Effects of NaOCl on Neuronal Excitability and Intracellular Calcium Concentration in Rat Spinal Substantia Gelatinosa Neurons

  • Recent studies indicate that reactive oxygen species (ROS) can act as modulators of neuronal activity, and are critically involved in persistent pain primarily through spinal mechanisms. In this study, we investigated the effects of NaOCl, a ROS donor, on neuronal excitability and the intracellular calcium concentration ($[Ca^{2+}]_i$) in spinal substantia gelatinosa (SG) neurons. In current clamp conditions, the application of NaOCl caused a membrane depolarization, which was inhibited by pretreatment with phenyl-N-tert-buthylnitrone (PBN), a ROS scavenger. The NaOCl-induced depolarization was not blocked however by pretreatment with dithiothreitol, a sulfhydryl-reducing agent. Confocal scanning laser microscopy was used to confirm whether NaOCl increases the intracellular ROS level. ROS-induced fluorescence intensity was found to be increased during perfusion of NaOCl after the loading of 2',7'-dichlorofluorescin diacetate ($H_2DCF$-DA). NaOCl-induced depolarization was not blocked by pretreatment with external $Ca^{2+}$ free solution or by the addition of nifedifine. However, when slices were pretreated with the $Ca^{2+}$ ATPase inhibitor thapsigargin, NaOCl failed to induce membrane depolarization. In a calcium imaging technique using the $Ca^{2+}$-sensitive fluorescence dye fura-2, the $[Ca^{2+}]_i$ was found to be increased by NaOCl. These results indicate that NaOCl activates the excitability of SG neurons via the modulation of the intracellular calcium concentration, and suggest that ROS induces nociception through a central sensitization.

  • The effects of the wall geometry on the spray-wall impingement process of a hollow-cone fuel spray emerging from a high-pressure swirl injector of the Gasoline Direct Injection (GDI) engine were investigated by means of a numerical method. The ized Instability Sheet Atomization (LISA) & Aerodynamically Progressed Taylor Analogy Breakup (APTAB) model for spray atomization process and the Gosman model were applied to model the atomization and wall impingement process of the spray. The calculation results of spray characteristics, such as a spray development process and a radial distance after wall impingement, compared with the experimental ones by the Laser Induced Exciplex Fluorescence (LIEF) technique. It was found that the radial distance of the cavity angle of 90$^{circ]$ after wall impingement was the shortest and the ring shaped vortex was generated near the wall after spray-wall impingement process.

  • Measurements of flame length, width and NOx emissions have been conducted to investigate the effect of an acoustic excitation on flame structure in turbulent hydrogen diffusion flames with coaxial air. The resonance frequency of oscillations was varied between 259 ,514 and 728 Hz with power rate of 0.405 and 2.88w. When these frequencies imposed to hydrogen flames, dramatic reduction of flame length and NOx emission was achieved. And acetone planar laser-induced fluorescence technique was used to measure a concentration of the near field of driven axisymmetric jet. The air-fuel stoichiometric line was plotted to investigate the mixing layer and development of air entrainment to fuel jet. Consequently, acoustic excitation on flame could enhance the air-fuel mixing resulting in abatement of NOx emission quantitatively.

  • 대기 중에서 HOx(OH, H $O_2$) 라디칼은 가장 중요한 산화제로서 광화학 반응의 핵심적 역할을 한다. 이러한 OH 라디칼의 정확한 농도를 측정하기 위한 여러 가지 시도가 있었으나, 그 존재량이 적고 수명이 짧기 때문에 측정에 많은 어려움이 있다. 간접적인 측정에 근거한 모델링과 계산적인 이론 값에 의하면 대류권에서의 전 지구적 평균 OH 라디칼의 농도는 대략 $10^{6}$molecules $cm^{-3}$이고, H $O_2$ 라디칼의 농도는 $10^{8}$molecules $cm^{-3}$정도로 추정된다. Impinging Atomization of Intermittent Gasoline Sprays (간헐 가솔린 분무의 충돌에 의한 미립화 촉진)

  • Experimental and analytical studies are presented to characterize the break-up mechanism and atomization processes of the intermittent- impinging-type nozzle. Gasoline jets passing through the circular nozzle with the outlet diameter of 0.4mm and the injection duration of 10ms are impinged on each other. The impingement of fuel jets forms a thin liquid sheet, and the break-up of the liquid sheet produces liquid ligaments and droplets subsequently. The shape of liquid sheets was visualized at various impinging velocities and angles using the planer laser induced fluorescence (PLIF) technique. Based on the Kelvin-Helmholtz wave instability theory, the break-up length of liquid sheets and the droplet diameter are obtained by the theoretical analysis of the sheet disintegration. The mean diameter of droplet is also estimated analytically using the liquid sheet thickness at the edge and the wavelength of the fastest growing wave. The present results indicate that the theoretical results are favorably agreed with the experimental results. The size of droplets decreases after the impingement as the impinging angle or the injection pressure increase. The increment of the injection pressure is more effective than the increment of the impinging angle to reduce the size of droplets.

Fast High-throughput Screening of the H1N1 Virus by Parallel Detection with Multi-channel Microchip Electrophoresis

  • A multi-channel microchip electrophoresis (MCME) method with parallel laser-induced fluorescence (LIF) detection was developed for rapid screening of H1N1 virus. The hemagglutinin (HA) and nucleocapsid protein (NP) gene of H1N1 virus were amplified using polymerase chain reaction (PCR). The amplified PCR products of the H1N1 virus DNA (HA, 116 bp and NP, 195 bp) were simultaneously detected within 25 s in three parallel channels using an expanded laser beam and a charge-coupled device camera. The parallel separations were demonstrated using a sieving gel matrix of 0.3% poly(ethylene oxide) ($M_r$ = 8,000,000) in $1{\times}$ TBE buffer (pH 8.4) with a programmed step electric field strength (PSEFS). The method was ~20 times faster than conventional slab gel electrophoresis, without any loss of resolving power or reproducibility. The proposed MCME/PSEFS assay technique provides a simple and accurate method for fast high-throughput screening of infectious virus DNA molecules under 400 bp.

Simultaneous measurement of velocity and temperature fields in micro-scale flow and its application to electrokinetic flow (마이크로 스케일 유동에서의 속도장 온도장 동시 측정 기법과 동전기 유동에의 적용)

  • In this paper, a technique of simultaneously measuring the velocity and the temperature in micro-scale flow is proposed. This method uses particle tracking velocimetry (PTV) for measuring the velocity and laser induced fluorescence (LIF) for measuring the temperature. To measure the accurate velocity and temperature, images for PTV and for LIF are separated by using two light sources and a shutter which is synchronized with a camera. By using only one camera, measurement system can be simplified and the error from complicate optical system can be minimized. Error analyses regarding the concentrations of fluorescent dye and particle and the light source fluctuation are also conducted. It is found that the error of the temperature and the velocity highly depends on the concentration of fluorescent particles which are used for PTV. This technique is applied to the simultaneous measurement of the velocity and the temperature in the electrokinetic flow. It is found that the velocity and temperature vary with the electric field strength and the concentration of electrolyte.

  • A direct photograph measurement technique was used to determine the spatial distribution of the spray droplet diameter in subsonic crossflow and it also obtain that SMD distribution by using PLLIF technique. The injector internal flow was classified as three modes such as a normal, cavitation, and hydraulic flip. The objectives of this research are getting a droplet distribution and drop size measurement of normal flow and compare with the other flow effects. Although the study showed visually that drop size were spatially dependent of Air-stream velocity, fuel injection velocity, and normalized distance from the injector exit length.(x/d, y/d) There are also difference characteristics between cavitation, hydraulic flip and the normal flow.

  • In the present study, oxygen transfer process across gas-liquid interface in a Y-shape micro-channel is quantitatively visualized using the micro laser induced fluorescence (${\mu}$-LIF) technique. Diffusion coefficient of Oxygen ($D_L$) is estimated based on the experimental results and compared to its theoretical value. Tris ruthenium (II) chloride hexahydrate was used as the oxygen quenchable fluorescent dye. A light-emitting diode (LED) with wavelength of 450 nm was used as the light source and phosphorescence images of fluorescent dye were captured by a CMOS high speed camera installed on the microscope system. Water having dissolved oxygen (DO) value of 0% and pure oxygen gas were injected into the Y-shaped microchannel by using a double loading syringe pump. In-situ pixel-by-pixel calibration was carried out to obtain Stern-Volmer plots over whole flow field. Instantaneous DO concentration fields were successfully mapped according to Stern-Volmer plots and DL was calculated as $2.0675{\times}10^{-9}\;m^2/s$.
