Bioremediation of metal contamination groundwater by engineered yeasts expressing phytochelatin synthase (Phytochelatin synthase 발현을 통한 효모의 중금속 처리에 관한 연구)

  • Heavy metal contamination has been increased in aqueous environments near many industrial facilities, such as metal plating facilities, mining operations, and tanneries. The soils in the vicinity of many military bases are also reported to be contaminated and pose a risk of groundwater and surface water contamination with heavy metals. The biological removal of metals through bioaccumulation has distinct advantages over conventional methods; the process rarely produces undesirable or deleterious chemical byproducts, it is highly efficient, easy to operate and cost-effective in the treatment of large volumes of wastewater containing toxic heavy metals. In addition, a recent development of molecular biology shed light on the enhancing the microorganism's natural remediation capability as well as improving the current biological treatment. In this study, characteristics of the cell growth and heavy metal accumulation by Saccharomyces cerevisiae strains expressing phytochelatin syntahse (PCS) gene were studied in batch cultures. The AtCRFI gene was demonstrated to confer substantial increases in metal tolerance in yeast. PCS-expressing cells tolerated more Cd$^{2+}$ than controls.

Lichen as Bioindicators: Assessing their Response to Heavy Metal Pollution in Their Native Ecosystem

  • Lichens play crucial roles in the ecosystems, contributing to soil formation and nutrient cycling, and being used in biomonitoring efforts to assess the sustainability of ecosystems including air quality. Previous studies on heavy metal accumulation in lichens have mostly relied on manipulated environments, such as transplanted lichens, leaving us with a dearth of research on how lichens physiologically respond to heavy metal exposure in their natural habitats. To fill this knowledge gap, we investigated lichens from two of South Korea's geographically distant regions, Gangwon Province and Jeju Island, and examined whether difference in ambient heavy metal concentrations could be detected through physiological variables, including chlorophyll damage, lipid oxidation, and protein content. The physiological variables of lichens in response to heavy metals differed according to the collection area: Arsenic exerted a significant impact on chlorophyll degradation and protein content. The degree of fatty acid oxidation in lichens was associated with increased Cu concentrations. Our research highlights the value of lichens as a bioindicator, as we found that even small variations in ambient heavy metal concentrations can be detected in natural lichens. Furthermore, our study sheds light on which physiology variables that can be used as indicators of specific heavy metals, underscoring the potential of lichens for future ecology studies.

Simultaneous Determination of Alkaline Earth Metal Ions by a Conventional High Performance Liquid Chromatographic System

  • A simultaneous determination method of alkaline earth metals was attempted with the conventional high performance liquid chromatographic system. Four cations, namely, magnesium, calcium, strontium and barium ion, were injected directly as aqueous solution into an eluent containing copper chloride solution and and were successfully separated and determined on a separating column (Zipax SCX, 4.6 mm i.d. ${\times}25$ cm length, Du Pont, USA) by using a variable wavelength UV detector. The linear calibration curves were obtianed in the range from $1.0{\times}10^{-4}M$ to $5.0{\times}10^{-4}M$ and the correlation coefficient of the calibration curve for each metal of magnesium and calcium in tap water. Alkaline earth metals were determined with the conventional high performance liquid chromatographic system.

Optoelectric properties of gate-tunable n-MoS2/n-WSe2 heterojunction with proper electrode metals

  • Two dimensional transition-metal dichalcogenides (TMDs) semiconductors are attractive materials for optoelectric devices because of their direct energy bandgap and transparency. To investigate the feasibility of transparent p-n junctions, we have fabricated a p-n heterojunction consisting of p-type WSe2 and n-type MoS2 flakes since WSe2 and MoS2 with proper electrode metals exhibit p-type and n-type behaviors, respectively. These heterojunctions exhibits gate-tunable rectifying behaviors and photovoltaic effects (ECE ~ 0.2%) indicating that p-n junctions were formed. In addition, photocurrent and photovoltaic effects were observed under light illumination, which were dependent on the gate voltage. In addition, the photocurrent mapping images indicate that the photovoltaic effects comes from the junction area. Possible origins of gate-tunability are discussed.

  • Oxidations of metal generate large quantity of thermal and light energies but no toxic pollutants, so that metals with high calorific values, such as beryllium, boron, aluminum, magnesium, and lithium, are possible to be used as clean fuels instead of fossil fuels. However, they are so explosive due to very high oxidation rates that they should be stabilized by their surface passivation with oxides, organics and inorganics. For reasonable use of energetic metal particles as solid fuel, therefore, some detail information, such as thermal properties, preparation and passivation methods, and application area, of the energetic metals is introduced in this manuscript.

  • Copper phthalocyanine(CuPc) thick film solar cells were fabgricated byspin coating and their photovoltaic behavior was studied. Polyvinylidene fluoride (PVdF) was used for the binder. Aluminum and indium were employed as electrode metals to form Schottky contact to CuPc layer. The cells showed rectifying J-V characteristics in the dark and photovoltaic effect associated with white light irradiation. The photovoltaic performance of the cells strongly depended on contact metals, in which the formation of oxide layer between binder layer and electrode interface affected the solar cell. Influnce of the CuPc layer thickness, CuPc/PVdF ratio on the photovoltaic performance of the cells were also examined.

ALC(Autoclaved Light-weight Concrete)를 이용한 생물학적 반응벽체에 관한 연구

  • The physical and chemical characteristics of ALC were analyzed and showed 2.2 of specific gravity and 9.05 of pH. The results of leaching tests with standard method for soil and waste indicated heavy metals(Cu, Cd, Pb, $Cr^{6+}$) were under maximum concentration level. The anaerobic digestion sludge was attached in the surface of ALC within 90 hours. As the results of batch test, pH of the ALC and Bio-ALC were decreased from initial pH of ALC to 8.7 and 7.8 respectively Also, the concentration of heavy metals was rapidly eliminated in the solution with the batch test. The result of column experiment indicates that the removal efficiency of ALC was showed 66% of T-P, 60% of T-N, and 67% of CODcr. Also, removal efficiency of Bio-ALC was slightly higher than that of ALC in T-N (64%) and CODcr (74%).

  • In case of ultrashort laser ablation of metals, the transfer of energy from the electronic system causing strong absorption of laser light to the lattice needs relaxation times of the order of some picoseconds. Under the above theoretical background, nickel was ablated using femtosecond, picosecond and nanosecond laser. As a result, nickel ablation by picosecond laser and femtosecond laser, which are called ultrashort laser, has similar machinability because of relaxation time of metals, whereas nanosecond Nd:YAG laser has lower absorption, higher thermalization effect in comparison with ultrashort laser.

  • High speed casting technology is an attractive method to increase the productivity of continuous casting. However, high speed casting causes flow instability of molten steel in a mold. In this study, Large Eddy Simulation (LES) has been performed to identify the characteristics of mold flow for various shapes of submerged entry nozzles. The LES code has been newly developed to efficiently compute the two-phase flow by using the Fractional Step Method (FSM) combined with the Volume of Fluid (VOF) method. The Immersed Boundary Method was used to implement the shape of the submerged entry nozzle. Three cases of discharge angle of the submerged entry nozzle were computed and compared. The current results shed light on improving shape design of a submerged entry nozzle.

Synthesis of TiCx Powder via the Underwater Explosion of an Explosive

  • In this study, a novel approach to the explosive synthesis of titanium carbide (TiC) is discussed. Nonstoichiometric $TiC_x$ powder was produced via the underwater explosion of a Ti powder encapsulated within a spherical explosive charge. The explosion process, bubble formation, and synthesis process were visualized using high-speed camera imaging. It was concluded that synthesis occurred within the detonation gas during the first expansion/contraction cycle of the bubble, which was accompanied by a strong emission of light. The recovered powders were studied using scanning electron microscopy and X-ray diffraction. Submicron particles were generated during the explosion. An increase in the carbon content of the starting powder resulted in an increase in the carbon content of the final product. No oxide byproducts were observed within the recovered powders.
