H2O2로 유발된 산화적 스트레스에 대한 브로콜리(Brassica oleracea var. italica) 잎 분획물의 PC12 cell 보호 효과 (PC12 Cell Protective Effects of Broccoli (Brassica oleracea var. italica) Leaf Fraction against H2O2-induced Oxidative Stress)

  • 브로콜리(Brassica oleracea var. italica) 잎의 n-hexane, chloroform, ethyl acetate, butanol 및 distilled water 분획물의 total phenolic 화합물 함량은 chloroform 분획물이 206.8 mg GAE/g으로 가장 높게 나타났으며, ABTS radical 소거활성 및 지질 과산화(MDA) 생성 억제효과 실험 결과에서 chloroform 분획물에서 가장 높은 활성을 나타내었다. in vitro 항산화 실험들에서 우수한 효과를 보인 chloroform 분획물을 이용하여 $H_2O_2$으로 유도한 산화적 스트레스에 대한 신경세포(PC12 cell)에서의 산화적 스트레스 생성 억제효과, 세포 생존율 그리고 세포막 손상 억제효과 역시 농도 의존적 경향을 나타내며 positive control 로 사용된 Vit.C와 유의적인 결과를 나타냈다. 마지막으로, HPLC 분석 결과 브로콜리 잎 chloroform 분획물에 존재하는 주요 phenolic 화합물은 feulic acid인 것으로 확인되었다. 본 연구 결과를 종합해볼 때, 생리활성 물질로서의 ferulic acid 등을 함유한 브로콜리 잎 chloroform 분획물은 in vitro 항산화 활성과 함께 신경세포 보호효과를 나타내는 고부가가치 기능성 소재로의 활용이 기대된다.

보중익기탕 가미방(補中益氣湯 加味方)과 선방패독탕(仙方敗毒湯)이 기도 점액의 분비와 뮤신 유전자발현에 미치는 영향 (Effects of Bojung-ikgitang-gamibang and Seonbang-paedoktang on Secretion of Airway Mucus and Expression of Mucin Gene)

  • Objectives In the present study, the author intended to investigate whether bojung-ikgitang-gamibang(BJGB) and seonbang-paedoktang(SBPT) significantly affect in vivo and in vitro mucin secretion from airway epithelial cells. Methods In vivo experiment, mice's mucin which is on a hypersecretion of airway mucin, mice's tracheal goblet cells in hyperplasia and mice's intraepithelial mucosubstances were exposed with SO2for3weeks. Effects of orally-administered BJGB and SBPT during 1 week on vivo mucin secretion and hyperplasia of tracheal goblet cells were assessed by using both enzyme-linked immunosorbent assay(ELISA) and staining goblet cells with alcian blue. In vitro experiment, confluent hamster tracheal surface epithelial(HTSE) cells were metabolically radiolabeled with 3H-glucosamine for 24hrs and chased for 30 min in the presence of each agent to figure out the effectiveness of 3H-mucin secretion. Total elution profiles of control spent media and treatment sample through Sepharose CL-4B column were analyzed. The effects of each agent on contractility of isolated tracheal smooth muscle and effects of each agent on MUC5AC gene expression in cultured HTSE cells were investigated. Also, possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Additionally, effects of BJGB and SBPT on both MUC5AC gene expression in cultured HTSE cells and TNF- or EGF-induced MUC5AC gene expression in human airway epithelial cells (NCI-H292) were investigated. Results (1) BJGB and SBPT inhibited hypersecretion of in vivo mucin. SBPT also inhibited the increase the number of goblet cells. However, BJGB did not affect the increase of number of goblet cells; (2) BJGB significantly increased mucin secretion from cultured HTSE cells, without significant cytotoxicity, and chiefly affected the 'mucin' secretion; (3) SBPT did not affect mucin secretion from cultured HTSE cells without significant cytotoxicity, and also did not affect the secretion of the other releseable glycoproteins; (4) BJGB and SBPT did not affect Ach-induced contraction of isolated tracheal smooth muscle; (5) SBPT significantly inhibit the expression levels of MUC5AC gene and BJGB significantly increased the expression levels of MUC5AC gene in both HTSE cells and NCI-H292 cells. Conclusions BJGB and SBPT can not only affect the secretion of mucin but also affect the expression of mucin gene. The author suggests that the effects BJGB and SBPT with their components should be further investigated and it is highly desirable to find from oriental medical prescriptions, novel agents which might regulate hypersecretion of mucin from airway epithelial cells.

가미통규탕(加味通竅湯)이 호흡기 뮤신 분비 및 기관 평활근 긴장도에 미치는 영향 (Effect of Gamitonggyu-tang on Secretion of Airway Mucin and Contractility of Tracheal Smooth Muscle)

However, it did not affect the increase the number of goblet cells (2) GTT significantly increased mucin release from cultured HTSE cells, without significant cytotoxicity (3) GTT chiefly affected the 'mucin' secretion and did not affect the secretion of the other releasable glycoproteins with less molecular weight than mucin (4) GTT did not affect Ach-induced contraction of isolated tracheal smooth muscle.Conclusions This result suggests that GTT can increase mucin secretion during short-term treatment (in vitro) whereas it can inihibit hypersecretion of mucin during long-term treatment (in vivo). The author suggests that the effect GTT with their components should be further investigated and it is valuable to find from oriental medical prescriptions, novel agents which might regulate mucin secretion from airway epithelial cells.

산화적 스트레스에 대한 석결명의 세포 보호 효과 (Cellular-protective effects of Nardotidis seu Sulculii Concha Extract against oxidative stress)

  • Objectives : This study investigated cellular-protective effects of Nardotidis seu Sulculii Concha water extract (NSCE) against oxidative stress induced by arachidonic acid (AA)+iron or tert-butylhydroperoxide (tBHP). Methods : In vitro, MTT assay was assessed for cell viability, and immunoblotting analysis was performed to detect expression of AMP-activated kinase (AMPK) signaling pathway and autophagy related proteins. In vivo, mice were orally administrated with the aqueous extract of NSCE of 500 mg/kg for 3 days, and then injected with CCl4 0.5 mg/kg body weight to induce acute damage. The level of liver damage was measured by serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) and lactate dehydrogenase (LDH) analysis. Results : Treatment with NSCE inhibited cell death induced by AA+iron and tBHP. NSCE induced the phosphorylation of AMPK, and this compound also induced the phosphorylation of LKB1, an upstream kinase of AMPK, and Acetyl-CoA carboxylase (ACC), a primary downstream target of AMPK. NSCE increased the protein levels of autophagic markers (LC3II and beclin-1) and decreased the phosphorylation of mammalian target of rapamycin (mTOR) and simultaneously increased the phosphorylation of unc-51-like kinase-1 (ULK-1) in time-dependent manner. Conclusions : NSCE has the ability 1) to protect cells against oxidative stress induced by AA+iron or tBHP. NSCE 2) to activate AMP-activated protein kinase (AMPK), and 3) to regulate autophagy, an important regulator in cell survival.

구아바 열매와 잎 열수 추출물의 신경세포 보호효과 (Neuronal Cell Protective Effects of Hot Water Extracts from Guava (Psidium guajava L.) Fruit and Leaf)

  • PC12 신경세포를 이용하여 구아바 열매와 잎 추출물이 $H_2O_2$로 유도된 신경세포 독성에 대한 보호 효과를 조사하였다. 구아바 열매와 잎 열수 추출물의 총페놀성 화합물 함량은 각각 11.75 및 293.25 mg/g이었고, gallic acid 함량은 각각 22.78 및 117.34 mg/100 g이었다. $H_2O_2$ 처리한 PC12 cell내의 활성산소 생성억제효과를 측정한 결과 구아바 잎열수 추출물에서 높은 활성산소 생성 억제효과를 보였다. MTT방법을 이용하여 $H_2O_2$로 유도된 PC12 신경세포에 대한 보호효과를 측정한 결과 구아바 열매와 잎 열수 추출물은 높은 신경세포 보호효과를 보였고, LDH release 실험결과 모든 농도에서 세포막 보호효과를 나타내었다. 본 연구 결과를 종합해 볼 때, 구아바 열매와 잎 열수 추출물의 항산화활성과 산화적 스트레스로부터의 신경세포 보호효과를 나타내어 퇴행성 뇌신경질환 등의 예방 및 개선을 위한 산업화 소재로서의 활용 가능성이 높다고 판단된다.

Proteomic Assessment of the Relevant Factors Affecting Pork Meat Quality Associated with Longissimus dorsi Muscles in Duroc Pigs

  • Meat quality is a complex trait influenced by many factors, including genetics, nutrition, feeding environment, animal handling, and their interactions. To elucidate relevant factors affecting pork quality associated with oxidative stress and muscle development, we analyzed protein expression in high quality longissimus dorsi muscles (HQLD) and low quality longissimus dorsi muscles (LQLD) from Duroc pigs by liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteomic analysis. Between HQLD (n = 20) and LQLD (n = 20) Duroc pigs, 24 differentially expressed proteins were identified by LC-MS/MS. A total of 10 and 14 proteins were highly expressed in HQLD and LQLD, respectively. The 24 proteins have putative functions in the following seven categories: catalytic activity (31%), ATPase activity (19%), oxidoreductase activity (13%), cytoskeletal protein binding (13%), actin binding (12%), calcium ion binding (6%), and structural constituent of muscle (6%). Silver-stained image analysis revealed significant differential expression of lactate dehydrogenase A (LDHA) between HQLD and LQLD Duroc pigs. LDHA was subjected to in vitro study of myogenesis under oxidative stress conditions and LDH activity assay to verification its role in oxidative stress. No significant difference of mRNA expression level of LDHA was found between normal and oxidative stress condition. However, LDH activity was significantly higher under oxidative stress condition than at normal condition using in vitro model of myogenesis. The highly expressed LDHA was positively correlated with LQLD. Moreover, LDHA activity increased by oxidative stress was reduced by antioxidant resveratrol. This paper emphasizes the importance of differential expression patterns of proteins and their interaction for the development of meat quality traits. Our proteome data provides valuable information on important factors which might aid in the regulation of muscle development and the improvement of meat quality in longissimus dorsi muscles of Duroc pigs under oxidative stress conditions.

호두 추출물의 항산화 활성과 신피질에서 세포 손상과 지질과산화 방지효과 (Antioxidant Activity of Extract from Walnut Uuglans sinensis Dode) and Its Protective Effect on Cell Injury and Lipid Peroxidation in Renal Cortical Slices)

  • 본 연구에서는 호두박 분획층의 항산화 활성을 검색하기 위해 MeOH, $CH_2Cl_2$, EtOAc, BuOH 그리고 $H_2O$ 분획으로 추출하여 수행하였고, 각 분획층의 산화적 세포손상과 지질 과산화의 방지 효과를 in vitro에서 확인하였다. 그 결과 호두박의 free radical (DPP보 radical) 소거 활성은 EtOAc층에서 가장높게 나타났고, 활성산소종 억제율과 peroxynitrite $(ONOO^-)$ 소거 활성은 $CH_2Cl_2$층을 제외한 모든 분획층에서 높게 나타났다. 신피질 절편에 t-BHP의 처리 시 LDH의 방출과 지질과 산화를 증가시켰고 이러한 변화는 호두의 MeOH, EtOAc, n-BuOH 분획에 의해서 완전하게 방지되었다. 이러한 결과는 호두 추출물이 t-BHP에 의한 신장 세포 손상에 효과적이고 이러한 효과는 항산화력에 의한 것으로 추측된다. 또한 이러한 결과는 호두 추출물에서 많은 질병의 원인이 되고 있는 산화적 스트레스를 방지할 수 있는 효과적인 약물 개발의 가능성을 시사한다

표고 유산균 발효물의 성분 및 간기능 보호 효과 (Chemical components and hepato-protective effect of Lentinula edodes fermented by lactic acid bacteria)

  • 본 연구에서는 기존에 인체에 유익한 효과가 있고 부작용 없이 장기간 안전하게 사용할 수 있으며 간보호 및 간기능 개선에 탁월한 효과가 있는 표고를 추출 및 농축하여 원유에 농도별로 첨가하여, 3종의 유산균을 이용하여 발효하였다. 표고의 농도를 달리하여 유산균을 배양하였을 때 면역관련 물질인 ergothioneine 및 β-glucan의 함량을 측정한 결과 ergothioneine함량은 30% 표고추출농축액에 L. plantarum이 첨가되어 배양된 발효물이 40.48 mg/100 g로 가장 높은 함량을 나타냈다. β-Glucan은 30% 표고추출농축액에 L. brevis가 첨가된 발효물이 13.94%로 가장 높은 함량이 나타났다. 두 면역관련 물질 모두 표고추출농축액 첨가량이 증가할수록 증가하는 경향을 나타냈다. SD rat을 간을 적출하여 일차 간세포로 분리해 발효물의 간보호 효과를 측정하였다. 표고추출농축액과 유산균 발효물을 10, 50, 100, 500 ㎍/mL을 일차 간세포에 처리하여 MTT assay 방법으로 세포독성을 측정한 결과, 모든 농도에서 독성을 보이지 않았다. 또한 10 mM 아세트아미노펜을 처리하여 독성을 유발한 일차 간세포에 시료를 처리하여 AST, ALT, LDH를 측정한 결과, 효소의 농도가 감소하여 간보호 기능에 효과가 있음을 확인하였다. 이중 L. brevis를 첨가하여 배양한 발효물 처리구에서 효소의 농도가 가장 감소한 것을 보았고, 가장 간보호 효과가 뛰어난 것을 확인하였다. 이상의 결과를 토대로 표고추출농축액에 유산균을 첨가하여 발효하였을 때, 기존의 표고보다 면역관련 물질의 함량이 증가하고, 간기능 보호에 더욱 효과가 있음을 입증하였다. 따라서 본 연구결과는 표고 유산균 발효물이 건강기능 소재로서 연구를 위한 기초 연구자료로 활용될 수 있 것으로 생각된다.
