In order to stabilze alloxan as a reagent for detection of amino acids by spot test, sugars and other reductants were added to the aqueous alloxan solution. It was found that lactose was the best for the purpose. The alloxan reagent containing lactose did not give color change on blank test and was very stable that there was no color change even it was allowed to stand in room temperature for several months. The color reaction with amino acids and some amines was not affected by lactose. This spot test for amino acids is in sensitivity as comparable to that of the previously reported methods and gave color reaction with proline and hydroxyproline to 1${\gamma}$ and 5${\gamma}$ respectively.
To investigate the energy, protein, lipid, and lactose intakes of breast-fed infants of lacto-ove-vegetarians, 25 infants(13 boys, 10 girls) were examined on the 0.5th, 1st, 2nd, and 3nd months of lactation. The amount of breast milk intake was determined by a test-weighing method, and the energy content of protein, lipid, and lactose was calculated using the Atwater factor. Thedaily energy intake for boys averaged 316, 436, 447, and 431kcal and that of girls averaged 284, 399, 401, and 390kcal during lactation, respectively. Average energy intake of boys from milk from 2nd week to the 3rd month postpartum was significantly higher than that of girls(p<0.05), because milk intake volume of boys was much higher than that of girls. The protein intake of boys on 2nd week-3rd month averaged 7.33g/day and that of girls averaged 6.29g/day. The lipid intake of 2nd week, months averaged 21.8g/day and that of girls averaged 20.1g/day. The lactose intake of boys averaged 45.48g/day, while that of girls averaged 41.09g/day. The mean intake of protein and lactose in boys was significantly higher than that of girls.
A strain YB-9 was isolated from soil as a producer of the extracellular ${\beta}-D-galactosidase$, which catalyzes the hydrolysis of lactose. The strain YB-9 was identified as Streptomyces sp. on the basis of its cultural, morphological and physiological properties. After treating culture supematant of the isolate with ammonium sulfate $(15{\sim}70%)$, the precipitated protein was used as a crude ${\beta}-galactosidase$ for analyzing its reaction properties with $para-nitrophenyl-{\beta}-D-galactoside$$(pNP-{\beta}Gal)$ and lactose as substrates. The {\beta}-galactosidase showed its maximal activity at pH $6.0{\sim}6.5$ and $60^{\circ}C$. The hydrolyzing activity of ${\beta}-galactosidase$ for both $pNP-{\beta}Gal$ and lactose was decreased by galactose. Its hydrolyzing activity for lactose was slightly decreased by glucose, but the activity for $pNP-{\beta}Gal$ was increased to 1.3-folds by glucose. Especially, its hydrolyzing activity was not affected for lactose and was increased to 1.6-folds for $pNP-{\beta}Gal$ by xylose.
Dextransucrase hyper-producing Leuconostoc mesenteroides B-512FMCM and dextransucrase constitutive mutants B-742CB and B-1355C catalyzed the transfer of glucose from sucrose to other carbohydrates which were present or were added to the reaction digests. When the acceptor was a maltose, gentiobiose, lactose or raffinose, there was produced a series of oligosaccharide acceptor products or single product based on the kinds of enzymes and reaction conditions. To obtain the quantitative information about the yield and the distribution of acceptor products and dextran two experimental parameters were studied: a) the ratio of acceptor to sucrose and b) the amount of enzyme at constant carbohydrate concentration (100 mM). As the amount of enzyme increased, the synthesis of acceptor products (of maltose or gentiobiose) increased, and the formation of dextran decreased. As the ratio of acceptor to sucrose increased, the amount of dextran and the number of acceptor-products decreased and the amount of acceptor-products increased. When maltose or gentiobiose was an acceptor, the glucose from sucrose was transferred to the C-6 hydroxyl group of the nonreducing-end glucose residue of accepters to give a homologous series of isomaltosyl dextrins. In case of lactose or raffinose, there was produced only one acceptor product from B-512FMCM dextransucrase reaction. In the lactose acceptor reaction, the glucose from sucrose was transferred to the C-2 hydroxyl of the reducing end glucose residue of lactose. To get a series of oligosaccharides from lactose or raffinose acceptor reaction we used B-742CB dextransucrase or B-1355C alternansucrase with 500 mM sucrose in reaction digest.
Purpose: The aim of this study was to investigate the age of onset and the prevalence of lactose malabsorption in early childhood in Korea. Methods: We conducted a study of lactose malabsorption by breath hydrogen test in healthy children aged between 25~96 months old. Standard lactose loading (2 g lactose/kg, maximum 40g) test was done in 129 children and cow's milk (10 mL/kg) loading in 126 children followed by breath sampling of 60 and 120 minutes after the loading. An increase above baseline of 20 ppm or more was used as a criteria for positive responses. Results: The prevalence of lactose malabsorption was 7% in 25~36 months old, 19% in 37~48 months old, 35% in 49~60 months old, 55% in 61~72 months old, 82% in 72~84 months old, 80% in 85~96 months old children. Only 1% of the children showed positive result in breath hydrogen test after the cow's milk challenge. Conclusion: The prevalence of lactose malabsorption was increased between 37 months and 60 months of age, reached to adult level of prevalence after 72 months of age. When physiological dose of lactose was used as the challenge, the number of lactose malabsorbers become clinically insignificant.
The purpose of this study is to investigate the contents of protein, fat, lactose and energy in the human milk and the intake of human milk and those nutrients in breast-fed infants from 16 lactating women in Seoul area during the first 5 months postpartum. Daily human milk intake was determined by test-weighing method and the milk from 16 mothers was analyzed for protein (semimicro Kjeldahl, N $\times$ 6.38), fat (milk checker), lactose (HPLC). Energy was calculated by Atwater and milk factor. The mean intake of human milk to the 5th month of lactation was 668 $\pm$ 186 $m\ell$/day. Mean protein contents of the human milk at 0.5, 12, 3, 4 and 5 months postpartum showed 1.48, 1.35, 1.17, 1.11, 1.15 and 1.10 g/100 $m\ell$ respectively. Lipid and lactose contents of the human milk during the first S months postpartum averaged 3.38 g/100 $m\ell$ and 6.79 g/100 $m\ell$. Energy contents at 0.5, 12, 3, 4 and 5 months postpartum showed 64.2, 66.1, 68.3, 60.1, 58.9 and 61.2 g/100 $m\ell$ respectively. Protein intake of infants from the human milk was 8.22 g/day which was 54.8% of Korean RDA. Lipid and lactose intake of infants were 23.0 and 45.6 g/day. Energy intake of infants was 422.3 kcal/day which was 84.4% of Korean RDA. Total energy content was almost same value regardless of Atwater or milk factor but the energy ratio was some different. Protein and solid contents decreased the course of lactation but lactose content was less changed. These results suggest that more research were required for the RDA.
To evaluate the effect of low lactose special formula (MF-1) for the treatment of acute diarrhea, we studied thirty nine infants who were admitted with acute diarrhea to the Department of Pediatics, Yeungnam University Hospital. 39 infants of under 6 months of age who had been fed on formula feeds were randomly allocated to receive either a diluted regular formula milk or a low lactose special formula. Each infant received intravenous rehydration during fasting for 6-12 hours. Group 1 (n=15) was fed half strength of regular formula (80-100cc/kg/day) for the first 24 hours, three quarters strength formula (100-120cc/kg/day) for the next 24 hours, and continued feeding with the full strength regular formula milk. Group 2 (n=24), who fed the same amount of milk as Group 1, continued feeding with the full strength low lactose special formula from the start of feeding. Male to female sex ratio was 1.6 to 1. The characteristics of infants on admission were comparable in the age, the duration of diarrhea and the stool frequency before admission, the degree of dehydration. There were no significant differences in the duration of hospitalization, changes in stool weight and stool frequency after admission between two groups.(p>0.05) The Body weight and skin fold thickness were increased in group 2 who fed low lactose formula, but those who fed diluted regular formula showed reduction of body weight and skin fold thickness ($64.2{\pm}51.4g$ vs $-11.4{\pm}52.2g$, $0.6{\pm}0.8mm$ vs $-0.1{\pm}0.3mm$ respectively) during hospitalization. (p<0.05) We conclude that low lactose special formula milk can be recommended instead of diluted regular formula for acute diarrhea treatment in infants.

A total of 125 pigs (5.8 kg BW, 21 d of age) were allotted in a completely randomized block design experiment. Dietary treatment added carbohydrate sources : corn starch, lactose, glucose, sucrose or dried whey in corn-soybean meal based diet. Each treatment has 5 replicates with 5 pigs per replicate. Lactose, sucrose and dried whey supported a better growth performance than starch and glucose (p < 0.05) during the first and second week postweaning. However, in the third week postweaning no difference was found in ADG and ADFI among treatment. For overall period, pigs fed lactose, sucrose and dried whey diets showed similar growth performance and were superior to starch and glucose. The gross energy digestibility in pigs fed lactose, sucrose and dried whey diets were similar and significantly higher than those fed glucose and starch diets (p < 0.05). DM digestibility was not significantly affected by other carbohydrate sources except starch. Pigs fed lactose, sucrose and dried whey showed the best nitrogen digestibility. In all nutrients digestibility, there was no significant difference among treatment except starch and glucose diet. DM excretion was lower in pigs fed lactose, sucrose and dried whey than pigs fed starch and glucose. In conclusion, it appeared that sucrose could be effectively incorporated in baby pig diet without sacrificing growth performance.
${\beta}$-Galactosidase was immobilized on chitosan bead by covalent bonding using glutaraldehyde. The characteristics of the immobilized enzyme were investigated. Maximum immobilization yield of 75% was obtained on chitosan bead. Optimum pH and temperature for the immobilized enzyme was 7.0 and $50^{\circ}C$, respectively. The immobilized enzyme showed a broader range of pH and temperature compared to a free one. A mathematical model for the operation of the immobilized enzyme in a packed-bed reactor was established and solved numerically. Under different inlet lactose concentrations and feed flow rate conditions, lactose conversion was measured in a packed-bed reactor. The experimental results of continuous operation in a packed-bed reactor were compared to theoretic results using Michaelis-Menten kinetics with competitive product inhibition and external mass transfer resistance. The model predicted the experimental data with errors less than 5%. Process optimization of continuous operation in a packed-bed reactor was also conducted. In a recirculation packed-bed operation, conversion of lactose was 97% in 3 hours. In a continuous packed-bed operation, the effect of flow rate and initial lactose concentration was investigated. Increasing flow rates and initial lactose concentration decreased the conversion of substrate.
The matrix affects the dissolution of furosemide, which is almost insoluble in the dissolution medium. In order to understand the effect of the matrix on the dissolution of furosemide, lactose, starch, $Avicel\;^{\circledR}pH\;101$, $Avicel\;^{\circledR}pH\;301$, $SiO_2$ and talc were used as the matrix and the solvent deposition method were used. The dissolution characteristics of four dissolution medium were compared to each other using various ratio of drug-to-matrix. The results are as follows: 1) Lactose was shown to be superior and talc was to be inferior to the other matrixes investigated. 2) A maximum dissolution rate and dissolution amount of furosemide were observed in 1 : 10 ratio of the drug-to-matrix. 3) $T_{80%}$ of 1 : 10 ratio of the drug-to-matrix in pH 7.2 was 1 min. from FM-lactose and 30 min. from FM-talc. $T_{50%}$ in pH 4.2 is 2 min. from furosemide-lactose and 150 min. from furosemide-talc. Total amount of furosemide in pH 1.2 at 30 min. were enhanced 13.3 fold in furosemide-lactose and 3.5 fold in furosemide-talc compared to the control. Diuretic action of those furosemide-lactose and furosemide-talc was also evaluated by monitoring changes in urinary excretion of sodium, potassium and urine volume in rat. The accumulated urine volume were enhanced 1.7 fold in furosemide-lactose (1.5) compared to the furosemide.
