DNAchip as a Tool for Clinical Diagnostics (진단의학 도구로서의 DNA칩)

  • The identification of the DNA structure as a double-stranded helix consting of two nucleotide chain molecules was a milestone in modern molecular biology. The DNA chip technology is based on reverse hybridization that follows the principle of complementary binding of double-stranded DNA. DNA chip can be described as the deposition of defined nucleic acid sequences, probes, on a solid substrate to form a regular array of elements that are available for hybridization to complementary nucleic acids, targets. DNA chips based on cDNA clons, oligonucleotides and genomic clons have been developed for gene expression studies, genetic variation analysis and genomic changes associated with disease including cancers and genetic diseases. DNA chips for gene expression profiling can be used for functional analysis in human eel Is and animal models, disease-related gene studies, assessment of gene therapy, assessment of genetically modified food, and research for drug discovery. DNA chips for genetic variation detection can be used for the detection of mutations or chromosomal abnormalities in cnacers, drug resistances in cancer cells or pathogenic microbes, histocompatibility analysis for transplantation, individual identification for forensic medicine, and detection and discrimination of pathogenic microbes. The DNA chip will be generalized as a useful tool in clinical diagnostics in near future. Lab-on-a chip and informatics will facilitate the development of a variety of DNA chips for diagnostic purpose.

MEMS Fabrication of Microchannel with Poly-Si Layer for Application to Microchip Electrophoresis (마이크로 칩 전기영동에 응용하기 위한 다결정 실리콘 층이 형성된 마이크로 채널의 MEMS 가공 제작)

  • We developed two kinds of the microchip for application to electrophoresis based on both glass and quartz employing the MEMS fabrications. The poly-Si layer deposited onto the bonding interface apart from channel regions can play a role as the optical slit cutting off the stray light in order to concentrate the UV ray, from which it is possible to improve the signal-to-noise (S/N) ratio of the detection on a chip. In the glass chip, the deposited poly-Si layer had an important function of the etch mask and provided the bonding surface properly enabling the anodic bonding. The glass wafer including more impurities than quartz one results in the higher surface roughness of the channel wall, which affects subsequently on the microflow behavior of the sample solutions. In order to solve this problem, we prepared here the mixed etchant consisting HF and $NH_4F$ solutions, by which the surface roughness was reduced. Both the shape and the dimension of each channel were observed, and the electroosmotic flow velocities were measured as 0.5 mm/s for quartz and 0.36 mm/s for glass channel by implementing the microchip electrophoresis. Applying the optical slit with poly-Si layer provides that the S/N ratio of the peak is increased as ca. 2 times for quartz chip and ca. 3 times for glass chip. The maximum UV absorbance is also enhanced with ca. 1.6 and 1.7 times, respectively.

  • ITU-R B.O. 1130-4 Digital System E adopted for Korean satellite DMB service is a multimedia broadcasting system based on DS-CDM-QPSK technique which broadcasts audio and video contents via the satellite or terrestrial gap-filler. However, Digital System E can not provide full loading services because the multi-channel interference (MCI) is increased due to the loss of orthogonality between signature waveforms in multipath fading channels. In this paper a chip equalizer using tap selection algorithm that enhances the receiving performance is proposed and compared to the conventional rake receiver for the satellite DMB system.

  • This paper presents a novel movable microneedle that becomes active when necessary to use. Conventional researches have been focused on the fabrication of microneedles and the interface with fluidic chip [1,2]. Therefore, we proposed an active microneedle to sample body fluids or deliver drugs in a controlled amount by actuating the needles. This allows us to keep the needles in operation only when necessary so that both the body skin and the needles can be protected from undesirable external disturbance while no operation.

Performance Assessment of Passive Micromixer using Numerical Analysis (수치해석을 이용한 패시브 마이크로 믹서의 성능평가)

  • A micromixer is a component of a lab-on-a-chip or microfluidic device that mixes two or more chemicals together(convergence). The purpose of this study is to assess the performance of passive micromixer of various shapes. Six shapes of micromixers were compared and three dimensional modeling was carried out to have the same hydraulic diameter. The commercial code, ANSYS Fluent, was used to simulate the internal mixing flow. A numerical analysis method is described in detail in this paper. The performance of the micromixer was compared with the mixing index and pressure drop. Consequently, the CDM-8T shape showed reasonable mixing performance and relatively low pressure drop.
