Light is an important factor that influences the growth and development of flowering plants. The present study investigated the effects of in vitro acclimatization to different light colors (white light (WL; control), blue light (BL; 447 nm), green light (GL; 519 nm), and red light (RL; 667 nm)) on the growth of petunia (Petunia hybrida) and of hardening cultivation of plant transferred form in vitro to a greenhouse under sunlight. Compared to the control, the shoot length and leaf width of Petunia increased by 42% and 11.7%, respectively, after acclimatization to BL and the shoot growth increased by 29.3% after acclimatization to RL. The chlorophyll and carotenoid contents after acclimatization to BL and GL were 16.7% and 11.3% higher, respectively, and 14.4% and 11.9% higher, respectively, than those in the control. During greenhouse cultivation, the shoot length increased by 16.7% and 11.3%, respectively, after acclimatization to BL and RL, respectively, and the leaf length and leaf width increased by 14.4% and 11.9%, respectively, after acclimatization to GL. While dry weight of root of GL and BL was not significant difference in vitro, increased by 59.0% and 22.9% ex vitro than that of WL. Thus, acclimatization to BL increased the shoot growth and leaf chlorophyll contents, and acclimatization to GL and RL enhanced shoot and root growth, in petunia.
Laying hens kept in different light and dark periods of the day at high ambient temperature of maximum $35^{\circ}C$ were challenged to $38.5{\pm}0.5^{\circ}C$ acute heat 3 hours daily for 7 consecutive days. They were found to have a significant (p<0.01) acclimatization response (rectal temperature) to heat stress during the dark period compared to those exposed to the same temperature during the light period. The blood pH was not significantly different. The partial pressure of carbon dioxide ($PCO_2$) was significantly high (p<0.01) except in day 4. Similarly the blood bicarbonate ($HCO_3$) concentration was significantly high (p<0.05) except day three and day four. Acute heat exposure in the first day increased the body temperature in both groups (Light and Dark) reaching $44^{\circ}C$, followed by gradual reduction in body temperature. The dark treated birds showed rapid reduction in body temperature ($42.88^{\circ}C$) and adaptation to high temperature during days 2-4 but that this was lost to some extent in days 6-8. However this was not obvious in the light treated birds. It is concluded that darkness reduce hyperthermia and enhance acclimatization responses during acute heat stress.
This study investigated the effect of cold acclimatization training on body composition including weight, fat mass, and muscle mass with 10 subjects (5 males and 5 females). During the 3-week acclimatization training program, they visited an artificial climate chamber ($15^{\circ}C$) 15 times and were exposed to cold environment with light clothing for 2 hours. Body composition was measured before and after cold training using bioelectric impedance analysis that was later compared by a paired t-test. In the process of thermoregulation, muscle contraction was accompanied by increased substrate metabolism for rising heat production. After cold training, the muscle mass increased and fat mass decreased significantly (p<.1, p<.05), subsequently the body composition changed. It was found that cold acclimatization training could be used as a treatment for obesity. It was suggested that further investigation on the long term effects of mild cold exposure using clothing and its potential applicability as an obesity treatment.
Photosynthetic rates of the large centric diatom Coscinodiscus granii were measured by means of multicolor variable chlorophyll fluorescence imaging, single cell $^{14}C$ assimilation, and optical $O_2$ sensor measurements during light acclimatization of cultures grown at five different irradiances: 50, 150, 235, 332, and $450{\mu}mol$ photons $m^{-2}\;s^{-1}$. Photo-acclimatization was evident from changes of cellular chlorophyll a content, growth rates, and light response curves. Each of the applied methods evaluates different parts and reactions in the photosynthetic apparatus, which makes a direct quantitative comparison of rates difficult, although a different degree of correlation were found between all three methods. However, when used in combination, they provide information about the internal relationship of photosynthetic pathways as well as the variation in photosynthetic capacity between individual cells within a single algal culture.
This study evaluated the influence of light quality and intensity during healing and acclimatization on the $CO_2$ exchange rate, growth, and morphogenesis of grafted pepper (Capsicum annuum L.) transplants, using a system for the continuous measurement of the $CO_2$ exchange rate. C. annuum L. 'Nokkwang' and 'Tantan' were used as scions and rootstocks, respectively. Before grafting, the transplants were grown for four weeks in a growth chamber with artificial light, where the temperature was set at $25/18^{\circ}C$ (light/dark period) and the light period was 14 hours $d^{-1}$. The grafted pepper transplants were then healed and acclimatized under different light quality conditions using fluorescent lamps (control) and red, blue, and red + blue light-emitting diodes (LEDs). All the transplants were irradiated for 12 hours per day, for six days, at a photosynthetic photon flux (PPF) of 50, 100, or 180 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. The higher PPF levels increased the $CO_2$ exchange rate during the healing and acclimatization. A smaller increase in the $CO_2$ exchange rates was observed in the transplants under red LEDs. At a PPF of 180 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, the $CO_2$ exchange rate of the transplants irradiated with red LEDs was lowest and it was 37% lower than those irradiated with fluorescent lamps. The $CO_2$ exchange rates of transplants irradiated with blue LEDs was the highest and 20% higher than those irradiated under fluorescent lamps. The graft take was not affected by the light quality. The grafted pepper transplants irradiated with red LEDs had a lower SPAD value, leaf dry weight, and dry matter content. The transplants irradiated with blue LEDs had longer shoot length and heavier stem fresh weight than those irradiated with the other treatments. Leaves irradiated with the red LED had the smallest leaf area and showed leaf epinasty. In addition, the palisade and spongy cells of the pepper leaves were dysplastic and exhibited hyperplasia. Grafted pepper transplants treated with red + blue LEDs showed similar growth and morphology to those transplants irradiated with fluorescent lamps. These results suggest that high-quality grafted pepper transplants can be obtained by healing and acclimatization under a combination of blue and red lights at a high PPF level.
접목 후 활착환경의 관리는 접목의 성공여부를 결정하는 중요한 요인이다. 일반적으로 활착초기 접목부위의 캘러스 분화를 촉진하고 식물체의 지나친 위조를 막기 위해 $25{\sim}30^{\circ}C$ 정도 온도와 90% 이상의 높음상대습도 조건하에서 관리하다가 활착이 진행됨에 따라 점차 온도 및 상대습도를 낮추고, 광 조사량을 늘려주는 환경관리방법이 제시되고 있다(Kim 등, 2001). 본 연구에서는 고추 접목묘의 활착 및 생육향상을 위한 활착단계별 환경조건을 제시하고자, 활착기간 중 온도, 상대습도 및 광 조건이 접목활착률 및 생육에 미치는 영향을 조사하였다. 암조건 하에서 총 6일간의 접목활착기간을 3단계로 나누어, 활착 1단계는 온도 $30^{\circ}C$, 상대습도 95% 조건하에서, 활착 2, 3단계에서는 온도($20^{\circ}C$, $25^{\circ}C$, 및 $30^{\circ}C$) 및 상대습도(75%, 85%, 및 95%) 조건을 달리 처리하였을 때, 접목활착률 및 생육을 조사하였다. 고추 접목묘의 활착을 위해서는 활착 초기 $30^{\circ}C$, 95% 상대습도 조건에서 2일정도 관리한 후, 이후 4일간은 온도와 상대습도 조건을 각각 $20{\sim}25^{\circ}C$, 75~85% 정도로 낮추어 주는 것이 바람직할 것으로 판단된다. 또한 접목활착기간 동안 온도 $30^{\circ}C$, 상대습도 85% 및 암 조건을 대조구로 하여, 활착기간 중 온도($25^{\circ}C$ 및 $30^{\circ}C$), 상대습도(65%, 75%, 및 85%) 및 광 조건(광 조사 유무, 광 조건 $45{\pm}2{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$)을 달리하여 처리하였 때, 접목 활착률은 처리간 차이가 없었으며 활착기간 중 저온 저습의 광조사 기간이 길수록 생육이 증가하는 경향을 보여, $25^{\circ}C$ 온도조건하에서 저광 조사 및 65% 까지의 저습 조건이 고추 접목묘의 활착 및 생육촉진에 효과적인 것으로 판단된다.
본 연구는 토마토 플러그묘의 접목후, 순화과정에서 단기간 광조사 여부가 접목 활착율과 묘소질에 미치는 영향을 구명하기 위하여 실시 되었다. 공시 품종으로 'B-Blocking', 'Kanbarune', 'High-Power'의 3종류 대목 품종에 방울토마토 'Choice' 품종을 각각 접목한 뒤, 활착실에서 10일간 적색LED 광조사구와 무처리(대조구)로 나누어 접목 활착율과 묘소질을 조사하였다. 적색 LED 광조사구의 접목 활착율은 대목품종에 관계없이 무처리구에 비해 높았다. 광조사구 접목 활착율은 세 품종 모두 90% 이상으로 품종간의 유의성이 없었으나, 무처리구의 접목 활착율은 35~72% 정도로 품종에 따른 차이가 매우 컸다. 활착기간 중 곰팡이병 이병율도 'Kanbarune'를 제외하고 광조사구가 무처리구에 비해 저하하는 경향을 보였으나, 품종간 유의차는 나타나지 않았다. 묘소질은 경경, 엽수, 엽록소 함량을 제외한 모든 생육지표에서 LED광 조사구가 우수한 결과를 나타내었으나, 처리에 따른 대목 품종간 유의차는 전반적으로 나타나지 않았다. 지하부 생육에서 평균 근경을 제외한 뿌리 표면적, 전 근장, root tip수는 광조사구가 무처리구에 비해 모든 품종에서 우세하였다.
In mass production of seed-potato plantlets, the processes for in vitro propagation and ex vitro acclimatization with a high cost should be improved by a culture system with environmental control using scaled-up culture vessels. The experiment was conducted to design a hydroponic culture system for enhancement of growth and development of seed-potato (Solanum tuberosum) plantlets cultured under photoautotrophic (without sugar in culture medium) conditions with controlled light intensity and ventilation rate. The culture system was consisted of scaled-up culture vessels, ventilation pipes, a multi-cell tray and an environmental control system (ECS) for optimum controlling in temperature, light intensity, ventilation rate, and culture-medium supply. Growth and development of the plantlets was significantly increased under the ECS compared with a conventional culture system (CCS) of photomixotrophic culture (with sugar in culture medium) using small scale vessels. For 21 days, leaf area of the plantlets was expanded more than 2 times, and number of internodes also approximately 4 times greate. under the ECS. In addition, the photoautotrophic growth in sweetpotato (Ipomoea batatas) and chrysanthemum (Chrysanthemum morifolium) plantlets was greater more than 2 times compared with the CCS.

To test the quality change of seedlings of three domestic medicinal plants raised in plug trays, a short term storage experiment was conducted. Seedlings were kept in growth chambers for two weeks at 4 or $8^{\circ}C$ temperature combined with 0 or $5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD. Quality of glasshouse-raised seedlings was assessed after two weeks of cold storage in the growth chamber and one week of acclimation in the greenhouse. After two weeks of storage in the growth chamber of Perilla frutescens var. acuta Kudo, plant height was the greatest in the treatment $8^{\circ}C$ combined with $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD. Internode length of P. frutescens var. acuta Kudo was the greatest in the treatment of $4^{\circ}C$ combined with $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD. After one week of acclimatization in a glasshouse, the growth and development, such as plant height, internode length and leaf size, were greater in the $8^{\circ}C$ combined with $5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD than in the other treatments. After two weeks of storage in the growth chamber of Sophora tonkinensis, plant height increased more in the treatment of $4^{\circ}C$ than $8^{\circ}C$. After one week of acclimatization in a glasshouse, number of leaves did not change in the treatment of $4^{\circ}C$ combined with $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD, but it increased in the other treatments. Leaf width increased more under the dark than light condition. Leaf length did not observably change in any treatments. After two weeks of storage in the growth chamber, plant height of Angelica gigas Nakai was the greatest in the treatment of $8^{\circ}C$. Number of leaves was the greatest in the treatment of $8^{\circ}C$. Leaf growth was greater under dark than light condition. These results suggested that optimal storage environment was $8^{\circ}C$ combined with $5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD for P. frutescens var. acuta Kudo, and $4^{\circ}C$ combined with $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD for S. tonkinensis and A. gigas Nakai. Hence, proper combination of temperature and PPFD were necessary for better storage, and acclimatization and growth, thereafter, of the plug seedlings of theses plant species.
본 연구는 사과 왜성대목 M.9와 M.26 조직 배양묘를 통기성 마개와 비통기성 마개의 MS배지에서 생장 특성을 비교하기 위해 시험되었다. 또한 배양묘의 생육 변화와 이 배양묘를 발근촉진제 침지 처리 후 기외 삽목을 하여 발근 및 순화를 동시에 시키기 위해 실시하였다. 배양병내의 $CO_2$와 에틸렌의 농도는 통기성 마개 배양병이 비통기성마개 배양병 보다 낮았다. 통기성 마개 배양병내의 온$.$습도 변화는 광조건에 따라 일정하게 반복되었다. 통기성 마개 배양묘의 기공은 외기에서 즉시 관찰 시 닫혀 있었으나 비통기성마개 배양묘의 기공은 외기에서 20분 후 관찰 시에도 열려 있었다. 통기성 마개 배양묘의 엽록체 지수와 잎 면적은 모두 비통기성 마개의 것 보다 높았다. 기외 삽목시 발근율은 M.9와 M.26 모두 300 mg/L IBA+ 3% sucrose 침지 처리 구에서 각각 86.5%와 85.4%로 가장 높았으며 순화율은 각각 71.7%와 70.0%로 가장 높았다. 통기성 마개 배양묘의 순화율은 비통기성마개 배양묘 보다 모든 침지 처리구에서 순화율이 비슷하거나 높았다.
