대청 추출물의 항산화 효소 활성 및 항균 효과 (Antioxidant Enzyme Activity and Antimicrobial Activity of Isatis tinctoria Extract)

  • 대청의 이용성 향상을 위한 자료 확보 측면에서 식물체 부위 및 추출 용매별에 따른 항산화 효소 활성 및 항균효과를 조사하였다. APX(Ascorbate Peroxidase) 활성은 줄기의 에탄올 추출물, 잎의 메탄올 추출물, 잎의 증류수의 추출물 순으로 높아 각각 1601.7, 1133.7 및 524.3(Unit/mg protein)을 나타냈다. CAT(Catalase) 활성은 꽃의 에탄올 추출물, 잎의 메탄올 추출물, 꽃의 증류수 추출물 순으로 높았는데, 각각 177.1, 120.8 및 55.4(Unit/mg protein)를 나타냈다. POD(Ascorbate Peroxidase) 활성은 꽃의 에탄올 추출물, 꽃의 메탄올 추출물, 줄기의 증류수 추출물 순으로 높았으며, 각각 27.1, 14.6 및 10.4(Unit/mg protein)를 나타냈다. SOD(superoxide dismutase) 활성은 뿌리의 증류수 추출물, 꽃의 메탄올 추출물, 뿌리의 에탄올 추출물 순으로 높았으며, 각각 90.8, 80.1, 75.5%를 나타냈다. 대청의 꽃 추출물은 Vibrio parahaemolyticus에 대해서만, 뿌리는 Staphy lococcus aureus에 대해서만, 줄기 추출물은 Bacillus subtilis, Escherichia coli, Staphy lococcus aureus에 대해서만 용매에 관계없이 항균활성을 나타냈다. 특히 대청 잎의 증류수 추출물은 Bacillus subtilis, Escherichia coli에 대해 높은 항균활성을 나타내어 저해환 직경이 각각 30.0 및 24.0 mm이었다. 이와 같은 결과는 약용식물로서 대청의 가치가 높음을 시사해 주었다.

Inhibition of TNF-α-mediated NF-κB Transcriptional Activity in HepG2 Cells by Dammarane-type Saponins from Panax ginseng Leaves

  • Panax ginseng (PG) is a globally utilized medicinal herb. The medicinal effects of PG are primarily attributable to ginsenosides located in the root and leaf. The leaves of PG are known to be rich in various bioactive ginsenosides, and the therapeutic effects of ginseng extract and ginsenosides have been associated with immunomodulatory and anti-inflammatory activities. We examined the effect of PG leaf extract and the isolated ginsenosides, on nuclear factor (NF)-${\kappa}B$transcriptional activity and target gene expression by applying a luciferase assay and reverse transcription polymerase chain reaction in tumor necrosis factor (TNF)-${\alpha}$-treated hepatocarcinoma HepG2 cells. Air-dried PG leaf extract inhibited TNF-${\alpha}$-induced NF-${\kappa}B$transcription activity and NF-${\kappa}B$-dependent cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) gene expression more efficiently than the steamed extract. Of the 10 ginsenosides isolated from PG leaves, Rd and Km most significantly inhibited activity in a dose-dependent manner, with $IC_{50}$ values of $12.05{\pm}0.82$ and $8.84{\pm}0.99\;{\mu}M$, respectively. Furthermore, the ginsenosides Rd and Km inhibited the TNF-${\alpha}$-induced expression levels of the COX-2 and iNOS gene in HepG2 cells. Air-dried leaf extracts and their chemical components, ginsenoside Rd and Km, are involved in the suppression of TNF-${\alpha}$-induced NF-${\kappa}B$ activation and NF-${\kappa}B$-dependent iNOS and COX-2 gene expression. Consequently, air-dried leaf extract from PG, and the purified ginsenosides, have therapeutic potential as anti-inflammatory.

Hydroxyapatite prepared from eggshell and mulberry leaf extract by precipitation method

  • Eggshell is a waste material after the usage of egg. In this work, biowaste chicken eggshells were used for preparing carbonated hydroxyapatite (HA) nanoparticles of high purity through aqueous precipitation method at room temperature. The eggshell-derived HA will be a cost-effective bioceramics for biomedical applications and an effective material-recycling technology. Additionally, mulberry leaf extract was used as a template to regulate the morphology, size and crystallinity of HA, and the effects of pH value were also examined. Characterization of the samples was performed by X-ray diffraction (XRD) and Fourier transform infrared (FT-IR) spectroscopy. Scanning electron microscopy (SEM) was used to determine the size, shape and morphology of HA. The results indicate that only one phase of HA were synthesized in the both absence and presence of mulberry leaf extract at pH of 7 and above, while DCPD or DCPA/DCPD phase was observed at pH 4 condition. The crystallite sizes of the HA samples obviously decreased when adding mulberry leaf extract as a template, while they decreased gradually as the solution pH levels increased. With increasing pH level from 7 to 14, the rod-like HA nanoparticles gradually changed to spherical shape at pH 14. Note that, the obtained product is Mg and Sr containing A- and B-type carbonate HA at alkaline pH and it can be a potential material for biomedical applications.

Studies on in-vivo Wound Healing Activity of Leaf Extract of Hypericum mysorense with Different Wound Model in Rats

  • The wound healing potential of the methanol extract of Hypericum mysorense Wight and Arn. Leaves (Family: Hypericaceae) was evaluated on different experimental models of wounds in rats. The methanol extract of leaves of Hypericum mysorense (HMM), in the form of ointment in two different concentrations (5% and 10% w/w ointment of aerial part extract in simple ointment base) was evaluated for wound healing potential in excision wound model and incision wound model in rats. Both the concentrations of the methanol extract ointment showed significant responses in both the wound types tested when compared with the control group. The effect produced by the extract ointment, in terms of wound contracting ability, wound closure time, regeneration of tissues at wound site, tensile strength of the wound and histopathological characteristics were comparable to those of a standard drug Nitrofurazone ointment.

상엽 추출물을 이용한 항당뇨 음료의 개발 (I) - 상엽 추출물의 제제화 탐색 - (Development of An Anti-Diabetic Functional Drink (I) - Screening of the Manufacturing of Mulberry Leaf Extract -)

  • 본 연구는 천연 자원인 뽕나무에서 혈당강하 효능을 가진 뽕잎(상엽)의 열수 추출법을 이용하여 상엽 추출물의 공정화 확립과 Formulation 개발을 위해 상엽 추출물과 silk peptide 및 한약재를 첨가한 시제품을 동물실험을 통하여 Formulation을 최종 결정하여 제품화하여 기능성 음료를 개발하고자 하였다. 연구 결과, 상엽 추출의 최적 조건은 상엽 크기는 3mm 이하로 하고, 2/3 정도 packing된 면포자루로 포장된 상엽을 사용하며, 상엽 사용량은 50~100 g/L, 추출 온도는 9$0^{\circ}C$, 추출 시간은 2 hr으로 하였으며 이때 수율은 20~25%였다. 또한 시료 배합에 따른 혈당강하 실험을 통하여 상엽 추출물과 silk peptide 의 농도는 각각 1%, 0.1%일 때 혈당강하 작용이 가장 우수하였으며 부재료로 사용된 둥글레는 상엽추출물의 10% 첨가한 경우가 혈당강하작용이 가장 컸으며, polydextrose도 효과가 있으므로 첨가하는 것이 바람직하다고 사료된다.

감나무 둥근무늬낙엽병균(Mycosphaerella nawae)의 배양적 특성 (Cultural Characteristics of Mcyosphaerella nawae causing Spotted Leaf Casting of Persimmon)

  • Mycosphaerella nawae, the causal organism, of spotted leaf casing disease of persimmon, was isolated from infected leaves showing typical symptom. The cultural characteristics of the fungus were compared on artificial media. Among 24 different combinations of culture media and supplements, oatmeal agar+persimmon leaf extract (PLE) and PAD+ PLE+streptomycin showed the highest rate of isolating as 57.1%. The best medium for mycelial growth of the fungus was PDA+persimmon leaf powder (PLP). The colony diameter was reached 47mm for 30 days at 2$0^{\circ}C$. PDA+PLE also supported good mcyelial growth showing 46mm of diameter in same condition. The optimum growth temperature of this fungus in PDA was recognized fairly low. The mycelial growth was higher at 2$0^{\circ}C$ than 15$^{\circ}C$. The variation of pH between pH 4 to pH 8 did not affect to the mycelial growth of the pathogen.

버어리종 잎담배의 화학성분에 의한 관능 특성 예측 (Prediction of Sensory Property from Leaf Chemical Property in Burely Tobacco)

  • This study was conducted to evaluate the prediction of sensory property of smoke from the leaf chemical property and characterize leaf chemical components for the best tobacco taste's leaves in burley tobacco. For analytical and sensory evaluations, sixteen grades were used. The major leaf chemical components to predict the sensory property of smoke were ether extract for tobacco-like, chloride for impact and total nitrogen/nicotine for irritation. Within ${\pm}20\;%$ range of difference, the predictable probabilities of sensory property of smoke from the leaf chemical properties were 100 % for tobacco-like, impact and irritation. As a result of K-means cluster analysis on the basis of tobacco taste, the desirable leaf chemical component contents were $6.5{\sim}6.8\;%$ in ether extract, $0.25{\sim}0.30\;%$ in chloride and $1.26{\sim}1.54$ in total nitrogen/nicotine ratio. This study suggest that the some regression equations may be useful to predict the sensory components of tobacco smoke from a few selected leaf chemical properties in burley tobacco and to select the burley tobacco leaves for enhance the tobacco taste of cigarette.

Effect of Alfalfa Plant Extracts on Germination and Early Seedling Growth of Forages

  • Alfalfa (Medicago sativa L.) plants have been reported to contain water-soluble substances that are autotoxic as well as allelopathic. Laboratory experiment through a petri-dish assay with imbibed seeds was conducted to evaluate both autotoxic and allelopathic effects of alfalfa leaf extracts on the germination and early seedling growth of alfalfa, red clover, crested wheatgrass, and Russian wildrye. Alfalfa seed germination was delayed dependent on extract concentration, with no difference in final germination at 72 hours. Root growth of alfalfa was stimulated up to 14% above control at very low concentrations of both leaf and stem extracts of alfalfa and was significantly reduced at extract concentration of more than 0.5g dry tissue/L (${gL}_{-1}$). Leaf extracts were generally more autotoxic for root growth than were stem extracts. Hypocotyl growth was not affected by all the concentrations of both leaf and stem extracts. Root length of legumes was more sensitive to the autotoxic chemicals from leaf extracts than was germination or shoot length. Hypocotyl growth of two legume plants and plant height of two grasses were not influenced by extracts. Seed germination and root growth of legumes were more inhibited by aqueous extracts of alfalfa leaf than were those of grasses. This result indicates autotoxic effect of alfalfa leaf extracts seems to be greater than allelopathic effect.

애플망고 잎의 추출물의 항산화 및 대식세포(RAW 264.7)에서 iNOS, COX-2 발현 저해 효과 (Antioxidant effect and iNOS, COX-2 Expression Inhibition on RAW 264.7 Cell of Mangifera indica L. Leafs)

  • 본 연구는 애플망고 잎 추출물의 항산화 및 항염증에 활성을 검증하여 연구하였다. 총 폴리페놀 함량은 Folin-Denis의 방법에 의해 측정하였다. 그 결과 애플망고 잎의 열수와 70% 에탄올 추출물은 각각 440.83±1.02, 475.63±1.3 mg/100 g TAE의 함량을 나타내었다. 항산화 활성을 측정하기위해 전자공여능 측정과 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) 라디칼 소거능 활성을 측정하였으며, 모든 추출물이 우수한 효과를 나타내었다. 대식세포내에서 애플망고 잎 추출물의 세포 생존율을 MTT assay의 방법으로 측정하였다. 세포의 독성이 나타나지 않은 농도구간에서 이하의 세포실험을 진행하였다. 항염증 활성을 효과적으로 측정하기 위해 LPS로 염증반응을 유도시키고 Griess assay를 이용해 nitric oxide를 측정하였다. 그 결과 애플망고 잎 추출물은 농도의존적으로 NO의 생산을 억제함을 나타내었다. LPS로 유도된 RAW 264.7세포에서 iNOS와 COX-2의 염증인자 생성억제를 mRNA발현 억제를 RT-PCR을 통해 확인하였다. 따라서 본 연구에서는 애플망고 잎 추출물의 항산화 및 항염증 활성의 우수한 효과를 확인하였고, 그 결과 천연소재로서의 유용성과 함께 화장품의 기능성소재로서의 활용가능성을 확인하였다.

구아바 잎 추출물의 총 폴리페놀과 플라보노이드 함량 및 항 염증 억제 효과 (Total Phenolic Compound, Total Flavonoid Compound And Anti-Inflammatory Inhibitory Effects of Psidium Guajava Leaf Extract)

  • 구아바 잎 추출물의 항산화 활성과 항염증 소재로서의 활용 가능성을 확인하고자 하였다. 구아바 잎 추출물의 총 폴리페놀 함량, 총 플라보노이드함량, RAW 264.7 세포에 대한 세포 독성 및 NO 생성 억제를 통한 항염증 효과, 피부에 대한 안전성 평가를 실시하였다. 본 실험 결과, 총 폴리페놀과 플라보노이드의 함량이 각 126.4 mg/g, 223.17 mg/g으로 높은 함량을 확인하였다. 대식세포인 264.7세포에 대한 세포 독성이 나타나지 않았으며 NO 생성을 농도 의존적으로 억제되는 것을 확인하였다. 구아바 잎 추출물을 함유한 크림을 제조하여 1차 첩포 테스트를 실시하여 피부 안전성을 확인한 결과 첩포 24 시간 후와 첩포 제거 24 시간 후에도 피부에 대한 자극은 확인되지 않았다. 이러한 결과를 종합해 볼 때 높은 총 폴리페놀과 플라보노이드의 함량으로 인한 항산화 활성이 우수하고, 세포에 대한 피부 독성이 적고 NO 생성 억제 효과가 확인됨에 따라 항산화 및 항염증 효과를 가진 소재로서의 활용 가능성을 확인하였다.
