cobalt chloride ($CoCl_2$) can mimic hypoxic conditions through transcriptional changes of some genes including hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$) and vascular endothelial growth factor (VEGF). This study evaluated the potential role of $CoCl_2$ preconditioning on multi-lineage differentiation of C3H/10T1/2, a murine MSC line to understand its possible molecular mechanisms in vitro. $CoCl_2$ treatment of MSCs markedly increased HIF-$1{\alpha}$ and VEGF mRNA, and protein expression of HIF-$1{\alpha}$. Temporary preconditioning of MSCs with $CoCl_2$ induced up-regulation of osteogenic markers including alkaline phosphatase, osteocalcin, and type I collagen during osteogenic differentiation, followed by enhanced mineralization. $CoCl_2$ also increased chondrogenic markers including aggrecan, sox9, and type II collagen, and promoted chondrocyte differentiation. $CoCl_2$ suppressed the expression of adipogenic markers including $PPAR{\gamma}$, aP2, and $C/EBP{\alpha}$, and inhibited adipogenesis. Temporary preconditioning with $CoCl_2$ could affect the multi-lineage differentiation of MSCs.

Mitochondria-Specific Monoclonal Antibodies in Eggs and Embryos of the Ascidian Halocynthia roretzi

  • Ascidian embryos have become an important model for embryological studies, offering a simple example for mechanisms of cytoplasmic components segregation. It is a well-known example that the asymmetric segregation of mitochondria into muscle lineage cells occurs during ascidian embryogenesis. However, it is still unclear which signaling pathway is involved in this process. To obtain molecular markers for studying mechanisms involved in the asymmetric distribution of mitochondria, we have produced monoclonal antibodies, Mito-1, Mito-2 and Mito-3, that specifically recognize mitochondria-rich cytoplasm in cells of the ascidian Halocynthia roretzi embryos. These antibodies stained cytoplasm like reticular structure in epidermis cells, except for nuclei, at the early tailbud stage. Similar pattern was observed in vital staining of mitochondria with DiOC2, a fluorescent probe of mitochondria. Immunostaining with these antibodies showed that mitochondria are evenly distributed in the animal hemisphere blastomeres at cleavage stages, whereas not in the vegetal hemisphere blastomeres. Mitochondria were transferred to the presumptive muscle and nerve cord lineage cells of the marginal zone in the vegetal hemisphere more than to the presumptive mesenchyme, notochord and endoderm lineage of the central zone. Therefore, it is suggested that these antibodies will be useful markers for studying mechanisms involved in the polarized distribution of mitochondria during ascidian embryogenesis.

Current situation and control strategies of H9N2 avian influenza in South Korea

  • The H9N2 avian influenza (AI) has become endemic in poultry in many countries since the 1990s, which has caused considerable economic losses in the poultry industry. Considering the long history of the low pathogenicity H9N2 AI in many countries, once H9N2 AI is introduced, it is more difficult to eradicate than high pathogenicity AI. Various preventive measures and strategies, including vaccination and active national surveillance, have been used to control the Y439 lineage of H9N2 AI in South Korea, but it took a long time for the H9N2 virus to disappear from the fields. By contrast, the novel Y280 lineage of H9N2 AI was introduced in June 2020 and has spread nationwide. This study reviews the history, genetic and pathogenic characteristics, and control strategies for Korean H9N2 AI. This review may provide some clues for establishing control strategies for endemic AIV and a newly introduced Y280 lineage of H9N2 AI in South Korea.

New distribution record of northern lineage plant of Stellaria filicaulis(Caryophyllaceae) from South Korea

  • A northern lineage plant, Stellaria filicaulis (Caryophyllaceae), was newly found in Yeoncheon-gun, Gyeonggi-do of South Korea. This species is distributed in China, Japan, Korea, Mongolia, and Russia. On the Korean Peninsula, St. filicaulis, however, has been known to grow in North Korea. Species identification was confirmed using morphological characteristics and DNA sequence data, while comparing with materials obtained from herbarium specimens. Stellaria filicaulis is distinguished from St. longifolia by having smooth surface of stem, petals about twice longer than sepals. On the neighbor-joining tree, St. filicaulis formed a clade, and the species is closely related to St. longifolia of the Parviflorae clade. Details of the morphological characters, the type specimens, voucher specimens data, and photographs of St. filicaulis in South Korea are presented. In addition, it is likely that a new habitat will be found by plant biodiversity field surveys through the middle part of the Korean Peninsula. Further research is needed to determine its population size, distribution, and threats, as well as identify appropriate locations for conservation collection of germplasm.

A Study on the Relationship between Kang Jeungsan and Jo Jeongsan Described in Chapter Two of Progress of the Order

  • Although English-language academic materials on Kang Jeungsan (강증산/姜甑山 1871-1909) are steadily increasing, the same cannot be said of Jo Jeongsan (조정산/道主 趙鼎山 1895-1958) who remains under-researched as figure of profound significance to the Korean new religion Daesoon Jinrihoe (대순진리회/大巡眞理會). Furthermore, in materials produced by Daesoon Jinrihoe that are later translated into English, the connections that exist between Kang Jeungsan and Jo Jeongsan are often reduced to a few representative examples, when, in fact, many additional examples could be provided for a more comprehensive understanding. Comprehending the basis for the first succession in the three-figure orthodox religious lineage of Daesoon Jinrihoe is crucial to task of properly differentiating Daesoon Jinrihoe from seemingly similar Korean new religions that enshrine Kang Jeungsan as their Supreme God. The research presented in this article, "A Study on the Relationship between Kang Jeungsan and Jo Jeongsan Described in Chapter Two of Progress of the Order," will provide readers with a thorough overview of the basis for Jo Jeongsan's successorship in the orthodox religious lineage of Daesoon Jinrihoe, through an in-depth exploration of Chapter Two of Progress of the Order from Daesoon Jinrihoe's main scripture, The Canonical Scripture. Furthermore, this article will include special explanatory notes to ensure that it can be easily read even by non-specialists.

Transcriptional and Epigenetic Regulation of Context-Dependent Plasticity in T-Helper Lineages

  • Th cell lineage determination and functional specialization are tightly linked to the activation of lineage-determining transcription factors (TFs) that bind cis-regulatory elements. These lineage-determining TFs act in concert with multiple layers of transcriptional regulators to alter the epigenetic landscape, including DNA methylation, histone modification and threedimensional chromosome architecture, in order to facilitate the specific Th gene expression programs that allow for phenotypic diversification. Accumulating evidence indicates that Th cell differentiation is not as rigid as classically held; rather, extensive phenotypic plasticity is an inherent feature of T cell lineages. Recent studies have begun to uncover the epigenetic programs that mechanistically govern T cell subset specification and immunological memory. Advances in next generation sequencing technologies have allowed global transcriptomic and epigenomic interrogation of CD4+ Th cells that extends previous findings focusing on individual loci. In this review, we provide an overview of recent genome-wide insights into the transcriptional and epigenetic regulation of CD4+ T cell-mediated adaptive immunity and discuss the implications for disease as well as immunotherapies.

Mutations in Streptomycin Resistance Genes and Their Relationship to Streptomycin Resistance and Lineage of Mycobacterium tuberculosis Thai Isolates

  • Background: Streptomycin (SM) is recommended by the World Health Organization (WHO) as a part of standard regimens for retreating multidrug-resistant tuberculosis (MDR-TB) cases. The incidence of MDR-TB in retreatment cases was 19% in Thailand. To date, information on SM resistance (SMR) gene mutations correlated to the SMR of Mycobacterium tuberculosis Thai isolates is limited. In this study, the mutations in rpsL, rrs, gidB, and whiB7 were investigated and their association to SMR and the lineage of M. tuberculosis were explored. Methods: The lineages of 287 M. tuberculosis collected from 2007 to 2011 were identified by spoligotyping. Drug susceptibility profiles were evaluated by the absolute concentration method. Mutations in SMR genes of 46 SM-resistant and 55 SM-susceptible isolates were examined by DNA sequencing. Results: Three rpsL (Lys43Arg, Lys88Arg, and Lys88Thr) and two gidB (Trp45Ter and Gly69Asp) mutations were present exclusively in the SM resistant M. tuberculosis. Lys43Arg rpsL was the most predominant SMR mutations (69.6%) and prevailed among Beijing isolates (p<0.001). No SMR-related mutation in was found rrs. The combination of rpsL and gidB mutations provided 76.1% sensitivity for detecting SMR in M. tuberculosis Thai isolates. whiB7 was not responsible for SMR in SM resistant isolates lacking rpsL and rrs mutations. The significance of the three gidB mutations, 276A>C, 615A>G, and 330G>T, as lineage signatures for Beijing and EAI were underscored. This study identified 423G>A gidB as a novel sub-lineage marker for EAI6-BGD1. Conclusion: Our study suggested that the majority of SMR in M. tuberculosis Thai isolates were responsible by rpsL and gidB polymorphisms constantly providing the novel lineage specific makers.

Dynamic Transcriptome, DNA Methylome, and DNA Hydroxymethylome Networks During T-Cell Lineage Commitment

  • The stepwise development of T cells from a multipotent precursor is guided by diverse mechanisms, including interactions among lineage-specific transcription factors (TFs) and epigenetic changes, such as DNA methylation and hydroxymethylation, which play crucial roles in mammalian development and lineage commitment. To elucidate the transcriptional networks and epigenetic mechanisms underlying T-cell lineage commitment, we investigated genome-wide changes in gene expression, DNA methylation and hydroxymethylation among populations representing five successive stages of T-cell development (DN3, DN4, DP, $CD4^+$, and $CD8^+$) by performing RNA-seq, MBD-seq and hMeDIP-seq, respectively. The most significant changes in the transcriptomes and epigenomes occurred during the DN4 to DP transition. During the DP stage, many genes involved in chromatin modification were up-regulated and exhibited dramatic changes in DNA hydroxymethylation. We also observed 436 alternative splicing events, and approximately 57% (252) of these events occurred during the DP stage. Many stage-specific, differentially methylated regions were observed near the stage-specific, differentially expressed genes. The dynamic changes in DNA methylation and hydroxymethylation were associated with the recruitment of stage-specific TFs. We elucidated interactive networks comprising TFs, chromatin modifiers, and DNA methylation and hope that this study provides a framework for the understanding of the molecular networks underlying T-cell lineage commitment.

本貫의 累層적 意味와 그 起原에 대한 歷史地理的 探索 (Multi-layers of Geographical Meanings in the Korean Bonkwan)

  • 사회역사지리학은 지리적 현상을 사회집단과 관련하여 인식하고 해석한다. 이 글은 한국 성씨집단을 지리적 사회집단으로 인식하고 그들의 본관 문제를 사회역사지리학의 시각에서 살핀 것이다. 필자는 현재의 본관 개념이 단일한 의미가 아닌 다양한 의미층을 갖는다고 인식하였다. 고려전기부터 본격적으로 등장한 우리 나라의 본관 개념에는 그 후 조선시기까지는 몇 개의 의미층이 누적되어 왔음을 알 수 있었고, 그 기원을 사회역사지리적 관점에서 해석하였다. 결과적으로 본관의 의미는 시대마다. 성씨집단에 따라 다양했음을 알 수 있었다. 이를 요약하면 실제로서의 본과, 기원으로서의 본관 권력에의 의지로서의 본관이다. 첫 번째 의미층은 <본관=거주 장소>의 의미로서 시기적으로 고려시기에 두 번째 의미층은 <본관= 기원 장소>의미로서 본관과 거주지가 분리된 상황 즉 여말선초에, 그리고 세 번째 의미층은 <본관= 상징 장소>라는 소위 권력에의 의지를 보여주는 허구적이고 상징적인 의미로서 조선 전기를 과도기로 하여 조선중기의 전란기이후에 형성된 것으로 보았다. 오늘날 우리나라 각 성씨집단들이 사용하고 있는 본관의 의미는 이상의 세 가지 의미중 한가지 이상으로 규정할 수있다. 그러나 구체적인 성씨집단별 본관의 의미 문제는 종족집단의 거주지 기원과 공간적 이주과정이라는 보다 넒은 주제의 틀에서 조명될 필요가 있다. 이것은 본관이 단순한 물리적 공간을 넘어선 컨텍스트상의 사회적 구성체이기 때문이다.

호남지역(湖南地域)의 왜계자료(倭系資料)와 전방후원형고분(前方後圓形古墳) (The Relics of Wae Lineage and the Keyhole-Shaped Mounds in the Honam Area)

  • 호남지역은 일제강점기부터 현재까지 한일 고고 고대사학 연구자 사이에서 항상 큰 관심을 받았던 지역이다. 근래 이 지역에서 왜계 고고자료 발견 사례가 양적 질적으로 급증하고 있기 때문에 선행 연구와 견해를 재검토할 필요가 커지고 있다. 전방후원형 고분은 기존에 13기로 알려졌지만 최근 발견 사례가 17기까지 증가했다. 한 곳에서 3기가 조영된 사실도 새롭게 밝혀졌으며, 또 일본의 고분시대(古墳時代)를 특징짓는 형상식윤(形象埴輪)이 처음으로 발견됐다. 호남지역 출토 왜계 고고자료에 대한 선행연구는 특정 자료나 특정 유적 유구에 한정된 사례가 많았다. 본고에서는 호남의 모든 왜계 고고자료를 대상으로 수량 고고자료의 종류 출토 위치를 파악한 후 그 변천을 추적하였다. 시기별 특징이나 변화상 등을 바탕으로 왜와 마한세력의 관계 의도 동향을 문헌 자료 등과 관련시켜 검토하였다. 호남지역 출토 왜계 고고자료는 5세기 전엽부터 증가 및 광범위한 분포를 보여주며 7세기 초까지 이어졌다. 4세기 말~5세기 전엽에는 왜계 무구와 왜계 매장주체부, 5세기 중엽~후엽에는 수혜기(須惠器)(계(系)), 5세기 말~6세기 전엽에는 전방후원형 고분과 왜계 횡혈식석실, 6세기 중엽~7세기 초에는 왜계 무구 석침이 중심을 이루어 시기마다 양상이 변화했음을 알 수 있었다. 4세기 말~5세기 전엽의 왜계 고고자료에 대해서는 이 시기에 집중된 백제와 왜의 왕래 기사를 근거로 외교 사절단을 수행했을 무인계층과의 관련성을 상정했다. 5세기 중엽~후엽에는 수혜기(須惠器)(계(系)) 토기가 호남 내륙부로 확산되었다. 물자의 집적에 유리한 지점에서 주로 출토되는 경향이 엿보인다. 전방후원형 고분의 피장자를 북구주(北九州) 제세력에 속한 왜인을 중심으로 한 교역자 집단으로 해석했다. 전방후원형 고분은 대체로 기본적으로 기존 수장묘가 조영되지 않던 지역에서 특별한 세력 기반 없이 돌연 출현했다. 그러나 재지인의 협조 없이 전방후원형 고분을 축조하기란 불가능했을 것이다. 이는 일본 현지와 호남지역 전방후원형 고분의 구축 기술 차이를 보아도 알 수 있다. 한편 일부 전방후원형 고분은 재지 수장묘와 동시기 아니면 직후에 조영되었다는 점에서 재지 집단과의 밀접한 관련성을 추정할 수 있다. 5세기 초 금관가야가 몰락하자 왜 제국(諸國)은 새로운 교섭 상대로 호남지역을 선택하였다. 그 배경에는 왜에서 구하기 어려운 (지하)자원이나 물자를 확보해야 할 절실한 필요가 있었을 것이다. 약 200년에 걸쳐서 사용되고 매장된 호남지역의 왜계 고고자료는 왜인의 다양한 교섭 노력과 관련된 것으로 판단된다.
