Isolation of auxotrophs and drug resistant mutants of Lentinus edodes (표고버섯의 영양요구성 및 약물내성주의 분리)

  • Auxotrophs and drug resistant mutants from the mycelia of Lentinus edodes were obtained by UV irradiation at survival rates of $0.024{\sim}2.45%$ and ethidium bromide (EtBr) enrichment after UV irradiation. The mutation rate was 0.40%, and back mutation rate was $4.81{\times}10^{-4}{\sim}8.46{\times}10^{-4}$. Various amino acid-, nucleic acid-, and vitamin-requiring auxotrophs were isolated. The concentrations of several fungicides, antibiotics and amino acid analogues inhibiting the growth of L. edodes were determined. The MIC values for cycloheximide, benomyl, and p-fluorophenylalanine were 2, 2000, and 1000 ug/ml respectively. Five p-fluorophenylalanine-resistant mutants and eight benomyl-resistant mutants were selected by UV irradiation.

Reutilization of Enokitake Cultural Waste as Lentinus edodes Cultivation Substrate

  • The availability of enokitake cultural waste for Lentinus edodes cultivation was investigated, although hardwood sawdust has traditionally been used as a substrate for this fungus. Firstly, physiochemical characteristics of cultural waste were analysed. Secondly, mycelial growth characteristics and fruiting yields of L. edodes on waste treated in some methods were determined. Physiochemical characteristics of enokitake cultural waste showed that the millwaste complex was a little degraded by enokitake fungus and suggested the probability that most component lost by enokitake could be rice bran. Mycelia of L. edodes grew and fruited well on waste supplemented by fresh rice bran and Quercus sawdust although didn't on waste only. Mycelial growths of these fungi on waste were accelerated when supplemented by rice bran to the percent of 40(w/w) but decreased or suppressed at above ratios(30, 40%, w/w). Supplementations of oak sawdust at above 40%(w/w) of the waste and rice bran at 20%(w/w) of the sawdust allowed such a good mycelial growth as to be selected as a pertinent mixing ratio for fruiting medium. A fruiting yield on enokitake cultural waste supplemented by oak sawdust (at 40% of the waste, w/w) and rice bran (at 20% of the sawdust, w/w) was not inferior to that on oak sawdust supplemented by rice bran only (at 20% of the sawdust, w/w). These results indicated strongly the potentiality of enokitake cultural waste as raw materials for shiitake cultivating substrates.

On the Mushroom Cultivation of Oak(Quercus spp.) Chip (참나무류(類) 칩을 이용(利用)한 표고버섯 재배(栽培))

  • To increase the production of mushrooms and the economic efficiency of mushroom cultivation, the pinchips(PC) and sawdusts(SD) of three Korean oak species were tested as cultivating media for Lentinus edodes with varing the composition of the media. The results are as follows. 1. Organic acids(tannic acid and citric acid), which were added in the PC medium to enrich the quality of mushrooms, did not increase the growths of the mycelia and the quality. 2. The mushrooms were able to be harvrested from six months after beginning the cultivation. No significant differences were found among the qualities of the mushrooms produced by various treatments. 3. The production rate of mushroom was relatively high with, 28.4~29.3% (700~730gr./2,500gr pack). The net profit estimated was about 1,450 won per pack.

  • The industrial procedure for the preparation of dietary fiber material from Lentinus edodes by drying and grinding process was developed. The chemical composition of dietary fiber of mycelia was as follows; crude protein: 16.16%, crude fat: 2.96%, crude ash: 3.25% and carbohydrates: 77.63%. The mycelia contained total dietary fiber of 54.5%. The 40 mesh pass of dietary fiber exhibited water-holding capacity 7.39g water/g and oil-holding capacity 2.03g oil/g, while $40{\sim}80$ mesh of dietary fiber had water-holding capacity 7.80g water/g and oil-holding capacity 2.77g oil/g.

Eeffect of Wood Xylem Flour in Liquid Culture on Mycelial Biomass of Lentinus lepideus and Lentinus edodes (목분 첨가 액체배양에 의한 잣버섯 및 표고 균사배양의 촉진 효과)

  • This study was carried out to investigate the promoting effect of wood flour on the mycelial growth of Lentinus lepideus and Lentinus edodes. To determine the optimal culture condition, we first examined the tissue origin of pine flour (Pinus densiflora) including needle, bark, root and xylem. Only the xylem-derived flour increased mycelial growth compared to no treatment control. The addition of the xylem flour (5 g/l) showed the highest increase and the glucose level in the basal medium was best at 10 g/l. The smaller particle size of the xylem flour showed the positive effect on mycelial growth; two-fold increase when supplemented with flour of which particle size is less than $106\;{\mu}m$ in diameter compared to $425\;{\mu}m$. The addition of the xylem flour continuously increased the mycelial production for 25 days while mycelia stopped growing within 15 days without the xylem flour. In addition, when woody flour obtained from the different tree species was applied to L. edodes mycelial culture, all treatments accelerated mycelial production compared to the control. Based on all results described above, we conclude that the supplementation of woody flour to culture medium may be an another promising way to increase mycelial production of economically important fungi.

Antiturmor Activities of Protein-bound Polysaccharide Extracted from Mycelia of Mushroom (버섯 균사체에 의한 암세포 성장억제 효과)

  • The purpose of this study is to observe the effect of protein-bound polysaccharide (PBP) on proliferation of Th1 cells and cytotoxicity of cancer cell. Mushrooms (Ganoderma lucidum, Agaricus blazei, Lentinus edodes, Coriolus versicolor and Phellinus linteus) were fractionated by 100$^{\circ}C$ hot water for 3hr. PBP was stimulated and proliferated Th1 cells most at 10 mg/$m\ell$ concentration and the percentage of cell proliferation was 40%. It was estimated cytotoxicity of PBP against 7 kinds of cancer cell lines. Antitumor activities of Agaricus blazei against P388D1 and L1210 (tumor cell lines) were 2.4% and 39.7% survival rate, and Lentinus edodes was 48.4% and 52.5% survival rate, respectively. PBP mixtures of Agaricus and Lentinus edodes prolonged (27∼40%) significantly the survival rate of mice intraperitoneally implanted with sarcoma 180.

Immunostimulating Activity by Protoplast Fusants between Ganoderma Iucidum and Lentinus edodes (영지와 표고의 융합체의 면역활성 증강작용)

  • On the inter-order protoplast fusants of Lentinus edodes and Ganoderma lucidum was the antitumor activity test performed and the fusant P22 was selected. The hot water extract of the cultured mycelia of P22 were purified by DEAE-cellulose chromatographya and the resulting purified fraction was designated as P22A. It was found to be a proteoglycan whose molecular weight was 47 kDa. When examined for immunopotentiation activity, P22A increased the number of colony forming unit in the bone marrow stem cells to 3-folds. It also potentiated the secretion of nitric oxide in activated macrophages to 2-folds. In humoral immune response, it increased the activities of the alkaline phosphatase in differentiated B cells to 1.6-folds and the number of plaque forming cells to 1.8-folds. In cellular immune response, it restored the depressed response of delayed type hypersensitivity in tumor bearing mice to normal level. These results suggest that P22A have potential to restore the decreased immune activity of the tumor bearing mice to normal level.

Anti-tumor Activity of Protein-bound Polysaccharides Extracted from Mycelia of Lentinus edodes (표고버섯 균사체로부터 추출한 단백다당체의 항암효과)

  • Protein-bound polysaccharides (PBP) were extracted from the mycellia of Lentinus edodes SR-1, and their anti-tumor activities and immunopotentiating properties were observed. The amounts of PBP needed to extend the doubling time twofold (1 unit) were found to be 1 mg for mouse leukemic cells $P_{388}\;and\;L_{1210}$; 4.4, 3.6 and 6.6 for bowel cancer cells, HCT-48, HRT-18, HT-29 respectively; and 2.6 mg for liver cancer cell, Hep G2. When $P_{388}\;and\;L_{1210}$ were treated with 4 mg of PBP, more than 90% of the cell number were reduced in 48 hours. However, 9 mg of PBP and 72 hrs of incubation time were needed to obtain the same effect for HRT-18, HT-29, and Hep G2. The significant reduction of cell size was observed as the amount of PBP and the incubation time increased. Mice spleen weight and plaque forming cell number increased when the cancer cells were treated with PBP.

  • This experiment was undertaken to investigate proper conditions for protoplast isolation from strains of Agaricus bisporus, Flammulina velutipes, Lentinus edodes, Pleurotus florida, Pleurotus ostreatus, Pleurotus sajor-caju and Volvariella volvacea. Combination of ${\beta}-D-glucanase$, novozym 234 and snail enzyme with 0.6M $MgSO_4$ in 0.05M Na-maleate buffer, pH 5.8 was the most effective for isolation of protoplasts. High yields of protoplasts were obtained from $4{\sim}5$ days old mycelia of P. florida and P. ostreatus on mushroom complete agar medium. Protoplasts were also obtained in good yield from other fungi but A. bisporus and V. volvacea. In the latter two cases protoplast release was observed; however, the yield was much lower than those of the other fungi.

Preparation of Mushroom Mycelia-cultured Traditional Meju with Enhanced Anticaricinogenicity and Sensory Quality (항암성과 향미가 개선된 재래식 버섯균사체메주의 제조)

  • Mushroom mycelia-cultured traditional meju (MTM) was prepared by inoculating 10% submerged-liquid culture of mushroom strains to five holes (1$\times$3 cm) per side of the traditionally-fermented meiu (10$\times$10$\times$10cm), followed by incubating additional 4 weeks at $25^{\circ}C$. Mushroom strains used were Neutari (Pleurotus ostreatus, PO), Yeongji (Ganoderma lucidum, GL), Synryeong (Agaricus blazei, AB), Ypsae (Grifola frondosa, GF), Pyogo(Lentinus edodes, PE), Dongchunghacho (Paecilomyces japonicus, PJ) and Sanghwang (Phellinus linteus PL). All MTMs showed an enhanced anticarcinogenicity against S-180 cell-induced mouse ascites cancer antimutagenicity against aflatoxin B$_1$ (AFB$_1$) and 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), and sensory qualities, relative to control meju. Such positive effects of MTM prepared with Sanghwang, Yeongji, or Synryeong were superior to those of MTM with Ypsae, Pyogo, Dongchunghacho, or Neutari.
