  • Fermented barley food was produced by the mixed culture fermentation with a tai-cults re system of a mold, a yeast and a lactobacillus. When Rhizopus delemar IFO 4746, Hansenular anomala IFO 0568 and Lactobacillus sp. L-5 were selected and cultivated on steamed barley at 3$0^{\circ}C$ for 2days and 37$^{\circ}C$ for 3days, the fermented product of good quality was obtained. During fermentation. changes in acidity, pH, water content and color of fermented barley were examined.

  • In this study of the simultaneous saccharification and fermentation (SSF) of paper sludge, fed-batch cultivation of Lactobacillus paracasei subsp. paracasei KLB58 was attempted to produce viable KLB58 cells and lactic acid. Optimal culture conditions, including the temperature and concentration of the supplemented enzyme, were examined in terms of lactic acid production and viable cell count. When the effects of culture temperature and $\beta$-glucosidase concentration were examined in fed-batch SSF, the highest viable cell counts and lactic acid production (i.e. 5$\times$$10^9$ CFU/ml and 45 g/L, respectively) were obtained at 37$^{\circ}C$ and 2 unit/ml of $\beta$-glucosidase.

Production and Characterization of Physiological Properties of Carotenoid from a Marine Bacterium Curtobacterium sp. (해양미생물로부터 Carotenoid의 생산 및 그 생리활성)

  • A marine bacterium producing carotenoid was isolated from the Yosu coastal area of South Korea, and has been recorded as MCPBK-1. It was identified as Curtobacterium sp.. The optimum conditions of marine carotenoid fermentation from Cutobacterium sp. were pH 7.0, a temperature of $25^{\circ}C$, 4 mM fructose as a carbon source, 0.07% tryptone as a nitrogen source, 0.5 mM $M^{+2}$ ion as a mineral source and $1{\;}\mu\textrm{M}$ of cyanocobalamine as a growth factor in a $7{\;}\ell$ jar-fermentor. 13.0 mg/ml of the marine carotenoid were produced under optimum conditions. The crude marine carotenoid isolated was composed of 5 different compounds, i.e : tunaxanthin(86.6%), diatoxanthin (7.1%), ${\beta}-carotene$ (2.1%), canthaxanthin(1.9%) and cynthiaxanthin (1.9%). Physiological properties including antibacterial activity, cytotoxic effect, antioxidative effect and free radical scavenging activity were characterized with the crude carotenoid, which exhibited no antibacterial activity against E. coli and Lactobacillus bulgaricus, but a strong cytotoxic effect against cancer cells such as HepG2 (Hepatocellular carcinoma, human, ATCC HB-8065) and HeLa (Cervical carcinoma, human, ATCC CCL-2) cells, the ratios of impediment were 86.4% and 39.2%, respectively. This carotenoid, also, expressed a strong antioxidative effect (83%) against CCL-13 (diploid, monotypic hepatocyte, human, ATCC CCL-13) and exhibited free radical scavenging activity (43.4%) when using at a concentration of $50{\;}\mu\textrm{g}/ml$ of the crude carotenoid.

Biological Probiotic Properties of Lactobacillus rhamonosus GG-4 Isolated from Infant Feces (유아분변으로부터 분리한 Lactobacillus rhamonosus GG-4의 생균제적 특성)

  • To develop probiotics, a kind of Lactobacillus sp. was isolated from infant feces. The bacterium was identified as Lactobacillus rhamnosus through 16S rDNA sequence analysis. The strain was a facultative anaerobe which grew better in aerobic conditions. The bacterium lowered the pH of the culture solution down to 2.4 during 48 hr in the MRS medium. The strain inhibited the growth of 6 pathogens - S. aureus, L. monocytogens, S. typhimurium, E. coli O-157, V. parahaemolyticus and P. aeruginosa. When the Lactobacillus were fed to chickens, along with commercial feed, for one month, amounts of $H_2S$ and $NH_3$ in the feces of the chicken decreased to 50% and 70%, respectively, compared to those of control group chickens. Amounts of other bad smells such as $(CH_3)SH$, $(CH_3)_2S$ and $(CH_3)_2S_2$ were not much different in the Lactobacillus-fed chickens compared to the control group. On the other hand, egg weights of the chickens fed Lactobacillus were higher by about $5{\pm}1\;g$ than those in the control group.

Quality characteristics and functionalities of Korean and Japanese spring Baechu cabbages and the kimchi prepared with such cabbages (한국산 및 일본산 봄배추와 이를 이용하여 제조한 김치의 품질특성과 기능성)

  • We examined the quality characteristics and functionalities of Korean and Japanese spring Baechu cabbages and the kimchi prepared with them. To study the physiochemical properties of the cabbages and the kimchis, we measured their water content, pH, acidity, microbial counts, and springiness. On the third week of the kimchi fermentation at $5^{\circ}C$, their sensory properties and in vitro DPPH radical scavenging and anticancer activities using AGS human gastric cancer cells were determined. The Japanese Baechu contained 97.1% water, and the Korean Baechu, 92.4%. The comparison of the textures of the raw Baechu and the brined Baechu showed that the Korean Baechu had higher springiness scores than the Japanese Baechu. After four-week fermentation, the springiness score of the kimchi with Korean Baechu was 53.5%, significantly higher than the 41.4% of the kimchi with Japanese Baechu. The kimchi prepared with Korean Baechu had a low total bacterial count but higher Lactobacillus sp. and Leuconostoc sp. counts than the kimchi with Japanese Baechu. The kimchi prepared with Korean Baechu had the highest overall acceptability score in the sensory evaluation test. The DPPH radical scavenging activity of the kimchi with Korean Baechu was 83.2%, and that of the kimchi with Japanese Baechu, 46.1%. When the AGS human gastric cancer cells were treated with the kimchis, the kimchi prepared with Korean Baechu showed a 45% cancer cell growth inhibition rate, and the kimchi with Japanese Baechu, 26%, at 1 mg/mL of methanol extracts. At the 2 mg/mL concentration, the kimchis with Korean Baechu and Japanese Baechu showed 97% and 74% inhibition, respectively. The Korean Baechu showed better quality than the Japanese Baechu, and the kimchi prepared with the Korean Baechu showed better kimchi quality and functionality than the Japanese Baechu.

Probiotic Properties of Lactic Acid Bacteria isolated from Feces and Kimchi (베트남인 분변 및 김치로부터 분리된 유산균의 프로바이오틱스 기능성 연구)

  • The purpose of this study was to investigate the probiotic properties and antioxidant capacity of lactic acid bacteria isolated from Vietnamese feces and the Korean traditional food kimchi. Six isolated strains were identified as Lactobacillus sp. by 16S rRNA sequencing. All strains showed good resistance to low pH (1.5, 2.0, and 3.0) and 0.3% oxgall bile acids. Culture filtrates from the six strains showed various antioxidant effects, including DPPH, ABTS, reducing power, and metal chelating ($Fe^{2+}$) activities. Two of the six Lactobacillus strains showed potential probiotic activity. Heat resistance and adhesion assays were conducted by mixing the selected strains, Lactobacillus acidophilus V4, Lactobacillus plantarum V7, and Lactobacillus paracasei DK121 isolated from kimchi. The results showed that the heat resistance of these strains was similar to that of a commercial strain, L. plantarum LP. In addition, a mucin attachment assay using the mixture of selected strains (V4, V7, and DK121) showed high binding activity to the mucous layer. In conclusion, a mixture of V4, V7, and DK121 shows promising probiotic activity and may be useful for the development of health-related products.

Rapid Processing of the Fish Sauce and Its Quality Evaluation (속성어간장 제조 및 품질 평가)

  • Changes in chemical characteristic, microflora, and sensory evaluation of fish sauce extracted at an interval of one week from fermented solution were investigated. pH was reduced from 6.0 to 4.5, and trimethylamine oxide from 132.5 to 87.2 mg/100g during fermenting periods. Trimethylamine increased from 5.6 to 50.2 mg/100g, and volatile basic nitrogen from 48.3 to 232.5 mg/100g. Bacterial flora isolated from the fish sauce were 70% Lactobacillus sp. and 13% Bacillus sp. Among the free amino acids, alanine, glutamic acid, valine, and methionine contents constitute 40% of the total free amino acids. Major non-volatile organic acid of the fish sauce was lactic acid (76%). Sensory evaluation results of the fish sauce were higher than the traditional soybean sauce after 28 days of fermentation.

Effects of extreme heat stress and continuous lighting on growth performance and blood lipid in broiler chickens (연속조명과 폭염 스트레스가 육계의 혈액지질 및 성장능력에 미치는 영향)

  • In this study, the effect of extreme heat diet on growth performance, lymphoid organ, blood immunoglobulin and cecum microflora change in broilers exposed to continuous lighting and extreme heat stress (EHS) was studied. Broilers raised under normal environment temperature ($25^{\circ}C$ or extreme heat stress temperature ($33{\pm}2^{\circ}C$, and consumed chow diet (CD) or extreme heat stress diet (EHSD). Five hundred Ross 308 day-old commercial broilers were arranged in a completely randomized block design of 5 treatment groups with 4 repetitions (25 heads per repetition pen). The broilers were divided into: T1 (normal environment+CD), T2 (EHS+CD), T3 (EHS+EHSD in which the tallow in CD was substituted by soy oil and contained 5% molasses), T4 (EHS+EHSD in which the tallow in CD was substituted by soy oil and contained 5% molasses, and 1.5 times more methionine and lysine than CD), and T5 (EHS+EHSD in which the tallow in CD was substituted by soy oil, contained 5% molasses, 1.5 times more methionine and lysine than CD, and 300ppm of vitamin C). The EHS significantly reduced the body weight gain and feed intake. The blood immunoglobulin, bursa of Fabricius, thymus, and spleen weight were significantly reduced when broilers were exposed to EHS. Compared to the normal environment temperature group, the cecum Lactobacillus sp. was low in the EHS treatment group, while Escherichia sp., Salmonella sp. and total aerobic bacteria in the EHS treatment group were high. A statistically significant difference was acknowledged between the treatment groups.

Standardization of Manufacturing Method and Lactic Acid Bacteria Growth and CO2 Levels of Nabak Kimchi at Different Fermentation Temperatures (나박김치의 제조 표준화 및 발효온도별 젖산균의 생육과 CO2 생성)

  • Ingredient ratio for making nabak kimchi and the manufacturing method were standardized from the available literatures. Fermentation properties and $CO_2$ production of the nabak kimchi were investigated during the fermentation at $5^{\circ}C,\;10^{\circ}C,\;and\;20^{\circ}C$. Standardized ingredients ratio of nabak kimchi that added 100 mL of water was as follows: 45.0 g baechu cabbage, 26.9 g radish, 1.9 g green onion, 1.0 g red pepper, 1.2 g crushed garlic, 0.9 g crushed ginger, 0.7 g red pepper powder. The standardized manufacturing method of nabak kimchi was as follows: washing ingredients, cutting radish and baechu cabbage $(2.5\times2.5\times0.5\;cm)$, salting for 20 min, washing and draining, pretreatment of ingredients, dissolving red pepper powder in water, blending, mixing, and adding the water to the mixed ingredients. Fermentations at $5^{\circ}C$ for 8 days, at $10^{\circ}C$ for 3 days, and at $20^{\circ}C$ for 1 day led to the acidity levels of $0.21\%,\;0.20\%,\;and\;0.31\%$, respectively. From the relationships between optimally ripened pH and acidity, nabak kimchi showed lower acidity of $0.20\~0.25\%$ with pH $4.2\~4.5.$ Like other kinds of kimchi, the Levels of Leuconostoc sp. were high specially at later stage of fermentation at low temperature $(5^{\circ}C)$. However, the levels of Lactobacillus sp. were low at $5^{\circ}C$. Nabak kimchi produced high levels of $CO_2$ in the initial fermentation period and followed by rapid decrease of $CO_2$ production with the fermentation. From the relationships between pH and $CO_2$ content, the highest $CO_2$ contents were found pH $4.0\~4.4$, 3.8 and 3.4 at $5^{\circ}C,\;10^{\circ}C$, and $20^{\circ}C$, respectively. This fact indicated that fermentation at $5^{\circ}C$ has the highest $CO_2$ content at optimally ripened pH of 4.3 and the fermentation at lower temperature such as $5^{\circ}C$ could extend the eatable time of nabak kimchi.
