Gene Expression Analysis of Zeaxanthin Epoxidase from the Marine Microalga Dunaliella tertiolecta in Response to Light/Dark Cycle and Salinity

  • Zeaxanthin is an important pigment in the photo-protection mechanism of microalgae. However, zeaxanthin epoxidase, an enzyme involved in the accumulation and conversion of zeaxanthin, has not been extensively studied in microalgae. In this work, we report the expression pattern of zeaxanthin epoxidase in Dunaliella tertiolecta (DtZEP) at different light and diverse salinity conditions. To confirm the responsiveness to light conditions, the ZEP expression pattern was investigated in photoperiodic (16 h of light and 8 h of dark) and continuous (24 h of light and 0 h of dark) light conditions. mRNA expression levels in photoperiodic conditions fluctuated along with the light/dark cycle, whereas those in continuous light remained unchanged. In varying salinity conditions, the highest mRNA and protein levels were detected in cells cultured in 1.5 M NaCl, and ZEP expression levels in cells shifted from 0.6 M NaCl to 1.5 M NaCl increased gradually. These results show that mRNA expression of DtZEP responds rapidly to the light/dark cycle or increased salinity, whereas changes in protein synthesis do not occur within a short period. Taken together, we show that DtZEP gene expression responds rapidly to light irradiation and hyperosmotic stress. In addition, ZEP expression patterns in light or salinity conditions are similar to those of higher plants, even though the habitat of D. tertiolecta is different.

  • Lhcb genes encoding light-harvesting chlorophyll-a/b binding (LHC) proteins of photosystem (PS) II were comprehensively characterized using the expressed sequence tag (EST) databases in the green alga, Chlamydomonas reinhardtii. The gene family was composed of eight Lhcb genes including four new genes, which were isolated and sequenced. The effects of light intensity on the levels of mRNAs accumulation of multiple Lhcb genes were studied under various conditions. The results indicate that Lhcb genes are coordinately regulated in response to light conditions, and repressed when the input light energy exceeded the requirement for $CO_2$ assimilation. The effects of high light on the expression of the Lhcb genes observed in the presence of an electron transport inhibitor, DCMU, and in mutants deficient in photosynthetic reaction centers suggest the presence of two alternative mechanisms for regulating the genes expression under high-light conditions.

Effects of light condition in the continuous mass cultivation of microalgae Scenedesmus dimorphus (미세조류 Scenedesmus dimorphus 연속 대량배양 운전에 있어 광 조건이 성장에 미치는 영향)

  • This study investigated the effects of the light conditions on the productivity of scenedesmus dimorphus in the continuous mass cultivation system. To compare the algal productivity according to the light conditions, S. dimorphus was cultivated continuously under the wide range of light intensity(200-600 PPFD) and various light wavelength(white light and red-blue mixed light). After 100 days of cultivation under the different light intensity, the productivity of S. dimorphus increased as light intensity decreased. So, the productivity was maximized as 100 mg/L/d when light intensity was 200 PPFD. In case of light wavelength, the productivity of S. dimorphus was enhanced about 20% with the white light compared to that of the red-blue mixed light. Consequently, the optimal light conditions for the continuous mass cultivation of S. dimorphus were 200 PPFD as light intensity and white light as light wavelength.

  • In general, LLC converters show great promise in applications that require high efficiency, especially under light load conditions. In particular, LLC converters feature wide gain capability with pulse-frequency modulation and zero voltage switching over entire load conditions. However, output voltage increases in light load conditions. In this study, Bode plot and impedance asymptotes analyses were conducted to obtain insights into the regulation characteristics of LLC converters under light load conditions. To improve the regulation characteristic of LLC converters, a new resonant tank with an additional capacitor is proposed. The design guideline for the proposed LLC converter is determined by the Bode plot and impedance asymptotes analyses. Therefore, the proposed LLC converter achieves the light load regulation while maintaining the advantages of typical LLC converters.

Development of a precision machining process for the outer cylinder of vacuum roll for film transfer (실험계획법을 통한 3.5인치 도광판의 두께 편차 최적화에 대한 연구)

  • In this study, experimental design methods were used to derive optimal process conditions for improving the thickness uniformity of a 0.40 mm, 3.5 inch light guide panel. Process mapping and expert group analysis were used to identify factors that influence the thickness of injection molded products. The key factors identified were mold temperature, mold temperature, injection speed, packing pressure, packing time, clamp force, and flash time. Considering the resin manufacturer's recommended process conditions and the process conditions for similar light guide plates, a three-level range was selected for the identified influencing factors. L27 orthogonal array process conditions were generated using the Taguchi method. Injection molding was performed using these L27 orthogonal array to mold the 3.5 inch light guide plates. Thickness measurements were then taken, and the results were analyzed using the signal-to-noise ratio to maximize the CpK value, leading to the determination of the optimal process conditions. The thickness uniformity of the product was analyzed by applying the derived optimum process conditions. The results showed a 97.5% improvement in the Cpk value of 3.22 compared to the process conditions used for similar light guide plates.

Effect of Light on Root Growth and Gravitropic Response of Phytochrome Mutants of Arabidopsis (Arabidopsis phytochrome mutant에서 빛이 뿌리 생장과 굴중성 반응에 미치는 영향)

  • Light, one of the environmental stimuli, is fundamental to the growth and development of plants. Red and far-red light are sensed using the phytochrome family of plant photoreceptors. To investigate the effect of light on root growth and gravitropism, we used the Arabidopsis phytochrome mutants grown in several light conditions. The root growth of $phyA$ reared in all light conditions except white light and was stimulated compared to the WT. The stimulation of root growth was obvious in $phyA$ grown in red light. On the other hand, the root growth of $phyB$ grown in all light conditions decreased, and the lowest rate of decrease was observed in $phyAB$ grown in white and red light. The gravitropic response of $phyA$ was stimulated compared to the WT when it was grown in all light conditions except far-red light. $PhyAB$ grown in all light conditions showed the inhibition of gravitropic response. The transcript level of ACS, one of the enzymes regulating ethylene biosynthesis, increased in $phyA$ grown in white and red light, but not in $phyA$ grown in far-red light. In conclusion, these results suggested that the $P_{fr}$ form of $phyB$ regulates the root growth and gravitropism.

Alteration of Media Composition and Light Conditions Change Morphology, Metabolic Profile, and Beauvericin Biosynthesis in Cordyceps bassiana Mycelium

  • Metabolic alterations of Cordyceps bassiana mycelium were investigated under the following culture medium and light conditions: dextrose agar supplemented with 0.5% yeast extract (SDAY) medium with light (SL), SDAY medium without light (SD), nut medium without light (ND), and iron-supplemented SDAY medium without light (FD). The levels of asparagine, aspartic acid, glutamic acid, glutamine, histidine, lysine, ornithine, and proline were significantly higher under SD and SL conditions. The levels of most of the alcohols, saturated fatty acids, unsaturated fatty acids, fatty acid esters, sterols, and terpenes were higher under the ND condition than in the other conditions, but beauvericin was not detectable under the ND condition. The FD condition was favorable for the enhanced production of aminomalonic acid, malic acid, mannonic acid, and erythritol. Thus, the metabolic characteristics of C. bassiana can be manipulated by varying the cultivation conditions, rendering this fungus potentially favorable as a nutraceutical and medicinal resource.

Evaluation and Application of Prediction Models for the Daylight Performance of a Light-Pipe System (광파이프 시스템의 채광성능 예측모델의 검증 및 적용)

  • The use of natural light has the potential for improving both the energy efficiency and indoor environmental quality in buildings. A light-pipe system can introduce daylight to spaces that would otherwise not be able to benefit from the advantages of daylight penetration. For the light-pipe system to be widely used in Korea, it is important to quantify its daylighting performance with due consideration regarding the effects imposed by the local climate conditions. This paper presents the evaluation results of existing semi-empirical models to predict daylighting performance of a light-pipe system. The evaluation of the existing models was based on the monitoring data obtained from a underground parking lot in which the light-pipe system was installed. Comparisons were made between the predicted and the monitored data obtained from the study. The results indicated that semi-empirical models which was developed using the experimental data obtained under the Korean climatic conditions had a good prediction performance. We also quantified the effects caused by sky conditions, solar altitudes, room dimensions, and the aspect ratio of a light-pipe system on both the daylighting performance of the light-pipe system and the indoor illuminance distributions of the space using the semi-empirical model. Finally, this paper provides the design guideline of the light-pipe system for its application to an underground parking lot space.

  • For the light guide plate of the TFT-LCD, there have been increasing demands for higher brightness, thin and light-weight design, and lower power consumption. To meet these demands, a micro-prism-type frontlight that integrates a prismatic sheet and a light-guiding plate has been developed. In this paper, the influences of processing conditions on the brightness were studied lot the injection molding of the light guide plate. Based on the experiment with an actual mold, the design of experiments and the neural network theory were used lot choosing the optimal processing parameters to increase the brightness and the uniformity. The verification experiment also showed that the brightness and the uniformity were increased dramatically with the chosen processing conditions.

Effects of Shading on the Growth and Chlorophyll Fluorescence under Agrivoltaic System Conditions

  • Agrivoltaic System (AVS) was introduced with the concept that it could generate electricity by using the extra light remain after crops use for photosynthesis in farm, which can earn additional income. However, crop yield was declined under the AVS condition due to the decrease in light energy. In the past, many researchers have been studied about crop states under shading conditions. However, the phenomenon of partial shading such as under the AVS is not well studied. In this study, to figure out the response of crop under the different light conditions, the electron transport rate (ETR) and non-photochemical quenching (NPQ) of rice was investigated using the chlorophyll fluorescence measurement. Also, physiological changes of crops under the shading conditions were investigated. The growth experiment under partial shading under AVS and overall shading which made of 35% shade cloth was conducted to understand the eco-physiological responses of rice to light in terms of the photosynthesis. Under the shading conditions, SPAD value and chlorophyll contents were higher, but the leaf thickness was lower than control. The overall shading condition show lower ETR than others during the growing season. In contrast, NPQ was higher than other treatments. This means the available light energy cannot contribute to photosynthesis under the shading condition.
