Effects of Leucine on in Vivo Protein Synthesis in Skeletal Muscles of Fed and Food-Deprived Rats (Leucine이 정상 또는 굶게 된 쥐의 골격근육의 단백질 생합성에 미치는 영향)

  • In vivo effects of leucine on skeletal muscle protein synthesis in fed and I-day food deprived young rats were examined. Animals assigned to leucine group were given a single i.p. injection of 80 or 160flmoles of leucine while control group animals were saline sham injected. The rate of protein synthesis was measured by the amount of $^{14}\textrm{C} incorporated into muscle protein after a single injection of $^{14}\textrm{C}-tyrosine, IO$\mu$ Ci/l00g B.W. Examined muscles were two different types of hind limb muscles. the oxidative solues and the glycolytic EDL and plantaris. Administered leucine elevated the concentration of free leucine in soleus muscles by 4-6.8 times the normal level. A massive dose of leucine, 160 flmoles, stimulated protein synthesis in the EDL and plantaris by 24 %, 29 % respectively of straved rats. The soleus of I-day food deprived rats and both types of muscles in fed rats did not respond to the injected leucine. The synthesis rate of the EDL and plantaris was supressed to one-half of the normal while the soleus that was not stimulated by leucine maintained a relatively normal rate, 78 %, of protein synthesis after I-day of food deprivation. Thus, in vivo stimulatory effect of leucine appears to be not a general phenomenon but to be related to the degree of catabolic condition developed by stress such as food deprivation. Although anabolic effects of leucine observed in this study was limited, any applicability of this special property of leucine to human subjects for the purpose of protein sparing in skeletal muscles remains to be examined.

Sugar and Amino Acid Transport in Yeast (효모세포의 당과 아미노산의 운반에 관한 연구 II)

  • Saccharomyces cerevisiae J170, a mutant, was used for $DL-^{14}C-leucine$ uptake during the sporulation and vegetative stage. $^{14}C-Leucine$ uptake into yeast cells appeared the highest at pH 6.0, indicating the same result of glucose transport, $^{14}C-Leucine$ uptake in sporulation period was higher than in growth phase, showing the evidence that leucine is more required for protein synthesis. This tendency has the evidence tht leucine is more required for protein synthesis. This tendency has the evidence that leucine is more required for protein synthesis. This tendency has been also supported from the result of Km values of leucine uptake in two stages of yeast. Leucine uptake was inhibited by 2,4-dinitrophenol in two stages of yeast. This means that leucine transport system is associated with energy dependent in both stages. The contents of all amino acid in growth phase cells were higher than those of sporulation stge cells, and those of methionine and tyrosine were showed in trace during the sporulation stage. In contrast, the content of glutamic acid in sporulation stage was compared with those of other amino acids.

  • The stimulatory effects of leucine on the activities of two soluble forms of brain glutamate dehydrogenase isoproteins (GDH I and GDH II) have been studied at various conditions. There were significant differences between GDH I and GDH II in their sensitivities to the action of leucine. When the effects of varied leucine concentrations on GDH activities were studied in the direction of reductive amination of 2-oxoglutarate with NADPH as a coenzyme, a marked activation was observed for both isoproteins at leucine concentrations up to 10 mM, whereas both isoproteins showed activation to a lesser extent with NADH as a coenzyme. The stimulatory effects of leucine on GDH activities in the direction of the oxidative deamination of glutamate were also observed, but to a much lesser extent. Leucine relieved the inhibition of GDH I by GTP and this resulted in an increase in the apparent activation by leucine in the presence of GTP. 2-Oxoglutarate was found to give rise to high substrate inhibition and leucine significantly reduced the substrate inhibition in the presence of $200\;{\mu}M$ NADH. Thus, the effects of leucine might be composed of a direct effect on the enzyme together with a relief of high substrate inhibition.

  • To determine the metabolism of leucine during the cuticle formation and the sclerotization process in Pieris rapae L., $U-^3$H-leucine or $U-^4$C-tyrosine is injected into the haemolymph of newly molted pupa through dorsal cuticle of heart area. The results show that leucine as a common amino acid participates in the synthesis of cuticle protein over the first 3 hr after ecdysis. It is also shown that leucine in the haemolymph at ecdysis is freely being moved between major internal organs during the short time period post-ecdysis, providing the evidence for some involvements including haemolymph protein synthesis and storage of fat body and gut in metabolism of leucine.

  • To analyze genetic effects on milk production traits, 164 unrelated Italian Jersey cows reared in 15 farms located in Southern Italy were characterized at the growth hormone locus. Using a Polymerase Chain Reaction-Restriction Fragment Length Polymorphism protocol all three possible genotypes for the Leucine/Valine polymorphism were identified even if in the considered population the number of homozygous animals (Leucine/Leucine = 36; Valine/Valine = 28) was smaller than Leucine/Valine (n = 100) cows; the Hardy-Weinberg equilibrium was verified. Statistical analysis showed that daily milk yield in the Leucine/Leucine genotype was higher (p<0.01) than in Leucine/Valine (22.76 kg vs. 16.96 kg ); cows with Leucine/Valine genotype differed significantly from Leucine/Leucine and Valine/Valine genotypes particularly in fat (4.95% vs. 4.13% and 4.82%, respectively) and protein content (4.00% vs. 3.47% and 3.79%, respectively). No significant effect on lactation length was observed.

Effects of Levels of L-Leucine Supplementation with Sub-optimal Protein in the Diet of Grower-finisher Broiler Chickens on Carcass Composition and Sensory Characteristics

  • An experiment involving 180 straight run one-day-Cobb broilers was conducted to evaluate the effect of supplementation of L-leucine with different levels of crude protein (CP) on carcass composition and sensory characteristics of broiler grower-finisher chickens. Six experimental diets comprising two levels of crude protein (CP) i.e., 20 and 20% with three levels of L-leucine i.e. 0, 0.5 and 0.67%, were offered to birds from 21-42 d of age. The birds were randomly divided into 36 experimental pens, 5 chickens in each pen, and there were 6 replicates under each diet. L-leucine supplementation did not affect the bone and lean, whereas fat was decreased (p<0.05) when L-leucine was added at 0.5%. Similarly, there were no significant differences (p>0.05) in the lean, fat and bone among chickens fed two levels of CP. No significant differences between dietary treatments were observed on any sensory characteristics affected by dietary L-leucine and CP. From this study, it is obvious that supplementation of up to 0.5% L-leucine reduced fat. However, other characteristics were not affected by supplementation of L-leucine. Similarly, reduction of body composition and sensory characteristics were not apparent on a diet low in CP.

  • In this study, three kinds of Korean snake wine (50 V% alcohol extracts) were determined by amino acid analyzer and were discussed as follows. 1. Salmosa Ju and Nungsa Ju were composed of 22 free amino acids: cysteic acid, aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystine, valine, methionine, iso-leucine, allo-iso-leucine, leucine, nor-leucine, tyrosine, phenylalanine, ${\beta}-alanine$, lysine, arginine, histidine, trytophan. 2. Doksa Ju were composed of 21 free amino acids which were all same as above except missing histidine. 3. The free amino acid composition were almost identical in Doksa Ju, Salmosa Ju and Nungsa Ju quantitatively. 4. The contents of cysteic acid, glutamic acid, glycine, alanine, valine, leucine and lysine were relatively high, on the other hand, methionine, allo-iso-leucine, nor-leucine, and tryptophan were trace amount in every snake wine. 5. Eleven unknowns of ninhydrin positives were identified in the every snake wine. 6. The free amino acids in snake wines were various in kind as compared with in beer, Japanese sake and Korean Tack Ju. Especially cysteic acid, allo-iso-leucine, nor-leucine and ${\beta}-alanine$ in snake wines were missed in every cereal wine.

Effects of Dietary L-leucine Levels in Low-lysine Diets on Growth Performance and Meat Quality Parameters in Finishing Duroc Pigs (저수준의 Lysine 사료에서 L-leucine 첨가수준이 비육후기 Duroc 품종 돼지의 성장 및 육질특성에 미치는 영향)

  • The objective of this study was to investigate effects of supplementation levels of L-leucine in low-lysine diets on growth performance and meat quality parameters in finishing Duroc pigs. A total of fifty-four pigs with an average initial weight of 74±1kg were allotted to one of three dietary treatments. Each treatment had six replications of three pigs per replicate. The treatment diets included 1)a corn-soybean meal basal diet containing lysine as low as 0.45% (CON), 2) basal diet plus 1.5% L-leucine (LEU 1.5), and 3) basal diet plus 3.0% L-leucine (LEU 3.0). No difference was found in ADG, ADFI and Feed/Gain among treatments. In carcass parameters, dressing precent and back-fat thickness were not affected by L-leucine levels, however, longissimus dorsi area tended to increase by supplementation of L-leucine. Marbling score was significantly higher (P<0.05) in LEU 1.5 and LEU 3.0 than in CON. In meat quality parameters, CIE L* of meat color significantly increased (P<0.05) and crude fat tended to increase by supplementation of L-leucine. However, there was no difference in water holding capacity, cooking loss and shear force value. This study suggests that dietary supplementation of L-leucine in low-lysine diets at the end of finishing period can produce high marbled pork which is preferred by Korean consumers.

The Development of Dimerized Chicken Recombinant Single-chain Fv (ScFv) Antibody Using Leucine Zipper Motif (Leucine Zipper Motif를 이용한 닭의 재조합 이량체 Single-chain Fv (ScFv) 항체의 개발)

  • Leucine zipper motif consists of multiple periodic leucine residues, which forms amphipathic alpha helix. The hydrophobic nature of leucine zipper motif can dimerize proteins which contain this motif. Leucine zipper motif addition at C-terminus of single-chain Fv (ScFv) antibody induces its dimerization. Since the dimeric ScFv antibody contains two antigen binding sites (bivalency) like Y-shaped complete antibody, it could increase avidity. As a result, it could show higher antigen binding activity than monomeric ScFv antibodies. Based on this concept, monomeric chicken 8C3 ScFv antibody previously developed from chicken hybridoma was dimerized by the addition of leucine zipper motif at C-terminus of ScFv antibody. The dimeric 8C3 ScFv antibody specifically reacted with Eimerian sporozoite which causes Avian Coccidiosis. As expected, dimeric 8C3 ScFv antibody showed 3-folds higher antigen binding activity than monomer due to increased avidity. In addition, protien yields of dimer expression were 2-folds higher than monomer.

  • Biotin 요구성 glutamic acid 생산능을 가진 Br-evibacterium sp.의 isoleucine 영양요구성 변이주가 다량의 L-leucine을 생성함을 발견하였다. 본균주에 관한 탄소원, 질소원, 무기염, vitamin, amino acid 등의 영양물질과 온도, 공기주입량, pH등에 관한 실험으로 다음과 같은 결과를 얻었다. 본균주는 탄소원, 질소원으로서 10% glucose와 4% $(NH_4)_2SO_4를$ 공합하여 $30^{\circ}C,$ 88시간 배양시킨 결과, 액중에 21 mg/ml의 L-leucine이 축적되었다. Fe$^{2+}$$Mn^{2+}$의 동시첨가는 L-leucine생성에 효과적이었으며 thiamine 역시 L-leucine 생성에 효과적이었다. 30ι jar fermentor 에서 폐당밀을 주원료로 한 실험결과, 폐당밀, $(NH_4)/SO_4,$ $KH_2PO_4,$ $MgSO_4ㆍ7H_2O로$ 구성된 액체배지에서 $30^{\circ}C,$ 36시간 배양시킨 것이, 27mg/ml의 L-leucine이 축적되었으며 이때의 대당수율은 16%이었다.
