Development of a Total Internal Reflection Fluorescence (TIRF) Microscopy for Precise Imaging the Drying Pattern of a Sessile Droplet (고착 액적 증발면의 정밀 관측을 위한 전반사 형광 현미경 기법 개발)

  • Compared to epifluorescence(EPI) microscopy which captures fluorescence from the entire depth of sample, total internal reflection fluorescence(TIRF) can selectively visualize only a single surface of it. TIRF uses a thin evanescent field generated by the total internal reflection of laser light on surface. However, conventional TIRF system are designed for total internal reflection to occur at the upper surface of sample, making them unsuitable for sessile droplet imaging. We designed a TIRF system suitable for a sessile droplet imaging by utilizing slide glass as a lightguide. We presented the details for constructing the TIRF system using a prism, slide glass, air slit, and optical trap. Then, we compared the TIRF with EPI by imaging the droplet with fluorescent particles during its drying process. As a result, TIRF allows us to distinctly visualize the drying pattern on the bottom surface of droplet.

  • Nitrogen-doped $TiO_2$ particles have been successfully prepared using titanium tetraisopropoxide as the Ti source and urea as the nitrogen source. As-prepared nitrogen-doped $TiO_2$ was characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), Brunauer-Emmett-Teller method (BET) and ultraviolet-visible light (UV-vis) absorption spectra techniques. Photocatalytic degradation of Methylene Blue (MB) has been carried out in both solar light (UV-vis) and the visible region (${\lambda}=420nm$). Nitrogen-doped $TiO_2$ exhibits higher activity than the commercial $TiO_2$ photocalyst, particularly under visible-light irradiation because bandgap of nitrogen-doped $TiO_2$ becomes remarkably decreased.

  • 본 연구에서는 고분자를 사용하여 만든 유기EL소자인 PLED (Polymer Light Emitting Diode)의 제조공정 변화에 따른 소자성능을 연구하였다. PLED의 제작은 크게 ITO 기판 제작, 발광층 및 전극 증착 등의 공정으로 나누어진다. ITO 기판은 사진식각공정으로, 발광층의 증착은 스핀코팅법으로, 전극은 진공증착법으로 각각 제작하였다. 코팅 시 스핀속도 및 점도 조절을 통하여 발광층의 두께를 조절하였고, 스핀코팅 후 건조방법에 따라서 표면의 uniformity와 발광특성을 비교해 보았다. 실험결과 특정 두께에서 발광특성이 우수하게 나타나는 것을 확인할 수 있었다. 그리고 건조방법에 따라 발광층의 표면 uniformity에 차이가 있었으며, 표면 uniformity에 따라 diode의 I-V 특성 경향이 달리 나타났다.

Photoresponsive Behavior of Liquid-Crystalline Networks

  • Freestanding azobenzene-containing liquidcrystalline network (LCN) films. with macroscopic uniaxial molecular alignment were prepared by insitu photopolymerization. By polarizing microscopy, fiber-like structures aligned in one direction were observed. Furthermore, with a confocal laser scanning microscope (CLSM), it was confirmed that the fiber-like structures were formed even in the bulk of the LCNs. Upon UV light irradiation to cause trans-cis photoisomerization of the azobenzene molecules, the LCNfilms underwent a significant and anisotropic bending toward the irradiation direction of UV light. When the bent LCNfilms were exposed to Vis light, unbending of the LCN films immediately took place and the initial flat LCN films were restored. This bending and unbending behavior of the LCN films could be repeated just by changing the wavelength of the irradiation light. It was suggested that the bending was induced by an absorption gradient which produced a volume difference between the front surface area and the bulk of the network films.

  • Nickel oxide-doped ACF and $TiO_2$ composites (NiO/ACF/$TiO_2$) were prepared by a sol-gel method. The composite obtained was characterized by BET surface area measurements, X-ray diffraction, transmission electron microscopy and energy dispersive X-ray analysis. A methylene blue (MB) solution under visible light irradiation was used to determine their photocatalytic activity. Excellent photocatalytic degradation of the MB solution was observed using the $TiO_2$, Ti-ACF and NiO/ACF/$TiO_2$ composite under visible light.

  • In this paper, a new positioning method with incoherence of white light is described and practically implemented to attach VCR heads on a drum very accurately. This method utilizes the Michelson Interferometer which uses white light with short coherence length as the light source to generate interference fringes only in case the optical path difference is shorter than about 2.mu.m. The course position of VCR heads and the fine are determined by appearance and visibility of interference fringes, respectively. The appearance are detected by an image processing technique using FFT(Fast Fourier Transform).

Photocatalytic Activity of EG-TiO2 Composite for Various Dye Solutions Under UV Light and Visible Light

  • Expanded graphite (EG) is synthesized by chemical intercalation of natural graphite (NG) and rapid expansion at high temperature, with titanium n-butoxide (TNB) used as titanium source by a sol-gel method to prepare EG-TiO$_2$ composite. The performances of the prepared EG-TiO$_2$ composite are characterized by BET surface area measurement, scanning electron microscopy (SEM), X-ray diffraction patterns (XRD) and energy dispersive X-ray analysis (EDX). To compare the photocatalytic activities of the EG-TiO$_2$ composite, three kinds of dye solutions, methylene blue (MB), methylene orange (MO) and rhodamine B (RhB), and two kinds of light source, UV light and visible light (VL), are used. Comparing the results, it can be clearly seen that the degradation of all of the dye solutions under irradiation by UV light is much better than that under irradiation by visible light, and the decomposition of MB solution was better than that of both of MO and RhB solution.

  • The secretory granul cells in the midgut epithelium of Blattella germanica L. were observed by the electron microscope. These secretory granul cells contain many electron dense granules, and granules are about $200{\AA}$ in diameter respectively. It is easy to distinguish 3 different types of granul cells based on their shapes, location, and staining intensity: 1) The light secretory granul cells and their nucleus are both round form and a number of mitochondria, vacuoles, and other cell organelles appear in the cytoplasm. 2) The other kind of light secretory granul cells are small and oval form but ceil organelles are not well developed in the cytoplasm. This granul cell is surrounded by a few regenerative cells ('nidi'). 3) Dark secretory granul cells are cone shaped, well stained, and endoplasmic reticulum, ribosomes, and a lot of secretory granules are found in the cytoplasm. They are all located in the basal portion of the midgut epithelium.

Characterization of Wavelength Effect on Photovoltaic Property of Poly-Si Solar Cell Using Photoconductive Atomic Force Microscopy (PC-AFM)

  • We investigated the effect of light intensity and wavelength of a solar cell device by using photoconductive atomic force microscopy (PC-AFM). The $POCl_3$ diffusion doping process was used to produce a p-n junction solar cell device based on a Poly-Si wafer and the electrical properties of prepared solar cells were measured using a solar cell simulator system. The measured open circuit voltage ($V_{oc}$) is 0.59 V and the short circuit current ($I_{sc}$) is 48.5 mA. Also, the values of the fill factors and efficiencies of the devices are 0.7% and approximately 13.6%, respectively. In addition, PC-AFM, a recent notable method for nano-scale characterization of photovoltaic elements, was used for direct measurements of photoelectric characteristics in local instead of large areas. The effects of changes in the intensity and wavelength of light shining on the element on the photoelectric characteristics were observed. Results obtained through PC-AFM were compared with the electric/optical characteristics data obtained through a solar simulator. The voltage ($V_{PC-AFM}$) at which the current was 0 A in the I-V characteristic curves increased sharply up to 1.8 $mW/cm^2$, peaking and slowly falling as light intensity increased. Here, $V_{PC-AFM}$ at 1.8 $mW/cm^2$ was 0.29 V, which corresponds to 59% of the average $V_{oc}$ value, as measured with the solar simulator. Also, while light wavelength was increased from 300 nm to 1,100 nm, the external quantum efficiency (EQE) and results from PC-AFM showed similar trends at the macro scale, but returned different results in several sections, indicating the need for detailed analysis and improvement in the future.

The Inactivation Effects of UV Light on Bacteriophage f2 (박테리오파지 f2에 대한 자외광선의 살균효과)

  • The effects of ultraviolet light on bacteriophage f2 were investigated to determine the inactivation kinetics and its mechanism. The 260nm light showed a little higher inactivation rate than the one of 300 nm. In this work, our main concern was whether structural and/or conformational changes in the protein capsid could occur by UV irradiation. The inactivation for the first 20 minutes irradiation was rapid with a loss of about 4 logs and followed by a slower rate during the next 40 minutes with no survival noted in the samples irradiated for 90 minutes or longer. The structural change of the protein capsid was examined by optical spectroscopic techniques and electron microscopy. The absorption spectra of the UV irradiated phages showed no detectable differences in terms of the spectral shape and intensity from the control phage. However, the fluorescence emission spectroscopic data, i.e. 1) fluorescence quenching of tryptophan residues upon irradiation of 300 nm light, 2) enhancement of fluorescence emission of ANS (8-aniline-1-naphthalene sulfonate) bound to the intact phages compared to the one in the UV-treated phages, and 3) decrease of energy transfer efficiency from tryptophan to ANS in the UV-treated samples, presented remarkable differences between the intact and UV-treated phages. Such a structural alteration was also observed by electron microscopy The UV-treated phages appeared to be broken and empty capsids. Therefore, the inactivation of the bacteriophage f2 by UV irradiation is thought to be attributed to the structural change in the protein capsid as well as damage in the viral RNA by UV irradiation.
