Comparative Hepatotoxicity Assessment of Cadmium and Nickel with Isolated Perfused Rat Liver(IPRL) (적출간 관류법을 이용한 카드뮴과 니켈의 간독성 비교)

  • Objectives : It is the objective of this study to compare hepatotoxicity of nickel chloride and cadmium chloride with each other through IPRL(Isolated Perfused Rat Liver) method. Methods : Biochemical indicator of hepatic function such as AST(aspartate aminotransferase), ALT(alanine aminotransferase), LDH(lactate dehydrogenase) and perfusion flow rate were used as the indicator of hepatotoxicity. Oxygen consumption rate were used as vability indicator. $300({\pm}50)g$ - weighted rats were allocated randomly to each group($0{\mu}M,\;50{\mu}M,\;200{\mu}M\;NiCl_2\;and\;CdCl_2$ exposure) by 5, totally 25. After Krebs-Ringer bicarbonate butler solution flowed into the penal vein and passed the liver cell, it flowed out of vena cava. Liver was administered with each $NiCl_2\;and\;CdCl_2$ of each concentration and observed with buffer solution sampling time. Butler which got out of liver was sampled and then biochemical indicator of hepatotoxicity was measured. Results : AST, ALT, and LDH in buffer increased with sampling time much more in $CdCl_2$ exposure group than $NiCl_2$ exposure group in both 50 and $200{\mu}M$ and statistical significance w3s verified with 2-way repeated ANOVA. Viability was decreased more and more in all substances during passed time. Conclusions : It is inferred that $CdCl_2$ has stronger hepatotoxicity than $NiCl_2$. IPRL method would be used widely for acute hepatotoxicity when considerating the benefit of it.

Study on Geochemical Behavior of Heavy Metals by Indigenous Bacteria in Contaminated Soil and Sediment (국내 일부 오염 토양 및 퇴적물 내 토착 미생물에 의한 중금속의 지구화학적 거동 연구)

  • Microbial control of the geochemical behavior of heavy metals (Cd, Cu, Pb, and Zn) and As in contaminated subsurface soil and sediment was investigated through activation of indigenous bacteria with lactate under anaerobic condition for 25 days. The results indicated that dissolved Cd, Pb and Zn were microbially removed from solutions, which was likely due to the formation of metal sulfides after reduction of sulfate by indigenous sulfate-reducing bacteria. Soils from the Dukeum mine containing a large amount of sulfate resulted in complete removal of dissolved As after 25 days by microbial activities, while there were gradual increases in dissolved As concentration in soils from the Hwabuk mine and sediments from the Dongducheon industrial area which showed low $SO_4{^2-}$ concentrations. Addition of appropriate carbon sources and sulfate to contaminated geological media may lead to activation of indigenous bacteria and thus in situ stabilization of the heavy metals; however, potential of As release into solution after the amendment should be preferentially investigated.

Analysis of the Physicochemical Characteristics and Sensory Properties in Makgeolli (시판 생막걸리의 이화학 성분과 관능적 특성 분석)

  • This study analyzed the physicochemical characteristics and sensory properties of makgeolli. Fifteen samples of 4 award winning makgeolli and 11 samples of non-award winning makgeolli in a Korea liquors contest were examined. Physicochemical analysis revealed a significant differences in the alcohol content, reducing sugar concentration, color difference, and volatile compounds between the award winning and non-award winning makgeolli (p<0.05). The award winning makgeolli was related to the high contents of alcohol and reducing sugar, bright color, and low contents of ethyl myristate. A total of 24 terms (appearance 2, odor 10, flavor and taste 8, and texture 4) were expressed in terms of makgeolli. Regarding the sensory property, the sweet, raspberry, pear, and yogurt smell were positively related to the award winning makgeolli. In addition, nuruk, savory smell, and savory flavor were positively related to the non-award winning makgeolli. A study of association between the volatile compounds and odor attribute revealed ethyl lactate to have a positive correlation with the raspberry smell (r=0.840) and pear smell (r=0.723) in the award winning makgeolli (p<0.01). The characteristics of makgeolli preferred by consumers are strongly related to the sweet taste and smell, bright color, and fruit flavor.

Effect of Dietary Supplementation of Terrapin or Snake Extract on Exercise Performance in Rats (용봉탕 또는 사탕(蛇湯) 보충식이가 횐쥐의 지구력 운동 수행능력에 미치는 영향)

  • Effect of terrapin or snake extract on forced-swimming capacity and related biochemical parameters of SD rats was evaluated. Treatment groups were fed diet supplemented with 5% terrapin extract or 1% snake extract for 4 weeks. After adaptation for 1 week, each group was divided into two subgroups: one group swam for 90 min (90-min subgroups), and the other swam until exhaustion (all-out subgroups). No significant difference was observed in swimming time until exhaustion among the groups. After swimming for 90 min, contents of fatigue factors of serum such as lactate, inorganic phosphate, creatine kinase, and ammonia were not significantly different (p < 0.05) between control and treatment groups, including all-out subgroups, except for lactate concentration. These results indicated that terrapin or snake extract had little effect on forced-swimming capacity of rats.

  • A rapid assay to determine respiration inhibition of Saccharomyces cerevisiae by chemicals was developed. S. cerevisiae was harvested with two different liquid media, yeast extract-peptone-dextrose (YPD) medium capable of occurring both glucose fermentation and mitochondrial respiration, and non-fermentable carbon-yeast extract (NFY) medium capable of occurring respiration only Wells in 96-well plate were loaded with each cell suspension and various concentrations of 46 fungicides with various modes of action. n NFY medium, the non-fermentable carbon source, ethanol (NFY-E medium), glycerol (NFY-G medium) or lactate (NFY-L medium), was used. After incubation for $1{\sim}3$ days, minimum inhibitory concentrations (MICs) of the chemicals were recorded in the media. Of the 46 inhibitors employed in this study, four inhibitors of fungal respiration by blockage of electron flux in the mitochondrial respiratory chain, azoxystrobin, kresoxim-methyl, metominostrobin, and trifloxystrobin, exhibited strong antifungal activity in all of NFY media, but no activity in YPD medium. In contrast to this, five N-trihalomethylthio fungicides showed much stronger antifungal activities in YPD medium than three NFY media. Eleven fungicides inhibited growth of S. cerevisiae in all media and the other 26 fungicides showed no antifungal activity in all media. Thus, our rapid and efficient in vitro method can be considered as an alternative assay system for respiration inhibitor.

Development of Polyclonal Antibodies to Abdominal and Subcutaneous Adipocyte for Reducing Body Fat of Rat (흰쥐 체지방 감소를 위한 지방부위별 특이 다클론 항체 개발 연구)

  • Polyclonal antisera against regional (abdominal and subcutaneous) fats were developed to reduce body fat in rats. Isolation and culture of abdominal and subcutaneous adipocytes of rats were performed for analyzing lactate dehydrogenase (LDH) concentration. At the level of 1:1,000 dilution, little antibody reactivity appeared in non-immunized serum whereas both of antisera against abdominal (AAb) and subcutaneous adipocyte plasma membrane proteins (SAb) had relatively strong reactivity till the level of 1:128,000 dilutions. Compared with regional fats, extremely low reactivities of AAb and SAb were detected with PMP of the organs (p<0.001). Both AAb and SAb were most strongly reacted with each adipocyte plasma memebrane proteins and showed statistically (p<0.01) higher cross-reactivities compared with non-immunized serum based on LDH analysis. In conclusion, these results may indicate that the present polyclonal antibodies against regional inedible adipocyte plasma membrane proteins are well developed and have safety in cross-reactivities with body organs.

4Effects of Turmeric (Curcuma longa L.) Supplementation on Creatinine and Hepatic Functional Enzyme Activities in Streptozotocin-induced Diabetic Rats (울금(Curcuma longa L.) 섭취가 당뇨성 흰쥐의 크레아티닌 및 간 기능 효소 활성에 미치는 영향)

  • The purpose of this study was to investigate the improvement effect of turmeric (Curcuma longa L.) on the hepatic functional enzyme and catalase activity of streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley (SD) male rats were divided into four groups (n=6), and fed experimental diets containing turmeric meal [basal diet+5% turmeric (BT), basal diet+STZ+5% turmeric (ST)], and control (basal diet, BD), BS groups (basal diet+STZ). Serum concentrations of creatinine and blood urea nitrogen (BUN) were significantly decreased (p<0.05) by 5% turmeric supplementation diet. The activities of akaline phosphatase (ALP), lactate dehydrogenase (LDH), aspartate transaminase (AST), alanine transaminase (ALT), amylase and lipase were decreased in the BD, BT and ST group than BS group. The catalase (CAT) activity was significantly increased (p<0.05) in turmeric supplementation diet (BT, ST group) than diabetic group (BS). Furthermore, the activities of amylase and lipase in the sera of turmeric diet group were significantly decreased (p<0.05). In vivo experiments with diabetic rats showed that ingestion of turmeric supplementation diet were effective in creatinine concentration, and hepatic functional enzyme activities.

Effects of using different roughages in the total mixed ration inoculated with or without coculture of Lactobacillus acidophilus and Bacillus subtilis on in vitro rumen fermentation and microbial population

  • Objective: This study aimed to determine the effects of different roughages in total mixed ration (TMR) inoculated with or without coculture of Lactobacillus acidophilus (L. acidophilus) and Bacillus subtilis (B. subtilis) on in vitro rumen fermentation and microbial population. Methods: Three TMRs formulations composed of different forages were used and each TMR was grouped into two treatments: non-fermented TMR and fermented TMR (F-TMR) (inoculated with coculture of L. acidophilus and B. subtilis). After fermentation, the fermentation, chemical and microbial profile of the TMRs were determined. The treatments were used for in vitro rumen fermentation to determine total gas production, pH, ammonianitrogen (NH3-N), and volatile fatty acids (VFA). Microbial populations were determined by quantitative real-time polymerase chain reaction (PCR). All data were analyzed as a 3×2 factorial arrangement design using the MIXED procedure of Statistical Analysis Systems. Results: Changes in the fermentation (pH, lactate, acetate, propionate, and NH3-N) and chemical composition (moisture, crude protein, crude fiber, and ash) were observed. For in vitro rumen fermentation, lower rumen pH, higher acetate, propionate, and total VFA content were observed in the F-TMR group after 24 h incubation (p<0.05). F-TMR group had higher acetate concentration compared with the non-fermented group. Total VFA was highest (p<0.05) in F-TMR containing combined forage of domestic and imported source (F-CF) and F-TMR containing Italian ryegrass silage and corn silage (F-IRS-CS) than that of TMR diet containing oat, timothy, and alfalfa hay. The microbial population was not affected by the different TMR diets. Conclusion: The use of Italian ryegrass silage and corn silage, as well as the inoculation of coculture of L. acidophilus and B. subtilis, in the TMR caused changes in the pH, lactate and acetate concentrations, and chemical composition of experimental diets. In addition, F-TMR composed with Italian ryegrass silage and corn silage altered ruminal pH and VFA concentrations during in vitro rumen fermentation experiment.

The Effects of Saururus chinensis Baill Extract Administration on the Blood Glucose, Electrolyte and Lipid Metabolism in STZ-Induced Hyperglycemic Rats (Streptozotocin 유도 당뇨성 흰쥐의 혈당, 전해질 및 지질대사에 삼백초 추출액이 미치는 영향)

  • The purpose of this study was to observe the effects of the feeding physiological activity substance in Saururus chinensis Baill extract on the improvement of the blood glucose, electrolyte (Na, K, CI) concentration and lipid metabolism in the serum of streptozotocin (STZ, 55 mg/kg B.W., I.P. injection)-induced hyperglycemic rats fed the experimental diets for 5 weeks. Concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglyceride (TG) and phospholipid (PL) in serum were significantly higher in the hyperglycemic group (group NSW), STZ (I.P.)+Saururus chinensis Baill leaf 3.5 g% extract group (group NSSL) and STZ (I.P.)+Saururus chinensis Baill root 3.5 g% extract group (group NSSR) than those in the control group (group Normal, basal diet+water). But the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, TG and PL in serum were remarkably lower in the groups NSSL and NSSR than those in the NSW, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the groups NSSL and NSSR were significantly higher than in the group NSW. Electrolyte and creatinine concentrations in serum were significantly lower in the groups NSSL and NSSR than those in the group NSW. The activities of aminotransferase (AST, ALT), creatine phosphokinase (CPK), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum were remarkably lower in the groups NSSL and NSSR than in the hyperglycemic group NSW. However, no significance was found in the comperative effect of the groups NSSL and NSSR. From the above results, it was suggested that Saururus chinensis Baill was effective in the improvement of the blood glucose, electrolyte, glucide and lipid metabolism in serum of STZ-induced hyperglycemic rats.

Effect of Anchovy Treated with Ethanol, Citric Acid and Dietary Calcium Supplements on Calcium Metabolism in Rats (주정과 구연산 및 식이성 칼슘소재를 처리한 멸치분말이 흰쥐의 칼슘대사에 미치는 영향)

  • This study was performed to investigate the effect of ethanol and citric acid-treated anchovy, caseino-phosphopeptides (CPPs), calcium lactate, and calcium phosphate as dietary calcium supplements on calcium metabolism in rats for 5 weeks. Experimental animals were randomly assigned to five treatments with 15 heads of SD male rats (mean body wt. of 100 g) in each group. The experimental diets were as follows; dried large anchovy powder (C) as control, ethanol+citric acid group (EC), ethanol+citric acid+cpps group (ECC), calcium lactate group (CL) and calcium phosphate group (CP), which were formulated with commercial semi-purified Chow diet, while maintaining the same level of calcium in all diets (1%) groups. The weight gain of EC group was significantly higher than ECC, CL and CP groups (p<0.05), food efficiency (FER) was not different. In vitro and in vivo calcium absorption rates of ECC group treated with citric acid and CPPs were 20.4 and 28.4%, respectively, and the highest among the experimental groups (p<0.05). The blood glucose levels of CL group (105.7 mg/dL) was significantly higher than control group (98.5 mg/dL). In terms of serum lipids, total-cholesterol concentration of EC group (75.1 mg/dL) was significantly higher than CP group (65.6 mg/dL) and triglyceride concentration of CP group (33.5 mg/dL) was the lowest (p<0.05). ALP activity and 057 level were not different among experimental groups. The serum calcium concentration of control group (C) was the lowest among groups (p<0.05). The femur weight of CP group was the lowest (p<0.05) and the femur length of ECC group is the longest (P<0.05). The bone density of CP group $(0.1116\;g/cm^2)$ was the lowest while ECC group $(0.1149\;g/cm^2)$ was the highest, and the bone density was increased by added CPPs. These data demonstrated that ECC group significantly increased in vitro and in vivo calcium absorption rate, serum Ca level, and the length and bone density of femur.
