Fine Structure of Epithelial Tissue in Korean Leech, Erpobdella lineata (한국산 거머리(Erpobdefla lineate)상피조직의 미세구조)

  • 한국산 거머리 각배obdeffo linea약의 상피조직을 배부상피(dorsal epithelium), 복부 상피(ventral epithelium) 측부상피(lateral epithelium) 별로 3부위를 광학현미경과 전자현미경으로 관찰한 결과 다음과 같은 결론을 얻었다. 전자현미경 관찰에서 거머리 상피조직은 3부위 모두 불규칙한 세포로 구성되었으며, 그들은 전자밀도가 높은세포와 중등도인세포 그리고 투명한세포 등 3종류로 구분되었다. 거머리의 불규칙한 상피조직 사이에서 두 종류의 분비성 과립인 a 및 b형과립과 색소성 과립인 D형과립이 관찰되었다 배부상피조직에서 관찰된 a형과립(66$\times$83 Um)은 복부와 측부에서 관찰된 과립(33H66 Um) 보다 형태가 훨씬 크고 그 수 도 두배정도 많이 관찰된 반면, b형과립은 모든 상피조직에서 동일한 크기와 형태를 보였지만 그 수는 배부와 복부에 비해 측부상피조직에서 두배정도 많이 관찰되었다 이상 분비성 과립들은 PAS-alcian blue(pH 2.5) 반응에서 모두 PAS에 강한 양성반응을 나타내어 중성점액다당류로 확인되었다. 거머리 몸마디를 이루는 체절은 배부와 복부에서 그 주름의 깊이와 넓이가 각각 다르게 나타나서(배부, 40 Um와 330 urn, 복부 100 urn와 150 urn), 복부의 깊은 체절이 발(foot)의 역활을 수행 하였다.

New Records of Seven Eupithecia (Lepidoptera: Geometridae) from Korea

  • The genus Eupithecia Curtis is the largest taxon in Lepidoptera (more than 1,500 species worldwide) and the adults of the genus are characterized by small in size, cryptically colored grayish and brownish forewing, and indistinct basal, ante- and postmedial transverse lines mostly with a definite discal spot on the forewing. Forty-four species have been reported so far in Korea. Herein, we present the first report on seven species of Eupithecia: Eupithecia rufescens Butler (1878), Eupithecia costiconvexa Inoue (1979), Eupithecia daemionata Dietze (1904), Eupithecia persuastrix Mironov (1990), Eupithecia actaeata Walderdorff (1869), Eupithecia suboxydata Staudinger (1897) and Eupithecia costimacularia Leech (1897). Diagnosis, descriptions and figures of the available species are provided.

  • Three species of Spilomelinae, Camptomastix septentrionalis Inoue, 1982, Omiodes indistinctus (Warren, 1892) and Piletocera aegimiusalis (Walker, 1859), are reported for the first time from Korea. Camptomastix septentrionalis is distinguished from Camptomastix hisbonalis (Walker, 1859) in the smaller body size, the lack of a clear discal spot on the forewings, and having the less protruding costa of the valva in the male genitalia and the smaller antrum in the female genitalia. Omiodes indistinctus is distinguished from Omiodes tristrialis(Bremer, 1864) in having the narrower forewings with the faded postmedian and subbasal lines and a densely-setose, sclerotized area on the terminal part of uncus dorsally in the male genitalia. Piletocera aegimiusalis is distinguished from Piletocera sodalis(Leech, 1889) in the larger body size and the lack of a pale yellow patch on the hindwings. Photos of the external and genital features are provided for the three newly-recorded spilomeline species.

Non-Pharmacological Treatments of Psoriasis in Persian Medicine a Narrative Review

  • Psoriasis is a chronic disease that has no definitive cure. In this review study, the main sources of Persian Medicine (PM) such as the Canon of Medicine (by Avicenna) and Al-Havi (by Rhazes) were assessed to identify non-pharmacological treatments for psoriasis. Several treatments that are recommended for this disease include nutritional advice, lifestyle modifications, and manipulation therapy such as wet cupping (Hijamah), leech therapy, and phlebotomy (Fasd). These recommendations may help to prevent recurrence and be useful in improving psoriasis. The efficacy of PM recommendations to improve psoriasis should be evaluated in future studies.

Catalogue of the Pyralidae of Korea (Lepidoptera) I. Evergestiinae and Pyraustinae (한국산 명나방과 목록 I. 새들명나방아과(신칭), 들명나방아과)

  • [ $\lceil$ ]한국산 명나방과(Pyralidae)에 관하여는 이미 필자(1976)에 의하여 언급된바와 같이 1889년 영국인 곤충학자 J.H. Leech가 처음으로 9종의 신종을 발표한데 이어 현재까지 300여종이 기록되어 왔다. 그러나 이들의 기록이 여러종류의 발표문에 산재해 있는 상태로 체계적인 정리가 이루어진바 없으며 학명뿐아니라 우리말 이름조차 갖가지로 쓰여지고 있어 곤충명 사용에 혼란이 야기되고 있는 실정이다. 이에 필자는 현재까지 발표되었던 종들을 조사하여 최근의 분류체계에 따라 학명 및 그들의 동의어(Synonym)를 정리함과 아울러 우리말 이름도 먼저 명명된 이름이나 또는 현재 많이 사용되고 있는 이름을 택하여 통일화하고 ( )안에 그 이명을 표시하였다. Evergestiinae(새들명나방아과, 신칭)은 현재까지는 주로 Pyraustinae(들명나방아과)에 포함되는 족(Tribe)으로 취급되어 왔으나 최근의 분류체계에 따라 독립된 아과로 취급하였다. 이들에 대해서는 Leech에 이어 일본인 Shibuya(1927년에 의하여 13종, 박세욱등 (1958, 1969)에 의하여 27종이 새로 기록되는등 1968년에 출판된 한국 동물명집 곤충편에는 94종이 수록되었다. 최근 필자(1976)에 의하여 우리나라 미기록종으로 발표된 24종과 기타 국내외의 문헌을 통한 기록의 근거를 기초로 작성된 이 목록에는 Evergestiinae에 1속 3종, Pyraustinae에 53속 123종이 포함되었으며 학명이 불분명하거나 채집의 근거가 불확실한 4종은 후미에 별도로 기재하였다. 분포란의 괄호안에 명시된 채집지는 필자에 의하여 채집 확인된 종들로서 그들의 표본은 농업기술연구소 곤충연구담당관실 표본실에 소장되어 있다. 다른 아과에 관하여는 정리가 완료되는대로 추후 발표할 예정이다.nemella hengsungica, Aphelenchoides besseyi 등으로 2아목 7과 11종이었다.는 내연가공한 옷생산에 사용되지 않는 것이 타당하겠다. 주아의 즙액에 의해서도 C. amaranticolor에 기계적 전염이 되었다. 7. C. amaranticolor 상에 계통분리된 마늘 모자이크 바이러스의 내열성은 $65^{\circ}-70^{\circ}C$, 희석한계는 $10^{-}2-10^{-3}$, 그리고 보존한계는 2 일이었다. 8. 마늘 모자이크 바이러스의 순화는 2회의 분획원심과 Sephadex gel filtration에 의해서 가능했다. 9. 전자현미경하에서 관찰한 마늘 모자이크 바이러스는 길이 1200-1225mu 폭 10-12mu의 사상이었다. 10. 혈청학적 미량침강 반응법에 의해 마늘잎에서뿐만 아니라 인편과 주아에서도 마늘 모자이크 바이러스의 검정이 가능했다. 11. 우리나라 5개 지방에서 수집한 마늘 종구 150개와 주아 30개에 대해 혈청학적방법으로 마늘 모자이크 바이러스의 감염률을 조사한 결과 $100\%$의 감염률을 보였다. 12. 마늘 모자이크 바이러스와 크기가 근사한 Potato Virus X. Potato virus Y, Potato virus S, Potato virus M 등과의 혈청학적 유연관계를 조사한 바, 마늘 모자이크 바이러스는 이들과 구별되는 다른 바이러스라고 생각된다. 13. 마늘의 모자이크 감염주에서 단일계통으로 분리하여 본 실험에 사용한 바이러스는 마늘의 바이러스 무감염주를 얻을 수가 없기 때문에 직접 마늘잎에 접종해서 모자이크톤의 병원이라는 것을 확인할 수 없었지만, 검정식물상의 반응, 혈청학적반응, 전자현미경적 관찰등의 간접적인 조사 결과로 미루어 미인록의 마늘모자이크 바이러스라고 생각된다

Treatment of Complete Scalp Avulsion with the Conditions of Unstable Vital Signs: A Case Report (불안정한 생체징후 하의 두피 결출상 환자의 치험례)

  • Purpose: Scalp avulsion is a life-threatening injury that may cause trauma to the forehead, eyebrows, and periauricular tissue. It is difficult to treat scalp avulsion as it may lead to severe bleeding. Therefore, emergency scalp replantation surgery is necessary, and we must consider the function, aesthetics, and psychology of the patients. A case of scalp avulsion leading to massive bleeding was encountered by these authors, which led to a failure to achieve the proper operation conditions in an adequate time period. Methods: A 49-year-old female was hospitalized due to having had her head caught in a rotatory machine, causing complete scalp avulsion which included the dorsum of the nose, both eyebrows, and ears. Emergent microsurgical replantation was performed, where a superficial temporal artery and a vein were anastomosed, but the patient's vital signs were too unstable for further operation due to excessive blood loss. Three days after the microanastomosis, venous congestion developed at the replanted scalp, and a medicinal leech was used. Leech therapy resolved the venous congestion. A demarcation then developed between the vitalized scalp tissue and the necrotized area. Debridement was performed 2 times on the necrotized scalp area. Finally, split-thickness skin graft with a dermal acellular matrix ($Matriderm^{(R)}$) was performed on the defective areas, which included the left temporal area, the occipital area, and both eyebrows. Results: The forehead, vertex, right temporal area, and half of the occipital area were successfully replanted, and the hair at the replanted scalp was preserved. As stated above, two-thirds of the scalp survived; the patient could cover the skin graft area with her hair, and could wear a wig. Conclusion: Complete scalp avulsion needs emergent replantation with microsurgical revascularization, but it often leads to serious vital conditions. We report a case with acceptable results, although the microanastomosed vessel was minimal due to the patient's unstable vital signs.

Reconstruction of Soft Tissue Defect of Lower Extremity with Anterolateral Thigh Perforator Flap (전외측 대퇴부 천공지 피판을 이용한 하지 연부조직 결손의 재건)

  • This study investigated the clinical application of anterolateral thigh (ALT) perforator flap in reconstruction of soft tissue defect of lower extremity. There were twenty-one patients who had been taken soft tissue reconstruction with anterolateral thigh perforator flap. There were 19 males and 2 females between 3 and 65 years (mean, 36 years). This study included 4 cases of pedatric case of under 10-year-old. All cases were a cutaneous flap. Flap size averaged $160\;cm^2\;(20{\sim}450\;cm^2)$. 19 cases were musculocutaneous perforator flaps and 2 were septocutaneous perforator. T-shaped pedicle were used to reconstruct and to preserve major artery of lower extremity in 2 cases. 19 cases flaps survived completely and 2 cases flap were marginal necrosis partially. There was venous congestion in one case of type of reverse island flap but that was improved after salvage procedure with leech. While the donor sites were closed directly in 5cases, 16 cases underwent skin graft. ALT flap is suitable for coverage of defects in lower extremity where have various condition and reliable in children as in adult.

A New Record of Notodontidae Moth, Neodrymonia marginalis in Korea

  • Herein, we report a new record of Notodontidae moth, Neodrymonia marginalis (Matsumura) in Korea. This species is characterized by grayish forewing that shows thick dark grayish band between basal and antemedial line, smooth postmedial line costally with blackish crescent line, dark grayish subtermen with dark brownish thick band costally, and whitish and black dot shaped discal dot. The female genitalia can be distinguished by the broad, plate-shaped postvaginalis, strongly sclerotized ductus dursae with three long spike shaped and several small spike shaped processes, and large, ovate corpus bursae with a large fan-shaped plate signum. Neodrymonia marginalis can be distinguished from N. deliana by the shape and color of the costal part of postmedial line and subtermen of forewing. The female genitalia of N. marginalis can be distinguished from those of N. deliana by the rectangular postvaginalis and more spike like processes of the anterior of ductus bursae. Up to now, four species of Neodrymonia are recorded in Korea: N. delia (Leech, 1889), N. coreana Matsumura, 1922, N. deliana Gaede, 1933, and Neodrymonia marginalis.

Genetic Stability of the Integrated Structural Gene of Guamerin in Recombinant Pichia pastoris

  • Genetic chracterstics of the structural gene of guamerin (a novel elastase inhibitor from Korean leech), integrated into the HIS4 locus of chromosomal DNA of Pichia pastoris along with the $\alpha$-factor leader sequence, were investigated. In the selected clone from candidates, two copies of the integration cassette including the structural gene copies of the integration cassette including the structural gene of guamerin were found in the integration site of the chromosomal DNA of P.pastoris. It was demonstrated that the integrated structural gene of guamerin was stable up to about 70 generations in the relay flask culture. Then, a high-cell-density culture could be fulfilled easily by DO-stat fed-batch culture, in which the cell growth and the recombinant guamerin production reached about 250 of OD600nm and 260 mg/l, respectively. Finally, it was revealed that the DNA sequence of the integrated structural gene of guamerin in P. pastoris was maintained correctly in the end of production cells of relay flask culture and high-cell-density culture.

Downstream Processing of Recombinant Hirudin Produced in Saccharomyces cerevisiae

  • A recombinant form of hirudin, a potent thrombin-specific inhibitor derived from the bloodsucking leech, was expressed as a secretory product in Saccharomyces cerevisiae under the control of GALl0 promoter and the mating factor $\alpha$pre-pro leader sequence. In an attempt to produce recombinant hirudin (r-Hir) of therapeutic purity in large quantities, the fed-batch fermentation was carried out by using this recombinant yeast, and subsequently downstream processing was developed with the preparative-scale column chromatography systems. About 234 mg/l of biologically active r-Hir was produced as a secretory product by the fed-batch fermentation strategy developed for an efficient downstream processing. Using a two-step chromatography process (an anion exchange chromatography followed by the reverse phase HPLC), the r-Hir was purified to>98% with an overall recovery yield of 84%. According to the N-terminal amino acid sequencing, the purified r-Hir was found to have the predicted N-terminal amino acid sequence. The biological activity of the purified r-Hir to inhibit thrombin was also identical to that of the commercial hirudin.
