Carbonic Maceration처리에 의한 Campbell Early 발효액의 감산 효과: 감산 관련 미생물의 분리 및 특성 (Deacidification Effect of Campbell Early Must through Carbonic-Maceration Treatment: Isolation and Properties of the Bacteria Associated with Deacidification)

  • Carbonic maceration 처리는 포도주 제조 시 사과산을 감소시키는 방법으로 사과산 감소의 원인 중 미생물의 영향을 알아보고자 사과산을 감소시키고 젖산을 생성시키는 미생물을 분리, 동정한 결과Lactobacillus brevis, Lactobacillus plantarum 및 Streptococcus thermophilus의 젖산균이 존재하는 것으로 나타났다. 분리된 균들은 대부분 당에서 젖산을 생성하는 균으로 알려져 있으며 사과산을 함유한 배지에서 균의 배양 중 사과산을 이용하지 않고 젖산을 생성하는 것으로 보아 주로 당을 이용하여 젖산을 생성하는 것으로 보인다. 사과산을 이용해 젖산을 생성시키는 대표적인 malo-lactic bacteria인 Oenococcus oeni 균은 본 실험에서는 동정되지 않았다. 따라서 carbonic maceration 처리 시 사과산의 감소는 포도에 자연적으로 생육한다는 malo-lactic bacteria나 감산 관련 미생물의 영향은 크게 받지 않는 것으로 판단되며, 젖산 함량의 증가는 당을 이용하는 다양한 젖산균에 의해 생성되는 것으로 판단된다.

Production of GABA (gamma amino butyric acid) by Lactic Acid Bacteria

  • Gamma-amino butyric acid (GABA) is a kind of pharmacological and biological component and its application is wide and useful in Korea specially, becoming aging society in the near feature. GABA is request special dose for the purposed biological effect but the production of concentrated GABA is very difficult due to low concentration of glutamic acid existed in the fermentation broth. To increase GABA concentrate using fermentation technology, high content of glutamic acid is required. For this reason, various strains which have the glutamic acid decarboxylase (GAD) and can convert glutamic acid to GABA, were isolated from various fermented foods. Most of GABA producing strains are lactic acid bacteria isolated from kimchi, especially added monosodium glutamate (MSG) as a taste enhancer. Optimizing the formulation of culture media and the culture condition, GABA conversion yield and amounts were increased. Finally GABA concentration of fermentation broth in batch or fed batch fermentation reached 660 mM or 1000 mM, respectively. Furthermore formulation of culture media for GABA production developed commercially. Many studies about GABA-rich product have been continued, so GABA-rich kimchi, cheese, yogurt, black raspberry juice and tomato juices has been also developed. In Korea many biological effects of GABA are evaluated recently and GABA will be expected to be used in multipurpose.

김치에서 분리한 저온성 젖산균의 배추즙에서의 배양 (Cultivation of Psychrotrophic Lactic Acid Bacteria Isolated from Kimchi in Korean Cabbage-Juice)

  • The purpose of this study was to predict the actions and roles of 10 strains of representative psychrotrophic lactic acid bacteria, in kimchi fermentation, which were Isolated from kimchi and identified as Leu. mesenteroides subsp. mesenteroides, Leu. mesenteroides subsp. dextranicum, Leu. lactic, Leu. paramesenteroides, Lac. bavaricus and Lac. homahiochii. For this, 0.01% of tactic starters were inoculated in germ free Korean cabbage-juice containing 2.5% NaCl, and then cultivated for 14 days at 1$0^{\circ}C$. All strains grew actively, and reached their stationary phase in 4 days. In death phase, the slopes of curves were much different each other by strains. The acidity increased rapidly between 2 and 4 days, and the pH decreased rapidly between 2 and 3 days. The total acidity was 0.5B~0.75%, the volatile acidity 0.04~0.18% and the pH 3.55~3.85, in final cultures. The cultures of Leuconostocs were better than those of lactobacilli on flavor test. It was thought that the ripening periods of kimchi would be much reduced, and that the over ripening would be also somewhat avoided, when these strains were used as starter bacteria for kimchi and the kimchi was fermented at low temperature.

탁주발효에 있어서 발효미생물군의 변동에 대하여 (A study on the microflora changes during Takju brewing)

  • In order to study ecology of microorganisms during Takju brewing, microflora changes were examined fromm the start to the sixth day of Takju fermentation in 24 hours intervals. Takju made from rice, flour and dried sweet potato in a liter volume open container at the laboratory and a sanple of Takju brewing factory were studied for their microflora and their changes during fermentationl together with a sample of Kokja. Results obtained were as follows ; 1. The followings were the identified microorganisms in Kokja. The molds ; Absidia spinosa, Aspergillus parasiticus. The yeasts ; Candida melinii, Candida Solani, Hansenula anomala. The bacteria ; Luctobacillus casei, Leuconostoc mesenteroides, Bacillus subtilis, Bacillus pumilus. 2. Torulopsis inconspicua, Lactobacillus casei, Leuconotoc mesenteroides, Bacillus subtilis, Bacillus pumilus were isolated from main mash of laboratory-made Takju samples. The yeast, Torupsis inconspicua which was not present in Kokja and, probably of a contaminant yeast, dominated the yeast flora of Takju mash of rice, flour and sweet potato of labotatory brewing. The laboratory brewing lost also always showed large population of lactic acid bacteria flora. 3. None of the wild yeasts which were present in Kokja appeared in Takju mashes. The Kokja appears to be of no use as the yeast source for Takju fermentation. Also the Kokja appears to be of not so effective amylolytic and proteolytic enzyme sources considering the microflora characteristics. Probably the major role of Kokja in Takju fermentation may be to contribute in taste formation. 4. Inoculation of Sacharomyces cerevisiae into the mash to the level of $10^7$ ml at the start of fermentation greatly changed the ecological aspects eliminating conditions of rather slow rising of natural contaminant yeast populaiton and fermentation which might give rise to prosperity of lactic acid and Bacillus bacteria that would be avoidable. 5. Examination of microflora of the large factory scale Takju fermentation showed the quite similar pattern of microflora and their changes to that of the cultured yeast-inoculated laboratory batch Takju fermentation. The cultured yeast dominated as the only predominant microflora, and the lactic acid bacteria flora were completely suppressed and aerobic bacteria, greatly. Probably this may be the regular microflora pattern of normal Takju fermentation. The role of lactic acid bacteria and aerobic bacteria in Takju fermentation may not be clear yet from this experiment alone.

사료용 생균제 개발을 위한 마늘 내성 유산균의 분리 (Isolation of Garlic Resistant Lactic Acid Bacteria for Feed Additives)

  • 마늘의 항균력에 저항성을 갖는 유산균을 분리하기 위해서 마늘 추출물이 첨가된 배지에서 생육하는 112 균주를 김치, 젓갈, 장아찌류로부터 분리하였다. 이 중에서 14균주에 대해서 내산성 및 내담즙성 그리고, 가축 병원성 세균인 Salmonella choleraesuis, Escherichia coli, Staphylococcus aureus, Shigella flesneri에 대해 항균력을 조사한 결과, 파김치 유래의 P'GW50-2 균주가 가장 우수한 특성을 지니고 있어 생균제로 이용 가능한 균주로 선발하였다. 선발된 균주는 16S rRNA 유전자의 염기서열 분석결과 Lactobacillus plantarum으로 동정되었고 L. plantarum TJ-LP-002으로 명명하였다. L. plantarum TJ-LP-002는 Bacillus cereus, S. aureus, Clostridium perfringens와 같은 Gram 양성균과 Aeromonas hydrophila, E. coli, Pseudomonas, Salmonella, Shigella와 같은 Gram 음성균에 대해서 비교적 넓은 항균활성을 나타내었다.

Pseudomonas sp. JH007에 의한 DL-2-Chloropropionic Acid로부터 D-Lactic Acid의 생산

  • For the production of D-lactic acid from DL-2-chloropropionic acid, about 80 strains of bacteria capable of assimilating DL-2-chloropropionic acid as a sole carbon and energy source were isolated from the soil. JH-007 strain that showed the higest productivity of D-lactic acid and didn't produce L-lactic acid from DL-2-chloropropionic acid was selected from them and identified as Pseudomonas sp. The optimal conditions for the production of D-lactic acid from DL-2-chloropropionic acid were examined. The resting cells of JH-007 cultured in LB medium containing 3 g/l of DL-2-chloropropionic acid were used as an enzyme source. The reaction mixtures for the maximal production of D-lactic acid were consist of 10 g/l of resting cells and 3 g/l of DL-2-chloropropionic acid in 125 mM sodium carbonate buffer. The optimal pH for the reaction was 10.0 and the optimal temperature was 30$\circ$C. When 1 g/l of DL-2-chloropropionic acid was added intermittently to the reaction mixture under the above condition, 5.72 g/l of D-lactic acid was produced after incubation of 5 hrs. This amount of D-lactic acid corresponded to a 98.4% yields and the optical purity was 99.8%.

젖산 발효제품에서 분리한 유산균의 내산성 비교 (Comparative Tests on the Acid Tolerance of Some Lactic-Acid-Bacteria Species Isolated from Lactic Fermented Products)

  • 유제품 30여종과 야채발효 제품 2종에서 Lactobacillus casei ssp.(5 strains), Lactobacillus acidophilus(2 strains), Lactobacillus delbrueckii ssp. bulgaricus(6 strains), Lactobacillus plantarum(3 strains), Streptococcus salivarius ssp. thermophilus(11 strains), and Streptococcus faecalis(2 strains)를 각각 분리하여 이들의 내산성과 담즙산 내성을 평가하였다. 초기생균수를 고려하여 공분산분석으로 내산성을 비교한 결과, L. casei에 속하는 균주들의 내산성이 가장 우수하였으며, L. bulgaricus와 균주들은 가장 낮은 내산성을 보였다(p<0.05). 담즙산 내성은 L. casei, L. plantarum, L. acidophilus, S. feacalis 균주들의 경우 비슷한 경향을 보였으나, L. bulgaridus와 S. thermophilus 그룹에 속하는 균주들의 담즙산 내성은 거의 없는 것으로 나타났다.

Rapid detection of beer-spoilage lactic acid bacteria: Modified hop-gradient agar with ethanol method

  • Hop-resistant lactic acid bacteria (LAB) are well-known, major beer-spoilage bacteria. The hop-gradient agar containing ethanol (c-HGA+E) method has been used to examine hop-resistance of beer-spoilage LAB. However, the selection of beer-spoilage bacteria by the c-HGA+E method is either too selective or too inclusive. Furthermore, it is accompanied by a complicated experimental procedure, high-cost and time. To overcome these disadvantages, the modified hop-gradient agar with ethanol (m-HGA+E) method was developed. The most remarkable modifications were the shape of the petri dish and the inoculation method for bacteria. The efficiency and validation of the m-HGA+E approach were proven by the formation of colonies at different hop concentrations in the bottom layer, co-culture with the bacteriocin producer and by PCR detection of hop-resistant genes. This study demonstrated that m-HGA+E is a rapid, economical, and easy method to monitor potential hop-resistant beer-spoilage LAB during the beer brewing process.

Effect of Lactobacillus acidophilus based probiotic product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs

  • The aim of this study was to evaluate the effect of Lactobacillus acidophilus probiotic (LAP) product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs. In total, 14 Beagle dogs with an average initial body weight of 10.19 ± 0.61 kg were randomly assigned into two dietary treatments,with and without LAP supplementation, for a 28-day feeding trial. At the end of the experiment, there was no significant (p > 0.05) difference in the concentration of serum total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), white blood cell (WBC), red blood cell (RBC), blood lymphocyte percentage, fecal hydrogen sulfide (H2S) and total mercaptans (R.SH) emission, and fecal coliforms counts. However, the serum concentrations of the triglyceride and fecal ammonia (NH3) emission of the LAP treatment were significantly (p < 0.05) decreased in the group compared with the CON dogs. Fecal total lactic acid bacteria counts were significantly (p < 0.05) increased in the LAP treatment. In conclusion, the supplementation of LAP in Beagle dog diets could decrease the blood triglyceride level and enhance the gut Lactobacillus count which may have positive effects on dogs.

유용미생물 (EM, Effective Microorganisms)의 활용 현황 (Current Status of EM (Effective Microorganisms) Utilization)

  • Effective Microorganisms (EM), a fermented medium developed by Professor Higa at the University of the Ryukyus, is a mixed culture containing dozens of microorganisms which are beneficial to nature including people, animals, plants and many microbial species in environment. EM is known to contain more than 80 kinds of anaerobic or aerobic microbes including photosynthetic bacteria, lactic acid bacteria, yeast, actinomycetes, fungi and so on, with yeast, lactic acid bacteria and photosynthetic bacteria as the main species of EM. Antioxidant effect generated by the concert of complex coexistence and coprosperity among these microbes is considered to be the main source of EM benefits. Currently, EM is earning an increasing attention with applications in agriculture, forestry, animal husbandry, fisheries, environment and medicine among others. At the same time, however, a quantitative interpretation of EM system based on a mixed culture model needs efforts from biochemical engineers for efficient production and further promotion of EM. In this paper, we describe the functions of major microbes in EM and current researches and applications of EM in agriculture, forestry, animal husbandry, fisheries, environment and medicine.
