벼의 생산력 분석(分析) -IV. 수기(受器)의 충전속도(充塡速度)와 수기(受器)-급기(給器) 관계(關係) (Analysis of Productivity in Rice Plant -IV. Sink-filling rate and sink-source relation)

  • IR667 계통(系統)(Suwon 213 및 214)과 장려품종(진흥(振興) 및 팔달(八達))의 수용기(受用器)(Sink) 및 급여기(給與器)(Source)의 크기와 그들간(間) 거리(距離), 동화산물의 형태(形態), 기상인자(氣象因子)와 관련(關聯) 수기(受器)(곡실(穀實))충전(充塡) 속도(速度)를 검토(檢討)하여 다음과 같은 결과(結果)를 얻었다. 1. IR667 계통(系統)은 장려 품종보다 최고수기충전속도(最高受器充塡速度)가 크고 빨리온다. 충전속도곡선(充塡速度曲線)의 양상(樣相)은 IR667 계(系)는 준봉형(峻峰型)을 장려품종은 구릉형(丘陵型)을 보인다. 2. 기상인자(氣象因子)는 충전곡선(充塡曲線)의 양상(樣相)에 큰 변화(變化)를 주지 못하는 것 같다. 3. 수기일급기간(受器一給器間) 기능거리(機能距離)(실거리(實距離)${\times}$각엽(各葉)의 생산기여율는 상위엽의존형(上位葉依存型)인 IR667 계(系)에서는 상위엽(上位葉)이 짧고 하위엽의존형(下位葉依存型)인 장려품중에서는 하위엽(下位葉)이 짧다. IR667의 상위엽의존성(上位葉依存性)이 유난히 짧은 제(第)1간장(稈長)(상부(上部)로부터)에 기인(基因)하는것 같다. 4. 간(稈)과 엽초의 유리당(遊離糖)/전분비(澱粉比)에 따라 IR667계(系)는 고당형(高糖型)으로 장려품종은 고전분형(高澱粉型)으로 분류(分類)되었으며 전이성(轉移性)(당(糖))과 비전이성(非轉移性) 동화산물(전분(澱粉))의 비(比)가 수기충전속도(受器充塡速度)와 관련되고 고당형(高糖型)이 생산효율이 높을 것으로 예상된다. 5. 군락구조(群落構造)의 각생산계층내의 균등도(均等度)가 클수록 곡실생산의 효율성이 높다는 가설(假說)을 제시(提示)하고 다수성(多收性) 및 안전성(安全性)과 관련(關聯)하여 검토(檢討)하였다. 6. 엽위별(葉位別) 엽신(葉身)의 N.P.K. 잔존률양상(殘存率樣相)이 진흥(振興)에서는 상부잔존형(上部殘存型)으로, IR667은 중앙부(中央部) 잔존형(殘存型)으로 구분(區分)되어 생산구조 보전형식(保存型式)의 원인(原因)으로 해석되었다. 7. 기관별(器管別) N.P.K 농도(濃度)와 함량분포비율(含量分布比率)에 의(依)하여 IR667은 엽초중심형으로 진흥(振興)은 엽신중심형(葉身中心型)으로 분류(分類)되었다. 8. 동일품종내(同一品種丙)에서 N.P간(間) N.P.K 각양분내(各養分內)에서 품종간(品種間) 수(穗)로의 전이율(轉移率)이 큰 경우 엽신잔존율(葉身殘存率)도 크다.

Morphological Traits of S598A Sweetpotato as an Industrial Starch Crop

  • Sweetpotato is one of the important starch crops, current more considered as an industrial crop rather than food because it has higher starch content (over 80% of biomass), it is used for bio resources for industrial area. In this study, we generated S598A (a mutant gene of oat phytochrome A) sweetpotato plant using Agrobacterium-transformation method. Morphological characteristics of S598A plant were compared with the wild type sweetpotato, S598A had darker green leaves, increased chlorophyll content higher than to two-fold, delayed leaf senescence, shorter plant height (60% shorter than that of the wild type), more number of leaves and petioles about 1.8-fold, shorter petiole length (30% shorter), 1.2-fold more branches and 1.6-fold thicker stem diameters. From this study, S598A plants with such phenotypic characteristics might be able to use the solar energy efficiently, to have increased tolerance to biotic and abiotic stresses and finally to increase productivity (not only starch yield but also root biomass yield). S598A sweetpotato lines are under field trials.

Treatment of ramie leaf β-amylase for preliminary purification

  • The thermal properties of ramie leaf ${\beta}$-amylase (RBA) were examined to develop a novel process for enzyme purification. The thermostability of RBA extract prepared from ramie leaf powder was examined at various temperatures. RBA activity decreased slightly, whereas other carbohydrate-active enzymes, such as $\small{D}$-enzyme, were rapidly inactivated during 30 min incubation at $60^{\circ}C$. When the heat-treated extract was incubated with various substrates, maltose was produced exclusively as the major product, whereas the untreated crude extract produced maltose and other maltooligosaccharides. In sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis, fewer protein bands were observed for the heat-treated extract than the untreated extract, indicating that the thermostable RBA was partially purified and other thermolabile enzymes were eliminated. Thus, the treatment of the RBA extract at $60^{\circ}C$ for 30 min resulted in 5.4-fold purification with a recovery yield of 90%.

Source-sink Relationships of Soybean as Influenced by Drought Stress during the Pod and Seed-developing Stage

  • This study was conducted to investigate the influence of drought stress during the pod developing and seed filling stage on source-sink relationships of soybean (Glycine max). Drought treatments were imposed by withholding water at the full-pod stage, 19 days after flowering, and then limited watering was relieved at 15 days after the initiation of drought treatment. Soybean seed yield was reduced by 39% mainly due to decreased pod number under drought stress, but the 100-seed weight was relatively less reduced. In spite of the 15-day drought during the full-pod stage, soybean produced good seeds showing similar l00-seed weight, protein, starch and soluble sugar content to those from the well-watered. Although drought during the full-pod stage caused source limitations; i.e. accelerated leaf senescence and reduced leaf soluble sugars, it did not cause limitations of other source characteristics such as SGR and leaf starch level. This is because the reduction in size of sinks, such as pod and seed abortions compensated for source limitations, resulting in balanced source-sink as expressed by LAR and the ratio of leaf area to seed dry weight. 전통장의 메주 제조에 관한 연구 (A Study on Making Meju (Molded Soybean) for Traditional Jang)

  • 서기 530년부터 1950년까지의 고조리서 32권에 수록된 장 만드는 방법 중 메주 만드는 방법을 언급한 225가지 중 재료의 가열방법, 메주의 형태, 메주를 띄우는데 사용하는 용기, 덮개 및 깔개를 분석하였다. 그 결과 전통 메주의 콩이나 전분질의 가열 방법은 찌기 57가지, 삶기 58가지, 볶아 삶기 21가지, 밥짓기 2가지의 순으로 나타났다. 메주의 형태는 알 41가지, 구형 27가지, 덩어리 22가지, 칼자루형 8가지, 납작형 6가지, 쪼가리형 4가지, 구멍형 1가지, 사각형 1가지의 순으로 나타났다. 고추장 메주 72가지 중 콩 가열 방법은 삶기 9가지, 찌기 6가지의 순이었고, 전분질의 가열방법은 떡찌기 19가지, 밥 짓기 11가지, 익반죽 삶기 5가지의 순으로 나타났다. 콩메주 띄우는 용기는, 가마니 섬 섶 49가지, 독 5가지, 둥구미 멱서리 14가지, 온돌 11가지, 소쿠리 채반 광주리 7가지, 독 5가지, 흙도랑 4가지, 시루 오쟁이 3가지, 바가지 2가지, 시루 2가지, 용수 1가지의 순으로 나타났다. 메주 띄우기용 덮개나 깔개는 짚 36가지, 닥나무잎 17가지, 멍석 거적 자리 15가지, 쑥 14가지, 솔잎 11가지, 콩잎 10가지, 도꼬마리잎 6가지, 북나무잎 6가지, 보릿짚 6가지, 뽕잎 6가지, 가랑잎 5가지, 띠풀 5가지, 삿자리 4가지, 헝겊 3가지, 개오동나무잎 2가지, 갈잎 1가지의 순으로 나타났다. 매달기는 5가지 밖에 없었다.

Lotus leaf alleviates hyperglycemia and dyslipidemia in animal model of diabetes mellitus

  • The purpose of this study was to investigate the effects of lotus leaf on hyperglycemia and dyslipidemia in animal model of diabetes. Inhibitory activity of ethanol extract of lotus leaf against yeast ${\alpha}$-glucosidase was measured in vitro. The effect of lotus leaf on the postprandial increase in blood glucose levels was assessed in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without lotus leaf extract (500 mg/kg) was administered to the rats after an overnight fast, and postprandial plasma glucose levels were monitored. Four-week-old db/db mice were fed a basal diet or a diet containing 1% lotus leaf extract for 7 weeks after 1 week of acclimation to study the chronic effect of lotus leaf. After sacrifice, plasma glucose, insulin, triglycerides (TG), total cholesterol (CHOL), high-density lipoprotein (HDL)-CHOL, and blood glycated hemoglobin levels were measured. Lotus leaf extract inhibited ${\alpha}$-glucosidase activity by 37.9%, which was 1.3 times stronger than inhibition by acarbose at a concentration of 0.5 mg/mL in vitro. Oral administration of lotus leaf extract significantly decreased the area under the glucose response curve by 35.1% compared with that in the control group (P < 0.01). Chronic feeding of lotus leaf extract significantly lowered plasma glucose and blood glycated hemoglobin compared with those in the control group. Lotus leaf extract significantly reduced plasma TG and total CHOL and elevated HDL-CHOL levels compared with those in the control group. Therefore, we conclude that lotus leaf is effective for controlling hyperglycemia and dyslipidemia in an animal model of diabetes mellitus.

광호흡 과정에서 생성된 Glutamate가 광합성 탄소대사에 미치는 영향 (Effects of Gultamate Synthesized during Photorespiration on Photosynthetic Carbon Metabolism)

  • The effects of ammonium ion and glutamate on CO2 fixation abilities and related carbon metabolism were investigated in pea (Pisum sativum L. cv. Sparkle) leaf discs under conditions favoring photorespiration (21% O2, 0.03% CO2) and nonphotorespiration (5% O2, 0.03% CO2). A concentration of more than 10 mM of NH4+ decreased the photosynthetic CO2 fixation and those inhibitory effects were more remarkable in 21% O2 than in 5% O2 conditions. The effect of glutamate on CO2 fixation was found to be independent of the O2 level, as glutamate increased the CO2 fixation under both 21% and 5% O2 conditions. L-methionine-dl-sulfoximine, an irreversible inhibitor of glutamate synthetase, however, inhibited the CO2 fixation markedly under 21% O2, but did not affect it under 5% O2 conditions. The treatment with NH4+ elevated the relative amounts of 14C incorporated into soluble components from 14CO2 with no relation to O2 levels, while glutamate increased 14C into insoluble components and neutral sugars. Glutamate, especially, seemed to stmulate the biosynthesis of starch under 5% O2 condition. These results indicated that NH4+ stimulated the degradation of sugar or starch and this proposal was confirmed by the increasing of pyruvate kinase activity in leaf discs treated with ammonium ion.

A Structured and Multi-cellular Model of Starch Biosynthesis in Potato

  • Recently, systems biology has been increasingly applied to gain insights into the complexity of living organisms. Many inaccessible biological information and hidden evidences fur example flux distribution of the metabolites are simply revealed by investigation of artificial cell behaviors. Most bio-models are models of single cell organisms that cannot handle the multi-cellular organisms like plants. Herein, a structured and multi-cellular model of potato was developed to comprehend the root starch biosynthesis. On the basis of simplest plant cell biology, a potato structured model on the platform of Berkley Madonna was divided into three parts: photosynthetic (leaf), non-photosynthetic (tuber) and transportation (phloem) cells. The model of starch biosynthesis begins with the fixation of CO$_2$ from atmosphere to the Calvin cycle. Passing through a series of reactions, triose phosphate from Calvin cycle is converted to sucrose which is transported to sink cells and is eventually formed the amylose and amylopectin (starch constituents). After validating the model with data from a number of literatures, the results show that the structured model is a good representative of the studied system. The result of triose phosphate (DHAP and GAP) elevation due to lessening the aldolase activity is an illustration of the validation. Furthermore, the representative model was used to gain more understanding of starch production process such as the effect of CO$_2$ uptake on qualitative and quantitative aspects of starch biosynthesis.

인삼 엽록체의 미세구조와 Photobleaching (Ultrastructural Feature and Photobleaching of ginseng Chloroplasts)

  • Ultrastructural and anatomical features of the leaf were studied in Panax ginseng C.A. Meyer(ginseng). The ginseng leaf poorly developed palisade tissue and the size of mesophyll cell was larger and the chloroplast density was lower than that of Glycine max (soyben). Ginseng chloroplast was filled with highly stacked grana and condensely-arrayed thylakoid, so the stroma space was hardly absorbed. However, ginseng mesophyll tissue and chloroplast array did not reduce light energy entering the mesophyll chloroplast, and the high LHCP/CP ratio of ginseng thylakoid resulted in the absorption of excess photon. It is reasonable to assume that 1O1-photogenearation by excess light energy partially resulted from the anatomical and ultrastructural characteristics of the ginseng leaf.

메밀(Fagopyrum esculentum) 꽃, 잎 추출건조물의 α-Amylase 효소활성 저해 ([ α ]-Amylase Inhibitory Activity of Flower and Leaf Extracts from Buckwheat (Fagopyrum esculentum))

  • 본 연구는 전분 가수분해효소인 ${\alpha}$-amylase의 효소활성 저해를 통해 식후 혈당조절제 개발 가능성을 확인하고자 메밀의 꽃, 잎, 줄기 및 곡립을 부위별로 분류하고 각각의 시료를 물, 40%, 70%, 100% 에탄올로 추출하고 건조한 추출건조물과 메밀건조 분말시료를 이용하여 in vitro에서 ${\alpha}$-amylase 활성 저해율을 측정하였다. 메밀부위별 물추출건조물(10%, w/w, starch basis)의 ${\alpha}$-amylase 활성 저해율은 꽃 66%, 잎 65%, 곡립 28%, 줄기 12% 순이었다. 이러한 결과는 추출 용매를 40%, 70%, 100% 에탄올로 하였을 때에도 유사하게 나타났다. 단지 잎추출건조물의 경우 70% 에탄올추출건조물의 효소활성 저해율이 100%와 40% 에탄올추출건조물보다 낮게 나타났다. 메밀 꽃과 잎 추출건조물을 기질의 0.1%, 0.5%, 1%, 2.5%, 5%, 10% 농도로 각각 첨가했을 때 에탄올로 추출한 꽃추출건조물의 경우 $0.5%{\sim}10%$ 농도에서 ${\alpha}$-amylase 활성을 약 90% 저해하였으며 이는 ${\alpha}$-amylase inhibitor인 acarbose와 유사한 수준이었다. 잎의 경우 100% 에탄올추출건조물에서 꽃과 비슷한 결과를 보였으며 40% 에탄올추출건조물에서는 꽃추출건조물보다 저해율이 다소 낮게 나타났다. 물추출건조물과 건조분말의 경우 에탄올추출물보다 꽃과 잎의 효소활성 저해율이 낮았다. 이러한 결과로 볼 때 메밀 꽃과 잎 추출건조물은 메밀 줄기 및 곡립 추출 건조물과 비교해서 효소활성 저해율이 높게 나타났으며, 물 추출이나 건조분말보다 100%, 40% 에탄올을 이용했을 때 추출건조물의 효소 저해율이 더욱 큰 것으로 나타나 이들 추출건조물은 식후 혈당조절제로 이용 가능성이 기대되는 소재로 사료된다.
