Chemical Composition and Size of Floating and Sunken Eggs of Olive Flounder Paralichthys olivaceus

  • Olive flounder (Paralichthys olivaceus) is one of the most commercially important species of farmed marine fish in Korea. Although techniques for rearing the larva of this species are improving, production costs are also increasing due to microbial influences and genetic degradation caused by successive culturing. Since the demand for healthy seed, which requires eggs of good quality of, is high, we examined the effects of nutrient composition on the size of P. olivaceus eggs. We analyzed floating (live) and sunken (dead) eggs of P. olivaceus from five different hatcheries for their size and amino and fatty acid composition. The sizes of eggs and oil globules from floating vs. sunken eggs were significantly different at p<0.05. No significant relationships were observed, however, among larval length, hatching percentage, and egg and oil globule size. The dry weight and amino acid levels of floating eggs were greater than those of sunken eggs (p<0.05), but no difference in fatty acid content was observed.

  • The effect of live food and commercial diet on sweetfish Plecoglossus altivelis culture was assessed by determining the daily growth rate and feed conversion ratio. After feeding for 8 weeks, the Moina macrocopa diet resulted in a higher growth rate of 5.101 compared to commercial (3.441) and Artemia (2.729) diets. In addition fish fed the M. macrocopa diet had the weight gain, averaging 41.29 g at the end of the experiment. The content of n-3 highly unsaturated fatty acids (HUFA) was highest in the commercial diet, while free fatty acids were not detected in the M. macrocopa diet. Triglycerides, the energy source in fishes, constituted 40.0% of the M. macrocopa diet, which was significantly higher than in the commercial and Artemia diets at 20.0 and 12.0%, respectively These results indicate that freshwater M. macrocopa can be used as feed for sweetfish larvae, even without docosahexaenoic acid (DHA) enrichment from Artemia.

  • The non-steroidal ecdysone agonist RH 5849 showed almost similar LC.o values( 18.1-26.5 ppm) at all stages of larval development of the tobacco cutworm, Spodoptera litura, when treated by a leaf-disk dipping method. The feeding-inhibition rate for the 4th instar larvae was dose-dependent, and simultaneously the weight gain of 3rd instar larvae ceased within 48 hour after feeding of the cabbage leafdisk dipped into RH 5849 4.2 ppm solution. The systemic larvicidal effect of RH 5849 was compared at cabbage and tobacco whole plant test. The $LC_{50}$ values below 20 ppm(mg/kg soil) was lasted for 15 days in cabbage, 30 days in tobacco respectively.

Influence of Temperature and Relative Humidity on the Rearing Performance and Disease Incidence in CSR Hybrid Silkworms, Bombyx mori L.

  • Fifth instar larvae of the new bivoltine hybrid (CSR2 ${\times}$ CSR5) were reared under different temperature and humidity viz., $20^{\circ}C$ and 85 ${pm}$ 5% R.H (T1), $25^{\circ}C$ and 70 ${pm}$ 5% R.H (T2-Optimum), $30^{\circ}C$ and 80% ${pm}$ 5 R.H (T3) and $35^{\circ}C$and 50 ${pm}$ 5% R.H (T4). The cocoon yield, cocoon characters and disease incidence were studied in normal (non infectious source, i.e control) rearing as well as in 1% infectious source of rearing. The results indicated that V instar larval duration was prolonged and cocoon weight was improved in T1. ERR and shell ratio were significantly improved and disease incidence was minimised in T2. Further significant difference was observed among the treatments with regard to spread of diseases.

  • A 6${\times}$4 top-cross set was made from the ten silkworm parental lines selected by the first principal component scores. They were also analysed for the relationship between the combining ability and the first principal component score. The highest general combining ability effects were detected in the parental lines of Japanese, N39 and chinese, C46, for the most quantitative characters in the study. The first principal component score of factors related to silk productivity in the parents was significantly and positively correlated to the general combining ability of the twelve characters such as cocoon yield, cocoon weight, cocoon shell weight, cocoon shell percentage, duration of the 5th instar larvae, total larval period, length of a bave, weight of a have, non-breaking length of a bave, non-breaking weight of a have, raw silk percentage, and neatness. Similarity distance (D$^2$) was related to the specific combining ability of the characters such as cocoon yield, non-breaking length of a bave, non-breaking weight of a have, non-breaking ratio of a bave, raw silk percentage, neatness. From the results, it is possible to predict the general combining ability effects by the principal component scores for the 12 characters of the parents related to silk productivity.

  • Series of studies were conducted to establish biological and chemical control method for Meloidogyne spp to medical herbs by applied of nematophagous fungi, Arthrobotrys spp, Fusarium spp, and egg parastic fungi, Paecilomyces lilacinus were applied for root-knot nematodes on medicinal herbs, Paeonia albiflora, Codonopsis lanceolata, Cnidium officinale. The results are as follow. In pot experiments, The no. of root gall and egg mass and larvae of Cnidium officinale. The results are as follow. In pot experiments. The no. of root gall and egg mass and larvae of Cnidium officinale, Codonopsis lanceolata, Paeonia japonica lowered in P. Lilacinus treated plots compare to untreated control plots. But A. thaumasia F. oxysporum treated plots were less effective. Effect of egg parasitic fungi and chemical treatment at divided root of Paeonia japonica after sterilized in pot were increased in the fresh weight, root weight, control effect in P. lilacinus treated plots as chemical, Carbo G treated plots compare to untreted control plots. I field experiment, the number of root gall, egg mass and nematode density of Paeonia were also suppressed in P. lilacinus treated plots. It was very effective continuous 2 years and transplanting time on Paeonia japonica infested soil with the M. hapla in field in both region, Chillgok and Euisung treated P. lilacinus as chemical treated plots. Soaking effect of insecticide for Paeonia japonica at diving shoot before transplating in pot were effective for 12hours immersion into 1,000 ppm of Benlate T + Mep Ec, Benlate T + Fenthion EC in pot and field Experiment. In the Examination of fungi activily on P.japonica field 1 year after soil treatment, Number of spore of P. lilacinus were 1,000~1,300 in 3 region except Euisung.

Effect of Mulberry Leaf Sprayed with Rutin and GABA on Silkworm Growth (Rutin 및 GABA 첨식이 누에 성장에 미치는 영향)

  • This experiment was carried out to investigate the feeding effect of rutin (quercetin-3-o-${\beta}$-rutinoside) and ${\gamma}$-aminobutyric acid(GAB) sprayed on mulberry leaves on silkworm growth. Amount of ingest, digest and feces showed no difference statistically except rutin. Rutin made 6% increase of amount of digest, compared to the control. However, rutin or GABA decreased maximaly silkworm weight by 28% during the experimental period. GABA showed body weight loss by 15%, rutin by 7% respectively, compared to the control. Rutin and GABA extended three days larval period and extended one day mounting period and decreased 15.4% and 91% in number of mounted silkworm compared with the control, respectively.

  • The storage proteins of the mulberry longicorn beetle, Apriona germari Hope, were purified and characterized. Three kinds of storage protein (SP1, SP2 and Sp3) were purified from the last instar larval hemolymph of A. germari by the FPLC techniques, anion exchange chromatography and gel permeation chromatography. The SP1, SP2 and SP3 have a native molecular weight of 480, 440 and 420 kDa, respectively. In the SDS-polyacrylamide gel electrophoresis analysis, these storage proteins are composed of a single protein subunit with molecular weight of 90, 85 and 80 kDa, respectively. This result showed that the storage proteins are hexameric protein. The SP1 and SP2 were stained with Schiffs reagent, but SP3 was not stained. It can be assumed that SP1 and SP2 are glycoprotein. Western blot analyses using the each of polyclonal antiserum against purified SP1, SP2 and SP3 showed that the three antibodies reacted with the each of SP bands, respectively. Also, antibodies against SP1 and SP3 cross-reacted with the SP3 and SP1, respectively. However, SP2 was not cross-reacted with these two antibodies. Also, antiserum against SP2 did not cross-reacted with the SP1 and SP3.

Light and Electron Microscopic Study on the Development of Nosema Bombyics Naegeli in the Midgut of Silkworm Bombyx Mori L.

  • Infection effect of $Nosema$ $bombyics$ on the midgut of silkworm $Bombyx$ $mori$ and subsequent appearance of spores and the performance of larvae was studied. Autopsy of larvae showed white pustules on the surface of midgut at 5 days of post infection (pi). At later stage, important organs like midgut, silk gland and gonads reduced in size and all these organs showed white pustules. Light microscope observation of pustules revealed enormous spores. Spore multiplication was at a faster rate in young larvae. Infection of the adult larvae resulted in pebrinized pupa and moths. Larval weight, cocoon weight and cocoon shell ratio reduced as the post infection period increased. Transverse sections of midgut showed $N.$ $bombycis$ infection limited to a few columnar cells at 3-5 days of pi. At 7 days pi, cell volume increased, cells were swollen and elongated. Heavily infected cells looked like sacks filled with parasite and the apical region of certain cells were bulging into the gut lumen. Later at 8-9 days of pi, spores or its developing stages leaked into the lumen either freely or enclosed within the globules of host cytoplasm. Besides columnar cells, development of $N.$ $bombycis$ was observed in the regenerative cells and rarely in goblet cells. Development of $N.$ $bombycis$ was also observed in both longitudinal and circular muscles at the late pi period. The histopathological changes, deformities and spore production time in the host were all influenced by the spore dosage and age of the host.

Breeding of New Productive Bivoltine Hybrid, CSR12 $\times$ CSR6 of Silkworm Bombyx mori L.

  • With an objective of evolving quantitatively and qualitatively superior bivoltine breeds/hybrids of silkworm Bombyx mori L. for tropical conditions, breeding work was initiated in Central Sericultural Research and Training Institute, Mysore during 1992 by utilizing two Japanese hybrids namely BNl8$\times$BCS25 and Shunrei$\times$Shogetsu along with Indian evolved breed, KA. The breed CSR12 which is characterized with sex-limited larval marking and white oval cocoons was evolved from the Japanese hybrid BNl8 ${\times}$ BCS25 by crossing with KA, while the breed CSR6 which is characterized with normal marking (marked larvae) and white dumbbell cocoons was extracted from the Japanese commercial hybrid Shunrei${\times}$Shogetsu through continuous inbreeding coupled with selection. After fixation, these breeds along with other newly evolved breeds were subjected to hybrid study under optimum environmental conditions in the laboratory for expression of full potential of the genotypes. These hybrids were evaluated by Multiple Trait Evaluation Index (Mano et al., 1993). The hybrid CSR12${\times}$CSR6 was selected based multiple trait evaluation index value. The hybrid CSR12$\times$CSR6 recorded survival of 96.0%, shell weight of 50.0 cg, shell ratio of 24.3%, raw silk percentage of 19.6, filament length of 1,216 m, boil off loss of 22.4% and renditta of 5.1. On the other hand, the control hybrid (KA ${\times}$ NB4D2) has recorded survival of 90.6%, shell weight of 42.1 cg, shell ratio of 20.4%, raw silk percentage of 15.9, filament length of 999 m, boil off loss of 24.8% and renditta of 6.3. The hybrid CSR12$\times$CSR6 was authorized during 1997 by Central Silk Board, Government of India for commercial exploitation during favourable months based on national level race authorization test.
