TESTING OF NEUROPEPTIDE Y INVOLVEMENT IN BLOOD FLOW REGULATION IN THE FELINE DENTAL PULP USING D-MYO-INOSITOL-1,2,6-TRISPHOSPHATE (D-myo-inositol-1,2,6-trisphosphate를 이용한 neuropeptide Y의 치수혈류 조절기능 연구)

  • The purpose of this study was to investigate whether D-myo-inositol-l,2,6-trisphosphate (PP56) can effectively antagonize vasoconstriction caused by neuropeptide Y in the dental pulp, and to understand involvement of neuropeptide Y in the regulation of microcirculation in the dental pulp with the aim of elucidating neurogenic inflammation. Experiments were performed on 7 cats anesthetised with sodium pentobarbital, and neuropeptide Y and a neuropeptide Y antagonist PP56 were injected close intra-arterially into the dental pulp. The probe of laser Doppler flowmeter was placed on the buccal surface of ipsilateral canine teeth to the drug administration and pulpal blood flow was measured. Intra-arterial injection of neuropeptide Y (1.3-$2.0\;{\mu}g$/kg) resulted in pulpal blood flow decrease of $37.73{\pm}5.73%$(mean${\pm}$SEM) (n=9). Intra-arterial injection of PP56(0.3 mg/kg) alone changed pulpal blood flow little by 1.03 % reduction. The effect of neuropeptide Y in the presence of PP56 resulted in significantly less decreases in pulpal blood flow ranging from $27.17{\pm}5.37$ to $16.63{\pm}3.48%$ from control as compared with neuropeptide Y alone(n = 13). In effect, PP56 attenuated pulpal blood flow caused by neuropeptide Y. Results of the present study have provided evidences that a non-peptide PP56 is capable of antagonizing vasoconstriction caused by neuropeptide Y in the feline dental pulp. In addition, they show functional evidences that neuropeptide Y plays an active role in modulating the microcirculation of the dental pulp.

  • This study was penonned to provide basic data that predict the usefulness of Gastrodiae rhizoma as food materials for oriental medicinal cuisine(YakSun). We measured the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats, following the intravenous injection of Gastrodiae rhizoma water extract The measurement was continually monitored by laser-doppler flowmeter and pressure transducer in anesthetized adult Sprague-Dawley rats for about two to two and half hours through the data acquisition system composed of MacLab and Macintosh computer. The results of this experiment were as follows. Gastrodiae rhizoma increased the changes of rCBF in rats significantly. The rCBF of Gastrodiae rhizoma did not change by pretreated propranolol, atropin, methylene blue and indomethacin. But the rCBF of Gastrodiae rhizoma was increased by pretreated L-NNA. Gastrodiae rhizaoma decreased the changes of BP significantly. The BP of Gastrodiae rhizoma did not change by pretreated propranolol, atropin, methylene blue and indomethacin. But the BP of Gastrodiae rhizoma was decreased by pretreated L-NNA. These results indicated that Gastrodiae rhizoma might increase the rCBF and the BP which related to nitric oxide synthesis. Also these results indicate that Gastrodiae rhizoma can be used as a safe and clinically applicable to diet therapy of cerebral cardiovascular disease.

Study on the Effects of Fructus Schizandrae on the Cerebral Blood Flow and Blood Pressure (뇌혈유와 혈압에 미치는 오미자의 효능에 대한 연구)

  • The purpose of this present study is to measure the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats. following the intravenous injection of drug. The measurement was continually monitored by laser-doppler flowmeter (Transonic Instrument, USA) and pressure tranducer(Grass, USA) in anesthetized adult Sprague-Dawley rats about for 2h to 2h and a half through the data acquisition system composed of MacLab and Macintosh computer. The result of this experiment was as following. 1. Fructus Schizandrae increased the changes of rCBF in rats, significantly. 2. The rCBF of Fructus Schizandrae did not change by pretreated propranolol. 3. The rCBF of Fructus Schizandrae did not change by pretreated atropine. 4. The rCBF of Fructus Schizandrae did not change by pretreated I-NNA. 5. The rCBF of Fructus Schizandrae was decreased by pretreated methylene blue. 6. The rCBF of Fructus Schizandrae did not change by pretreated indomethacin. 7. Fructus Schizandrae decreased the changes of blood pressure significantly. 8. The BP of Fructus Schizandrae did not change by pretreated propranolol. 9. The BP of Fructus Schizandrae did not change by pretreated atropine. 10. The BP of Fructus Schizandrae did nol change by pretreated I-NNA. 11. The BP of Fructus Schizandrae was decreased by pretreated methylene blue. 12. The BP of Fructus Schizandrae did not change by pretreated indomethacin. These results indicate that Fructus Schizandrae can increase the rCBF and decrease the BP, that related to guanylyl cyclase activity.

Effects of Joongjeo($TE_3$) Supplementation Aekmoon($TE_2$) Draining on Changes in Cerebral Blood Flow and Blood Pressure in Normal Rats (수소양삼초경(手少陽三焦經) 정격(正格)의 자경보사(自經補瀉)(중저 보(中渚 補), 액문 사(液門 瀉)) 자침(刺鍼)이 정상 흰쥐의 뇌혈류량(腦血流量) 및 혈압(血壓)에 미치는 영향(影響))

  • Objectives : Joongjeo($TE_3$) Supplementation Aekmoon($TE_2$) Draining is a method belongs to Ohaeng-acupuncture, using directional supplementation and draining. Methods : This study was designed to investigate the effects of $TE_3$ supplementation $TE_2$ draining on changes in cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) in normal rats. For these reasons, the present author investigated rCBF and MABP using laser doppler flowmeter in normal rats. In addtion, the present author also investigated action mechanisms of $TE_3$ supplementation $TE_3$ draining on changes in rCBF and MABP too. Results : In this results, $TE_3$ supplementation $TE_2$ draining elevated rCBF in time-dependent manner, but MABP levels decresed by $TE_3$ supplementation $TE_2$ draining. Pre-treatment with indomethacin (IDM), an inhibitor of cyclooxygenase, inhibited increase of rCBF effectively. But pre-treatment with methylene blue(MTB), an inhibitor of guanylate cyclase, decreased rCBF levels. In addition, pre-treatment with IDM also decreased MABP levels, but pre-treatement with MTB increased MABP levels. Conclusions : In conclusion, these results suggest that $TE_3$ supplementation $TE_2$ draining is effective to treat patient with disease related to cerebral ischemia, because $TE_3$ supplementation $TE_2$ draining can increase rCBF. In addition, the mechanisms are thought to be related to guanylate cyclase pathways.

Protective Effects of Cardiotonic Pills(CP) on Neuroglia Cells Against Oxidative Stress and the Effects on Regional Cerebral Blood Flow in Normal Rats (심적환(心適丸)이 산화적 손상에 따른 신경교세포 보호효과 및 국소 뇌혈류량 변화에 미치는 영향)

  • Objectives: SimJeok-Hwan(CP, Cardiotonic Pills) was made to treat patients with coronary arteriosclerosis, angina pectoris and hyperlipidemia. This study was designed to investigate the effects of CP on Proliferation rates neuroglia cells and protective effect of CP against oxidative stress, and also investigate the effects on regional Cerebral Blood Flow(rCBF) in normal rats. Methods: In this experiment, effects of CP on proliferation rates of neuroglia cells were measured using modified MTT methods. Oxidative stress was induced by treatment with 200 mM of hydrogen peroxide for 2 hr. rCBF and MABP were measured using Laser doppler flowmeter. Results: Treatment with CP elevated proliferation rates in C6 cells. In addition, CP protected cell death of C6 cells induced by oxidative stress. In results, rCBF was elevated by treatment with CP in normal rats. But, Mean Arterial Blood Pressure(MABP) did not affected by CP. In addition, the elevation of rCBF was blacked by pre-treatment with 1 mg/kg of indomethacin effectively. On the other hand, pre-treatment with 0.01 mg/kg of methylene blue did not affect rCBF level in normal rats. Conclusions: In conclusion, these results suggest that CP can act as anti-oxidant to protect neuroglia cells and also suggest that CP can elevate rCBF, which are involved in cyclooxygenase pathway.

Physiological and Psychological Effects of Vibroacoustic Stimulation to Scapular and Sacrum of Supine Position

  • Objective: This research measured physiological and psychological effects of Vibroacoustic stimulation(VA) to scapular and sacrum of supine position on the mattress. Background: When vibroacoustic stimulation applies to human body, it has a positive influence on physiological and psychological effects by stimulating the organs, tissues and cells of whole body. Method: This experiment was conducted to 10 normal males in two conditions: no stimulation and vibroacoustic stimulation. No stimulation experiment was executed as a supine position for 30 minutes without any vibrational stimulus, while vibroacoustic stimulation was transmitted by the vibrational speaker, which uses 40Hz frequency. Subjects had a laser Doppler flowmeter probe in scapular, sacrum, and also had 8 channel electroencephalogram(EEG) measurement sensor in the scalp. Blood pressure and skin temperature were measured in two conditions with an underlying posture for 30 minutes. Additionally, blood flow rate and EEG were measured before and after for two minutes on two conditions. Results: According to the vibroacoustic stimulation, blood flow rate and skin temperature were increased, while blood pressure was decreased. When using vibroacoustic stimulation compared to no stimulation, blood flow rate went approximately two times higher, and skin temperature also higher 3~4 times. Furthermore, the relative alpha power of brain wave was significantly increased when we applied to vibroacoustic stimulation. Conclusion: This experiment tested the VAT embedded in mattress in two conditions. According to this experiment, VAT decreases blood pressure, improves not only a physiological effect on blood flow rate as well as skin temperature, but also psychological functions by increasing relative alpha power. Application: The results of the publishing trend analysis might help physiological and psychological effects of vibroacoustic stimulation.

Changes in regional cerebral blood flow(rCBF) and blood pressure(BP) were measured in rats, following the intravenous injection of processed Gastrodiae rhizoma water extract. In its processing, we used rice water, Sderotium Poriae Cocos and Radix Ligustici Chuanxiaong. The rCBF and BP measurements were continually monitored by a laser-doppler flowmeter and a pressure transducer in the anesthetized adult Sprague-Dawley rats for approximately about two to two and a half hours, through a data acquisition system composed of a MacLab and Macintosh computer. The results of the experiment are as follows: the processed Gastrodiae rhizoma significantly increased changes in rCBF in the rats. The rCBF with processed Gastrodiae rhizoma did not change by pretreatment with propranolol, atropin, methylene blue, and indomethacin. But the rCBF of the processed Gastrodiae rhizoma was increased by pretreatment with L-NNA. The processed Gastrodiae rhizoma significantly decreased the changes in BP. However, BP with the processed Gastrodiae rhizoma did not change by pretreatment with propranolol, atropin, methylene blue and indomethacin. On the other hand, BP decreased with the processed Gastrodiae rhizoma pretreatment with L-NNA. These results indicate that processed Gastrodiae rhizoma might increase the rCBF and the BP which are related to nitric oxide synthesis. Also these results indicate that the used of processed Gastrodiae rhizoma in safe, as well as clinically applicable in diet therapy for cerebral related disease and hypertension.
