Transient uidA expression was used to optimize parameters required for biolistic transformation of suspension cells of Easter lily, Lilium longiflourm. Maximum uidA expression occurred following bombardment with gold particles as compared to tungsten. A 3hr pre-treatment of suspension cells with 0.125M osmoticum resulted in a 1.5X increase in uidA expression. A helium pressure of 1550 psi combined with a particle travelling distance of 6cm resulted in maximum uidA expression as compared to either 1100, 1200, or 1800 psi. Transient transformation resulted in up to 493 uidA expressing cells/Petri plate. For stable transformation suspension cells of Lilium longiflorum, were co-bombarded with plasmid DNA containing cucumber mosaic virus (CMV) replicase under the rice actin (Act1) promoter and either the bar or PAT genes under the cauliflower mosaic virus (CaMV 355) promoter. Ten regenerated plants contained the transgene as analyzed by PCR, and two of the ten plants were confirmed to contain the transgene by Southern hybridization. The two transgenic plants were independent transformants, one containing the bar gene and the other both the CMV replicase and bar genes. Plants were sprayed at the rosette stage and found to be resistant to 1000 mg/L of phosphinothricin (Trade name-Ignite) indicating expression of the bar gene throughout the leaves when bar was under control of the CaMV 35S promoter.
A method of collection and long-term storage of viable lily (Lilium longiflorum) pollen grains were developed for their in vitro growth and transformation in consistency. Petroleum ether, n-heptane, cyclohexane and benzene, as pollen collection medium, were determined less toxic to pollen growth in vitro than others tested. Pollen grains, however, lost their growth activity if stored in these solvents more than a week, So, a serial performance, that is, pollen grain collection in these solvents, air-drying and immediate transfer to low temperature condition was determined desirable for keeping the viability much longer. Pollen grains from this storage showed a successful transformation in vitro with a cDNA encoding tissue plasminogen activator (TPA) protein using Agrobacterium via vacuum infiltration according to western blotting analysis.
Supernumerary B chromosomes were found in Lilium amabile (2n = 2x = 24), an endemic Korean lily that grows in the wild throughout the Korean Peninsula. The extra B chromosomes do not affect the host-plant morphology; therefore, whole transcriptome analysis was performed in 0B and 1B plants to identify differentially expressed genes. A total of 154,810 transcripts were obtained from over 10 Gbp data by de novo assembly. By mapping the raw reads to the de novo transcripts, we identified 7,852 differentially expressed genes (log2FC > |10|), in which 4,059 and 3,794 were up-and down-regulated, respectively, in 1B plants compared to 0B plants. Functional enrichment analysis revealed that various differentially expressed genes were involved in cellular processes including the cell cycle, chromosome breakage and repair, and microtubule formation; all of which may be related to the occurrence and maintenance of B chromosomes. Our data provide insight into transcriptomic changes and evolution of plant B chromosomes and deliver an informative database for future study of B chromosome transcriptomes in the Korean lily.

Eighteen accessions of Curcuma germplasm were evaluated for use as cut flowers or potted plants. Curcuma species or cultivars that have colorful showy bracts and long inflorescence longevity can be produced as cut flowers or potted plants. For cut flower uses, following cultivars were recommended: C. alismatifolia 'Chiangmai Pink', 'Tropic Snow', 'Chiangmai Dark Pink', 'Lady Di', and 'Siam Violet Lily' with long flower stalk length (40~55 cm) and long vase life. For potted plant production, C. alismatifolia 'Cambodian Scarlet', 'Cambodian Red', C. sparganifolia 'Siam ruby', and C. thorelii 'Chiangmai Snow' producing short flower stalk length (about 10~20 cm) were evaluated.
Plants regenerated from in vitro cultures carry chromosomal variations, especially in long-term culture. Reducing the duration of plant tissue culture is one of the ways to reduce genetic and epigenetic changes. In this study, we reduced the duration of long-term culture and repeat subculture using small bulblets derived from bulb scales in two lily cultivars. The adventitious bulblets derived from bulb-scale tissue were cultured on three different media containing Murashige and Skoog (MS) basal medium supplemented with 1 g/L Charcoal, MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone with or without Charcoal, respectively. About seven weeks later, the number of newly propagated multiple shoots in the two media, A and B media, showed little differentiation. Compared to both media, the number of propagated multiple shoots increased 5-fold in MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone without Charcoal (C medium). The number of propagated multiple shoots ranged from 5 to 6 and 4 to 6 with an average of 5 in TropicalPink and GreenStar cultivars, respectively. The flow cytometric measurements indicated no variation in the ploidy level between control and in vitro propagated plants.

Viral disease symptoms were investigated in the field grown Longiflorum hybrid cultivars, and the damages caused by infection with Lily mottle virus (LMoV) and Cucumber mosaic virus (CMV) were assessed by comparing growth of plants produced from seeds of Longiflorum hybrid cultivar both infected by artificial inoculation and free from infection with theses viruses. Dominant symptom caused by spotaneous infection with LMoV and CMV in the field was mottle combined with chlorotic stripe on leaves. LMoV developed brownish necrotic lesion on floral leaves. The incidence of viral disease by mixed infection with LMoV, CMV or Lily symptomless virus (LSV) in the filed grown Longiflorum hybrid cultivar, cultivated for more than 6 years, was 80 to 84 percent. In comparison with virus-free plants, plants doubly infected with CMV and LMoV by artificial inoculation decreased stem length by 14 percent and fresh weight by 38 percent. In conclusion, flower quality and the stem length of Longiflorum hybrid cultivar were affected by LMoV and CMV infection.

This experiment was carried out to establish the screening methods for the selection of Zantedeschia aethiopica cultivars resistant to calla lily soft rot caused by Pectobacterium carotovorum subsp. carotovorum. A center of leaf and petiole tissues of four Z. aethiopica cultivars were wounded and inoculated with three different inoculum concentrations, $1{\times}10^7cfu/mL$, $1{\times}10^8cfu/mL$, and $1{\times}10^9cfu/mL$, of EccNHRI-21 isolate. And they were evaluated at 4, 18, and 26 hours after inoculation. The lesion of maceration was developed well in the leaf tissues inoculated with $1{\times}10^9cfu/mL$ concentration. And evaluation of resistance in 18 hours after inoculation was correlated with field resistance positively. Using this method, ten Z. aethiopica commercial cultivars and four wild types were screened. 'Crowbrough' and 'White Cutie' were selected as highly resistant genotypes and 'Mont Blanc' and 'Silky White' as resistant genotypes. 'Wedding March' and 'Kiwi Blush' were evaluated as moderate resistant and the others including 'Childsiana' were susceptible. And all of four wild types were evaluated as more than moderate resistant. In spite of control through cultural, biological and chemical methods, the use of resistant cultivars is most efficient to overcome calla lily soft rot. This newly developed screening method was helpful for breeding new varieties resistant to calla lily soft rot.

In order to determine the relationship between water content and flower qualities of oriental hybrid lily cv. 'Siberia' cut flower, flowers were subjected to dry and cold storage at $5^{\circ}C$ for 3, 6, and 12 days and subsequently exposed to ambient temperature ($26^{\circ}C$) in bottles with water for up to 16 days. Flowers stored at $22^{\circ}C$ in dry condition for 3 days were used as the control. Changes in fresh weight, moisture content, water balance, flowering stages, osmolality and vase life of cut flowers were observed. Flowers treated with cold and dry storage had higher moisture content compared to control sample. However, this trend was evident only for 3-day cold and dry stored sample during the whole storage period. The fresh weight of cut flowers increased gradually when the samples were transferred to ambient temperature in water bottles and then declined steadily before reaching the peak in between 6-8 days of vase life. However, the changes of fresh weight of control sample were substantially faster than samples pre-treated with cold and dry storage. This was also correlated with the water balance of cut flower as it reached the minus (-) value in 6-8 days of vase life at ambient temperature. Cut lily flowers showed high osmolality values corresponding with the duration of dry storage regardless of low or higher temperature. However, osmolality had no effect on vase life since flower stem absorbed water rapidly at the end of dry storage period. Our vase life results suggest that cold and dry storage of lily cut flowers for a certain period could ensure longer vase life at ambient temperature. It was observed that prolonging the storage period at cold and dry condition for more than a week significantly increased bud abortion, reduced longevity of flowers and reduced the vase life of cut flowers. On the other hand, the shorter cold and dry storage treatment delayed the bud opening and senescence of the flowers, thus, slowering the normal maturation and aging. Results indicated that dry and cold storage at $5^{\circ}C$ for 3 days was effective in maintaining and preserving overall quality and vase life at ambient condition of oriental hybrid lily cut flowers.
Packaging of export cut lilies reduces physical damage during distribution like dropping, shock, vibration, compression and serves to protect from necrosis, microbial contamination and decomposition. Study on which packaging materials must be selected is necessary because it serves to direct effect maintaining quality and the degree of freshness. newspaper, perforated OPP film, Oriented Nylon film (ON), punched OPP film, OPP films are used in this study. Plant senescence of lilies was happened rapidly during storage at $25^{\circ}C$ rather than at $5^{\circ}C$. Also, water absorption, fresh weight at during storage $25^{\circ}C$ rather than at $5^{\circ}C$ In addition, fresh weight of lilies storage at $25^{\circ}C$ appears higher than at $5^{\circ}C$ all of the packaging materials except OPP because water absorption of lily storage at $5^{\circ}C$ appears higher than at $25^{\circ}C$, thereby suppressing the fresh weight reducing Also, effect on biological activity of lilies from shape of packaging material appears higher during storage at $5^{\circ}C$ than at $25^{\circ}C$. Compared with each packaging materials, newspaper packaging is the smallest effect on changes in biological activity of cut lilies during storage. Thus, the results demonstrate that flowering date are affected mainly by the storage temperature rather than packaging materials.
Effects of traditional fumigation (methyl bromide; MB) and combined fumigation (methyl bromide/phosphide; $MB/PH_3$) on the disinfestation, and phytotoxicity and longevity of cut flowers (lily and rose) were investigated. Combined fumigation with low concentrations of $7/3g/m^3\;(MB/PH_3$) at $10^{\circ}C$ immediately after transportation from harvested place was found to be much more effective in disinfestation of aphid, mite, and thrips than fumigation with $MB\;48g/m^3$ only at $20^{\circ}C$ after simulated transportation for 2 days. Quality and longevity of cut flowers measured after fumigation were better in flowers treated by combined fumigation ($7/3g/m^3,\;MB/PH_3$) at $10^{\circ}C$ for 2.5-3 hrs, irrespective of pretreatment with pulsing solution immediately after transporation, even though phytotoxicity varied with cut flowers fumigated. On the other hand, fumigation with $MB\;48g/m^3$ only at $20^{\circ}C$ after simulated transportation for 2 days showed tendency of enhanced insect disinfestation, but caused phytotoxicity to both lily and rose, regardless of pretreatment and moreover, reduced the longevity of the cut flowers.
