Immunomodulating Activity of DW-116, A New Quinolone Antibiotic

  • DW-116, [1-(5-fluoro-2-pyridyl)-6-fluoro-7-(4-methyl-1-piperazinyl)-1,4-dihydro-4-oxoquino-line-3-carboxylic acid hydrochloride}, is a new quinolone antibiotic with a broad antibacterial spectrum against G(+) and G(-) bacteria. DW-116 was evaluated for the immunomodulating activities, which is one of the efforts to investigate the mechanism of action related to the good in vivo antibacterial efficacy. The results of in vitro studies revealed there was no statistically significant increase in B and T lymphocyte proliferation. But the results of in vivo studies showed that the number of plaque forming cells (PFC), the amount of polyclonal antibodies and delayed-type hypersensitivity (DTH) were significantly increased after the repeat administration with 12 and 60 mg/kg of DW-116. Taken together, these results proposed that immunostimulting effect of DW-116 could be one of the action mechanisms for demonstrating in vivo antibacterial activities under these experimental conditions.

Studies on Protective Effect of Da-9601, Artemisia asiatica Extract, on Acetaminophen- and $CCI_{4}$induced Liver Damage in Rats

  • The hepatoprotective effect of DA-9601, a quality-controlled extract of artemisisa asiatica, on liver damage induced by acetaminophen (APAP) and carbon tetrachloride ($CCI_{4}$) was investigated by means of serum-biochemical, hepatic-biochemical, and histopathological examinations. Doses of Da-9601 (10, 30, or 100 mg/kg) were administered intragastrically to each rat on three consecutive days i.e. 48 h, 24 h and 2 h before a single administration of APAP (640 mg/kg, i.p.) or $CCI_{4}$ (2 ml/kg, p.o.). Four h and 24 h after hepatotoxin treatment, the animals were sacrificed for evaluation of liver damage. Pretreatment of Da-9601 reduced the elevation of serum ALT, AST. LDH and histopathological changes such as centrilobular necrosis, vacuolar degeneration and inflammatory cell infiltration dose-dependently. Da-9601 also prevented APAP- and $CCI_{4}$-induced hepatic glutathione (GSH) depletion and $CCI_{4}$-induced increase of hepatic malondialdehyde (MDA), a parameter of lipid peroxidation, in a chemically induced liver injury by complex mechanisms which involve prevention of lipid peroxidation and preservation of hepatic GSH.

Chitosan-Based Film of Tyrothricin for Enhanced Antimicrobial Activity against Common Skin Pathogens Including Staphylococcus aureus

  • Chitosan-based film-forming gel is regarded as a promising vehicle for topical delivery of antimicrobial agents to skin wounds, since it protects from microbial infection and the cationic polymer itself possesses antibacterial activity. In this study, possible synergistic interaction against common skin pathogens between the cationic polymer and tyrothricin (TRC), a cyclic polypeptide antibiotic, was investigated, by determining the concentration to inhibit 90% of bacterial isolates (MIC). The addition of the polysaccharide to TRC dramatically reduced the MIC values of TRC by 1/33 and 1/4 against both methicillin-resistant and methicillin-susceptible Staphylococcus aureus, respectively. The synergism of TRC and chitosan combination against both strains was demonstrated by the checkerboard method, with a fractional inhibitory concentration index below 0.5. Moreover, co-treatment of TRC and chitosan exhibited antibacterial activity against Pseudomonas aeruginosa, due to the antibacterial activity of chitosan, whereas TRC itself did not inhibit the gram-negative bacterial growth. These findings suggested that the use of chitosan-based film for topical delivery of TRC could be an alternative to improve TRC antimicrobial activity against strains that are abundant in skin wounds.

Analysis of DA-6034, a New Flavonoid Derivative in Biological Fluids by HPLC (HPLC를 이용한 생체 시료중의 새로운 플라보노이드 유도체인 DA-6034의 분석)

  • A high performance liquid chromatographic method was developed for the determination of DA-6034 in biological fluids using internal standard. Plasma containing DA-6034 and inter nal standard was extracted by liquid-liquid extraction at an acidic pH. After evaporation of the organic layer, the drug and internal standard were reconstituted with mobile phase and injected into the column. They were separated by high performance liquid chromatography on inertsil ODS II column at 334 nm. The detection limit of DA-6034 in plasma was 0.02 ${\mu}$g/ml. In this method, the range of recovery and coefficients of variation were 96-110% and 0.40-3.78%, respectively. There was no interference from endogenous substances. Urine and bile were analysed using the deproteinization method and the detection limit of DA-6034 was 1${\mu}$g/l.

  • This study was performed to determine whether the $LH{\beta}$-subunit gene missense mutation is present in Korean infertile patients with 46,XX POF women. The variants of $LH{\beta}$ exon 2 (Trp 8Arg; TGG to CGG and Ile15Thr; ATC to ACC) were studied in forty-four 46,XX idiopathic POF and 54 nonpregnant women. The $LH{\beta}$ exon 2 variants were more frequent in POF patients (20.5%) than nonpregnant (16.7%) women (p>0.05). POF patients with the variant was slightly higher than nonpregnant women with the variant.

Quantitative Assessment Strategy for Determining the Exposures to Volatile Organic Chemicals in Chemistry Laboratories (화학실험실의 휘발성유기화합물 노출에 대한 정량적 평가전략)

  • Working in a research laboratory means exposure to a wide range of hazardous substances. Several studies indicated that laboratory workers, especially working with chemicals, might have an increased risk of certain cancers. However, exposure assessment data in laboratory settings are scarce. This study was performed to examine several approaches for quantitatively assessing the exposure levels to volatile organic compounds (VOCs) among workers in chemistry laboratories. The list of 10 target VOCs, including ethanol, acetone, 2-propanol, dichlormethane, tetrahydrofuran, benzene, toluene, n-hexane, ethyl acetate, chloroform, was determined through self-administered questionnaire for six chemistry research laboratories in a university, a government-funded research institute, or private labs. From September to December 2008, 84 air samples were collected (15 area samples, 27 personal time weighted samples, 42 personal task-basis short-term samples). Real time monitors with photo ionization detector were placed during the sampling periods. In this study, benzene was observed exceeding the action levels, although all the results were below the American Conference of Governmental Industrial Hygienists (ACGIH) Threshold Limit Value (TLV). From the air sampling results, we concluded that (1) chemicals emitted during experiments could directly affect to neighbor office areas (2) chemical exposures in research laboratories showed a wide range of concentrations depending on research activities (3) area samples tended to underestimate the exposures relative to personal samples. Still, further investigation, is necessary for developing exposure assessment strategies specific to laboratories with unique exposure profiles.

An-isotropic Corrosion Behavior of A Marine Steel with Cold Rolling

  • Microstructure of a marine steel with a modified AISI-1004 composition was controlled by cold rolling and heat treatment, which corrosion behavior in an artificial sea water was electrochemically determined for the each deformation direction. The lowest corrosion rate of the surface normal to the rolling direction is related t the (111) fiber structure. Additional annealing at $550^{\circ}C$ for 24 hours improves the corrosion rate which is related to re-crystallization and reduction of (111) concentration.

  • The efficacy of the CA-2-MP120 vaccine, a cell culture-attenuated strain of virulent porcine reproductive and respiratory syndrome virus (PRRSV), was assessed in pigs. Despite the persistence of viremia in all vaccinated animals during the immunization period, the virus was not detected in vaccinated pigs following challenge. Furthermore, no pigs in the vaccinated group shed PRRSV nasally, orally or rectally throughout the experiment. Moreover, histopathological lung and lymph node lesions in the immunized group were much milder than those in the unimmunized and challenged group. These results indicated that CA-2-MP120 can provide effective protection against virulent wild-type PRRSV-2.

  • This paper describes effects of visual information influencing sound localization in the peripheral visual Held under dynamic conditions. Presentation experiments of an audio-visual stimulus were carried out using a movie of a moving patrol car and its siren sound. The tallowing results were obtained: first, the sound image on the timing at the beginning of the presentation was more strongly captured by the visual image than that at the end, i.e., the "beginning effect" was occurred; second, in the peripheral regions, the "beginning effect" was strongly appeared in near the fixation point of eyes.

Task-based Exposure Assessment among Laboratory workers in Organic Synthesis Laboratories (유기합성실험실 연구자의 단위작업별 노출 평가)

  • Objective: Significant concerns have been raised over chemical exposure and potential health risks such as increased cancer mortality among laboratory workers. The aim of this study was to investigate the overall exposure and unit task exposure levels of researchers in organic synthesis laboratories at universities. Methods: Seventy-seven personal Time-weighted average(TWA) samples and 139 task-based samples from four organic synthesis laboratories at two universities were collected over three days. The concentrations of acetone, chloroform, dichloromethane(DCM), diethyl ether, ethyl acetate, n-hexane, tetrahydrofuran(THF), benzene, toluene, and xylene were determined using the GC-FID. Results: The most frequently used chemicals in the laboratories were acetone, DCM, n-hexane, methanol, and THF. Carcinogens such as benzene, chloroform, and DCM were used in one or more laboratories. The TWA full-shift exposures of researchers to acetone was the highest(ND-59.3 ppm). Benzene was observed above the occupational exposure limit in 18-40% of the samples. The levels of exposure to organic solvents were statistically different by task(p<0.05), while washing task was the highest. Washing was not perceived as a part of the real lab tasks. Rather it was considered as simple dish-washing or experimental preparation and performed in an open sink where exposure to organic solvents was unavoidable. TWAs and task-based concentrations were compared by substance, which suggests that TWA-based assessment could not reflect short-term and high concentration exposures. Conclusions: Laboratory workers may be exposed to various organic solvents at levels of concern. TWA-based measurement alone cannot guarantee holistic exposure assessment among lab workers as their exposures are very dependent on their tasks. Further investigation and characterization for specific tasks and overall chronic exposures will help protect lab workers from unnecessary exposure to chemicals while they perform research.
