Light and Scanning Electron Microscope Observatt-ons on Sexual Dimorphism in Pupa of Mullberry silkworm, bombyx Mori Linn (Lopidoptera : Bombycidae)

  • Under Light and Scanning electron microscope, the pupal morphology of mulberry silkworm Bombyx mori Linn. revealed the prothorax and metathorax, well developed mesothorax, less defined last pair of sporacle, well exposed prothoracic femora and wing pads approaching the anterior margin of Ab III. The important sex separating characters viz, wegiht, antennal elevations, intersegmental lines and genings have discussed. Further, two separate openings bursa copulatrix and ovipositional opening were observed, performing different functions in abult moth.

Polarization State of Scattered Light in Apertureless Reflection-mode Scanning Near-Field Optical Microscopy

  • We studied the polarization state in an apertureless scanning near-field microscopy (a-SNOM) operating in reflection mode by using three-dimensional Finite-difference Time-domain (FDTD) method. As a result, the electric field around tip apex in the near-field region enhanced four times stronger than the incident light for ppolarization when the tip-sample separation was 10 nm. We find that the p- and s-polarization state is maintained for the scattered light when the probe is perpendicular to the sample. When the probe is not perpendicular to the sample, the polarization state of scattered light will rotate an angle that equals to the inclination angle of probe with p-polarization illumination. On the other hand, the polarization state will not rotate with s-polarization illumination.

Biomineralization on the stalk of the eustigmatophyte Pseudocharaciopsis (Eustigmatophyceae)

  • The stalks of the eustigmatophyte $Pseudocharaciopsis$ $minuta$ were examined by light and scanning electron microscopy. Light microscopy revealed orange-red granules at the base of the stalk. Energy dispersive X-ray microanalysis of the bases indicated that they were mainly composed of manganese. Manganese has not been previously reported from eustigmatophytes. This study indicates that the Eustigmatophyceae needs further study into many aspects of the biology.

Recent Developments in Correlative Super-Resolution Fluorescence Microscopy and Electron Microscopy

  • The recently developed correlative super-resolution fluorescence microscopy (SRM) and electron microscopy (EM) is a hybrid technique that simultaneously obtains the spatial locations of specific molecules with SRM and the context of the cellular ultrastructure by EM. Although the combination of SRM and EM remains challenging owing to the incompatibility of samples prepared for these techniques, the increasing research attention on these methods has led to drastic improvements in their performances and resulted in wide applications. Here, we review the development of correlative SRM and EM (sCLEM) with a focus on the correlation of EM with different SRM techniques. We discuss the limitations of the integration of these two microscopy techniques and how these challenges can be addressed to improve the quality of correlative images. Finally, we address possible future improvements and advances in the continued development and wide application of sCLEM approaches.

조직내 Cryptococcus neoformans의 전자현미경적 관찰 (Ultrastructure of Cryptococcus neoformans in the Skin Tissue)

  • 전신성 Cryptococcosis로 판명된 4세 남아의 피부 생검조직에서 발견된C. neoformans의 미세구조를 광학현미경 관찰과 함께 보고 하였다. 광학현미경 관찰에서는 각종 만성 염증 세포의 침윤과 함께 다수의 구형의 organism을 관찰할 수 있었다. 전자현미경적으로도 난원형의 세포가 gelatinous 또는 filamentous한 capsular material에 둘러 싸여 단세포 혹은 여러개의 세포가 모여 있는 상태로 관찰되었으며 budding중인 상태의 것도 관찰되었다. 세포막은 고전사 밀도의 여러층으로 구성되어 있었으며 한개의 핵과 핵인이 관찰되었고 세포질내에는 mitochondria, ribosome, lipid bodies, vacuole등과 함께 plasma membrane의 infolding으로 형성된 mesosome-like structure도 관찰되었다.

고양이에서 유발된 장액성중이염시 중이점막의 미세구조에 관한 연구 (Ultrastructure of the tympanic mucosa in experimentally produced serous otitis media)

  • Serous otitis media is closely related with auditory tube function, but its etiology and pathogenesis are not clearly defined yet. So we tried to prove the theory of hydrops ex vacuo via the experimental study with cats by means of obstructing the pharyngeal orfice of the auditory tube and observe the serial changes in tympanic mucosa through light and electron microscopy. The results are as follows; 1. We confirmed the production of serous otitis media with auditory tube obstruction and have a new understanding of auditory tube function in middle ear aeration. 2. The effusion in serous otitis media was produced from the next day of experiment and increased till the fourteenth day, but decreased after the spontaneous perforation of ear drum. 3. Through the light microscopy, we observed the increasement of the sercretory cells including goblet cells, epithelial hyperplasia, capillary proliferation and invasion of inflammatory cells. 4. Through the elctron microscopy, we observed the protrusion of secretory cells, blobs in cilia, loss of cilia, increasement of vesicles, vacuoles and dense bodies in ciliated cells and Invasion of inflammatory cells. With above results, we concluded that aeration through auditory tube is the most important factor in serous otitis media and presumed the effusion was secreted by secretory cells.

Monocrotaline을 투여한 백서 폐의 초기 혈관병변에 관한 주사전자현미경적 관찰 (Scanning Electron Microscopic Observation on Early Vascular Lesion in Rat Lung Administered with Monocrotaline)

  • An experimental study was performed to observe the early effects of monocrotaline on pulmonary vascular system by means of light microscopy and scanning electron microscopy, attempting to expore the mechanism behind the process of pulmonary hypertension. Experimental animal(Sprague-Dawley male rats ; 150-200g B. W.) were intra-peritoneal administered with 100mg/kg B. W. monocrotaline. Authors observed light microscopically various gradational increase of wall thickness in pulmonary muscular and non-muscular arteries in duration from 2 weeks to 5 weeks after monocrotaline administration and the changes were more sever in the latter than the former. The scanning electron microscopy shows severe and diffuse endothelical cell swelling, microvilli and microbleb formation since 1 hour after monocrotaline administration and during the course, after 5 hours the severity of endothelial cell damage was prominent with presence of fibrin, webs, platelet thrombi and white cell adherence. It was concluded that the monocrotaline primarily induced severe and diffuse endothelial cell damage of pulmonary arteries and laterly added the participation of platelets, which attributed to the pathogenesis of monocrotaline induced pulmonary vascular lesions in relation to pulmonary hypertension.

Nano-Resolution Connectomics Using Large-Volume Electron Microscopy

  • A distinctive neuronal network in the brain is believed to make us unique individuals. Electron microscopy is a valuable tool for examining ultrastructural characteristics of neurons, synapses, and subcellular organelles. A recent technological breakthrough in volume electron microscopy allows large-scale circuit reconstruction of the nervous system with unprecedented detail. Serial-section electron microscopy-previously the domain of specialists-became automated with the advent of innovative systems such as the focused ion beam and serial block-face scanning electron microscopes and the automated tape-collecting ultramicrotome. Further advances in microscopic design and instrumentation are also available, which allow the reconstruction of unprecedentedly large volumes of brain tissue at high speed. The recent introduction of correlative light and electron microscopy will help to identify specific neural circuits associated with behavioral characteristics and revolutionize our understanding of how the brain works.

인체 피부에 기생한 Leishmania tropica의 전자현미경적 관찰 (Ultrastructural Observations of a Human Cutaneous Leishmaniasis)

  • 중동지방에 다녀온 48세의 한국인 남자 피부에 발생한 leishmania증 1예에 대하여 광학 및 전자현미경적 관찰을 하여 문헌 고찰과 아울러 보고하였다. 광학현미경적으로는 조직구 침윤을 주로 하는 만성 육아종성염증반응을 보였고 Giemsa염색상 다수의 leishmania충체를 조직구내에서 관찰할 수 있었다. 전자현미경적으로는 난원형의 단세포 또는 분열중인 충체가 조직구내에 존재하였고 충체는 이중막으로 싸였으며 그 바로밑에 microtubule이 배열되고 세포질내에는 신장된 mitochondrion내에 kinetoplast가 존재하고 그 전방에 flagella가 위치하였으며 기타 다수의 ribosome과 드물게 Golgi complex등을 관찰할 수 있었다. 이와같은 특징은 Leishmania tropica의 promastigote stage와 일치하였다. Leishmania증은 원칙적으로 열대병이나 열대지방과의 접촉이 빈번함에 따라 앞으로 우리나라에서도 보다 많은 증예가 발생할 것으로 사료된다.

  • 우기자혹응애 (Eriophyes kuko KISHIDA)의 기생에 의하여 구기자 나무 (Lycium Chinens MILL) 잎에 형성되는 흑 (mite gall) 의 행태적 변화를 광학현미경 및 전자현미경으로 관찰한 결과는 다음과 같다. 1. 광현적 관찰 : 구기자혹응애가 구기자나무의 어린잎의 뒷면 표피세포에 침입되면 먼지 표피세포가 이상비대를 하면서 혹이 형성되기 시작한다. 혹이 성장함에 따라 붕상조직과 해면상조직의 세포도 이상비대를 하게되고, 점차적으로 각 조직의 세포는 이상비대로 인한 불규칙한 형태적 변화를 일으켜 조직의 구별이 힘들게 된다. 이상비대한 혹세포의 핵은 건전세포의 핵보다 크며, 혹이 성장함에 따라서 핵의 크기도 커진다. 2. 전현적 관찰 : 혹이 성장함에 따라 혹세포의 mitochondria, golgi-apparatus. plastid 등은 퇴 화 및 붕괴되고, 특히 큰 흑의 이상비대한 혹세포에는 성상체 (star body) 와 환상 (ring form) 의 구조등이 나타난다.
