Acute Toxicity of Leuconostoc citreum GJ7 Isolated from Kimchi in Mice (김치로부터 분리한 Leuconostoc citreum GJ7의 마우스에 대한 급성독성)

  • The present study was carried out to investigate the potential acute toxicity of Leuconostoc citreum GJ7 (Leu. citreum GJ7), a lactic acid bacterium isolated from Kimchi, in ICR male and female mice. The test article was administered to the mice orally or intraperitoneally. Mortality rates, clinical findings, and body weight changes were monitored for the 2 weeks following administration. The results showed that in 50% of the cases, lethal doses ($LD_{50}$) of Leu. citreum GJ7 were determined as >5, 000 mg/kg (p.o.) and >2500 mg/kg (i.p.) in both sexes. There were no significant changes in general conditions, body weights clinical signs and any gross lesions between vehicle control and Leu. citreum GJ7-treated groups. Hence, it is suggested that Leu. citreum GJ7 does not induce any significant acute toxicity in ICR mice.

  • Synthesis of $(D-Phe^7\;-Leu^8)$ bradykinin and bradykinin by solid phase method using a new reaction vessel was carried out. Coupling was performed by dicyclohexylcarbodiimide. After cleavage with dried HBr the peptides were purified by high pressure liquied chromatography. Their purify was assayed by paper and thin layer chromatography, melting point and amino acid analysis. $(D-Phe^7\;-Leu^8)$ bradykinin and bradykinin were incubater in vitro endopeptidase $({\alpha}-chymotrysis)$ and exopeptidase(carboxypeptidase A, leucine aminopeptidase) in order to study the degradation pattern of peptides. $(D-Phe^7\;-Leu^8)$ bradykinin and bradykinin were rapidly degradated by ${\alpha}-chymotrypsin$ and carboxypeptidase A $(D-Phe^7\;-Leu^8)$ bradykinin and bradykinin coution$(D-Phe^7\;-Leu^8)$ bradykinin and bradykinin contain imino peptide bound from proline at N-terminal and therefore they were not attacted by leucine aminopeptidase.

Isolation and Characterization of Exopolysaccharide Producing Lactic Acid Bacteria from Kimchi (김치로부터 Exopolysaccharide 생성 유산균의 분리 및 특성 규명)

  • Three slime-forming lactic acid bacteria were isolated from Kimchi and shown to produce viscous exopolysaccharides (EPS) in sucrose media. The isolated strains, GJ2, C3 and C11, were identified as Leuconostoc kimchii, Leuconostoc citreum and Leuconostoc mesenteroides, respectively, by examining their metabolic characteristics and determining their 16S rDNA sequences. Leu. kimchii GJ2, Leu. citreum C3 and Leu. mesenteroides C11 exhibited high viability (maintained initial viable cell count of $10^8$ CFU/ml) in 0.05 M sodium phosphate buffer (pH 3.0) for 2 h, in artificial gastric juice for 2 h and in 0.3% oxgall for 24 h. When tested, Leu. kimchii GJ2, in particular, displayed antimicrobial activity against pathogenic microorganisms. Leu. kimchii GJ2, Leu. citreum C3 and Leu. mesenteroides C11 produced 21.49 g/l, 16.46 g/l and 22.98 g/l EPS, respectively, in sucrose (5%) medium. The amount of purified EPS extracted from Leu. kimchii GJ2, Leu. citreum C3 and Leu. mesenteroides C11 was 14.61 g/l, 7.73 g/l and 4.77 g/l, respectively. Although the EPS produced by Leu. kimchii GJ2, Leu. citreum C3 and Leu. mesenteroides C11 differed in viscosity, TLC and HPLC analysis revealed that each contained only one type of monosaccharide, glucose. The average molecular mass of EPS produced by Leu. kimchii GJ2 was 306,606 Da.

Mutation Analysis of Wilson Disease Gene: Arg778Leu Mutation in Korean Children (윌슨 유전자의 돌연변이 분석: 한국 윌슨병 환자에서의 Arg778Leu 돌연변이)

  • Background: Wilson disease (WD) is an autosomal recessive disorder of copper transport and characterized by degenerative changes in the brain, liver dysfunction, and Kayser-Fleischer rings due to toxic accumulation of copper. Since the identification of Wilson disease gene (ATP7B), more than 80 mutations have been detected among the different ethnic groups. Methods: Twenty three children with Wilson disease were included in this study. They were all diagnosed by low serum ceruloplasmin and increased 24 hour urinary copper excretion with characteristic clinical findings. We analysed WD gene mutation by assessing the nucleotide sequence of exon 7, 8, 9 and 10 including intron-exon boundaries of ATP7B gene from genomic DNA. Results: Arg778Leu mutation was identified in 16 WD patients; three were homozygous and 13 were heterozygous for this mutation. Of the 46 alleles, 19 alleles had a Arg778Leu mutation (19/46=41%). Homozygote patients had neurologic forms of WD. Arg778Leu mutation was not found among 50 normal healthy persons. Conclusion: Arg778Leu mutation is a common mutation in Korean WD gene. Arg778Leu mutation screening might be used as a useful supplementary diagnostic test in some patients to confirm Wilson disease in Korea.

Cultural Characteristics of Psychrotrophic Lactic Acid Bacteria Isolated from Kimchi (김치에서 분리한 저온성 젖산균의 배양특성)

  • The cultural characteristics of 60 strains of psychrotrophic lactic acid bacteria which were isolated from kimchi, a Korean traditional fermented vegetable food, and identified as Leuconostoc mesenteroides subsp. mesenteroides, Leu. mesenteroides subsp. dextranicum, Leu. paramesenteroides, Leu. lactis, Lactobacillus bavaricus and Lac. homohiochii were tested. All strains grew at $5^{\circ}C,\;10^{\circ}C\;and\;37^{\circ}C$ in tomato glucose broth, but not at $45^{\circ}C$. The optimum growth temperature of Leu. mesenteroides and Lactobacillus sp. were $33{\sim}34^{\circ}C\;and\;34{\sim}36^{\circ}C$, respectively. All strains of Leu. mesenteroides and Lactobacillus sp. grew at 4.8 and 4.2 of initial pH, but not at 4.0. The final pH of Leu. mesenteroides and Lactobacillus sp. in glucose broth were $3.84{\sim}4.10\;and\;3.82{\sim}3.99$, respectively. None of the 60 strains clotted milk nor reduced litmus in litmus milk. All strains of Leu. mesenteroides and Lactobacillus sp. grew in tomato glucose broth containing 7% ethanol or 6.5% NaCl, but not in the broth containing 15% ethanol or 10% NaCl. All strains grew in tomato glucose broth containing 40% bile juice and survived in the artificial gastric juice of pH 3.5. Furthermore, all strains of Leu. mesenteroides survived in the artificial gastric juice of pH 3.0. Since many strains of lactic acid bacteria tested in this study showed differences in several physiological characteristics from those described in Bergey's Manual of Systematic Bacteriology, it was considered that further tests would be necessary to clarify their positions in taxonomic system.

  • We tested the pre-treatment of alumina, leucine, alumina mixed with leucine at ratios of 9:1 (Al-Leu (9:1)) and 5:1 (Al-Leu (5:1)), and alumina mixed with sodium lactate at a ratio of 5:1 (Al-Lac (5:1)) on aged fingerprints (1, 7, 14, and 30 days after natural fingerprinting) before cyanoacrylate (CA) fuming to improve the development efficacy. As a result of the experiment, fingerprints with pre-treatment of alumina, leucine, Al-Leu (9:1), Al-Leu (5:1), and Al-Lac (5:1) showed better development efficacy (area and minutiae) than fingerprints without pre-treatment. Therefore, this modified CA fuming method improved the development efficacy with five pretreatments.

Anti-fatigue effect of fermented porcine placenta through the regulation of fatigue-associated inflammatory cytokines

  • Fatigue is a common complaint and affects the quality of life in modern people. Physical stress may induce activation of certain immune cells. Fermented porcine placenta (FPP) has been used to alleviate fatigue. Inflammatory cytokines are produced by physical stress and results in symptoms of fatigue. However, the role of FPP on fatigue-associated inflammatory cytokine production has not been elucidated yet. Thus, we estimated the anti-fatigue effect of FPP and its active components, leucine (Leu) and lysine (Lys) in activated RAW264.7 macrophages and forced swimming test (FST) fatigue animal model. Pretreatment with FPP, Leu, or Lys significantly inhibited the lipopolysaccharide (LPS)-induced tumor necrosis factor-α, interleukin (IL)-1β, and IL-6 production without inducing cytotoxicity on LPS-stimulated RAW264.7 macrophages. FPP, Leu, or Lys inhibited the production of nitric oxide and downregulated the expression of inducible nitric oxide synthase on LPS-stimulated RAW264.7 macrophages. Furthermore, caspase-1 activities increased by LPS were significantly reduced by FPP, Leu, or Lys. In the FST, inflammatory cytokine levels of the mice administrated with FPP, Lys, and Leu were significantly reduced compared with the control group at 21 days. Collectively, these results show that anti-fatigue effect of FPP and its active components, Leu and Lys might be derived from the down-regulating of inflammatory mediators.

Influences of Cultural Temperature on Growth Rates of Lactic Acid Bacteria Isolated from Kimchi (배양온도가 김치에서 분리한 젖산균의 증식속도에 미치는 영향)

  • Growth curves of seven strains of lactic acid bacteria isolated from Kimchi were graphed during cultivation at 1$0^{\circ}C$, 2$0^{\circ}C$, 3$0^{\circ}C$ and 4$0^{\circ}C$ in filter sterilized Chinese cabbage juice, and then lag time and generation time at each conditions were calcurated. At 3$0^{\circ}C$, the lag time of Leu. mesenteroides subsp. dextranicum was 168 min(minutes), Leu. mesenteroides subsp. mesenteroides 204 min, Leu. paramesenteroides 612 min, Lac. bavaricus 258 min, Lac. homohiochii 228 min, Lac. plantarum 270 min and Lac. brevis 264 min. And at this temperature, the generation times of Leu. mesenteroides subsp. dextranicum and Leu. mesenteroides subsp. mesenteroides were all 36 min, Lac. bavaricus 33 min, Lac. homohiochii 39 min, Lac. plantarum 66 min, Lac. brevis 42 min and Leu. paramesenteroides 162 min. As cultural temperature was lowed from 3$0^{\circ}C$ to 1$0^{\circ}C$, all strains showed remarkable prolongations in lag time and in generation time, and the prolongations were most conspicuous in Lac. plantarum. At 1$0^{\circ}C$, 2$0^{\circ}C$ and 3$0^{\circ}C$, both the lag time and the generation time of Leu. mesenteroides subsp. mesenteroides were shorter than those of Lac. plantarum. But at 4$0^{\circ}C$, this pattern was completely inverted. As a whole lower temperatures were more favorable for the growth of Leu. mesenteroides subsp. mesenteroides, while higher temperatures were for Lac. plantarum.

Purification and Characterization of the Anabolic Acetolactate Synthase III from Serratia marcescens ATCC 25419

  • The anabolic acetolactate synthase III was purified to homogeneity from Serratia marcescens using DEAE-Sepharose, Phenyl-Sepharose, and hydroxylapatite column chromatography The native molecular weight of the enzyme was approximately 165 kDa. The enzyme is composed of two large and two small subunits with molecular weights of 64 and 15 kDa, respectively. The N-terminal sequence of the large and small subunit of the enzyme was Ser-Ala-Thr-Pro-Gln-Pro-Ser-Thr-Arg-Phe-Thr-Cys-Ala-Gln-Leu-Ile-Ala-His-Leu and Met-Leu-Gln-Pro-Gln-Asp-Lys-Pro-Gln-Val-Ile-Leu-Glu-Leu-Ala-Val-Arg-Asn-His-Pro-Gly-Val-Met-Ser-His-Val, respectively. The optimum pH and pI value were 7.5 and 5.5, respectively The $IC_{50}$ values were $20\;{\mu}M$ and $14\;{\mu}M$ for valine and herbicide SU7, respectively. The substrate specificity ratio, R value, was determined to be approximately 40, which suggests that this enzyme prefers the formation of $\alpha$-aceto-$\alpha$-hydroxybutyrate leading to the synthesis of isoleucine.

Kinetic Study of Thermolysin-Catalyzed Synthesis of N-(Benzyloxycarbonyl)-L-Phenylalanyl-L-Leucine Ethyl Ester in an Ethyl Acetate Saturated Aqueous System

  • The kinetics of the thermolysin-catalyzed synthesis of N-(benzyloxycarbonyl)-L-phenylalanyl-L-leucine ethyl ester (Z-Phe-LeuOEt) from N-(benzyloxycarbonyl)-L-phyenylalanine (Z-Phe) and L-leucine ethyl ester (LeuOEt) in an ethyl acetate saturated aqueous system in a batch operation were studied. The kinetics for the synthesis of Z-Phe-LeuOEt were expressed using a rate equation for the rapid equilibrium random bireactant mechanism. The four kinetic constants involved in the rate equation were determined numerically by the quasi-Newton method so as to fit the calculated results with the experimental data. Within the pH and temperature range examined, the $K_{cat}$ value for the synthesis of Z-Phe-LeuOEt reached a maximum at pH 7.0 and $45^{\circ}C$, whereas the affinity between Z-Phe and thermolysin reached a maximum at pH 6.0 adn $40^{\circ}C$. The inhibitory effect of Z-Phe on the condensation reaction decreased as the pH and temperature decreased. In contrast, they affinity between LeuOEt and thermolysin remained unchanged within the pH and temperature range examined. Therefore, it was concluded that the protonation state of the carboxyl groups. of Z-Phe was more imprtant than that of the amono groups of LeuOEt for the synthesis of Z-Phe-LeuOEt in the present solvent system. The equilibrium yield at pH 6.0 and $30^{\circ}C$ was 8% higher than that at pH 7.0 and $40^{\circ}C$, although the rate was much slower. This result suggested that the affinity between the enzyme and the substrate rather than the overall rate was a more important factor affecting the equilibrium yield, when the peptide synthesis was carried out in a product-precipitation system.
