Cytohistological Study of Abnormal Cell Division of Arabidopsis Stem Infected with Geminivirus (Geminivirus에 감염된 Arabidopsis 줄기의 이상세포분열에 관한 세포조직학적 연구)

  • The internal structures of Arabidopsis thaliana infected with beet curly top virus (BCTV) were studied by light microscopy. Hyperplasia was observed in the inflorescence stems of Arabidopsis thaliana ecotype Sei-O at 2 weeks after BCTV-Logan inoculation and callus was induced on symptomatic tissues at 4 weeks after virus inoculation. The infection processes were revealed as follows: hyperplasia of phloem tissue, necrosis of hyperplastic phloems, lacuna formation of necrotic tissues, elongation and enlargement of cortex and epidermal cells surrounding the lacuna formed phloem tissues, induction of cell division in the enlarged cortex and epidermal cells, and induction of callus tissue. Callus formation on Arabidopsis was caused by the virus infection, and virus inclusion body was observed in both phloem and callus tissue by azure-A staining.

  • The first laboratory culture of host-parasite system of Prorocentrum minimum- Amoebophrya sp. was established by single cell isolation method. Here, we report the life cycle stages of the parasitic dinoflagellate. Amoebophrya sp. of the red tide dinoflagellate P. minimum as observed by light and epifluorescence microscopy. Infections developed inside the nucleus of P. minimum. The trophont developed to occupy almost all the intracellular space of the host at its late stage. The fully developed trophont finally ruptured through the host cell. “Vermiform stage”, the free-swimming extracellular lift cycle stage is followed by another stage for the sudden release of many individual dinospores. Our laboratory strain of the host-parasite system for P. minimum, a causative species fur the huge red tides in spring and summer in Korean coastal waters, could be a useful living material for the in situ biological control of harmful algal blooms.

Production and Characterization of Beta-lactoglobulin/Alginate Nanoemulsion Containing Coenzyme Q10: Impact of Heat Treatment and Alginate Concentrate

  • The aims of this research were to produce oil-in-water ${\beta}$-lactoglobulin/alginate (${\beta}$-lg/Al) nanoemulsions loaded with coenzyme $Q_{10}$ and to investigate the combined effects of heating temperature and alginate concentration on the physicochemical properties and encapsulation efficiency of ${\beta}$-lg/Al nanoemulsions. In ${\beta}$-lg/Al nanoemulsions production, various heating temperatures (60, 65, and $70^{\circ}C$) and alginate concentrations (0, 0.01, 0.03, and 0.05%) were used. A transmission electron microscopy was used to observe morphologies of ${\beta}$-lg/Al nanoemulsions. Droplet size and zeta-potential values of ${\beta}$-lg/Al nanoemulsions and encapsulation efficiency of coenzyme $Q_{10}$ were determined by electrophoretic light scattering spectrophotometer and HPLC, respectively. The spherically shaped ${\beta}$-lg/Al nanoemulsions with the size of 169 to 220 nm were successfully formed. The heat treatments from 60 to $70^{\circ}C$ resulted in a significant (p<0.05) increase in droplet size, polydispersity, zeta-potential value of ${\beta}$-lg/Al nanoemulsions, and encapsulation efficiency of coenzyme $Q_{10}$. As alginate concentration was increased from 0 to 0.05%, there was an increase in the polydispersity index of ${\beta}$-lg/Al nanoemulsions and encapsulation efficiency of coenzyme $Q_{10}$. This study demonstrates that heating temperature and alginate concentration had a major impact on the size, polydispersity, zeta-potential value and encapsulation efficiency of coenzyme $Q_{10}$ in ${\beta}$-lg/Al nanoemulsions.

Analysis of Soot Particle Morphology Using Rayleigh Debye Gans Scattering Theory (RDG 산란 이론을 이용한 그을음 탄소 입자의 형상 분석)

  • Soot particles generated by fossil fuel combustion normally have fractal morphology with aggregates consisting of small spherical particles. Thus, Rayleigh or Mie elastic light scattering theory is not feasible for determining the fractal properties of soot aggregates. This paper describes a detailed process for applying Rayleigh-Debye Gans (RDG) scattering theory to effectively extract the morphological properties of any nano-scale particles. The fractal geometry of soot aggregates produced from an isooctane diffusion flame was observed using ex situ transmission electron microscopy (TEM) after thermophoretic sampling. RDG scattering theory was then used to analyze their fractal morphology, and various properties were calculated, such as the diameter of individual soot particles, number density, and volume fraction. The results show indiscernible changes during the soot growth process, but a distinct decreasing trend was observed in the soot oxidation process. The fractal dimension of the soot aggregates was determined to be around 1.82, which is in good agreement with that produced for other types of fuel. Thus, it can be concluded that the value of the fractal dimension is independent of the fuel type.

Effect of Heat Treatment on Mechanical Reliability of Solder Joints in LED Package (LED 패키지 솔더 접합부의 기계적 신뢰성에 미치는 열처리의 영향)

  • We studied the effect of heat treatment on the microstructures and mechanical strength of the solder joints in the Light Emitting Diode (LED) packages. The commercial LED packages were mounted on the a flame resistance-4 (FR4) Printed Circuit Board (PCB) in the reflow process, and then the joints were aged at $125^{\circ}C$ for 100, 200, 300, 500 and 1000 hours, respectively. After the heat treatment, we measured the shear strength of the solder joints between the PCB and the LED packages to evaluate their mechanical property. We used Pb-free Sn-3.0Ag-0.5Cu solder to bond between the LED packages and the PCBs using two different surface finishes, Electroless Nickel-Immersion Gold (ENIG) and Electroless Nickel-Electroless Palladium-Immersion Gold (ENEPIG). The microstructure of the solder joints was observed by a scanning electron microscope (SEM). (Cu,Ni)6Sn5 intermetallic compounds (IMCs) formed between the solder and the PCB, and the thickness of the IMCs was increased with increasing aging time. The shear strength for the ENIG finished LED package increased until aging for 300 h and then decreased with increasing aging time. On the other hand, in the case of an ENEPIG finished LED package, the shear strength decreased after aging for 500 h.

Methodologies for Inhalation Exposure Assessment of Engineered Nanomaterial-containing Consumer Spray Products (분사형 소비자 제품 중 나노 물질의 흡입 노출 평가 방법)

  • Objective: This study aimed to review the methodologies for evaluation of consumer spray products containing engineered nanomaterials (ENM), particularly focusing on inhalation exposure. Method: Literature on the evaluation methods for aerosolized ENM exposure from consumer spray products were collected through academic web searching. Common methodologies used in the literature, including research reports and academic articles, were also introduced. Results: The number of ENM-containing products have shown a considerable increase over recent years, from 54 in 2005 to 1,827 in 2018. Currently there is still discussion over the existing regulations with regard to product safety. Analysis of both ENM suspensions in the products and their aerosols is important for risk assessment. Comparison between the phases suggests how the size and concentration of particles change during the spray process. To analyze the ENM suspensions, dynamic light scattering, electron microscopy techniques, and inductively coupled plasma with mass spectrometry were used. In the aerosol monitoring, direct-reading instruments have been used to monitor the aerosols and conventional active sampling is used together to supplement the lack of real-time monitoring. There are also some models for estimating inhalation exposure. These models may be used to estimate mass exposure to nanomaterials contained in consumer products. Conclusion: Although there is no standardized method to evaluate ENM exposure from consumer products, many concerns about ENM have emerged. Every potential measure to reduce exposure to ENM from spray product use should be implemented through a precautionary recognition.

Effect of High Voltage Pulsed Electric Fields on Extraction of Purple Sweet Potato Pigment (자색 고구마 색소의 추출에 미치는 고전압 펄스 전기장의 영향)

  • High voltage pulsed electric fields (PEF) is a promising technology for the nonthermal extraction of effective components from biological materials. Plant cells were ruptured with PEF at ambient or refrigerated temperature for a short treatment time of second or microsecond. Treatments of coarsely ground purple sweet potato (PSP) with PEF(30 kV/cm, 500 Hz) resulted in maximum extraction yield of 65% as compared with 45-50% for control. An increase in electric field strength (from 10kV/cm to 35kV/cm) and frequency (from 100Hz to 500Hz) resulted in increased amount of extracted pigments, but treatment time is not affected on pigment extraction. Starch granules were not detected and large intracellular spaces were visible between the cells on light and scanning election microscopy of PEF treated PSP. This result suggests that PEF have potential to use on extraction of pigments from plant cells.

Synthesis and characterization of CaZrO3:Pr3+ phosphor by skull melting method (스컬용융법에 의한 CaZrO3:Pr3+ 형광체 합성 및 특성분석)

  • Ca1-xZrO3:xPr phosphor with perovskite structure was successfully synthesized by using skull melting method. The crystal structure, morphology and optical properties of synthesized phosphor were characterized using X-ray diffraction (XRD), scanning electron microscopy (SEM), ultraviolet fluorescence reaction and photoluminescence. The XRD results indicated that single crystals of CaZrO3:Pr3+ belongs to orthorhombic perovskite system. The synthesized phosphor could be excited by UV light (254 nm) and the emission spectra results indicated that green luminescence of CaZrO3:Pr3+ due to charge transfer transition 3P03H4, 3P13H5 and 3P03H5 at 506, 536 and 548 nm was dominant.

Preparation of Hydrated Liquid Crystalline Vesicles Containing High Content of Ceramide using a Solubilizer (가용화제를 활용한 세라마이드 고함량의 수화 액정형 베시클 개발)

  • Hydrated liquid crystalline vesicles containing a high content of ceramide were prepared by constituting an optimal composition in which ceramides can be mutually self-associated with phospholipid and cholesterol. From the result of manufacturing various vesicles with different component composition, when the edge activator sodium deoxycholate (SDOC) and the solubilizer PEG-60 hydrogenated castor oil (HCO 60) were mixed to form vesicles, the smallest nano-sized particles were produced and the vesicle dispersion solution was weakly acidic and maintained the most stable state. In addition, it was confirmed through polarized light microscopy and thermal analysis that the addition of SDOC and HCO 60 had an effect on the inhibition of crystallinity of lipid components such as ceramide. The stability of the vesicle dispersion solution was maintained without change in appearance and viscosity even after long-term storage at high temperature for eight weeks.
