The timing of poly(3-Hydroxybutyrate) (PHB) biosynthesis was controlled by varying the agitation speed of a stirred tank fermentor during the pH-stat fed-batch culture of recombinant Escherichia coli strain GCSC 6576 harboring pSYL107. Using a concentrated whey solution containing ca. 200 g/l lactose as the nutrient feed, the PHB content was only 57% after 35h due to volumetric limitation of the fermentor. However, by limiting the oxygen by maintaining the agitation speed at 300 rpm, the final PHB content increased to 70% after 70h with a cell concentration of 15 g/l. When the agitation speed was increased up to 500 rpm, a cell concentration of 31 g/l with 80% PHB was obtained after 52h. A further increase in the maximum agitation speed increased the cell concentration, PHB concentration, and PHB productivity, however, the PHB content decreased to 56-58%.

This study was conducted to assay the relationship between the characteristics of watershed and water quality, and to evaluate water quality characteristics of the classified types by TSI deviation analysis with the collected data from 490 reservoir. Relatively shallow depth (<5m) reservoirs out of selected 490 appeared to be eutrophic. The mean TP concentration in reservoirs with the PFA+UFA/watershed area of above 30% was ${\geq}0.1$ mg $L^{-1}$. The mean TN concentration in reservoirs with the PFA/watershed area of above 25% was ${\geq}2.6$ mg $L^{-1}$. Based on the TSI deviation analysis, water quality parameters in TYPE III reservoirs were in high concentration compared to other reservoirs types. Characteristics of Type III generally showed eutrophic, small DA/RA ratio, shallow depth, and large paddy field and upland field to watershed ratio compared to other types of reservoirs. Both water quality and morpho-physical parameters, Type I and II reservoirs were similar with the exceptions of BOD and chi. ${\alpha}$ concentration. Phosphorus in Type I reservoirs was not the primary limiting factor on algal growth, but significant decrease chl. ${\alpha}$ concentration with the increasing TN/TP indicated that phosphorus was the possible secondary limiting factor. Overall results indicated that type of land use, such as PFA and UFA area in watershed, was important parameters for the assessment of water quality characteristics, and phosphorus was limiting nutrient on algal growth in 490 reservoirs.
The increase of population and industrial activities had brought into eutrophication in the Nakdong river. A remarkable acceleration of eutrophication brought about serious problems for water supply. Therefore, for the purpose of conservation of water quality in the Nakdong river it is necessary to control nutrients. MBOD method was use to evaluate algal growth limiting factor and algal growth potential in the Nakdong river from June to August 1994. The modified biochemical oxygen demand(MBOD) depends on the amount of available inorganic nutrient and organic substrate during 5 day incubation in the dark at 20$^{circ}C$. The MBOD assay depends on inorganic nutrients such as P and N as well as reduced carbon and called the MBOD, the MBOD-P, and the MBOD-N, respectively. The results of bioassay by MBOD(Modified BOD) method showed that the MBOD, MBOD-P and MBOD-N value were found to be in the ranges of 3.8∼96.0 mg$O_2$/l, 5.6∼94.0 mg$O_2$/l and 42.0∼220 mg$O_2$/l, respectively. And the the bioassay value was found to be the highest in Koryong area and the lowest in Waekwan area throughout the Nakdong river. The variations of MBOD-P and MBOD-N value showed similar tendencies to the variations of phosphorus and nitrogen value, respectively. By MBOD method, the relationships of MBOD, MBOD-P and MBOD-N value were MBOD ≒ MBOD-P 《 MBOD-N. The MBOD value was nearly equal to the MBOD-P value, and the MBOD-N value was 3 to 20 times more than the MBOD-P value, approximately. Therefore, in the Nakdong river, phosphorus was the limiting factor for algal growth during summer season. Algal growth potential (AGP) assay using Cochlodinium polykrikoides was conducted in Saryang Island coast where C. polykrikoides red tide occurred annually from July to October 1998. The effects of macro- and micro-nutrients on the growth of C. polykrikoides were specifically evaluated by the algal assay method. Two different types of growth response of C. polykikoides for the addition of nutrients were clearly obseued. For both before and after C. polykrikoides occurrence, the growth of C. polykikoides was significantly stimulated by the addition of either nitrate or ammonium of $50{\mu]M$ with phosphate of $5{\mu}M$. The addition of a single nutrient had no clear effect on the growth of C. polyhikoides and the addition of trace metals, vitamins, and EDTA etc. did not stimulate the algal growth, also. This result indicates that both N and P potentially limited the growth of C. polyhikoides in this period. However, during a bloom of C. polyhikoides, the growth was unlikely to be stimulated by the addition of both macro- and micro-nutrients. At that time the nutrient concentration of Saryang Island coast was $24.33{\mu}M$ for ammonium, $1.61{\mu}M$ for phosphate, and $0.58{\mu}M$ for nitrate, respectively. The concentrations of nutrients increased, on average, 8.2-fold for ammonium and 4.8-fold for phosphate, decreased 3.3-fold for nitrate compared to both before and after the red tide. This result shows that the growth of C. polykikoides was not limited by the nutrients during the bloom in September. Therefore, our results suggest that the C. polykrikoides red tide may outbreak especially when the water is fertilized due to the increased N and P.
In order to understand the spatial and temporal variations of nutrients and factors controlling their distribution in Gwangyang Bay, this study was carried out bimonthly from June 2001 to July 2003, Inorganic silicate and nitrate concentrations ranged from $0.04{\mu}M\;to\;69.5{\mu}M(avg.\;12.9{\mu}M)$, and from $0.12{\mu}M\;to\;42.2{\mu}M\;(avg.\;7.83{\mu}M)$, respectively. Silicate concentrations measured just after the typhoon were the highest with an average of $43.2{\mu}M$ at the surface layer in June 2001, whereas the highest nitrate concentration $(avg.\;37.0{\mu}M)$ was observed in the surface layer in July 2003. River runoff apparently influenced variations in silicate and nitrate concentrations (r=0.701 and 0.728, p<0.000, respectively) as well as salinity (r=-0.628, p<0.000). Phosphate concentrations ranged from $0.24{\mu}M\;to\;5.70{\mu}M\;(avg.\;1.34{\mu}M)$ and were highest at stations 5, 6, and 7, near a fertilizer plant with an average of $2.01{\mu}M$. On the basis of N/P and Si/N molar ratios, limiting nutrients have varied temporary and spatially. During 2001-2002, nitrogen was a limiting nutrient in the study area, and Phosphate was limited when a large volume of freshwater flowed into the bay. Silicate was limited when the high standing crops of phytoplankton occurred in the whole study area throughout 2003, and in the inner bay in February and August 2002. During the study period, factors controlling the distribution of nutrients might be summarized as follows; 1) inflow of freshwater by heavy rain accompanied by typhoons and frequent rainfall in summer, showing high concentrations of silicate and nitrate, 2) release of high phosphate concentrations from the fertilizer plant located in the south of Moydo to adjacent stations, 3) release of nutrients from bottom sediment, 4) magnitude of occurrence of phytoplankton standing crops.

Skeletal muscle is of major economic importance since it is finally converted to meat for consumers. The increase in meat production with low costs of production may be achieved by optimizing muscle growth, whereas a high meat quality requires, among other factors, the optimization of intramuscular glycogen and fat stores. Thus, research in energy metabolism aims at controling muscle metabolism, but also liver and adipose tissue metabolism in order to optimize energy partitioning in favour of muscles. Liver is characterized by high anabolic and catabolic rates. Metabolic enzymes are regulated by nutrients through short-term regulation of their activities and long-term regulation of expression of their genes. Consequences of liver metabolic regulation on energy supply to muscles may affect protein deposition (and hence growth) as well as intramuscular energy stores. Adipose tissues are important body reserves of triglycerides, which result from the balance between lipogenesis and lipolysis. Both processes depend on the feeding level and on the nature of nutrients, which indirectly affect energy delivery to muscles. In muscles, the regulation of rate-limiting nutrient transporters, of metabolic enzyme activities and of ATP production, as well as the interactions between nutrients affect free energy availability for muscle growth and modify muscle metabolic characteristics which determine meat quality. The growth of tissues and organs, the number and the characteristics of muscle fibers depend, for a great part, on early events during the fetal life. They include variations in quantitative and qualitative nutrient supply to the fetus, and hence in maternal nutrition. During the postnatal life, muscle growth and characteristics are affected by the age and the genetic type of the animals, the feeding level and the diet composition. The latter determines the nature of available nutrients and the rate of nutrient delivery to tissues, thereby regulating metabolism. Physical activity at pasture also favours the orientation of muscle metabolism, towards the oxidative type. Consequently, breeding systems may be of a great importance during the postnatal life. Research is now directed towards the determination of individual tissue and organ energy requirements, a better knowledge of nutrient partitioning between and within organs and tissues. The discovery of new molecules (e. g. leptin), of new molecular mechanisms and of more powerful techniques (DNA chips) will help to achieve these objectives. The integration of the different levels of knowledge will finally allow scientists to formulate new types of diets adapted to sustain a production of high quality meat with lower costs of production.
A CRD experiment with thirty growing cross bred calves were assigned at random to three treatments rations. 1) $T_0$, 0% Urea + 20% M. O. cake, 2) $T_1$, 1% Urea + 10% M. O. cake and 3) $T_2$, 2% Urea + 0% M. O. cake to develop a rice straw based ration for ruminants. Sweetish odour and yellowish colour were observed in good recovered silage. Organic matter varied from 87.45% to 89.63% whereas crude protein varied from 14.0% to 14.5% in each treatment. No significant differences were found among the nutrient composition of the ration. The dry matter in take (DMI) and dry matter digestibility was higher in $T_0$ (0% Urea) than those of ration containing 1% ($T_1$) and 2% Urea ($T_2$). The organic matter digestibility decreases with increasing doses of urea. The crude protein & nitrogen-free-extract digestibility were found higher in the ration $T_1$ containing 1% urea whereas crude fibre digestibility and available metabolizable energy (ME) were higher in $T_0$ containing no urea as compared to $T_1$ and $T_2$. Total digestible nutrient (TDN) decreases with the increase of urea level. The highest feed efficiency was found in $T_0$ having no urea and lowest was in $T_2$. The animals gained in weights from each ration. Highest gain in weight was found in $T_0$ ration, then followed $T_1$, and $T_2$. This is due to natural protein available in M. O. cake only. It is concluded that supplemetation of urea or M. O. cake with readily available energy source as molasses upto 20% of total dietary dry matter in a complete ration may increase the intake of low quality fibrous roughage only when nitrogen and mineral are not limiting factor.

We measured seedling growth, foliar nutrient and extract concentrations of 3-year-old Ginkgo biloba seedlings growing in a nursery following a single fertilization with nitrogen (N), phosphorus (P) and nitrogen plus phosphorus (N+P) fertilizers. Fertilization did not change foliage, stem and root biomass of the seedlings except for the high N+P treatment, Foliar N and P concentrations following fertilization varied according to the amount of fertilizers. In general, foliar N and P concentrations increased with fertilization, but fertilization with 400kg N/ha and 100kg P/ha decreased foliar N and P concentrations, respectively. Seedling growth and foliar nutrient concentrations showed that N and P were the growth-limiting nutrients in our study site. It was found that fertilization reduced the concentrations of secondary metabolites (Ginkgo flavon glycosides and terpene lactones) in foliages. It seemed there was a relationship between foliage biomass production and secondary chemicals in G. biloba seedlings.

A monitoring study has been conducted to identify hydrologic conditions, water quality and nutrient loading characteristics of small watershed in Juam Lake. Climate data of the watershed were collected; flow rate was measured and water quality sampling was conducted at the watershed outlet for this study. Water quality data revealed that T-P concentrations meet I grade of lake water quality standard during non-storm period, but degraded up to II-III grade of lake water quality standard during storm period. The observed T-N concentrations always exceeded lake water quality standard. Therefore, T-P was identified as limiting chemical constituent for eutrophication of Juam Lake. T-P concentration of non-storm period also revealed that point source pollution is not serious in the watershed. Three year monitoring results showed that the observed T-N losses were $10.85\~18.88$ kg/ha and T-P losses were $0.028\~0.323$ kg/ha during six month (Mar. - Oct.), respectively. Major portion of runoff amount discharged by a few storm events a year and nutrient load showed apparent seasonal variation. Huge runoff amounts were generated by intense storms, which make application of water treatment or detention facilities ineffective. Monitoring results confirmed that water quality improvement by abating nonpoint source pollution in rural watershed of monsoon climate should be focused on source control. T-P losses from paddy field seemed to consist of significant amount of total load from study watershed. Therefore, management of drainage from paddy field is considered to be important for preventing algal blooming problem in Juam Lake.
Kinetic analysis was important to develope the biological nutrient removal process effectively. In this research, anoxic-anaerobic-aerobic system was operated to investigate kinetic behavior on the nutrient removal reaction. Nitrification and denitrification were important microbiological reactions of nitrogen. The kinetics of organic removal and nitrification reaction have been investigated based on a Monod-type expression involving two growth limiting substrates : TKN for nitrification and COD for organic removal reaction. The kinetic constans and yield coefficients were evaluated for both these reactions. Experiments were conducted to determine the biological kinetic coefficients and the removal efficiencies of COD and TKN at five different MLSS concentrations of 5000, 4200, 3300, 2600, and 1900 mg/L for synthetic wastewater. Mathematical equations were presented to permit complete evaluation of the this system. Kinetic behaviors for the organic removal and nitrification reaction were examined by the determined kinetic coefficient and the assumed operation condition and the predicted model formulae using kinetic approach. The conclusions derived from this experimental research were as follows : 1. Biological kinetic coefficients were Y=0.563, $k_d=0.054(day^{-1})$, $K_S=49.16(mg/L)$, $k=2.045(day^{-1})$ for the removal of COD and $Y_N=0.024$, $k_{dN}=0.0063(day^{-1})$, $K_{SN}=3.21(mg/L)$, $k_N=31.4(day^{-1})$ for the removal of TKN respectively. 2. The predicted kinetic model formulae could determine the predicted concentration of the activated sludge and nitrifier, investigate the distribution rate of input carbon and nitrogen in relation to the solid retention time (SRT).
