Separation of soil Organic Debris using Sucrose-ZnCl2 Density Gradient Centrifugation

  • The active fraction of soil organic matter, which includes organic debris and light organic fraction, plays a major role in nutrient cycling. In addition, particulate organic matter is a valuable index of labile soil organic matter and can reflect differences in various soil behaviors. Since soil organic matter bound to soil mineral particles has its density lower than soil minerals, we partitioned soil organic matter into debris ($<1.5g\;cm^{-3}$), light fraction ($1.5-2.0g\;cm^{-3}$), and heavy fraction ($>2.0g\;cm^{-3}$), based on high density $ZnCl_{2-}$ sucrose solutions. Generally, partitioned organic bands were clearly separated, demonstrating that the $ZnCl_{2-}$ sucrose solutions are useful for such a density gradient centrifugation. The available gradient ranges from 1.2 to $2.0g\;cm^{-3}$. Although there was not a statistically meaningful difference in organic debris and organomineral fractions among the examined soils, there was a general trend that a higher content of organic debris resulted in a higher proportion of light organomineral fraction. In addition, high clay content was associated with increased fraction of light organomineals. Partitioning of soil organic carbon revealed that carbon content is reduced in the heavy fraction than in the light fraction, reflecting that the light fraction contains more fresh and abundant carbon than the passive resistant fraction. It was also found that carbon contents in the overall organic matter, debris, light fraction, and heavy fractions may differ considerably in response to different farming practices.

Comparison Analysis on Characteristics and Components of Various Waste Plastic Pyrolysis Oils by Vacuum Distillation Method (감압증류를 통해 분획된 폐플라스틱 열분해유의 특성평가와 구성성분 비교분석 연구)

  • Globally, the amount of waste plastics has been occurring to environmental problems. As a result, it is necessary to research methods that utilize waste plastic pyrolysis oil (WPPO) produced by pyrolysis. One such method being studied is utilizing WPPO as a naphtha feedstock. In this study, five types of WPPO were analyzed to determine whether they can be used as raw materials for naphtha. Because of their wide boiling point range, the WPPOs were fractionated into light and heavy fractions through vacuum distillation, and the separation and purification techniques were analyzed using GC-VUV to determine the content of paraffin, olefin, and other compounds. All WPPOs showed high olefin content regardless of the source and fraction. Aromatic and paraffin content varied depending on the source, and oxygen and other compounds also varied significantly by source and fraction. In addition, the light fraction showed a carbon distribution similar to that of naphtha, whereas the heavy fraction showed a carbon distribution of C11 ~ C14. In conclusion, additional processes and raw material selection are required to utilize waste plastic pyrolysis oil as a raw material for naphtha.

Soil Organic Matter Fractions in Upland Soil under Successive Application of Animal Manure Composts (밭 토양에서 가축분퇴비 연용시 토양 유기물 Fraction)

  • The objective of this study was to analyze change of soil organic matter fraction from a range of livestock manure compost that differed in their total C, N content and C quality, to gain a better understanding of their influence on soil organic matter. The chicken (CHM), pig (PIM), and cow (COM) manure-based composts, and manure-sawdust-based composts (CHMS, PIMS, and COMS) were applied annually to the upland soil with $3Mg\;C\;ha^{-1}$ during 4 years. After 4 years, the soil carbon content was increased to 25-30 and 40% for manure-based compost and manure-sawdust-based compost compared to control. In the all treatments, the content of light fraction C was sharply increased after second year. The content of light fraction C in the manure-sawdust-based compost was higher than in the manure-based compost. By contrast, the content of heavy fraction C was higher in the manure-based compost than in the manure-sawdust-based compost. These results indicate that stabilization of carbon applied from microbiological process was faster in the manure-based compost than in the manure-sawdust-based compost.

Studies on the Isolation and Immunochemical Properties of SIgA from Human and Bovine Milk (인유(人乳) 및 우유(牛乳)로부터 Secretory Immunoglobulin A의 분리(分離) 및 면역화학적(免疫化學的) 특성(特性)에 관(關)한 연구(硏究))

  • These experiments were carried out to isolate SIgA from human and bovine milk. The immunochemical properties of SIgA from human and bovine milk were examined by Gel filtration, DEAE and SDS-PAGE. Double Immunodiffusion, and Immunoelectrophoresis. The results obtained are as follows: 1. Human SIgA was purified from colostrum of Korean women by repeated gel filtration on Sephadex G-200 and Sepharose 6B, but bovine SigA was not cleary purified from bovine colostrum of Holstein cows by anion exchange chromatography using DEAE-Sephadex A-50 and gel filtration on Sephadex G-200 and Sepharose 6B. 2. The immunochemical properties of fractions from gel filtration on the Sephadex G-200 and Sepharose 6B column as assessed by Immunoelectrophoresis and double Immunodiffusion to identify the presence of IgM in first peak fraction, and the presence of pure SIgA in second peak fraction. However, Bovine SigA rich fraction from bovine colostrum of Holstein cows contained a large amount of $IgG_1$-dimer in addition to SIgA. 3. The fragments of reduced bovine colostrum SIgA rich fraction were estimated to have molecular weights of secretory component, heavy chain and light chain (75,000-80,000, 50,000-60,000, 25,000-27,000 daltons) by SDS-PAGE, respectively. Those were similar to the molecular weight of reduced SIgA from human milk.

Recovery of Lipids from Chlorella sp. KR-1 via Pyrolysis and Characteristics of the Pyrolysis Oil (Chlorella sp. KR-1 열분해에 의한 지질 회수 및 열분해 오일 특성 분석)

  • Lipids in microalgal biomass were recovered by using pyrolysis method. The pyrolysis experiments of two Chlorella sp. KR-1 samples, which have triglyceride contents of 10.8% and 36.5%, respectively were carried out at $600^{\circ}C$ to investigate the effects of lipid contents in the cells on the reaction characteristics. The conversion and liquid yield of the lipid-rich sample were higher than those of the lipid-lean sample since its carbon to hydrogen ratio was low. There were low molecular weight organic acids, ketones, aldehydes and alcohols in the liquid products from both KR-1 samples, but the pyrolysis oil of the lipid-rich sample was abundant in free fatty acids, particularly palmitic acid, oleic acid and stearic acid while the content of nitrogen containing organic compounds was low. The microalgal pyrolysis oil had two layers composed of the light hydrophobic fraction and the heavy hydrophilic fraction. The light fraction might be originated from triglycerides and the heavy fraction might be from carbohydrates and proteins. In the light fraction of the liquid products, there were considerable linear alkanes such as pentadecane and heptadecane as well as free fatty acids, implying that deoxygenation reaction including decarboxylation was occurred during the pyrolysis. The yield of the liquid products from the pyrolysis of the KR-1 sample having triglyceride content of 36.5% was 56.9% and the light fraction in the liquid products was 68.2%. Also more than 80% of the light fraction was free fatty acids and pure hydrocarbons, thus showing that most triglycerides could be extracted in the form of suitable raw materials for biofuels.

  • Analysis and separation of constituent materials of old car have been performed by using the industrial shredding line. For this aim, three old cars made by domestic automobile manufacturers, Sonata II, Sephia and Prince were chosen and delivered in pressed form without engine, tires and doors, etc. Shredding line was substantially composed of pre- and main-shredder. cyclone, magnetic separation, eddy current separation and man-power separation. From the separation of shredder products, iron scrap was observed to be the major material of old car accounting for 60.1 % of total weight and non-ferrous metals involving Al, Cu and Zn, etc. were about 2%. Light fluff, about 90% of total fluff product, was comprised with plastic, fiber and sponge, etc. and the fraction of 5 cm undersize in light fluff was 70.5%. In case of heavy fluff, however. rubber and plastic were found to be the major constituent materials of it. Among the constituent materials of fluff, plastic showed the highest calorific value, more than 10,000 cal/gr and leather and rubber showed relatively high chlorine content, 10.3 and 2.55 wt%, respectively.

Process Development of Pyrolysis Liquefaction for Waste Plastics (폐플라스틱의 열분해 유화기술 개발)

  • The target of this work was the process development of demonstration plant to produce the high quality alternative fuel oil by the pyrolysis of mixed plastic waste. In the first step of research, the bench-scale units of 70 t/y and the pilot plant of 360 t/y had been developed. Main research contents in this step were the process performance test of pilot plant of 360 ton/year and the development of demonstration plant of 3,000 t/y, which was constructed at Korea R & D Company in Kimjae City. The process performance of pilot plant of 360 t/y showed about 80% yield of liquid product, which was obtained by both light gas oil(LGO) and heavy gas oil(HGO), The boiling point range distribution of LO product that was mainly consisting of olefin components in PONA group appeared at between that of commercial gasoline and kerosene. On the other hand, HO product was mainly paraffin and olefin components and also appeared at upper temperature distribution range than commercial diesel. Gas product showed a high fraction of $C_3\;and\;C_4$ product like LPG composition, but also a high fraction of $CO_2$ and CO by probably a little leak of process.

  • Modern passenger cars with diesel engines are equipped with DOC(diesel oxidation catalyst) for the purpose of reducing HC and CO in the exhaust stream. Cold start exhaust emissions pose troubles here as on gasoline engine vehicles. As a result, some of the diesel passenger cars roll off todays the assembly lines with WCC(warm-up catalytic converter). Oxidation characteristics of the particulates in WCC is analyzed in this study by EEPS(engine exhaust particulate size spectrometer). The maximum number of PM is found to come out of WCC in sizes near 10nm when an HSDI diesel engine is operated under the conditions of high speed and medium to heavy load. When the temperature of the WCC exceeds $300^{\circ}C$, the number of PM smaller than 30 nm in diameter sharply increases upon passing through the WCC. Total mass of emitted PM gets reduced downstream of the WCC under low speed and light load conditions due to adsorption of PM onto the catalyst. Under conditions of high speed and medium to heavy load, the relatively large PM shrink or break into fine particles during oxidation process within the WCC, which results in more mass fraction of fine particles downstream of the WCC.

  • Broadband impedances for the 3-GeV pohang light source-II (PLS-II) storage ring have been numerically estimated using a full three dimensional finite-difference time-domain code, CST particle studio. The total broadband impedance of all the vacuum components was estimated as 0.256 ohm, which is a small fraction of the total machine impedance budget.

Characterization of Wintertime Atmospheric Aerosols in Seoul Using PIXE and Supplementary Analyzers

  • Particle Induced X-ray Emission (PIXE) and Elemental Analysis Syztem (EAS) were applied to the investiga-tion of the Characteristics and sources of wintertime atmospheric aerosols in Seoul. Atmospheric aerosols were collected by both fine and coarse fractions using a two-stage filter pack sampler from Kon-Kuk university during the winter season of 1999. PIXE was applied to the analysis of the middle and heavy elements with atomic numbers greater than 14(Si) and EAS was applied to the measurement of the light elements such as H, C and N. The fact that 64.2% of mass of fine particles in Seoul consists of the light elements (N, C , and H) suggests that the measurement of light elements is extremely important. The average mass concentration is Seoul was 38.6$\mu\textrm{g}$m(sup)-3. Elements such as Ca, Fe, Mg, and Ti appeared to have very low Fine/Coarse ratios(0.1∼0.4), whereas che-mical components related to anthropogenic sources such as Br, V, Pb, and Zn were observed to accumulate in the fine fraction. In the Asian Dust Storm(ADS) event, the concentation of soil components increased dramatically. Reconstruction of the fine mass concentrations estimated by a newly revised simple model was fairly in good agreement with the measured ones. Source identification was attempted using the enrichment factor and Pearsons coefficient of correlation. The typical elements derived from each source could be classified by this method.
